Human papillomavirus (HPV) is the most common sexually transmitted agent worldwide. Prevalence varies according to the geographic regions, and is highest in developing countries. Geographic differences exist also in the detection rate of oncogenic types in malignant cervical lesions. In this study, the prevalence of HPV infection as well as the spectrum of HPV types was evaluated in Italian and immigrant women of the urban area of Rome. Several risk factors (age at first intercourse, number of partners, smoking, pregnancy, age at first pregnancy, contraception, education, and menarche) were taken into consideration. Overall, there was a high prevalence of HPV infection in the two groups studied. No significant differences were observed in the spectrum of HPV types detected. HPV 16 and 18 were the types detected more frequently in both groups. Interestingly, HPV 54 and 70 were found only in the immigrants. Whether this finding reflects a recent introduction of these HPV types in the population studied remains to be established. Monitoring of HPV types in the population is advisable, especially in countries like Italy which is a destination and a gateway for immigrants directed towards north and central Europe. The introduction of high risk HPV variants may have a clinical impact and affect the diagnostic procedures.

The new THELMA code, including a thermal-hydraulic (TH) and an electro-magnetic (EM) model of a cable-in-conduit conductor (CICC), has been developed. The TH model is at this stage relatively conventional, with two fluid components (He flowing in the annular cable region and He flowing in the central channel) being particular to the CICC of the International Thermonuclear Experimental Reactor (ITER), and two solid components (superconducting strands and jacket/conduit). In contrast, the EM model is novel and will be presented here in full detail. The results obtained from this first version of the code are compared with experimental results from pulsed tests of the ENEA stability experiment (ESE), showing good agreement between computed and measured deposited energy and subsequent temperature increase.

The role of p16INK4aas a marker of HR-HPV and in the diagnosis of CIN has been well established, but its predictive value in the clearance of the virus after CIN treatment and its use as a prognostic marker of cervical cancer has not been studied. A series of 302 archival samples, including 150 squamous cell carcinomas (SCCs) and 152 CIN lesions, were subjected to immunohistochemical staining for p16INK4aand HPV testing using PCR with three primer sets (MY09/11, GP5+/GP6+, SPF). Follow-up data were available of 88 SCC patients, and 67 of the CIN lesions had been followed-up with serial PCR after conization. HR-HPV types were closely associated with CIN (OR 19.12; 95%CI 2.31–157.81) and SCC (OR 27.25; 95%CI 3.28–226.09). There was a significant linear relationship between the lesion grade and intensity of p16INK4astaining (p = 0.0001). The expression of p16INK4awas also closely related to HR-HPV (p = 0.0001). p16INK4astaining was a 100% specific indicator of CIN, with 100% PPV, and showed 83.5% sensitivity and 80.1% PPV in detecting HR-HPV. However, p16INK4astaining did not predict clearance/persistence of HR-HPV after treatment of CIN. Similarly, despite a slightly more favorable survival in women with strong/intense p16INK4astaining in univariate analysis, p16INK4aexpression was not an independent prognostic predictor in multivariate survival (Cox) analysis. After adjustment for p16INK4astaining, HR-HPV, histological grade, International Federation of Gynecology and Obstetrics (FIGO) stage, and age, only the last two were significant prognostic predictors (p = 0.0001 and p = 0.003, respectively). The present data confirm the role of p16INK4aas a highly specific marker of CIN and HR-HPV type, but expression of this protein does not seem to be of any prognostic value in cervical cancer or in predicting the clearance of HR-HPV after treatment of CIN. We speculate that different subgroups of cervical cancer are characterized by aberrant p16INK4a/cyclin D/Rb pathways that are due to different mechanisms that can be mutually exclusive.

