Inflammation resolution is an essential process for preventing the development of chronic inflammatory diseases. However, the mechanisms that regulate inflammation resolution in psoriasis are not well understood. Here, we report that ANKRD22 is an endogenous negative orchestrator of psoriasiform inflammation because ANKRD22-deficient mice are more susceptible to IMQ-induced psoriasiform inflammation. Mechanistically, ANKRD22 deficiency leads to excessive activation of the TNFRII-NIK-mediated noncanonical NF-κB signaling pathway, resulting in the hyperproduction of IL-23 in DCs. This is due to ANKRD22 being a negative feedback regulator for NIK because it physically binds to and assists in the degradation of accumulated NIK. Clinically, ANKRD22 is negatively associated with IL-23A expression and psoriasis severity. Of greater significance, subcutaneous administration of an AAV carrying ANKRD22-overexpression vector effectively hastens the resolution of psoriasiform skin inflammation. Our findings suggest ANKRD22, an endogenous supervisor of NIK, is responsible for inflammation resolution in psoriasis, and may be explored in the context of psoriasis therapy.

Dual metal atom catalysts with nitrogen coordination are emerging as promising materials that can be substituted for platinum group-based catalysts employed for the oxygen electroreduction reaction. Proper introduction of nanoparticles (NPs) in the presence of single atoms (SAs) to construct binuclear or multinuclear sites is conducive to tailoring the electronic configuration and further realizing moderate binding strength between catalytic sites and O-containing intermediates. In this work, a versatile route for synthesizing FeCu–N4sites with doping of small Fe and Cu bimetal nanoparticles co-encapsulated in nitrogen-doped porous carbon nanosheets (FeCuSNP@FeCuSA/NC) is provided for use as a bifunctional cathode catalyst in a zinc–air battery (ZAB). The experimental data and theoretical analysis indicate that the small Fe/Cu nanoparticles and FeCu SAs synergistically act to accelerate oxygen reduction reaction and oxygen evolution reaction activity under alkaline conditions. As a result, the obtained FeCuSNP@FeCuSA/NC displayed competitive half-wave potential (E1/2= 0.883 V vs.RHE) that is 36 and 15 mV higher than that of its FeSNP@FeSA/NC and CuSNP@CuSA/NC counterparts, respectively. In addition, the FeCuSNP@FeCuSA/NC-based ZAB exhibited a high specific discharge capacity of 808.8 mAh g−1, maximum power density of 187.3 mW cm−2at 325.8 mA cm−2, and excellent cycling stability at 10 mA cm−2(180 h).

Molecular catalysts with precisely designed structures and abundant metal-nitrogen active sites have garnered significant attention for their ability to efficiently perform electroreduction of carbon dioxide (ERCD). However, the catalytic efficiency and selectivity are significantly limited by the fixed electronic structure of the metal-nitrogen structure, resulting in low production efficiency of more valuable products. This work presents the electronic regulation of cobalt phthalocyanine (CoPc) through in situoxidation of N-doped Ti3C2Txnanosheets (CoPc/o–N–MXene). The resulting material exhibits high activity and tailorable selectivity. The study confirmed a strong interaction between Co-N4active sites and N–MXene after oxidation, resulting in a “Co–N–MXene” bond. This bond contributes to the transformation of intermediate ∗CO into ∗HCO, leading to a high selectivity of ERCD to methanol. This work provides a perspective on the mechanism of ERCD to methanol and an avenue for the synthesis of effective catalysts.

