2. Clinical significance of peripheral blood-derived inflammation markers combined with serum eotaxin-2 in human colorectal cancer

- Abstract
ABSTRACTTo investigate the value of serum eotaxin-2, neutrophil/lymphocyte ratio (NLR), and platelet/lymphocyte ratio (PLR) in the diagnosis and prognosis of colorectal cancer (CRC). The association between different clinicopathological characteristics and eotaxin-2, NLR, and PLR in different patient groups was evaluated. The combined detection indicator and the combined detection's predictive effect on distant metastasis were examined. The receiver operating characteristic (ROC) curve was drawn to evaluate the efficacy of combined detection. The association between eotaxin-2, inflammation markers, and postoperative complications was assessed. Multivariate analysis to investigate the factors affecting the prognosis of patients with CRC. We detected a marked positive correlation between NLR and PLR (p < 0.05, r= 0.209). The AUC of serum eotaxin-2 combined with inflammation markers was 0.889, which was higher than single diagnosis. Compared with the single eotaxin-2 test, the combined detection of eotaxin-2 and inflammation markers might improve the specificity of CRC assessment. In univariate analysis, age, surgical method, high eotaxin-2, and high NLR were associated with postoperative complications. In multivariate analysis, age (≥ 60 years), high eotaxin-2, and high NLR were independent risk elements influencing postoperative complications of CRC. The distant metastasis, TNM staging -Ⅳ stage, NLR ≥ 3.18, and PLR ≥ 193 were independent factors affecting the prognosis of patients with CRC. The combined detection of eotaxin-2 and inflammatory markers has a particular value in improving the diagnosis of CRC, predicting distant metastasis, and guiding the frequency of reexamination after radical resection of CRC.

3. Adaptive Event-Triggered Fuzzy Control for Unreliable Networked Control Systems with Time-Varying Delay

- Abstract
This paper aims to investigate an adaptive event-triggered control scheme for unreliable networked control systems with data loss, transmission delay, and time-varying delay described by Takagi–Sugeno (T–S) fuzzy model. In order to achieve higher computational efficiency, better robustness, and higher control accuracy, an adaptive event-triggered controller based a sampling-state error mechanism is proposed. The proposal of this scheme extends the adaptive range of the event-triggered controller. On the basis of linear matrix inequality (LMIs), a new stability criterion is attained under the Lyapunov–Krasovskii functional (LKF) theory. Compared with the previous results, the scheme is less conservative. By using a numerical system and a continuous stirred tank reactor (CSTR) system, the quantitative experimental results indicate that the number of sampling in the controller is reduced, thus verifying the effectiveness and superiority of this control method.

4. Continuous Hydrogenation of Maltose over Raney Ni in a Trickle-Bed Reactor

- Abstract
Maltitol has been identified as an important value-added chemical with versatile applications and is typically manufactured via the catalytic hydrogenation of maltose, which is in urgent need of upgrading from batch to continuous operation to boost production efficiency. Here we propose a viable continuous approach for maltose hydrogenation in a trickle-bed reactor applying a millimeter-scale Raney Ni. The parametric study was performed at various temperatures, liquid flow rates, initial maltose concentrations, pressures, and hydrogen flow rates to maximize the catalytic performance and optimize the product distribution. Under the optimal reaction conditions, generally excellent conversion of maltose (91.0%) and a high yield of maltitol (87.9%) were obtained. Furthermore, a continuous hydrogenation of concentrated maltose proceeded properly, providing a slight reduction in maltose conversion but satisfactory space time yield of maltitol (0.150 gmaltitolgcat–1h–1). Detailed kinetics and density functional theory calculations were combined to unravel the reaction mechanism of maltose hydrogenation. The continuous flow system based on a trickle-bed reactor proved to have high durability and remarkable substrate suitability, thus demonstrating a reliable scale-up capability for the industrial utilization of biomass-derived sugars.

