Trace element concentrations in toenail clippings have increasingly been used to measure trace element exposure in epidemeological research. Conventional methods such as inductively coupled plasma mass spectrometry (ICP-MS) and high-performance liquid chromatography ICP-MS (HPLC-ICP-MS) are commonly used to measure trace elements and their speciation in toenails. However, the impact of the removal of external contamination on trace element quantification has not been thoroughly studied. In this work, the microdistribution of trace elements (As, Ca, Co, Cu, Fe, K, Mn, Ni, Rb, S, Sr, Ti, and Zn) in dirty and washed toenails and the speciation of As in situin toenails were investigated using synchrotron X-ray fluorescence microscopy (XFM) and laterally resolved X-ray absorption near edge spectroscopy (XANES). XFM showed different distribution patterns for each trace element, consistent with their binding properties and nail structure. External (terrestrial) contamination was identified and distinguished from the endogenous accumulation of trace elements in toenails─contaminated areas were characterized by the co-occurrence of Co, Fe, and Mn with elements such as Ti and Rb (i.e., indicators of terrestrial contamination). The XANES spectra showed the presence of one As species in washed toenails, corresponding to As bound to sulfhydryl groups. In dirty specimens, a mixed speciation was found in localized areas, containing AsIII–S species and AsVspecies. ArsenicVis thought to be associated with surface contamination and exogenous As. These findings provide new insights into the speciation of arsenic in toenails, the microdistribution of trace elements, and the effectiveness of a cleaning protocol in removing external contamination.

Environmental context The marine ecosystem is highly elevated in arsenic in comparison to the terrestrial environment. More than 100 different naturally occurring organoarsenic compounds have been found in different marine organisms. In order to understand how highly toxic AsHCs are formed and bioaccumulate in different marine organisms, large scale monitoring of AsHCs from a range of different laboratories is necessary. Despite their toxicity, AsHCs have not been extensively studied due in part to the lack of low-cost analytical methods and the lack of commercially available standards. Rational This study aims to develop a new analytical method for the quantitative determination of arsenic-containing hydrocarbons (AsHCs) with gas chromatography–mass spectrometry (GC-MS). The new method is an alternative approach to determine these compounds in environmental samples. Methodology AsHCs were extracted and purified from fish oil and seaweed samples. Due to the thermal conversion of oxo-AsHCs to their trivalent forms in the injection port of the GC, the conversion is incomplete resulting in low signal intensities and large carry overs. We therefore chemically reduced AsHCs to their trivalent forms and analysed them with GC-MS, gas chromatography–tandem mass spectrometry (GC-MS/MS) and high performance liquid chromatography–inductively coupled plasma mass spectrometry (HPLC-ICPMS). The possibility for compound independent quantification with commercially available standards was investigated. Results After optimisation of conditions for reduction of AsHCs, these compounds could be determined in marine samples using authentic standards with the new method. Results showed good agreement with results from HPLC-ICPMS. The novel GC-MS method showed compound independent quantification using triphenylarsine (TPA) as a commercially available standard. Discussion With the developed method we could demonstrate the applicability of GC-MS for the quantitative determination of AsHCs in marine samples for the first time. The compound independent quantification with TPA opens up a quantification for AsHCs species for which no standards are commercially available. The broad applicability might enhance screening for these toxic compounds in the environment.

Environmental context Arsenic contamination has a disproportionate effect on marine ecosystems. Organisms such as some marine macroalgae, which accumulate potentially toxic elements from the surrounding environment, have developed an internal conversion process that is not yet fully understood. Are arsenic-containing sugars a product of detoxification, or simply the result of phospholipid degradation? Rationale Arsenosugars (AsSugar) account for the majority of total arsenic in common seaweed species, yet it is unclear whether these are formed through some detoxification pathway for inorganic arsenic or are precursors/degradation products of arsenic-containing phospholipids in the cell wall. Methodology Temporal and intra-thallus variations in water-soluble arsenic were measured by HPLC-ICP-MS, as well as total non-polar and polar arsenic-containing lipids by ICP-MS in Laminaria digitata to offer potential insight into the origins of arsenosugars. Water-soluble speciation with and without freeze-drying were also compared to determine whether freeze-drying changes the water-soluble As speciation. Results In general, lower levels of total As were detected in the samples collected in May (39.2–74.5 mg kg−1 ) compared to those collected in February (72.6–151 mg kg−1 ). The concentration of arsenate was found to consistently increase along the thallus from the holdfast/stipe (0.78–1.82 mg kg−1 ) to the decaying fronds (44.4–61.0 mg kg−1 ) in both months, and AsSug-SO3 was the dominant AsSugar in the majority of samples. The extraction efficiency was lower in fresh samples (64–77%) than in freeze-dried (95–116%) from the same month. Water-soluble, polar AsLipids, and residual As concentrations, were generally highest in February, and the non-polar AsLipids accounted for Discussion Our results suggest that the arsenosugars are not a product of arsenic detoxification, but a by-product of normal biological activity. It is probable that the arsenosugars are bound to the cell membrane within the Laminaria digitata cells, and lyophilisation is required to release them quantitatively. Future research should focus on speciation of polar lipid-soluble As extracted from fresh samples to determine if the lower extraction efficiency observed in this study is due to the As being in an unextractable form, i.e. lipids, and thus is not removed from cells during water-based extractions.