Toxoplasma gondii(T. gondii) infection causes severe zoonotic toxoplasmosis, which threatens the safety of almost one-third of the human population globally. However, there is no effective protective vaccine against human toxoplasmosis. This necessitates anti-T. gondiivaccine development, which is a main priority of public health. In this study, we optimized the adjuvant system 04 (AS04), a vaccine adjuvant constituted by 3-O-desacyl-4′-monophosphoryl lipid A (a TLR4 agonist) and aluminum salts, by packing it within natural extracts of β-glucan particles (GPs) from Saccharomyces cerevisiaeto form a GP–AS04 hybrid adjuvant system. Through a simple mixing procedure, we loaded GP–AS04 particles with the total extract (TE) of T. gondiilysate, forming a novel anti-T. gondiivaccine GP–AS04–TE. Results indicated that the hybrid adjuvant can efficiently and stably load antigens, mediate antigen delivery, facilitate the dendritic uptake of antigens, boost dendritic cell maturation and stimulation, and increase the secretion of pro-inflammatory cytokines. In the mouse inoculation model, GP–AS04–TE significantly stimulated the function of dendritic cells, induced a very strong TE-specific humoral and cellular immune response, and finally showed a strong and effective protection against toxoplasma chronic and acute infections. This work proves the potential of GP–AS04 for exploitation as a vaccine against a range of pathogens.

Radionuclide extraction from wastewater is a long-term process, in which the study on the reuse and decomposition of adsorbents provides the ability to complete the post-treatment after adsorption. Herein, A novel biodegradable amidoxime chitosan has been synthesized through one-step without crosslinking agent and characterized by FT-IR, SEM, XPS, TGA and element analysis. The batch adsorption experiments of U(VI) and Th(IV) on AO-CTS adsorbent were studied and maximum adsorption of U(VI) and Th(IV) were 97 and 56 mg/g, respectively. The U(VI) and Th(Ⅳ) can be effectively desorbed from the AO-CTS materials at low acidity, The AO-CTS can be reused 6 times without reducing absorbency for U(VI) and Th(Ⅳ). When finish the adsorption process, the AO-CTS can be degraded by lysozyme at room temperature, there were no toxic or harmful substances are produced.

Solar-driven interfacial evaporation is a promising sustainable strategy to address global water scarcity. However, developing large-area, high-performance, multifunctional anti-fouling, and energy-using solar steam collection systems remains a significant challenge. Here, inspired by nature, we designed and prepared 3D-Felt fabric evaporators with hydrophilic MXene and chitosan&dopamine modified concave array structures, featuring multilayer mesh porous structures and adjustable concave sizes. Benefiting from the unique structural advantages, this evaporator has high light absorption ability (98.1%), balance between interfacial heating and water supply rate, effective vapor diffusion and low water evaporation enthalpy. The 3D evaporator achieves an evaporation rate of up to 2.69 kg m−2h−1under one-sun illumination, an impressive photothermal conversion efficiency of 97.09%, and an outdoor evaporation volume of up to 11.64 kg m−2. It exhibits high antibacterial (efficiency 99%), salt resistance (200 h in 15 wt% NaCl solution) and oil contamination resistance (underwater oil contact angle of 150°), allowing for long-term stable water purification. Importantly, the integrated solar evaporation system simultaneously provides effective desalination, self-sustaining irrigation for agriculture, and thermal energy-powered electricity generation. This work offers practical demonstration and good purifying performance, inspiring new paradigms for developing high performance solar vapor technology.

Although flexible and multifunctional textile-based electronics are promising for wearable devices, it is still a challenge to seamlessly integrate excellent conductivity into textiles without sacrificing their intrinsic flexibility and breathability. Herein, the vertically interconnected conductive networks are constructed based on a meshy template of weave cotton fabrics with interwoven warp and weft yarns. The two-dimensional early transition metal carbides/nitrides (MXenes), with unique metallic conductivity and hydrophilic surfaces, are uniformly and intimately attached to the preformed fabric via a spray-drying coating approach. Through adjusting the spray-drying cycles, the degree of conductive interconnectivity for the fabrics is precisely tuned, thereby affording highly conductive and breathable fabrics with integrated Joule heating, electromagnetic interference (EMI) shielding and strain sensing performances. Interestingly, triggered by the interwoven conductive architecture, the MXene-decorated fabrics with a low loading of 6 wt % (0.78 mg cm–2) offer an outstanding electrical conductivity of 5 Ω sq–1. The promising electrical conductivity further endows the fabrics with superior Joule heating performance with a heating temperature up to 150 °C at a supply voltage of 6 V, excellent EMI shielding performance, and highly sensitive strain responses to human motion. Consequently, this work offers a novel strategy for the versatile design of multifunctional textile-based wearable devices.