5. AMFFNet: Asymmetric Multiscale Feature Fusion Network of RGB-NIR for Solid Waste Detection

- Abstract
Multimodal fusion object detection aims to improve detection accuracy by integrating information from multiple modalities. RGB image-based object detection makes finely sorting solid waste according to material challenging. This article built a dual-camera acquisition platform using a line-scan color camera and a hyperspectral camera to collect RGB and hyperspectral images. In order to use RGB images and hyperspectral images for feature fusion more effectively, we propose an asymmetric multiscale feature fusion network (AMFFNet) based on RGB-near-infrared (NIR) multisensor fusion technology. Specifically, we designed a hyperspectral image convolution unit (HICU) to fully extract multiscale features from hyperspectral images. Second, we concatenate the hyperspectral feature maps and the feature maps output by feature pyramid networks (FPNs) of the RGB image feature extraction stage to achieve asymmetric multiscale feature fusion. In addition, a dimensionality reduction strategy (DRS) is proposed to remove the bands that are redundant and have a low signal-to-noise ratio in hyperspectral images. Ablation studies have confirmed the effectiveness of AMFFNet components, and we have conducted extensive integration experiments on the article-based solid waste dataset. The experimental results and analysis show that AMFFNet based on mask region-based convolutional neural network (Mask RCNN), faster RCNN, and RetinaNet detectors outperforms the original model by 5.05%, 3.57%, and 6.36% on AP at IoU = 0.5 and by 3.1%, 2.05%, and 3.73% on COCO’s standard AP metric, and the parameters of the network increased by 0.47–0.56 M. In conclusion, the proposed method effectively improves the performance of the object detection model for fine identification of solid waste.

6. Are public—private partnerships still an answer for social infrastructure? A systematic literature review

- Abstract
Social infrastructure has become an important element for measuring national economic development and social benefits that are usually financed in the form of government grants, private investment, and public—private partnerships (PPPs). However, social infrastructure PPPs have attracted considerable public debate due to their low profitability and complex operational demands. This study aims to answer the question, “are PPPs still an answer for social infrastructure?”, from the perspectives of the body of knowledge, application status, and prospects. Initially, an iterative search and review procedure and a scientometric analysis were performed to systematically screen the literature and to structure the body of knowledge of the social infrastructure PPPs literature. Furthermore, the application status and trends were analyzed to further explore the studied countries/regions, application sectors, and research topics. Results show that PPPs still have valuable application potential for delivering social infrastructure despite their mixed results across different topics. Six main research themes and the corresponding research trends were also identified, namely, financing and economics viability, risk management, performance management, contract and relationship management, governance and regulation, and facilitating and hindering factors. These findings offer practitioners and researchers a comprehensive overview and constructive guidance on social infrastructure PPPs.

- Abstract
Isolation of antigen-specific plasma B cells had been challenging until the recent arrival of the Beacon platform. Leveraging light-sorting technology, Beacon can perform high-throughput screening of plasma B cells on a chip to sort single cells with the desired antigen specificity. Here, we present a protocol for isolating antigen-specific plasma B cells from immunized mice using Beacon, sequencing the encoded B cell receptors (BCRs), and cloning and expressing the resulting antibodies. This protocol can easily be extended to human samples.

8. Lysine deficiency impaired growth performance and immune response and aggravated inflammatory response of the skin, spleen and head kidney in grown-up grass carp (Ctenopharyngodon idella)

- Abstract
This dissertation was primarily focused on the immune response, inflammatory response and molecular mechanisms in the skin, head kidney and spleen of grown-up grass carp (Ctenopharyngodon idella). Six iso-nitrogen diets differing in lysine concentrations (5.6, 8.5, 11.6, 14.4, 17.5 and 20.7 g/kg) were fed to 540 grass carp (164.85 ± 0.79 g) for 60 d. After that, grass carp were challenged by Aeromonas hydrophilafor 6 d. This study revealed that lysine deficiency (1) suppressed the growth performance of the fish and decreased their ability to resist skin lesion morbidity, (2) impaired the immune organ's immune response by decreasing the gene expressions of mucin, liver-expressed antimicrobial peptide (LEAP)-2B, β-defensin-1 and LEAP-2Aand the production of antibacterial compounds of grown-up grass carp, and (3) aggravated the inflammatory response of immune organs in the fish by increasing the gene expressions of pro-inflammatory cytokines (interferon γ2 [IFN-γ2], tumor necrosis factor α [TNF-α], interleukin [IL]-15, IL-17D, IL-12p40, IL-6and IL-8) and down-regulating anti-inflammatory cytokines (IL-11, transforming growth factor β1 [TGF-β1], IL-10and IL-4/13A), which were tightly correlated with signal transducer and activator of transcription (STAT)1 and STAT3 signaling pathway, respectively. The different phenomenon in the skin, spleen and head kidney of fish may be correlated with the difference in gene subtype. In addition, using quadratic regression analysis of percent weight gain (PWG), skin lesion morbidity, and the lysozyme activities in the spleen and head kidney, the dietary lysine requirements for grown-up grass carp were estimated to be 13.58, 13.51, 14.56 and 14.18 g/kg, respectively.