Environmental context Arsenic can be found in all environmental compartments in a large number of chemical forms of varying toxicity. We performed an inter-laboratory comparison study focusing on quantitation of some of the most toxic arsenic forms in seven different biological materials and found very good agreement among the submitted results. Certification of the studied materials will provide suitable quality control samples for environmentally relevant concentrations of arsenic in food products and biota. Rationale Arsenic (As) speciation analysis in biological matrices has been performed for several decades; however, there are very few matrix certified reference materials available for the validation of analytical methods. The literature data on the mass fractions of As species in the existing certified reference materials are inconsistent and suggest method extraction dependency. Methodology In the present study, an international round-robin study was organised to identify the possible sources of discrepancies in quantitation of several As species in different matrices of biological reference materials: one plant tissue, three marine and three terrestrial biological tissues. Each participating laboratory was provided with a set of identical calibration standard solutions, and analysed the samples by following a common group extraction method as well as using an in-house protocol. Results The results showed that significant biases can be introduced by insufficient verification of the analyte’s mass fractions in the calibration standard solutions. The choice of extraction method seems to have very little impact on the quantitation of As species in the studied plant and terrestrial biological tissues. However, following a prescribed extraction method led to significant reduction of uncertainties in more complex samples such as marine animal tissues. Discussion Some differences in the mass fractions of As species extracted from marine animal tissues in water (with and without H2 O2 ) were observed and should be further investigated. Despite the variety of extraction methods used, very good agreement between reported mass fractions was achieved and the combined consensus values will be published as certified reference values of As species in the studied materials.

Environmental context Intensive aquaculture is the main source of fisheries products. Thus, investigating the food safety of these products and the environmental impacts of the farms on mangroves is necessary. A shrimp productive cycle was evaluated with a focus on trace element accumulation in shrimps and effluent. The results revealed secure levels of elements in the final product; however, the effluent produced in farms is an important source of contamination to mangrove environment. Rationale Aquaculture systems have increased in the last years due to the high demand for seafood consumption, this could impact the environment and subject fisheries to accumulation of toxic elements. To understand some parameters of food safety and environmental impact, the present study evaluated the concentration of trace elements (Al, As, Cd, Co, Cr, Cu, Mn, Mo, Pb, Se, V and Zn) throughout the production cycle of shrimp. Methodology About 50 shrimps per cycle were collected in a shrimp farm in Brazil and their trace element contents were determined by inductively coupled plasma–tandem mass spectrometry (ICP-MS/MS) and microwave-induced plasma–optical emission spectroscopy (MIP-OES). Results At their final lifecycle stage, shrimp samples present a content (mg g−1 ) of elements following the trend: Cu (102 ± 12) > Al (20.06 ± 4.24) > Zn (14.82 ± 2.46) > Mn (6.24 ± 0.94) > As (2.65 ± 0.42) > Se (0.932 ± 0.140) > Co (0.380 ± 0.05) > Mo (0.254 ± 0.03) > V (0.204 ± 0.02). Discussion The content of Cd and Pb are in allowance with Brazil and USA legislation for crustaceans, however, the content of As is 3–4-fold higher than that allowed by guidelines in all stages of the growth of the shrimp. The final effluent of the shrimp’s pond into the mangroves shows a high mass fraction of Zn and Mn that could be a source of contamination. Some correlations between some elements in the shrimp samples were found, such as As–Se, Se–Co and Se–V. This study was a scoping experiment to study the content of trace elements throughout the farming cycle of shrimps, encouraging the researcher to undergo a wide survey to evaluate the environmental impact of aquaculture shrimp farming.

Although several per- and polyfluoroalkyl substances (PFAS) have been banned and classified as substances of very high concern by the European Chemicals Agency, similar chemicals remain widely used compounds to date. Even though more than 4700 PFASs may occur in the environment, only 40–50 compounds are routinely determined in targeted analysis by ESI-MS using isotopically labeled standards. Nontargeted analysis using high resolution (HR) molecular mass spectrometry suffers from a lack of data mining algorithms for identification and often low ionization efficiency of the compounds. An additional problem for quantification is the potential lack of suitable species specific standards. Here, we demonstrate the usefulness of a hard ionization source (ICP-MS/MS) as a fluorine-specific detector in combination with ESI-MS for the identification of fluorine containing compounds. Simultaneous hyphenation of HPLC-ICP-MS/MS with HR-ESI-MS is applied to evaluate biodegradation products of organofluorine compounds by sewage sludge. The data are analyzed in a nontarget approach using MZmine. Due to the fluorine-specific detection by ICP-MS/MS, more than 5000 peaks (features) of the ESI-MS were reduced to 15 features. Of these, one was identified as a PFAS degradation compound of fluorotelomer alcohol (8:2 FTOH) without using targeted analysis. The feasibility of the detection of organofluorine metabolites using a fluorine-specific detection was demonstrated using a model compound and can thus be applied to new experiments and unknown organofluorine containing samples in the future.

