Doping high-zinc aluminium alloys with Ti builds in-situ composites reinforced with ternary aluminides Ti (Al,Zn)3 with a significantly grain-refined matrix. In a series of studies, Ti was introduced with Al-6 wt% Ti (AlTi6) and Zn-4 wt% Ti (ZnTi4) master alloys in amount to contribute about 3 wt% Ti in the examined alloys. The alloy microstructure has been studied using light microscopy, SEM / EDS and XRD measurements. The observed significant refinement of the alloys matrix should lead to improvement in ductility, while the in-situ reinforcement should improve tensile strength and wear properties.

Increased grain yield (GY) is the primary breeding target of wheat breeders. We performed the genome-wide association study (GWAS) on 168 elite winter wheat lines from an ongoing breeding program to identify the main determinants of grain yield. Sequencing of Diversity Array Technology fragments (DArTseq) resulted in 19,350 single-nucleotide polymorphism (SNP) and presence-absence variation (PAV) markers. We identified 15 main genomic regions located in ten wheat chromosomes (1B, 2B, 2D, 3A, 3D, 5A, 5B, 6A, 6B, and 7B) that explained from 7.9 to 20.3% of the variation in grain yield and 13.3% of the yield stability. Loci identified in the reduced genepool are important for wheat improvement using marker-assisted selection. We found marker-trait associations between three genes involved in starch biosynthesis and grain yield. Two starch synthase genes (TraesCS2B03G1238800and TraesCS2D03G1048800) and a sucrose synthase gene (TraesCS3D03G0024300) were found in regions of QGy.rut-2B.2, QGy.rut-2D.1, and QGy.rut-3D, respectively. These loci and other significantly associated SNP markers found in this study can be used for pyramiding favorable alleles in high-yielding varieties or to improve the accuracy of prediction in genomic selection.

In the lifetime of a plant, flowering is not only an essential part of the reproductive process but also a critical developmental stage that can be vulnerable to environmental stresses. To ensure survival during drought, plants accelerate the flowering process, and this response is known as “drought escape.” HvGAMYB—transcription factor associated, among others, with flowering process and anther development in barley—has also an important role in developmental modification and yield performance in plants subjected to stressed conditions. Due to the fact that information about the mechanisms associated both with the flowering acceleration and the anther or pollen disruption is limited, the exploration of the potential HvGAMYBrole in flower development may shed light on pollen and spike morphology formations in plants grown under unfavorable water conditions. The aim of this study was to characterize differences in responses to drought among early- and late-heading barley genotypes. These two subgroups of plants—differentiated in terms of phenology—were analyzed, and traits linked to plant phenotype, physiology, and yield were investigated. In our study, the drought stress reactions of two barley subgroups showed a wide range of diversity in terms of yield performance, anther morphology, chlorophyll fluorescence kinetics, and pollen viability. The studied plants exhibited different yield performances under control and drought conditions. Moreover, the random distribution of genotypes on the biplot showing variability of OJIP parameters in the second developmental point of our investigation revealed that prolonged drought stress caused that among early- and late-heading plants, the studied genotypes exhibited different responses to applied stress conditions. The results of this study also showed that the HvGAMYBexpression level was correlated positively with traits associated with lateral spike morphology in the second developmental point of this investigation, which showed that this association occurred only under prolonged drought and highlighted the drought stress duration effect on the HvGAMYBexpression level.