- Abstract
Selenium (Se), an indispensable trace element in broiler chickens, is closely associated with the growth and development of skeletal muscles. However, the role of Se in the proliferation of broiler myoblasts and its specific biological mechanisms have not been elucidated. In the present study, an in vitro growth model of broiler pectoral myoblasts cultured with Se (Na2SeO3) for 24 h was established. Using light microscopy, Cell Counting Kit-8 (CCK-8) assay, and flow cytometry, we found that compared to the control (Con) group, Se supplemental level obviously promoted myoblast proliferation and prevented cell cycle arrest from the G1 phase to the S + G2 phase. Through intracellular reactive oxygen species (ROS) generation detection, western blotting, and quantitative reverse transcription-polymerase chain reaction (qRT-PCR), the study showed that the reduced ROS production caused by Se supplementation significantly decreased PTEN expression and activated the PI3K/AKT signaling pathway in myoblasts, thereby promoting the P53/P21/CyclinD1-regulated cell cycle progression, as well as the expression of proliferation-related myogenic regulatory factors (MRFs). Our findings support the potential of Se to maintain the proliferative capacity of chicken myoblasts and emphasize the importance of Se intake in regulating skeletal muscle growth and development in poultry.

10. Dietary manganese supplementation decreases hepatic lipid deposition by regulating gene expression and enzyme activity involved in lipid metabolism in the liver of broilers

- Abstract
This study aimed to characterize the effects of different dietary forms of supplemental manganese (Mn) on hepatic lipid deposition, gene expression, and enzyme activity in liver fat metabolism in 42-d-old broiler chickens. In total 420 one-day-old Arbor Acres (AA) broilers (rooster:hen = 1:1) were assigned randomly based on body weight and sex to 1 of 6 treatments (10 replicate cages per treatment and 7 broilers per replicate cage) in a completely randomized design using a 2 (sex) × 3 (diet) factorial arrangement. The 3 diets were basal control diets without Mn supplementation and basal diets supplemented with either Mn sulfate or Mn proteinate. No sex × diet interactions were observed in any of the measured indexes; thus, the effect of diet alone was presented in this study. Dietary Mn supplementation increased Mn content in the plasma and liver, adipose triglyceride lipase (ATGL) activity, and ATGLmRNA and its protein expression in the liver by 5.3% to 24.0% (P< 0.05), but reduced plasma triglyceride (TG), total cholesterol, and low-density lipoprotein (LDL-C) levels, liver TG content, fatty acid synthase (FAS) and malic enzyme (ME) activities, mRNA expression of sterol-regulatory element-binding protein 1 (SREBP1), FAS, stearoyl-coA desaturase (SCD), and ME, as well as the protein expression of SREBP1 and SCD in the liver by 5.5% to 22.8% (P< 0.05). No differences were observed between the 2 Mn sources in all of the determined parameters. Therefore, it was concluded that dietary Mn supplementation, regardless of Mn source, decreased hepatic lipid accumulation in broilers by inhibiting SREBP1 and SCD expression, FAS and ME activities, and enhancing ATGL expression and activity.Dietary manganese supplementation reduced hepatic lipid accumulation in broiler chickens by modulating lipid metabolism-related gene expression and enzyme activity in the liver.Dietary manganese supplementation regulates lipid deposition in broiler chickens, with the liver being a significant site of lipid metabolism. This study investigated the effects of different dietary forms of supplemental manganese on hepatic lipid deposition, gene expression, and enzyme activity in the liver fat metabolism of broiler chickens. The results showed that dietary manganese supplementation decreased the hepatic lipid accumulation of broilers by inhibiting the expression of sterol-regulatory element-binding protein 1 (SREBP1) and stearoyl-coA desaturase (SCD), as well as fatty acid synthase (FAS) and malic enzyme (ME) activities, and enhancing the expression and activity of adipose triglyceride lipase (ATGL). This reduction in excessive fat production will help improve poultry health and mitigate losses in the poultry industry.

11. Zinc proteinate with moderate chelation strength enhances zinc absorption by upregulating the expression of zinc and amino acid transporters in primary cultured duodenal epithelial cells of broiler embryos