Within the framework of the R&D studies for the International Thermonuclear Experimental Reactor (ITER) project, the first full size NbTi conductor sample was fabricated industrially and tested in the SULTAN facility (Villigen, Switzerland). This sample (PF-FSJS), which is relevant to the poloidal field coils of ITER, is composed of two parallel straight bars of conductor, connected at the bottom through a joint designed according to the CEA twin-box concept. The two conductor legs are identical except for the use of different strands: a nickel plated NbTi strand with a pure copper matrix in one leg, and a bare NbTi strand with a copper matrix and internal CuNi barrier in the other leg. The two conductors and the joint were extensively tested as regards DC and AC properties. This paper reports on the test results and analysis, stressing the differences between the two conductor legs and discussing the impact of the test results on the ITER design criteria for the conductor and joint. While joint DC resistance, conductors and joint AC losses fulfilled the ITER requirements, neither conductor could reach its current sharing temperature at relevant ITER currents, due to instabilities. Although the drop in temperature is slight for the CuNi strand cable, it is more significant for the Ni plated strand cable.

The growth-related gene product beta (GRObeta) is a small chemoattractant cytokine that belongs to the CXC chemokine family, and GRObeta receptors are expressed in the brain, including the cerebellum. We demonstrate that rat cerebellar granule neurones express the GRObeta receptor CXCR2. We also show that, in addition to the known stimulation of a phosphoinositide-specific phospholipase C, GRObeta activates both neutral (N-) and acidic (A-) sphingomyelinases (SMase) and the stress-activated c-Jun N-terminal kinase 1 (JNK1). Although both exogenous ceramide and bacterial SMase stimulate JNK1, GRObeta-induced JNK1 activation is an event probably independent of ceramide generated by A-SMase, since it is maintained in the presence of compounds that block A-SMase activity. This is the first report on the activation of the SMase pathway by chemokines.

Although glucuronidation is considered a necessary step in aromatic amine-induced bladder cancer, the specific enzymes involved are not known. This study assessed the capacity of five different human recombinant UDP-glucuronosyltransferases expressed in COS-1 cells to glucuronidate benzidine, its metabolites and 4-aminobiphenyl. [¹⁴C]UDP-glucuronic acid was used as co-substrate. UGT1A1, UGT1A4 and UGT1A9 each metabolized all of the aromatic amines. UGT1A9 exhibited the highest relative rates of metabolism with preference for the two hydroxamic acids, N-hydroxy-N-acetylbenzidine and N-hydroxy-N,N′-diacetylbenzidine. UGT1A9 metabolized 4-aminobiphenyl ~50% faster than benzidine or N-acetylbenzidine. UGT1A4 N-glucuronidated N′-hydroxy- N-acetylbenzidine at the highest relative rate compared with the other transferases. UGT1A6 was effective in metabolizing only four of the eight aromatic amines tested. UGT1A1 demonstrated more extensive metabolism of the hydroxamic acid, N-hydroxy-N,N′-diacetylbenzidine, and the ring oxidation product, 3-OH-N,N′-diacetylbenzidine, than it did for the other six amines. UGT2B7 was the only product of the UGT2 gene family examined and it metabolized all the aromatic amines at similar low relative levels compared with a preferred substrate, 4-OH-estrone. The K_m values for N-acetylbenzidine metabolism by UGT1A1 and UGT1A4 were 0.37 ± 0.14 and 1.8 ± 0.4 mM, respectively. The O-glucuronide of 3-OH-N,N′-diacetylbenzidine was not hydrolyzed during a 24 h 37°C incubation at either pH 5.5 or 7.4. Likewise, the O-glucuronide of 3-OH-benzidine was stable at pH 7.4, with 52% remaining at pH 5.5 after 24 h. These results suggest the following relative ranking of transferase metabolism: UGT1A9 > UGT1A4 > > UGT2B7 > UGT1A6 ≈ UGT1A1. The relative pH stability of O-glucuronides is consistent with a role in detoxification and excretion of aromatic amines, while the acid lability of N-glucuronides is consistent with delivery of these amines to the bladder epithelium for activation, resulting in DNA adducts which may lead to mutations.