A new determination method was developed for the measurement of methylmercury (Me-Hg) and inorganic mercury (i-Hg) in biological samples using high-performance liquid chromatography-inductively coupled plasma-mass spectrometry (HPLC-ICP-MS) following alkaline extraction. Mercury species in biological samples were extracted with 10% (w/w) tetramethylammonium hydroxide (TMAH) solution at 80°C for 2 h. Methylmercury was completely separated from i-Hg by adamantyl type and octadecylsilyl type columns within 6 and 4 min using isocratic elution, respectively. The detection limits (3(3σ)) of adamantyl and octadecylsilyl columns using the proposed system were 0.08 and 0.13 ng g−1(as Hg), respectively. Inorganic Hg completely separates from Me-Hg without tailing. The proposed determination methods were applied to several biological certified reference materials (CRMs). The measurement results of Me-Hg obtained by the present method were in good agreement within the expanded uncertainties (k= 2) with the certified values. The analytical precision (n= 3) of Me-Hg was less than 2%, and the recoveries of Me-Hg and i-Hg were 101 ± 1 and 103 ± 3%, respectively. In addition, this method enables the determination of Me-Hg and i-Hg for 20 samples in 1 h.

The inductively coupled plasma mass spectrometry (ICPMS) has been attracting increasing attention for many applications as an element-selective chromatographic detector. A major and fundamental limitation in coupling ICPMS with liquid chromatography is the limited compatibility with organic solvents, which has so far been addressed via a tedious approach, collectively referred to as the “organic ICPMS mode”, that can decrease detection sensitivity by up to 100-fold. Herein, we report 1,2-hexanediol as a new eluent in high-performance liquid chromatography–ICPMS which enables avoiding the current limitations. Unlike commonly used eluents, 1,2-hexanediol was remarkably compatible with ICPMS detection at high flow rates of 1.5 mL min–1and concentrations of at least 30% v/v, respectively, under the standard conditions and instrumental setup normally used with 100% aqueous media. Sensitivity for all tested elements (P, S, Cl, Br, Se, and As) was enhanced with 10% v/v 1,2-hexanediol relative to that of 100% aqueous media by 1.5–7-fold depending on the element. Concentrations of 1,2-hexanediol at ≤30% v/v were superior in elution strength to concentrations at >90% v/v of the common organic phases, which greatly decreases the amount of carbon required to elute highly hydrophobic compounds such as lipids and steroids, enabling detection at ultra-trace levels. The proposed approach was applied to detect arsenic-containing fatty acids in spiked human urine, and detection limits of <0.01 μg As L–1were achieved, which is >100-fold lower than those previously reported using the organic ICPMS mode. Nontargeted speciation analysis in Allium sativumrevealed the presence of a large number of hydrophobic sulfur-containing metabolomic features at trace levels.

The mechanisms of arsenic (As) hyperaccumulation in Pteris vittata, the first identified As hyperaccumulator, are unknown. We investigated the interactions of arsenate and phosphate on the uptake and distribution of As and phosphorus (P), and As speciation in P. vittata. In an 18-d hydroponic experiment with varying concentrations of arsenate and phosphate, P. vittata accumulated As in the fronds up to 27,000 mg As kg(-1) dry weight, and the frond As to root As concentration ratio varied between 1.3 and 6.7. Increasing phosphate supply decreased As uptake markedly, with the effect being greater on root As concentration than on shoot As concentration. Increasing arsenate supply decreased the P concentration in the roots, but not in the fronds. Presence of phosphate in the uptake solution decreased arsenate influx markedly, whereas P starvation for 8 d increased the maximum net influx by 2.5-fold. The rate of arsenite uptake was 10% of that for arsenate in the absence of phosphate. Neither P starvation nor the presence of phosphate affected arsenite uptake. Within 8 h, 50% to 78% of the As taken up was distributed to the fronds, with a higher translocation efficiency for arsenite than for arsenate. In fronds, 49% to 94% of the As was extracted with a phosphate buffer (pH 5.6). Speciation analysis using high-performance liquid chromatography-inductively coupled plasma mass spectroscopy showed that >85% of the extracted As was in the form of arsenite, and the remaining mostly as arsenate. We conclude that arsenate is taken up by P. vittata via the phosphate transporters, reduced to arsenite, and sequestered in the fronds primarily as As(III).

In Bihar state, India, the cure rate of antimonial compounds (eg, sodium stibogluconate) in the treatment of visceral leishmaniasis (VL) has fallen from more than 85% to less than 50%. This reduction has been attributed to long-term, widespread misuse of antimonial drugs within the Indian private health-care system. We aimed to test the hypothesis that exposure to arsenic in drinking water in this region has resulted in antimony-resistant Leishmaniaparasites.
