Several fundamental research and applications in biomedicine and microfluidics often require controlled manipulation of suspended micro- and nanoscale particles. Speckle tweezers (ST) by incorporating randomly distributed light fields have been used to control micro-particles with refractive indices higher than their medium and to perform manipulation tasks such as guiding and sorting. Indeed, compared to periodic potentials, ST represents a wider possibility to be operated for such tasks. Here, we extend the usefulness of ST into micro-particles of low index with respect to the surrounding. Repelling of such particles by high intensity regions into lower intensity regions makes them to be locally confined, and the confinement can be tuned by changing the average grain intensity and size of the speckle patterns. Experiments on polystyrenes and liposomes validate the procedure. Moreover, we show that ST can also manipulate the nano-particle (NP)-loaded liposomes. Interestingly, the different interactions of NP-loaded and empty liposomes with ST enable collective manipulation of their mixture using the same speckle pattern, which may be explained by inclusion of the photophoretic forces on NPs. Our results on the different behaviors between empty and non-empty vesicles may open a new window on controlling collective transportation of drug micro-containers along with its wide applications in soft matter.

Autobiographical memory distortions are a key feature of posttraumatic stress disorder (PTSD). In this proof-of-concept randomized controlled trial (N= 43), we evaluated an autobiographical memory flexibility intervention, MemFlex. We aimed to determine whether the mechanism-focused intervention, which aims to improve autobiographical memory processes, may also affect other cognitive predictors of PTSD and potentially reduce PTSD symptoms in Iranian trauma survivors diagnosed with PTSD. Results indicated significant, moderate to large between-groups effect sizes in favor of MemFlex, relative to wait-list control, for the targeted cognitive mechanism of autobiographical memory flexibility and PTSD symptoms. A large, significant effect was also observed on maladaptive posttraumatic cognitions—a strong predictor of PTSD prognosis, which is a key target of high-intensity cognitive therapies for PTSD. Findings support future completion of a scaled-up trial to evaluate treatment efficacy of MemFlex for PTSD to determine whether MemFlex may offer a culturally adaptive, low-cost, low-intensity intervention able to improve cognitive mechanisms of PTSD.

Investigating real-time phenomena in bio-polymers has received much attention because of their increasing demands in polymer substitution. The 3D morphometry of polymer surfaces may be very impactful in such studies. Here, digital holographic microscopy (DHM) is applied for quantitative measurement of the rare morphological changes of UV-A and UV-C exposed nanocomposites during their incubation with excess water. By reconstructing the recorded successive digital holograms, the time evolution of the swelled regions of the samples is derived. Our results clearly show that the higher water swelling of UV-A irradiated starch/kefiran/ZnO may be attributed to its higher hydrophilicity.

Reduced ability to retrieve specific autobiographical memories is a well-defined feature of posttraumatic stress disorder (PTSD), and science-driven interventions have emerged to improve memory specificity and thereby symptoms. However, research in depressed samples indicates that the ability to flexibly move between retrieval of specific and general memory types (i.e., memory flexibility) may more accurately conceptualize autobiographical memory deficits in emotional disturbance. In this study, we evaluated memory specificity and memory flexibility in Iranian trauma survivors (N= 63) with and without PTSD relative to community control participants. Trauma-exposed participants had experienced a serious road-traffic accident. Results indicated that individuals with PTSD experienced reduced memory specificity and memory flexibility relative to trauma-exposed participants and community control participants. A small sample size limits the strength of conclusions, although good statistical power was obtained. Findings suggest that reduced memory flexibility may be a transdiagnostic marker of emotional disturbance and support further development of memory flexibility interventions for PTSD.

Scaffold-based bone tissue engineering aims to develop 3D scaffolds that mimic the extracellular matrix to regenerate bone defects and damages. In this paper, we provide a laser speckle analysis to characterize the highly porous scaffold. The experimental procedure includes in situ acquisition of speckle patterns of the bone scaffold at different times under preserved environmental conditions, and follow-up statistical post-processing toward examining its internal activity. The activity and overall viscoelastic properties of scaffolds are expressed via several statistical parameters, and the variations in the computed parameters are attributed to time-varying activity of the samples during their internal substructure migration.

