We have previously shown that individuals with high depression scores demonstrate impaired behavioral and neural responses during social learning. Given that depression is associated with altered dopamine (DA) and serotonin (5-HT) functioning, the current study aimed to elucidate the role of these neurotransmitters in the social learning process using a dietary depletion manipulation. In a double-blind design, 70 healthy volunteers were randomly allocated to a 5-HT depletion (N= 24), DA depletion (N= 24), or placebo (N= 22) group. Participants performed a social learning task during fMRI scanning, as part of which they learned associations between name cues and rewarding (happy faces) or aversive (fearful faces) social outcomes. Behaviorally, 5-HT depleted subjects demonstrated impaired social reward learning compared to placebo controls, with a marginal effect in the same direction in the DA depletion group. On the neural level, computational modeling-based fMRI analyses revealed that 5-HT depletion altered social reward prediction signals in the insula, temporal lobe, and prefrontal cortex, while DA depletion affected social reward prediction encoding only in the prefrontal cortex. These results indicate that 5-HT depletion impairs learning from social rewards, on both the behavioral and the neural level, while DA depletion has a less extensive effect. Interestingly, the behavioral and neural responses observed after 5-HT depletion in the current study closely resemble our previous findings in individuals with high depression scores using the same task. It may thus be the case that decreased 5-HT levels contribute to social learning deficits in depression.

This study sought to determine the spectrum of brain lesions seen in heart failure (HF) patients and the extent to which lesion type contributes to cognitive impairment.

Introduction:Acute ischemic cardiac injury predisposes for cognitive impairment, dementia and depression. Beyond altered cerebral blood flow and neurohumoral activation, recent positron emission tomography data pathophysiologically suggest acute and chronic neuroinflammation, which is accompanied by astroglial activation, after acute experimental myocardial infarction (MI).Hypothesis:Peripheral serum biomarkers indicating neuronal and glial involvement are altered in the year after first acute MI.Methods:Within the investigator-initiated, prospective, multicenter diagnostic ETiCS study, we quantified serum levels of neurofilament light chain (NfL) and glial fibrillary acidic protein (GFAP) using ultra-sensitive molecular immunoassays serially in the year after first ST-elevation MI (STEMI). Cardiac magnetic resonance imaging enabled precise detection of infarction volume and ejection fraction.Results:Sufficient biomaterial was available from 45 STEMI patients (aged 28 to 78 years, median 56 years, 11% female). Median serum NfL (quartiles) at hospital admission was 10.6 (7.2, 14.8 pg/ml) and median GFAP 63.8 (47.0, 89.9 pg/ml). Absolute levels of NfL (ρ=0.78; p<0.001) and GFAP (ρ=0.56; p<0.001) correlated to patient age at all time-points. While NfL remained unaltered in the course of one year, GFAP after STEMI increased in the first 3 months after MI with a mean change of +10.6±25.0 pg/ml (p=0.007), and remained elevated after 6 months (+12.4±32.9 pg/ml; p=0.015) and 12 months (+12.6±31.2 pg/ml; p=0.010) compared to baseline. Larger relative infarction volume (ρ=0.41; p=0.009) and higher peak creatine kinase levels (ρ=0.39; p<0.001), but not ejection fraction, correlated to increases of GFAP and NfL, respectively.Conclusions:Our findings suggest subtle glial damage after STEMI which correlates to infarction volume. Further studies need to evaluate the valence of serum GFAP as a peripheral biomarker of cognitive decline after MI.

Activation of innate immunity, especially infiltration of monocytes, is critical for proper wound healing and scar formation after myocardial infarction (MI). Therefore, we tested the hypothesis that interleukin-13 (IL-13), which influences the differentiation of monocytesmacrophages and has profibrotic properties, modulates wound healing and remodeling after MI.

The World Health Organization (WHO) classification of myeloproliferative neoplasms (MPNs) comprises several entities including essential thrombocythemia (ET); primary myelofibrosis (PMF); and MPN, unclassifiable (MPN,U). Differential diagnosis between ET and early, prefibrotic PMF can be challenging but is critical because clinical course and outcome vary considerably between these entities. We have previously shown that the transcription factor nuclear factor erythroid 2 (NF-E2) is aberrantly expressed in MPN patients. Here we demonstrate that NF-E2 is mislocalized in PMF cells and that aberrant NF-E2 localization discriminates statistically highly significantly between ET and PMF. A threshold of 20% nuclear NF-E2 staining was cross-validated by “.682+ bootstrapping.” Moreover, this cutoff correctly classifies diagnostic bone marrow biopsies of MPN,U patients specified upon follow-up as ET or PMF with 92% accuracy. Because interobserver concordance between independent pathologists was high (Spearman’s rank correlation coefficient, 0.727), we propose that quantitative NF-E2 immunohistochemistry represents a diagnostic tool that can reliably support a differential diagnosis between ET and PMF.

Dibenzocycloheptanones (dibenzotropones) were prepared by Brønsted acid mediated intramolecular alkyne-carbonyl metathesis (ACM) reactions. The cyclization precursors are readily available by Sonogashira reaction of 2-bromobenzoyl chloride with terminal alkynes, followed by Suzuki reactions with benzaldehydes. The ACM reactions are highly modular and atom economic and allow for the construction of two regioisomeric series of dibenzotropones.

Introduction:Multiple myeloma (MM) relapse is common and may eventually lead to highly refractory/relapsed MM (RRMM). Therefore, novel treatment combinations are crucially needed for this highly challenging subgroup of patients (pts). The aim of the here presented phase I/II IIT was to test the tolerability and activity of a novel, so-called VBDD schedule within an outpatient regimen for extensively pretreated RRMM pts. In addition to Bortezomib, Doxorubicin and Dexamethason, which are all active and approved drugs in RRMM, Vorinostat has shown promising anti-tumor effects as a histone deacetylase inhibitor (HDACi). It inhibits the enzyme activity of HDAC 1, 2, 3 and 6, thereby allowing the activation of tumor suppressor genes. MM cells have been shown to escape from bortezomib treatment by formation of aggrosomes which desensitize cells to proteasome inhibitors and by microtubule mediated protein shuttling to lysosomes, where proteins are degradaded in order to prevent cytotoxic stress and ultimately escape from apoptosis (Fig.A). Albeit vorinostat has shown moderate activity when given alone, it has promising additive effects when combined with other antimyeloma agents, and was therefore used as an add-on agent within this RRMM regimen as it blocks microtubule coppling and aggrosome building and thereby may antagonize escape mechanisms in bortezomib-refractory pts.