Over the last few years, a great deal of effort has been devoted to solving the problem of power and particle handling in divertors, which has been recognized as a critical issue for the operation of a magnetic fusion reactor. In particular, the choice of materials for plasma facing components has been examined with a view to developing heat and erosion resistant materials for divertor target plates. A large database on the behaviour of low-Z (carbon or beryllium) materials in tokamaks is available, while for high-Z materials there is little experience in the present generation of magnetic fusion devices. Frascati Tokamak Upgrade (FTU), a high field compact tokamak, has devoted part of its experimental campaign to studying the plasma characteristics when its limiter material is changed from the usual Inconel (nickel) to molybdenum and tungsten. Siliconization of the machine has also allowed the comparison of plasma performance when a relatively low-Z (silicon) ion is the dominant impurity. In this article, results are reported concerning the plasma operation, the differences in plasma characteristics and radiation losses, the impurity generation mechanisms and the relative impurity concentrations in the core plasma. A simulation of the experimental results, made with a self-consistent edge-core coupled model is presented, in order to provide evidence of the main physics mechanisms responsible for the observed behaviour

Plasma behaviour in the high density regime has been investigated on the Frascati Tokamak Upgrade (FTU). The items particularly addressed are density limit, MARFE characteristics and fuelling efficiency. With gas puffing, a maximum line average density of 3.2*10²⁰ m^-3 has been obtained in the ohmic regime at q=5.1, corresponding to 1.7 times the Greenwald limit; while with pellet injection, a value of 3.7*10²⁰ m^-3 has been reached at q=3.5. The density limit appears to be connected with the impurity content and edge parameters, so the best results are obtained with very clean plasmas and peaked electron density profiles. The MARFE phenomenon always appears beyond a critical density that depends on the total input power and the effective charge: emissivities in the range of 2 to 12 MW/m³ have been measured in this highly radiating region. The fuelling efficiency, starting from a value of about 50% at low density, progressively deteriorates, falling to 10% near the density limit: this behaviour is interpreted as being due to a decrease of the scrape-off layer transparency to incoming neutrals

Three methods of density modulation have been used in FTU discharges (gas puffing, pellet injection and current rampup). The analysis of particle balance confirms that the neoclassical theory is not adequate and the experimental results obtained by the three methods are used to verify the validity of an empirical expression for the radial particle flux. Such an expression is relevant for the design of future experiments and constitutes a constraint for any satisfactory theory

Cerebellar granule cells grown in the presence of a serum complex differentiate but are resistant to the lethal action of excitatory amino acids. When these cells are grown also in the presence of insulin-like growth factor I (IGF-I) they become fully susceptible to the toxic, lethal action of glutamate. The glutamate-sensitizing action of IGF-I is dependent on concentration (half-maximal effect at 2-4 ng/ml) and time (half-maximal effect at 2-4 days in vitro) and is paralleled by the appearance of functionally active, glutamate-activated, Ca2+ channels and of voltage-gated Na+ and late K+ channels. IGF-I-induced glutamate sensitivity is rapidly reversible (t1/2 = 30-60 min) after removal of this somatomedin. The action of IGF-I is not mimicked by IGF-II, nerve growth factor, basic or acidic fibroblast growth factor, platelet-derived growth factor, or tumor necrosis factor alpha. We postulate that the constitutive phenotype of cerebellar granule cells is glutamate-resistant and becomes responsive to excitatory amino acids under the action of epigenetic cues among which IGF-I may be one of those operative in vivo.

WE present the first comparative study of the uptake of metal ions by neurones, performed for Zn, Cr, Co, Mo, Al, Ni, Mn and Cd. The study reveals substantial differences in the uptake of different metals, under similar exposure procedures. In particular, we found very large uptakes for aluminium and molybdenum. We also found significant effects of excitatory substances, in particular kainate, as stimulants of uptake of some of the metals.

WE tested a new approach to cell decapping on rat cerebellar neurones, and observed its effects on cell topography by atomic force microscopy (AFM). The results clearly demonstrate the effectiveness of our decapping approach, and also the ability of AFM to reveal fine details of the decapped cells. Specifically, varying the conditions and duration of the decapping process modifies the extent of the decapping. Such a method can be used to investigate the cytoplasm with surface sensitive techniques.