Mueller matrix microscopy (MMM) is a powerful approach to probe microstructural and optical information of many important specimens (e.g., tissue and bacteria), which otherwise cannot be obtained directly from intensity or spectral images. Achieving high lateral resolution in MMM, similar to other microscopy approaches, remains a challenge. Here, we extend the idea of microsphere (MS) -assisted microscopy into MMM toward resolution-enhanced polarimetric imaging. The goal is achieved by insertion of a transparent MS in the working distance of the imaging microscope objective in the optical train of an MMM system. We experimentally show that an MS close to the sample in MMM may increase the resolution beyond the intrinsic diffraction limit of the system by redirecting the higher spatial frequencies of the sample into the acceptance cone. In order to be a case in point, the experiment is conducted on a standard holographic diffraction grating with 1 µm line-width, which is beyond the diffraction limit of a 10× objective. Two-dimensional images of the Mueller matrix and some of the widely used quantitative polarimetric parameters of the sample are calculated and compared in the two cases before and after insertion of MS. The proposed arrangement is easy to implement and has the potential to serve as a high-resolution polarimetric microscope for visualizing the polarization characteristics of the microscopic objects.

In this paper, we show that laser speckle analysis (LSA) can provide valuable information about the structure of crumpled thin sheets. Crumpling and folding of slender objects are present in several phenomena and in various ranges of size, e.g., paper compaction, cortical folding in brains, DNA packing in viral capsids, and flower buds, to name a few. The analysis of laser speckles, both numerical and graphical, is a source of information about the activity of biological or non-biological materials, and the development of digital electronics, which brought the ease of image processing, has opened new perspectives for a spectrum of LSA applications. LSA is applied on randomly crumpled and one-, two-, and three-times folded papers, and appreciable differences in LSA parameters are observed. The methodology can be applied for easy-to-implement quantitative assessment of similar phenomena and samples.

In this paper, we measure the proximity effect of a flat wall on the sedimentation of colloidal particles by the use of digital holographic microscopy (DHM). DHM provides quantitative phase contrast and three-dimensional (3D) imaging in arbitrary time scales that are suitable to investigate various phenomena, including dynamic behavior of colloids. 3D information is obtained by post-processing of the recorded digital holograms through the angular spectrum propagation approach for numerical reconstruction. We have measured the trajectory and velocity of spherical colloids in various distances from a flat wall while they sediment under gravity. The results are in agreement with theoretical studies of Goldman et al. [Chem. Eng. Sci.22, 637 (1967)CESCAC0009-250910.1016/0009-2509(67)80047-2]. We observed an increase in the drag force due to the proximity to the flat wall, which was effective for distances up to several particle sizes. The presented method is capable of being used for different applications in study of colloidal dynamics where 3D information in real time is required.

Scanning probe microscopy techniques are widely applied as three-dimensional (3D) metallography methods in studying metallic microstructures. While providing high lateral and vertical resolutions, these techniques suffer from a limited field-of-view, which is a crucial requirement in many metallurgy studies. Moreover, they are complex and expensive, and need a complicated specimen preparation procedure. In this paper, we utilized reflective digital holographic microscopy for 3D microstructural characterization of metallic surfaces within an adjustable field-of-view and a resolution close to diffraction-limit. Polished and etched CK45 plain carbon steel and AISI 304 stainless steel samples have been studied with the presented method. The experimental results show the capability of the method for metallurgic granular and intergranular studies. The method has the potential to be used in a variety of metallurgic phenomena such as crystal plasticity, microstructural oxidation, and intergranular corrosion.

