The effect of the anti-dopaminergic drug haloperidol (6 mg/day i.m. for 2 days) on the blink reflex elicited by electric stimulation of supraorbital nerves was investigated in 12 adult volunteers. A significant increase in amplitude of the early component (R1) of the reflex was observed. This increase disappeared within 5 days of stopping the drug. Possible neural systems which might be involved in these effects are considered.

In cats with mesencephalic decerebration sparing the cerebral peduncles and ablation of sensorimotor cortex, changes in firing of single neurons of caudate nucleus (CD), putamen (PU), globus pallidus (GP) and entopeduncular nucleus (EN) were studied following stimulation of the ipsilateral medullary pyramidal tract (MPT). Cells in CD and PU were not extensively influenced by impulses backfired from MPT (14.7% and 18.7%, respectively). Conversely, a larger number of GP cells (28.1%) and especially EN cells (46.9%) exhibited pronounced changes in their firing following MPT stimulation. The MPT-induced effects on CD and PU were either inhibition or excitations, the latter appearing at latencies greater than 11 ms. The responses observed in GP and EN cells were most frequently excitations, some of which appeared with latencies below 5 ms.

In unanesthetized neuraxis intact cats microstimulation of the interpositus nucleus (IN) which activated a single flexor or extensor muscle in limbs, was used to investigate changes of unitary discharges of rubrospinal (RST) cells. Recordings were made from sites the stimulation of which excited the same muscle activated by the IN (agonist cells), its antagonist (antagonist cells) or heteronymous muscles (heteronymous cells).

Motor effects produced by microstimulation of restiform body (RB) were studied in acute unanesthetized cats, using tungsten electrodes for stimulating the peduncle and bipolar steel electrodes for recording muscular activity (EMG). The main results were the following.1.Threshold microstimulation (18.24 µA±8.77 S.D.) of effective foci within RB elicited single muscle contractions of ipsilateral limbs, primarily of forelimb; overthreshold activation (32.83 µA±9.25 S.D.) of the same points produced complex movements in 61.54% of cases that involved muscles of shoulder, neck, and trunk.2.Single muscle contractions exhibited a mean latency (20.09 msec ± 2.04 S.D.) which was significantly longer than that shown by complex movements (10.00 msec±3.10 S.D.). Furthermore, a decrease in frequency of stimulating train below 300 Hz and a reduction in duration below 30 msec caused a steep rise of threshold for single muscle responses that was not observed when studying complex movements.3.Acute RB interruption between stimulating electrode and cerebellum abolished single muscle contractions; conversely, complex movements remained unmodified even when the RB was lesioned in cats chronically submitted to interruption of brachium conjunctivum (BC).4.The pathway involved in promoting RB induced single muscle activation includes interpositus nucleus, BC and rubrospinal tract.

Encoding of information by neuronal populations implies a correlation of neural activity across cells. Therefore recent evidence for correlated activity in different parts of the nervous system has been interpreted as a possible mechanism for functional coupling or coordinated information processing. We compared the activity in the mossy fiber projection, the dorsal spinocerebellar tract (DSCT) with the responses of Purkinje cells in the spinocerebellum for evidence of correlated activity. A principal component analysis of post-stimulus spike activity revealed temporal patterns of correlated Purkinje cell activity in all parts of the spinocerebellum evoked by rapid, small amplitude passive joint rotations in anesthetized cats. The same basic temporal patterns were induced in DSCT neurons by joint rotations and also by direct nerve stimulation, showing that the patterning resulted from centrally generated rather than peripheral timing. The evidence implies that spinal timing generators, activated by mechanical stimulation of the ipsilateral hindlimb, transmit temporally correlated activity to the cerebellum via spinocerebellar pathways resulting in a coherent modulation of activity in diverse areas of the cerebellar cortex thereby providing the basis for a functional coupling.

The efficacy of type la synapse on alpha-motoneurons of soleus and lateral gastrocnemius muscles has been investigated, using the H-reflex technique, in athletes engaged in sports requiring very rapid and intense contractions (sprinters and volley-ball players) as well as in non-trained subjects. It has been observed, in both muscles, that the ratio between the mean value of the maximal reflex response (Hmax) and the mean value of the maximal direct response (Mmax) elicited upon electrical stimulation of the tibial nerve is significantly smaller in athletes trained for explosive-type movements than in non-trained subjects. This difference in the Hmax:Mmax ratio was dependent on a smaller amplitude of Hmax and not on a greater amplitude of Mmax. No significant differences were observed between sprinters and volley-ball players. In both trained and non-trained subjects, soleus and lateral gastrocnemius muscles displayed significant differences in Hmax: Mmax ratio and Mmax amplitude but not in Hmax amplitude. Since the H-response is considered to be due mainly to activation of the smallest motoneurons in the motoneuronal pools, the difference in Hmax amplitude and Hmax:Mmax ratio between athletes and non-trained subjects could have been dependent on a lower incidence of these motoneurons in the athletes. This is in accord with the mechanical needs of muscles during explosive-type power training. Although this difference ,ay have been wholly determined genetically, the possibility is discussed as to whether the lower incidence in sprinters and volley-ball players of small motoneurons could have been related to a training-induced transformation of small and slow motoneurons into large and fast ones.

A theoretical model is described which in response to combinations of poissonian pulse trains with different mean frequencies on three independent incoming lines, generated output signals simulating spontaneous discharges of thalamic ventrobasal (VB) neurones during sleep and wakefulness. Some dynamic neuronal properties as refractoriness, facilitation, short term memory were simulated and characteristics of response to single pulses on different lines properly selected to reproduce those exhibited by VB neurones upon artificial stimulation of thalamic afferent systems. The data obtained from the model are briefly discussed in relation to possible contributions of specific and nonspecific afferent systems in producing spontaneous VB discharges characteristic of different levels of vigilance.