- Abstract
Recent study showed that zinc (Zn) and amino acid transporters may be involved in enhancing Zn absorption from Zn proteinate with moderate chelation strength (Zn-Prot M) in the duodenum of broilers. However, the specific mechanisms by which Zn-Prot M promotes the above Zn absorption are unknown. Therefore, in this study, 3 experiments were conducted to investigate specific and direct effects of Zn-Prot M and Zn sulfate (ZnS) on Zn absorption and expression of related transporters in primary duodenal epithelial cells of broiler embryos so as to preliminarily address possible mechanisms. In experiment 1, cells were treated with 100 μmol Zn/L as ZnS or Zn-Prot M for 20, 40, 60, 80, 100, or 120 min. Experiment 2 consisted of 3 sub-experiments. In experiment 2A, cells were treated with a Zn-unsupplemented basal medium (Control) or the basal medium supplemented with 100 or 200 μmol Zn/L as ZnS or Zn-Prot M for 60 min; in experiment 2B, cells were treated with a Zn-unsupplemented basal medium (Control) or the basal medium supplemented with 200 μmol Zn/L of as the ZnS or Zn-Prot M for 120 min; in experiment 2C, cells were treated with a Zn-unsupplemented basal medium (Control) or the basal medium supplemented with 400 or 800 μmol Zn/L as ZnS or Zn-Prot M for 120 min. In experiment 3, cells were treated with a Zn-unsupplemented basal medium (Control) or the basal medium supplemented with 400 μmol Zn/L as ZnS or Zn-Prot M for 120 min. The results of experiment 1 indicated that the minimum incubation time for saturable Zn absorption was determined to be 50.83 min using the best fit line. The results in experiment 2 demonstrated that a Zn concentration of 400 μmol/L and an incubation time of 120 min were suitable to increase the absorption of Zn from Zn-Prot M compared to ZnS. In experiment 3, Zn absorption across cell monolayers was significantly increased by Zn addition (P < 0.05), and was significantly greater with Zn-Prot M than with ZnS (P < 0.05). Compared to the control, Zn addition significantly decreased Zn transporter 10 and peptide-transporter 1 mRNA expression levels and increased y + L-type amino transporter 2 (y + LAT2) protein abundance (P < 0.05). Moreover, protein expression levels of zrt/irt-like protein 3 (ZIP3), zrt–irt-like protein 5 (ZIP5), and y + LAT2 were significantly greater for Zn-Prot M than for ZnS (P < 0.05). These findings suggest that Zn-Prot M promote Zn absorption by increasing ZIP3, ZIP5 and y + LAT2 protein expression levels in primary duodenal epithelial cells.Zinc proteinate with moderate chelation strength increased the zinc absorption in primary duodenal epithelial cells of broiler embryos by promoting the protein expression levels of ZIP3, ZIP5, and y + LAT2 compared to zinc sulfate.Our previous studies demonstrated that zinc (Zn) proteinate with moderate chelation strength (Zn-Prot M) exhibited the greatest bioavailability compared to the inorganic Zn sulfate (ZnS) and other organic Zn sources with either weak or strong chelation strength in broilers. Our recent study further showed that Zn and amino acid transporters could be potentially involved in promoting the absorption of Zn as Zn-Prot M in the duodenum of broilers. Nevertheless, further in vitro experiments are required to reveal the specific mechanisms by which Zn-Prot M promotes Zn absorption, where it is necessary first to investigate the specific and direct effect of Zn-Prot M on Zn absorption and the expression of Zn and amino acid transporters compared to that of ZnS. Therefore, we performed an in vitro study and found that Zn-Prot M increased Zn absorption and protein expression levels of the zrt–irt-like protein 3 (ZIP3), zrt–irt-like protein 5 (ZIP5), and y + L-type amino transporter 2 (y + LAT2) compared to ZnS, suggesting that ZIP3, ZIP5, and y + LAT2 might be involved in promoting the absorption of Zn from Zn-Prot M in the primary cultured duodenal epithelial cells of broiler embryos.

14. Cancer stem cell vaccine inhibits metastases of primary tumors and induces humoral immune responses against cancer stem cells

- Abstract
The inability to target cancer stem cells (CSC) may be a significant factor contributing to treatment failure. We have developed a strategy to target the CSC populations in melanoma and squamous cell carcinoma using CSC lysate-pulsed dendritic cells (DCs). The CSC-DC vaccine was administered in the adjuvant setting after localized radiation therapy of established tumors. Using mouse models we demonstrated that DCs pulsed with CSCs enriched by virtue of their expression of the CSC marker ALDH (termed CSC-DC) significantly inhibited tumor growth, reduced development of pulmonary metastases and prolonged survival. The effect was associated with downregulation of chemokine (C-C motif) receptors CCR7 and CCR10 in tumor cells and decreased expression of the chemokine (C-C motif) ligands CCL21, CCL27 and CCL28 in lung tissue. The CSC-DC vaccine significantly reduced ALDHhighCSC frequency in primary tumors. Direct targeting of CSCs was demonstrated by the specific binding of IgG produced by ALDHhighCSC-DC vaccine-primed B cells to ALDHhighCSCs, resulting in lysis of these target CSCs in the presence of complement. These data suggest that the CSC-DC vaccine approach may be useful in the adjuvant setting where local and systemic relapse are high after conventional treatment of cancers.