In this paper, we use a glass microsphere incorporated into a digital holographic microscope to increase the effective resolution of the system, aiming at precise cell identification. A Mirau interferometric objective is employed in the experiments, which can be used for a common-path digital holographic microscopy (DHMicroscopy) arrangement. High-magnification Mirau objectives are expensive and suffer from low working distances, yet the commonly used low-magnification Mirau objectives do not have high lateral resolutions. We show that by placing a glass microsphere within the working distance of a low-magnification Mirau objective, its effective numerical aperture can be increased, leading to super-resolved three-dimensional images. The improvement in the lateral resolution depends on the size and vertical position of microsphere, and by varying these parameters, the lateral resolution and magnification may be adjusted. We used the information from the super-resolution DHMicroscopy to identify thalassemia minor red blood cells (tRBCs). Identification is done by comparing the volumetric measurements with those of healthy RBCs. Our results show that microsphere-assisted super-resolved Mirau DHMicroscopy, being common path and off-axis in nature, has the potential to serve as a benchtop device for cell identification and biomedical measurements.

In the last decade, the transport of intensity has been increasingly used in microscopy, wavefront sensing, and metrology. In this study, we verify by simulation and experiment the use of the transport of intensity equation (TIE) in the accurate testing of optical aspheric surfaces. Guided by simulation results and assuming that the experimental setup parameters and the conic constants are known, one can estimate an appropriate defocusing distance Δz that leads to an accurate solution of the TIE. In this paper, this method is verified through the construction of a non-nulled experiment for testing the 2D profile of an aspheric surface. The theoretical method and experimental results are compared to validate the results. Finally, to validate the TIE methodology, the phase distribution obtained by TIE is compared with the phase distribution obtained by a Shack–Hartmann sensor.

Starch/kefiran/ZnO nanocomposite films may exhibit different physicochemical properties depending on the distribution of ZnO nanoparticles. As a result of UV exposure, the hydrophobicity of the nanoparticles may be modified, resulting in their dispersion in the polymer matrix. The aim of this paper is to characterize starch/kefiran/ZnO nanocomposite films using dynamic speckle analysis. In this experiment, speckle patterns of the nanocomposite are acquired in situ under controlled moisture, pressure, and temperature conditions. This is followed by a statistical postprocessing procedure to determine the deformation pattern of the nanocomposite. A numerical analysis of the successive speckle patterns is used to determine the time evolution of sample deformation. There is a correlation between the intensity and contrast of speckle patterns and the temporal alteration of the polymer. Several factors have been considered to examine the structural evolution of the nanocomposite, including time history speckle pattern, co-occurrence, graphical speckle contrast, roughness parameter, auto-correlation, and Shannon entropy. The variation and overall viscoelastic properties of the nanocomposites are expressed via several statistical parameters. The changes in the computed parameters are attributed to the time-varying activity of the samples during their higher hydrophilicity.

This research investigated the influence of culture and posttraumatic stress disorder (PTSD) on global autobiographical remembering (Study 1a) and on the phenomenological properties (Study 1b) and memory-content variables (Study 1c) of trauma-specific autobiographical remembering. Australian, British, and Iranian trauma survivors with and without PTSD completed the Autobiographical Memory Test, Self-Defining Memory Task, and Autobiographical Memory Questionnaire and provided trauma- and negative-memory narratives. We found that there were pan-cultural deficits and distortions in the global autobiographical remembering of participants with PTSD (Study 1a). In addition, the presence of PTSD moderated the usual effect of culture on the phenomenological properties of the trauma memory (Study 1b). Finally, participants with PTSD, regardless of cultural background, had significantly fewer expressions of autonomy and self-determination in their autobiographical remembering than did those without PTSD (Study 1c). The findings suggest that pan-culturally, individuals with PTSD have similar disruptions and distortions in their autobiographical remembering.

We present an integrated optical system for three-dimensional (3D) imaging of micrometer-sized samples, while immobilizing and manipulating the samples by means of an optical fiber trap. Optical traps allow us to apply and measure pico-Newton-sized forces, and perform detailed measurements of micrometer-sized dielectric systems in the field of biology. The integrated 3D system can be used as a major tool in the field of biophysics. The trap is built using a tapered optical fiber to enhance the effective numerical aperture of the fiber. The trapping system is mounted on a conventional microscope, in which the two eyepieces’ output ports are used as the paths of an off-axis self-referencing digital holographic microscopy (DHM) setup. The trap is calibrated using a high-speed camera, and trap stiffness is determined through the power spectrum method. The compact setup provides an elegant apparatus for temporally stable DHM for 3D imaging of optically controlled samples. Three-dimensional information and quantitative phase contrast images of the trapped samples are obtained by postprocessing the recorded digital holograms. Experiments were performed on lipids and red blood cells. Quantitative phase contrast images and temporal evolution of optical thickness of trapped samples are presented.