The imbalance of T lymphocyte subsets substantially conduces to disturbed intestinal immune system and succeeding colonic tissue damage in inflammatory bowel diseases. It is considered that regulation of phytochemicals on cytokine production potentially provides a broad prospect for the exploitation of immunomodulatory agents. Here, we reported that oral administration of feruloylated oligosaccharides (FOs) effectively alleviated mice colitis disease induced by dextran sulfate sodium (DSS). FOs decreased the percentage of T helper (Th)17 cells and downregulated the production of Th17-specific cytokines. In contrast, FOs increased the percentage of regulatory T (Treg) cells and elevated the production of Treg-specific cytokines in colons of DSS-challenged mice. These results indicated that FOs restored the immunologic equilibrium of Th17 and Treg subsets, hereby ameliorating the deterioration of colitis. Furthermore, FOs diminished the secretion of interleukin (IL)-23 and IL-6 but enhanced the transforming growth factor-β1 (TGF-β1) in dendritic cells in vitro and in vivo, which contributed to the restoration of Th17 and Treg cells immune balance. The mechanistic analysis showed that the regulation of FOs on IL-23 and IL-6 was associated with the nuclear factor-κ-gene binding signaling pathway and TGF-β1 with mitogen-activated protein kinase-activator protein 1 signaling pathway. Taken together, oral administration of FOs exerted potent immunomodulatory effects against mice colitis via restoring the immune balance of Th17 and Treg cells.

Amino acid (aa) metabolism is closely correlated with the pathogenesis of psoriasis; however, details on aa transportation during this process are barely known. Here, we find that SLC38A5, a sodium-dependent neutral aa transporter that counter-transports protons, is markedly upregulated in the psoriatic skin of both human patients and mouse models. SLC38A5 deficiency significantly ameliorates the pathogenesis of psoriasis, indicating a pathogenic role of SLC38A5. Surprisingly, SLC38A5 is almost exclusively expressed in dendritic cells (DCs) when analyzing the psoriatic lesion and mainly locates on the lysosome. Mechanistically, SLC38A5 potentiates lysosomal acidification, which dictates the cleavage and activation of TLR7 with ensuing production of pro-inflammatory cytokines such as interleukin-23 (IL-23) and IL-1β from DCs and eventually aggravates psoriatic inflammation. In summary, this work uncovers an auxiliary mechanism in driving lysosomal acidification, provides inspiring insights for DC biology and psoriasis etiology, and reveals SLC38A5 as a promising therapeutic target for treating psoriasis.

Abstract: Solar steam generation technology has emerged as a promising approach for seawater desalination, wastewater purification, etc. However, simultaneously achieving superior light absorption, thermal management, and salt harvesting in an evaporator remains challenging. Here, inspired by nature, a 3D honeycomb-like fabric decorated with hydrophilic Ti3C2Tx(MXene) is innovatively designed and successfully woven as solar evaporator. The honeycomb structure with periodically concave arrays creates the maximum level of light-trapping by multiple scattering and omnidirectional light absorption, synergistically cooperating with light absorbance of MXene. The minimum thermal loss is available by constructing the localized photothermal generation, contributed by a thermal-insulating barrier connected with 1D water path, and the concave structure of efficiently recycling convective and radiative heat loss. The evaporator demonstrates high solar efficiency of up to 93.5% and evaporation rate of 1.62 kg m−2h−1under one sun irradiation. Moreover, assisted by a 1D water path in the center, the salt solution transporting in the evaporator generates a radial concentration gradient from the center to the edge so that the salt is crystallized at the edge even in 21% brine, enabling the complete separation of water/solute and efficient salt harvesting. This research provides a large-scale manufacturing route of high-performance solar steam generator.