ABSTRACTOne of the main signal transduction pathways that modulate plant growth and stress responses, including drought, is the action of phytohormones. Recent advances in omics approaches have facilitated the exploration of plant genomes. However, the molecular mechanisms underlying the response in the crown of barley, which plays an essential role in plant performance under stress conditions and regeneration after stress treatment, remain largely unclear. The objective of the present study was the elucidation of drought-induced molecular reactions in the crowns of different barley phytohormone mutants. We verified the hypothesis that defects of gibberellins, brassinosteroids, and strigolactones action affect the transcriptomic, proteomic, and hormonal response of barley crown to the transitory drought influencing plant development under stress. Moreover, we assumed that due to the strong connection between strigolactones and branching the hvdwarf14.dmutant, with dysfunctional receptor of strigolactones, manifests the most abundant alternations in crowns and phenotype under drought. Finally, we expected to identify components underlying the core response to drought which are independent of the genetic background. Large-scale analyses were conducted using gibberellins-biosynthesis, brassinosteroids-signaling, and strigolactones-signaling mutants, as well as reference genotypes. Detailed phenotypic evaluation was also conducted. The obtained results clearly demonstrated that hormonal disorders caused by mutations in the HvGA20ox2, HvBRI1, and HvD14genes affected the multifaceted reaction of crowns to drought, although the expression of these genes was not induced by stress. The study further detected not only genes and proteins that were involved in the drought response and reacted specifically in mutants compared to the reaction of reference genotypes and vice versa, but also the candidates that may underlie the genotype-universal stress response. Furthermore, candidate genes involved in phytohormonal interactions during the drought response were identified. We also found that the interplay between hormones, especially gibberellins and auxins, as well as strigolactones and cytokinins may be associated with the regulation of branching in crowns exposed to drought. Overall, the present study provides novel insights into the molecular drought-induced responses that occur in barley crowns.

Bacteria inject effector proteins into host cells to manipulate cellular processes that promote disease. Since bacteria deliver minuscule amounts of effectors only into targeted host cells, it is technically challenging to capture effector-dependent cellular changes from bulk-infected host tissues. Here, we report a new technique called effector-inducible isolation of nuclei tagged in specific cell types (eINTACT), which facilitates affinity-based purification of nuclei from Arabidopsisplant cells that have received Xanthomonasbacterial effectors. Analysis of purified nuclei reveals that the Xanthomonaseffector XopD manipulates the expression of Arabidopsisabscisic acid signalling-related genes and activates OSCA1.1, a gene encoding a calcium-permeable channel required for stomatal closure in response to osmotic stress. The loss of OSCA1.1causes leaf wilting and reduced bacterial growth in infected leaves, suggesting that OSCA1.1promotes host susceptibility. eINTACT allows us to uncover that XopD exploits host OSCA1.1/abscisic acid osmosignalling-mediated stomatal closure to create a humid habitat that favours bacterial growth and opens up a new avenue for accurately elucidating functions of effectors from numerous gram-negative plant bacteria in native infection contexts.

BackgroundMapping the breakpoints in de novo balanced chromosomal translocations (BCT) in symptomatic individuals provides a unique opportunity to identify in an unbiased way the likely causative genetic defect and thus find novel human disease candidate genes. Our aim was to fine-map breakpoints of de novo BCTs in a case series of nine patients.MethodsShallow whole-genome mate pair sequencing (SGMPS) together with long-range PCR and Sanger sequencing. In one case (BCT disrupting BAHD1and RET) cDNA analysis was used to verify expression of a fusion transcript in cultured fibroblasts.ResultsIn all nine probands 11 disrupted genes were found, that is, EFNA5, EBF3, LARGE, PPP2R5E, TXNDC5, ZNF423, NIPBL, BAHD1, RET, TRPS1and SLC4A10. Five subjects had translocations that disrupted genes with so far unknown (EFNA5, BAHD1, PPP2R5E, TXNDC5) or poorly delineated impact on the phenotype (SLC4A10,two previous reports of BCT disrupting the gene). The four genes with no previous disease associations (EFNA5, BAHD1, PPP2R5E, TXNDC5), when compared with all human genes by a bootstrap test, had significantly higher pLI (p<0.017) and DOMINO (p<0.02) scores indicating enrichment in genes likely to be intolerant to single copy damage. Inspection of individual pLI and DOMINO scores, and local topologically associating domain structure suggested that EFNA5, BAHD1and PPP2R5Ewere particularly good candidates for novel disease loci. The pathomechanism for BAHD1may involve deregulation of expression due to fusion with RETpromoter.ConclusionSGMPS in symptomatic carriers of BCTs is a powerful approach to delineate novel human gene–disease associations.