Myelin figures (MFs) are cylindrical multilamellar lipid tubes that can be found in various healthy and diseased living cells. Their formation and dynamics involve some of the most mysterious configurations that lipid molecules can adopt under certain conditions. They have been studied with different microscopy methods. Due to the frequent coiling of their structure, the usual methods of microscopy fail to give precise quantitative information about their dynamics. In this paper, we introduced Digital Holographic Microscopy (DHM) as a useful method to calculate the precise dynamical volume, thickness, surface and length of the myelin figures. As an example of DHM imaging of myelin figures, their structure and growth rate in the presence and absence of temperature gradient have been studied in this work. We showed that the thickness of a myelin figure can be changed during the first few seconds. However, after approximately ten seconds, the thickness stabilizes and does not alter significantly. We further studied the effect of the thermal gradient on the length growth. The calculation of the length growth from the measurement of the myelin figure volume shows that the length (L) grows in time (t) as L∝tat the early stage of the myelin protrusion in both the presence and the absence of the thermal gradient. However, thermal gradient facilitates the growth and increases its rate.

Crimean-Congo haemorrhagic fever (CCHF) virus is a tick-borne member of the genus Nairovirus, family Bunyaviridae. CCHF virus has been isolated from at least 31 different species of ticks. The virus is transmitted through the bite of an infected tick or by direct contact with CCHF virus-infected patients or the products of infected livestock. This study was conducted to determine the rate of CCHF virus infection in ticks in the district of Zahedan, in the province of Sistan and Baluchistan, southeastern Iran. A total of 140 ticks were collected from Sistan and Baluchistan. Reverse transcriptase-polymerase chain reaction (RT-PCR) was used for the detection of the CCHF virus genome in the tick population. This genome was detected in 4.3% of ticks collected from livestock of different regions of Zahedan. The infected tick genera belonged to Hyalommaand Haemaphysalis. Although in the epidemiology of CCHF virus Hyalommaticks are considered to be the most important vectors and reservoirs, the virus has also been reported to occur in other genera of ticks, which conforms to the current data in our study from Sistan and Baluchistan. Given that animals are common hosts for Hyalommaand Haemaphysalis, regular monitoring programmes for livestock should be applied for CCHF virus control.

Interference of light has numerous metrological applications because the optical path difference (OPD) can be varied at will between the interfering waves in the interferometers. We show how one can desirably change the optical path difference in diffraction. This leads to many novel and interesting metrological applications including high-precision measurements of displacement, phase change, refractive index profile, temperature gradient, diffusion coefficient, and coherence parameters, to name only a few. The subject fundamentally differs from interferometry in the sense that in the latter the measurement criterion is the change in intensity or fringe location, while in the former the criterion is the change in the visibility of fringes with an already known intensity profile. The visibility can vary from zero to one as the OPD changes by a half-wave. Therefore, measurements with the accuracy of a few nanometers are quite feasible. Also, the possibility of changing the OPD in diffraction allows us to use Fresnel diffraction in Fourier spectrometry, to enhance or suppress diffracted fields, and to build phase singularities that have many novel and useful applications.

Digital holographic microscopy (DHM) provides a non-destructive and quantitative phase contrast imaging that is suitable for high resolving investigations of living cells. On the other hand, for many applications including cell analysis the immobilization of the sample under study is a crucial task. Optical trap is an elegant candidate to immobilize the micro samples. In this work, DHM is integrated with an optical trapping setup. This combination is of particular advantage for quantitative visualization of three dimensional structures that are trapped by laser beam. The recorded hologram by CCD can be post-processed to three dimensional (3D) reconstruction of the trapped objects.
