The growth factors BDNF and GDNF are gaining more and more attention as modulators of synaptic transmission in the mature central nervous system (CNS). The two molecules undergo a regulated secretion in neurons and may be anterogradely transported to terminals where they can positively or negatively modulate fast synaptic transmission. There is today a wide consensus on the role of BDNF as a pro-nociceptive modulator, as the neurotrophin has an important part in the initiation and maintenance of inflammatory, chronic, and/or neuropathic pain at the peripheral and central level. At the spinal level, BDNF intervenes in the regulation of chloride equilibrium potential, decreases the excitatory synaptic drive to inhibitory neurons, with complex changes in GABAergic/glycinergic synaptic transmission, and increases excitatory transmission in the superficial dorsal horn. Differently from BDNF, the role of GDNF still remains to be unraveled in full. This review resumes the current literature on the interplay between BDNF and GDNF in the regulation of nociceptive neurotransmission in the superficial dorsal horn of the spinal cord. We will first discuss the circuitries involved in such a regulation, as well as the reciprocal interactions between the two factors in nociceptive pathways. The development of small molecules specifically targeting BDNF, GDNF and/or downstream effectors is opening new perspectives for investigating these neurotrophic factors as modulators of nociceptive transmission and chronic pain. Therefore, we will finally consider the molecules of (potential) pharmacological relevance for tackling normal and pathological pain.

Cistus x incanusL. is a Mediterranean evergreen shrub used in folk medicine for the treatment of inflammatory disorders but the underlying mechanisms are not fully understood. We therefore investigated the anti-inflammatory effects of an ethyl acetate fraction (EAF) from C. x incanusL. leaves on lipopolysaccharide (LPS) activated RAW 264.7 macrophages. HPLC analysis revealed myricetin and quercetin derivatives to be the major compounds in EAF; EAF up to 1 µM of total phenolic content, was not cytotoxic and inhibited the mRNA expression of interleukin-6 (IL-6) and cyclooxygenase-2 (COX-2) (p< 0.05) and the production of prostaglandins E2(PGE2) (p< 0.05). Meanwhile, EAF triggered the mRNA expression of interleukin-10 (IL-10) and elicited the nuclear translocation of nuclear factor erythroid 2-related factor 2 (Nrf2), as well as the expression of its main target gene, heme oxygenase-1 (HO-1) (p< 0.05). These data indicate that EAF attenuates experimental inflammation via the inhibition of proinflammatory mediators and at least in part, by the activation of Nrf2/HO-1 pathway. These effects are likely due to myricetin and quercetin derivatives but the role of other, less abundant components cannot be excluded. Further studies to confirm the relevance of our findings in animal models and to highlight the relative contribution of each component to the anti-inflammatory activity of EAF should be conducted.

Slug catcher is an important and costly equipment in the up-stream assets, but is the less supported by research or commercial simulation software in terms of design and sizing criteria.

The article presents a study which analyzed the impact of sealed and unsealed soils by urbanization. The study examines the four pavement types including impermeable, porous, and permeable and the key plants and soil parameters including soil moisture, soil oxygen, and soil temperature. Study results reveal the effects of pavements during establishment on soil and plant traits including gas diffusion between soil and atmosphere, root morphology, and leaf physiology.