Structural modification of ternary aluminium – zinc – coper alloys influences their wear properties. In a series of studies, Al – 30 wt. % Zn – 3 wt. % Cu alloy (Al-30Zn-3Cu) has been doped with 1 wt.% Mn introduced with AlMn-based master alloys. The alloy microstructure and mechanical properties have been studied using light and SEM/EBSD microscopy, and measurements of wear resistance and dimensional changes. Reducing Cu content to 3 wt.% and doping with 1 wt.% Mn allows obtaining alloy of significantly refined grains, improved wear resistance and preserved high dimensional stability.

Drought has become more frequent in Central Europe causing large losses in cereal yields, especially of spring crops. The development of new varieties with increased tolerance to drought is a key tool for improvement of agricultural productivity. Material for the study consisted of 100 barley recombinant inbred lines (RILs) (LCam) derived from the cross between Syrian and European parents. The RILs and parental genotypes were examined in greenhouse experiments under well-watered and water-deficit conditions. During vegetation the date of heading, yield and yield-related traits were measured. RIL population was genotyped with microsatellite and single nucleotide polymorphism markers. This population, together with two other populations, was the basis for the consensus map construction, which was used for identification of quantitative trait loci (QTLs) affecting the traits. The studied lines showed a large variability in heading date. It was noted that drought-treatment negatively affected the yield and its components, especially when applied at the flag leaf stage. In total, 60 QTLs were detected on all the barley chromosomes. The largest number of QTLs was found on chromosome 2H. The main QTL associated with heading, located on chromosome 2H (Q.HD.LC-2H), was identified at SNP marker 5880–2547, in the vicinity of Ppd-H1gene. SNP 5880–2547 was also the closest marker to QTLs associated with plant architecture, spike morphology and grain yield. The present study showed that the earliness allele from the Syrian parent, as introduced into the genome of an European variety could result in an improvement of barley yield performance under drought conditions.

The quality of wheat depends on a large complex of genes and environmental factors. The objective of this study was to identify quantitative trait loci controlling technological quality traits and their stability across environments, and to assess the impact of interaction between alleles at loci Glu-1and Glu-3on grain quality. DH lines were evaluated in field experiments over a period of 4 years, and genotyped using simple sequence repeat markers. Lines were analysed for grain yield (GY), thousand grain weight (TGW), protein content (PC), starch content (SC), wet gluten content (WG), Zeleny sedimentation value (ZS), alveograph parameter W (APW), hectolitre weight (HW), and grain hardness (GH). A number of QTLs for these traits were identified in all chromosome groups. The Glu-D1locus influenced TGW, PC, SC, WG, ZS, APW, GH, while locus Glu-B1affected only PC, ZS, and WG. Most important marker-trait associations were found on chromosomes 1D and 5D. Significant effects of interaction between Glu-1and Glu-3loci on technological properties were recorded, and in all types of this interaction positive effects of Glu-D1locus on grain quality were observed, whereas effects of Glu-B1locus depended on alleles at Glu-3loci. Effects of Glu-A3and Glu-D3loci per se were not significant, while their interaction with alleles present at other loci encoding HMW and LMW were important. These results indicate that selection of wheat genotypes with predicted good bread-making properties should be based on the allelic composition both in Glu-1and Glu-3 loci, and confirm the predominant effect of Glu-D1dallele on technological properties of wheat grains.