To study the cellular signals underlying the regulatory mechanisms involved in maintenance of sarcomeric integrity, we have used quail skeletal muscle cells that reach a high degree of structural maturation in vitro, and also express a temperature-sensitive mutant of the v-Src tyrosine kinase that allows the control of differentiation in a reversible manner. By immunofluorescence and electron microscopy we show that v-Src activity in myotubes leads to an extensive cellular remodeling which affects components of the sarcomeres, the cytoskeleton network and the triad junctions. We have previously shown that activation of v-Src causes a selective dismantling of the I-Z-I segments coupled to the formation of aggregates of sarcomeric actin, alpha-actinin and vinculin, called actin bodies. We now show that intermediate filaments do not participate in the formation of actin bodies, while talin, a component of costameres, does. The I-Z-I segments are completely dismantled within 24 hours of v-Src activity, but the A-bands persist for a longer time, implying distinct pathways for the turnover of sarcomeric subdomains. Immunofluorescence labeling of markers of the triad junctions demonstrates that the localization of the alpha 1 subunit of the dihydropyridine receptor is disrupted earlier than that of the ryanodine receptor after tyrosine kinase activation. Furthermore, the location of junctional sarcoplasmic reticulum and transverse tubule membranes is maintained in myotubes in which the I-Z-I have been removed and the regular disposition of the intermediate filaments is disrupted, supporting a role for sarcoplasmic reticulum in the proper positioning of triad junctions. Altogether these results point to a tyrosine kinase signaling cascade as a mechanism for selectively destabilizing sarcomere subdomains and their tethering to the cytoskeleton and the sarcolemma.

The aim of the present study was to assess whether endogenous and newly synthesized glutamate can be released from differentiating cultured cerebellar granule cells in a way compatible with a neurotransmitter role. Granule cells from 8-day-old rat cerebella were grown in basal Eagle's medium with 10% fetal calf serum for 2-12 days in vitro(DIV), then washed with Krebs-Ringer medium, and labeled for 45 min with tracer amounts of radioactive glutamine. Subsequently, the release of endogenous glutamate and of newly formed radioactive glutamate was measured in basal conditions and upon depolarization with elevated K+concentration or veratridine. At 2 DIV, the release of endogenous and newly synthesized glutamate evoked by high K+concentration was small and Ca2+independent, but it progressively and steadily increased (up to 8- to 10-fold) and became Ca2+dependent (up to 80-85%) at later stages (4, 8, and 12 DIV). Veratridine was almost ineffective with cells at 2 DIV but greatly increased glutamate release (endogenous and neosynthesized) at 8 DIV, and its action was totally antagonized by tetrodotoxin. The level and synthesis of glutamate remained fairly constant in cells from 2 to 12 DIV. γ-Aminobutyric acid synthesis from radioactive glutamine was about 3% of that of glutamate, and γ-aminobutyric acid release (endogenous and neosynthesized) was not measurable. Aspartate synthesis was about 10% of that of glutamate, and the high K+concentration-evoked release of this amino acid was modest and scarcely affected by Ca2+. Neither high K+concentration nor veratridine was able to induce glutamate release from confluent cerebellar astrocyte cultures at 14 DIV, although the level and synthesis of the amino acid were comparable to those in granule cells. In conclusion, the data show that a stimulus-coupled release of endogenous and neosynthesized glutamate is progressively expressed by cerebellar granule cells differentiating in culture, and this strongly supports the concept that glutamate is the neurotransmitter of these cells.

EXPERIMENTAL evidence of the preferential uptake of aluminium by GABAergic neurones and glial cells was provided by synchrotron spectromicroscopy studies. We observed rat cerebellar cultures enriched for GABAergic neurones or glial cells exposed to aluminium ions, detecting the presence and identifying the chemical status of aluminium on cell structures.