Shrubs are often used for side slope greening and protection in the urban landscape. Only a few species are commonly used in the European city environment, bestowing upon cities a monotonous appearance. The aim of this 3-year trial, set up at Fondazione Minoprio (Vertemate con Minoprio; Como, Italy), was to evaluate the performance of 25 shrub species grown on a slope during three seasons (2007-09). Moreover, to evaluate the influence of weeds on plant growth and the weeding time, two mulches (biodegradable textile and polypropylene fabric) were used in comparison with bare soil (control). Shrubs were planted in late Spring 2007. To simulate urban conditions, no pruning or disease control were applied. Irrigation was carried out only in the driest periods of the first summer. Plant height and percent cover were measured every 2 months, whereas plant phenology and state of health were recorded weekly. Chlorophyll fluorescence and chlorophyll content were evaluated on two different drought-tolerant species during the third growing season. At the end of the trial shoot biomass was measured and root characteristics (root density and specific root length) were determined. Weeds were removed twice in the first and third years and three times in the second year. Time for weed removal was recorded for each experimental plot. Results show that the highest growth (height and biomass) was detected in mulched plots, probably because of both limited weed competition for water and nutrients and lower water loss by evaporation. Plants grown in bare soil showed higher root density and finer roots compared with mulched plants; this may be explained by the necessity of the plant root system to explore the soil to reach for water. Differences in growth, groundcovering, and root characteristics were observed among species. As a result of higher mulching cost and the poorer root characteristics of mulched plants, bare soil and fast growing shrubs should be used to limit weed competition and assure a satisfactory slope greening and consolidation. This research evaluated the effectiveness of biodegradable mulches for weed control in container-grown 'Martin' giant arborvitae (Thuja plicata) and measured the effects of these mulches on evaporation and substrate temperature. The experiment was carried out in the 2008 and 2009 growing seasons. Four biodegradable mulching materials were tested and compared with a chemical control (oxadiazon) and a non-mulched/non-treated control. Two levels of overhead irrigation were evaluated: 1) daily irrigation to container capacity (well watered) and 2) daily irrigation to 30% of container capacity (water stressed). Two weed management regimes were used: 1) hand weeding three times during the growing season and 2) no weeding until the end of the growing season. Plants were potted in 3-L containers and arranged in a split-split plot design in an experimental nursery. Ornamental shoot dry weight was measured at the end of the growing season. Weed shoot dry weight per container was recorded after each hand weeding. Water content per pot (as a percentage of water-holding capacity) was measured by weighing containers every 2 hours during the day. Substrate temperature was measured in the warmest period of the day. Mulches limited weed growth to the same extent as the chemical control. In 2008, mulched plants resulted in a higher shoot dry weight than non-treated and non-mulched plants, while in the second year, no differences were observed. The black color of the 3-L containers was probably the main factor driving substrate temperature increase, indicating mulching materials did not affect substrate temperatures. In both experiments, container water content was unaffected by mulching materials. Results seem to demonstrate that transpiration is the main component of water loss from container-grown giant arborvitae plants. [ABSTRACT FROM AUTHOR]

Root fracture is a combined injury of cementum, dentin, and pulp. Many of these traumas remain untreated, mistreated, or overtreated. It leads to a more complicated treatment in case of tooth loss. Many different treatment procedures, with a very changeable success rate, have been proposed for years to treat teeth with root fractures. The objective of the following clinical studies was to evaluate the clinical effectiveness of implants placed in fresh extraction sites to treat teeth with horizontal root fracture.

Ex vivospinal cord slice cultures (SCSC) allow study of spinal cord circuitry, maintaining stimuli responses comparable to live animals. Previously, we have shown that mesenchymal stem/stromal cell (MSC) transplantation in vivoreduced inflammation and increased nerve regeneration but MSC survival was short-lived, highlighting that beneficial action may derive from the secretome. Previous in vitrostudies of MSC conditioned medium (CM) have also shown increased neuronal growth. In this study, murine SCSC were cultured in canine MSC CM (harvested from the adipose tissue of excised inguinal fat) and cell phenotypes analysed via immunohistochemistry and confocal microscopy. SCSC in MSC CM displayed enhanced viability after propidium iodide staining. GFAP immunoreactivity was significantly increased in SCSC in MSC CM compared to controls, but with no change in proteoglycan (NG2) immunoreactivity. In contrast, culture in MSC CM significantly decreased the prevalence of βIII-tubulin immunoreactive neurites, whilst Ca2+transients per cell were significantly increased. These ex vivoresults contradict previous in vitroand in vivoreports of how MSC and their secretome may affect the microenvironment of the spinal cord after injury and highlight the importance of a careful comparison of the different experimental conditions used to assess the potential of cell therapies for the treatment of spinal cord injury.

There is a growing body of evidence that flavonoids do not primarily function as UV-B screening pigments in photoprotection. Recent findings support the idea that excess light stress, irrespective of the relative proportions of the solar wavebands reaching the leaf surface, up-regulates the biosynthesis of dihydroxy B-ring-substituted flavonoid glycosides, as a consequence of and aimed at countering the generation of ROS. Intriguingly, the very conditions that lead to the inactivation of antioxidant enzymes can also up-regulate the biosynthesis of antioxidant flavonoids, which suggests flavonoids constituting a secondary ROS-scavenging system in plants exposed to severe/prolonged stress conditions. H2O2may diffuse out of the chloroplast at considerable rates and be transported to the vacuole, the storing site for flavonoids, by tonoplast intrinsic proteins, under severe excess light conditions. We suggest that the unanticipated key role of the vacuole in the ROS homeostasis might be mediated by flavonoids.