In plant cells, calcium-dependent protein kinases (CDPKs) are important sensors of Ca2+flux resulting from various environmental stresses like cold, drought or salt stress. Previous genome sequence analysis and comparative studies in Arabidopsis (Arabidopsis thalianaL.) and rice (Oryza sativaL.) defined a multi-gene family of CDPKs. Here, we identified and characterised the CDPK gene complement of the model plant, barley (Hordeum vulgareL.). Comparative analysis encompassed phylogeny reconstruction based on newly available barley genome sequence, as well as established model genomes (e.g. O. sativa, A. thaliana, Brachypodium distachyon). Functional gene copies possessed characteristic CDPK domain architecture, including a serine/threonine kinase domain and four regulatory EF-hand motifs. In silico verification was followed by measurements of transcript abundance via real-time polymerase chain reaction (PCR). The relative expression of CDPK genes was determined in the vegetative growth stage under intensifying drought stress conditions. The majority of barley CDPK genes showed distinct changes in patterns of expression during exposure to stress. Our study constitutes evidence for involvement of the barley CDPK gene complement in signal transduction pathways relating to adaptation to drought. Our bioinformatics and transcriptomic analyses will provide an important foundation for further functional dissection of the barley CDPK gene family.

High-yielding capacity of the modern barley varieties is mostly dependent on the sources of semi-dwarfness associated with the sdw1/densolocus. The objective of the study was to identify quantitative trait loci (QTLs) associated with the plant height and yield potential of barley recombinant inbred lines (RILs) grown under various soil moisture regimes. The plant material was developed from a hybrid between the Maresi (European cv.) and CamB (Syrian cv.). A total of 103 QTLs affecting analysed traits were detected and 36 of them showed stable effects over environments. In total, ten QTLs were found to be significant only under water shortage conditions. Nine QTLs affecting the length of main stem were detected on 2H-6H chromosomes. In four of the detected QTLs, alleles contributed by Maresi had negative effects on that trait, the most significant being the QLSt-3H.1-1 in the 3H.1 linkage group. The close linkage between QTLs identified around the sdw1/densolocus, with positive alleles contributed by Maresi, indicates that the semi-dwarf cv. Maresi could serve as a donor of favourable traits resulting in grain yield improvement, also under water scarcity. Molecular analyses revealed that the Syrian cv. also contributed alleles which increased the yield potential. Available barley resources of genomic annotations were employed to the biological interpretation of detected QTLs. This approach revealed 26 over-represented Gene Ontology terms. In the projected support intervals of QGWSl-5H.3-2 and QLSt-5H.3 on the chromosome 5H, four genes annotated to ‘response to stress’ were found. It suggests that these QTL-regions may be involved in a response of plant to a wide range of environmental disturbances.

Hydroxamates (HX) are major secondary metabolites synthesized by rye and are responsible for some of the unique properties of this cereal, including good tolerance of biotic and abiotic stresses and allelopathy. Recently, five genes encoding enzymes taking part in HX biosynthesis have been sequenced and characterized, which was the starting point to undertake the present study. Association analysis of the content of six HX–HBOA (2-hydroxy-1,4-benzoxazin-3-one), GDIBOA (2,4-dihydroxy-1,4-benzoxazin-3(4H)-one glucoside), DIBOA (2,4-dihydroxy-1,4-benzoxazin-3(4H)-one), GDIMBOA (2,4-dihydroxy-7-methoxy-1,4-benzoxazin-3(4H)-one glucoside), DIMBOA (2,4-dihydroxy-7-methoxy-1,4-benzoxazin-3(4H)-one) and MBOA (6-methoxy-benzoxazolin-2(3H)-one) in the above-ground parts of plants and roots was performed on a population consisting of 102 and 121 diverse inbred lines, in 2013 and 2014, respectively. Altogether, 48 single nucleotide polymorphisms (SNPs) were found to be associated with the content of at least one HX: 20 SNPs were associated with HX synthesized in the above-ground parts of rye plants (AG-SNP), and 28 were associated with HX synthesized in the roots (R-SNP). The highest number of SNPs was present in genes ScBx1(9) and ScBx5(11). The majority of SNPs were affected by environmental factors, except for two: ScBx4_1702 associated with GDIBOA and MBOA contents, and ScBx5_1105 associated with HBOA content in roots.