When postnatal rat cerebellar cells were cultured in a chemically defined, serum-free medium, the only type of astrocyte (defined by the expression of the glial fibrillary acidic protein, GFAP) present was unable to accumulate gamma-[3H]aminobutyric acid (GABA), did not express surface antigens recognized by two monoclonal antibodies, A2B5 and LB1, and showed minimal proliferation. In these cultures, nonneuronal A2B5+, LB1+ stellate cells exhibiting "neuron-like" [3H]GABA uptake formed cell colonies of increasing size and were GFAP-. After about one week of culturing, the A2B5+, LB1+, GABA-uptake positive cell groups became galactocerebroside (GalCer) positive. Immunocytolysis of the A2B5+ cells at 3 and 4 days in vitro prevented the appearance of the A2B5+, LB1+, GABA-uptake positive cell colonies, and also of the GalCer+ cell groups. If 10% (vol/vol) fetal calf serum was added to 6-day cultures, the A2B5+, LB1+, GABA-uptake positive cell groups expressed GFAP and not GalCer. If the serum was added to the cultures 2 days after lysing the A2B5+ cells, only A2B5-, LB1-, GABA-uptake negative astrocytes proliferated. It is concluded that the putative fibrous astrocytes previously described in serum-containing cultures (which had a stellate shape and were A2B5+, LB1+, GABA-uptake positive) derive from bipotential precursors that differentiate into oligodendrocytes (GalCer+) in serum-free medium or into astrocytes (GFAP+) in the presence of serum, while the epithelioid A2B5-, LB1-, GABA-uptake negative astrocytes originate from a different precursor not yet identified.

The characterization (Ritter, J.K., Chen, F., Sheen, Y. Y., Tran, H.M., Kimura, S., Yeatman, M.T., and Owens, I. S. (1992) J. Biol. Chem. 267, 3257-3261) of the single-copy UGT1 gene complex locus encoding both bilirubin and phenol UDP-glucuronosyltransferases (transferase) has been critical to the determination of genetic defects in Crigler-Najjar patients. The complex (UGT1A-UGT1M) codes for at least two bilirubin, three bilirubin-like, and eight phenol transferase isozymes. In the 5' region, a minimum of 13 different exons 1, each with an upstream promoter, are arrayed in series with 4 common exons in the 3' region of the locus. Each exon 1 encodes the amino terminus of a transferase, and the common exons encode the common carboxyl terminus of each isoform. Although a deleterious mutation in a common exon inactivates the entire locus, a deleterious mutation in an exon 1, as we report here for the UGT1A gene in a Crigler-Najjar Type I patient, affects the amino terminus of that single isoform. Recessively inherited mutant alleles for the predominant bilirubin isozyme, the HUG-Br1 protein, substituted Arg for Gly at codon 276 (G276R) in exon 1 of UGT1A abolishing a conserved di-glycine. The mutant HUG-Br1-G276R protein expressed in COS-1 cells had no detectable bilirubin glucuronidating activity at either pH 7.6 or 6.4. Although each of the bilirubin-type isozymes contains a conserved peptide between residues 270 and 288, all UDP-glucuronosyltransferases contain a di-glycine at approximately position 276-277, making it strictly conserved. Structure-function relationship was studied by site-directed mutations of the HUG-Br1 cDNA; G276A, G276Q, G276E, G276I, and P270G mutants were inactive, and V2751- and P285G-altered transferases expressed normal activity. Conservation of residues between the related baculoviral ecdysone UDP-glucosyltransferase and the UDP-glucuronosyltransferases confirms the critical role of the Gly-276 as well as other residues.

Kelletinin A [ribityl-pentakis (p-hydroxybenzoate)] (KA), a natural compound isolated from the marine gastropodBuccinulum corneum, showed antiviral activity on the human T-cell leukemia virus type-1 (HTLV-1) and antimitotic activity on HTLV-1-infected MT2 cells. KA inhibited cellular DNA and RNA synthesis, without influencing protein synthesis, and interfered with viral transcription by reducing the levels of high molecular weight transcripts. Finally, the compound inhibited HTLV-1 reverse transcriptase in vitro.

Two cases of unusual valgus and lateral stem displacement in cementless total hip replacement are reported. A certain number of mechanical conditions must have been present to cause this unusual prosthesis displacement. The Authors suggest that the stem's design was one of these factors.