The presented work is aimed at determining thermal diffusivity, thermal conductivity and heat capacity coefficients of silica quartz bentonite foundry sand. The values of the above thermo-physical properties were determined for temperature range of about 30 - 450 °C using the Casting measuring method [1-. The results obtained during the examinations presented in the paper can be useful when formulating boundary conditions in numerical models of heat and mass transfer in the system: casting mould ambient. The Casting method allows preserving real conditions during the experiment, i.e. contact of the mass with liquid metal and solidifying casting, and the obtained results are in a good agreement with the mean values available in literature. From the obtained results it follows that examinations should be also focused on thermo-physical properties vs. mass density dependency.

Spontaneous intracranial haemorrhage is one of the most dramatic neurological disasters. The source of haemorrhage is linked to the pathology of the arterial wall or is unknown. Because the risk of haemorrhagic stroke increases with age, we tried to investigate the relationship between age and rupture pressure of cerebral arteries. In the presented study, 51 segments of large cerebral arteries (at the level of the circle of Willis and its incoming and outgoing branches) were obtained from 33 cadaver brains aged 12–86 years. The segments were pressurized up to the rupture. The rupture pressure was noted. The highest observed pressure was 4.3atm in specimen aged 24 years. The lowest pressure 1.19atm was observed in specimen aged 80 years. The mean value of rupture pressure of all investigated segments was 2.28atm.

Abstract: Flavones and isoflavones are a major group of phenolic secondary metabolites which occur in leaves of narrow leafed lupine (Lupinus angustifolius) either as free aglycones or in a form of glycosides and malonyl-glycosides. Profiles of phenolic compounds in leaves of seedlings infected with anthracnose causing fungus Colletotrichum lupini were compared to those of healthy plants. A HPLC with diode array UV detector was used as the analytical method and identification of these secondary metabolites was confirmed with a HPLC/MSn instrument. Isomers of several target compounds differing in the glycosilation and/or malonylation pattern were detected in the studied samples. However, the application of standard HPLC with C18 columns resulted in the co-elution of several glyconjugates in single chromatographic peaks whereas for isoflavonoid aglycones complete resolution was achieved. Lupine plants grown in a greenhouse were either sprayed with the C. lupini spore suspension or the suspension was spotted on to wounded leaves. Profiles of the isoflavones were altered in result to infection with both methods. In particular, the concentration of isoflavone free aglycones detected in extracts from diseased plants was substantially increased in all of the studied samples. However, the pattern of these compounds depended on the age of lupine leaves as well as on the method of infection. Synthesis of luteone and 2′-hydroxygenistein was enhanced in the youngest leaves of plants sprayed with spores as well as in wound-infected leaves. Wighteone synthesis was induced mainly in older leaves of plants sprayed with the spore suspension.

Despite research the role of the M34T and V37I variants of GJB2 in causing hearing impairment (HI) remains controversial. Our purpose was to test a hypothesis that M34T and V37I are pathogenic but have distinct features resulting in a reduced penetrance. We screened for known GJB2/GJB6 mutations 233 Polish consecutive unrelated subjects with non‐syndromic, sensorineural HI who were previously found to carry 35delG mutation on one chromosome. The most frequent mutations were also analyzed in ∼1,000 controls. We found that M34T and V37I were significantly (P ≪ 10−6) overrepresented among patients, but their penetrance was estimated as 1/10 relative to mutations of undisputed pathogenicity. This finding apparently could not be explained by low degree of HI associated with M34T and V37I since another mutation causing comparably mild HI (L90P) did not have reduced penetrance. Subsequent analyses showed that the patients with M34T/35delG and V37I/35delG had significantly later onset of HI than patients with other genotypes (P < 10−6) including the L90P/35delG (P = 0.006). Also, among these patients (but not others) a strong correlation between the degree of HI and its duration was found (r = 0.79, P < 10−5). We tentatively suggest that M34T and V37I might cause mild HI characterized by relatively late onset and progression. © 2007 Wiley‐Liss, Inc.

The investigated material consisted of the core collection of the Polish Pisumgene bank at Wiatrowo and represented a described, monohybrid variation (type lines for characters/genes and their initial lines, tester lines with markers and lines from wild taxa, 266 accessions in total). A polymorphism in 18 isozymic loci was analyzed (Lap-1, Px-1, Aat-p, Aat-m, Aat-c, Pgm-c, Pgm-p, Gpi-c, Idh, Acp-1, Skdh, Est-1, Est-2, Est-3, Aldo, 6Pgd-p, 6Pgd-c, Dia-1). The practical outcome of the analysis was a selection of lines with a high number of fast or slow isozymic alleles, useful as tester lines with markers for chromosome mapping. A polymorphism was stated in all investigated loci with 2 to 4 alleles ranging from very fast, fast, slow to very slow. Fast and slow alleles occurred in all loci. In 11 loci the incidence of fast alleles was higher than slow ones. Conversly, slow alleles occurred more frequently than fast ones in 5 loci. Additionally, there was a low incidence of a very fast allele in 7 loci and of a very slow allele in 3 loci. A slow allele was rare in 2 loci. Primitive lines and land races were a source of these rare alleles. It is suggested to consider a frequent allele of a given locus found in a group of wild lines as a so-called wild type of Pisum.

Sodium dodecyl sulphate-polyacrylamide gel electrophoresis of reduced seed globulins covered 141 accessions of Lathyrus sativusand the following related species: L. amphicarpos, L. blepharicarpus, L. cicera, L. gorgoni, L. marmoratus, L. pseudociceraand L. stenophyllus. In the total examined material 71 globulin polypeptide bands were distinguished. The number of polypeptide bands detected in particular species varied from 14 in L. amphicarposto 29 in L. sativus; all the species under study showed intraspecific variation due to individual variation and/or differences among accessions. Electrophoretic data were subjected to statistical analysis using hierarchical UPGMA grouping. Frequency distribution of polypeptide bands in L. sativusshowed some correlations with geographical origin and certain seed characteristics (seed coat colour, seed weight) of the studied accessions. As regards interspecific relationships, the studied species showed to be distantly related taxa except for the rather closely allied L. ciceraand L. marmoratus. The obtained results are compared with the electrophoretic seed albumin data reported earlier for the same taxa.

Electrophoretic analysis of seed albumins covered 66 Lathyrus accessions representing L. amphicarpos, L. blepharicarpus, L. cicera, L. gorgoni, L. marmoratus, L. pseudocicera and L. stenophyllus. The number of major albumin bands recorded in particular species varied from two in L. amphicarpos to eight in L. gorgoni. Apart from L. amphicarpos, all the species under study showed intraspecific variation. In total, 21 bands were distinguished. The results were compared with the corresponding data reported earlier for L. sativus. All the data were statistically analysed. The species examined in this study proved to be distantly related to L. sativus. Distinctness of particular Lathyrus species was quite marked except for L. cicera and L. marmoratus which produced similar electrophoretic seed albumin patterns.

Electrophoretic analysis of seed albumins covered 105 Lathyrus sativus accessions originating from different geographical regions. Four major albumin bands were responsible for the detected variation. Due to differences in the number and combination of these bands, eleven electrophoretic seed albumin patterns were distinguished; four of them were rare phenotypes found in less than 1% of the 817 examined individuals. Electrophoretic data were subjected to statistical analysis using hierarchical UPGMA grouping and biplot techniques. Generally, no correlation was found between occurrence of either the seed albumin patterns or the major albumin bands and geographical origin of the forms under study. On the other hand, data concerning frequency distribution of the major albumin bands tended to separate two groups of accessions, white-seeded with large seeds and coloured-seeded with relatively small seeds; coloured-seeded accessions with rather large seeds mixed with the above groups.
