We studied the reactivity of T and B cells against a soluble form of the glycoprotein of vesicular stomatitis virus (VSV-G) which was expressed in a transgenic mouse (line 23) under the control of the hormone regulated β-lactoglobulin promoter. Transgenic mice expressed VSV-G in the thymus, spleen, mammary gland and lung. VSV-G transcripts in the thymus varied with age, i.e., expression was high early in life and decreased with age. VSV-G transgenic mice immunized with recombinantvacciniavirus expressing VSV-G exhibited normal VSV-G-specific IgM levels, but a 30-fold reduction in IgG response, indicating functional VSV-G-specific B cell activity but impaired T helper cell responses. Interestingly, VSV-G-specific T helper cell activity was reduced only early (4–10 weeks) and late in life (>40 weeks) but was normal in between. Double transgenic mice expressing VSV-G and a VSV-G-specific TCR (line 23×7) demonstrated that TCR transgenic CD4+T cells were partially deleted in early life, but then gradually repopulated the periphery and remained constant. These findings suggest that in line 23 two different mechanisms regulated levels of the immune response: clonal reduction/deletion of VSV-G-specific T cells during early life followed by peripheral anergy at a later stage.

The ‘protein only’ hypothesis postulates that the prion, the agent causing transmissible spongiform encephalopathies, is PrP(Sc), an isoform of the host protein PrP(C). Protease treatment of prion preparations cleaves off approximately 60 N‐terminal residues of PrP(Sc) but does not abrogate infectivity. Disruption of the PrP gene in the mouse abolishes susceptibility to scrapie and prion replication. We have introduced into PrP knockout mice transgenes encoding wild‐type PrP or PrP lacking 26 or 49 amino‐proximal amino acids which are protease susceptible in PrP(Sc). Inoculation with prions led to fatal disease, prion propagation and accumulation of PrP(Sc) in mice expressing both wild‐type and truncated PrPs. Within the framework of the ‘protein only’ hypothesis, this means that the amino‐proximal segment of PrP(C) is not required either for its susceptibility to conversion into the pathogenic, infectious form of PrP or for the generation of PrP(Sc).

We have examined the expression of the recombination activating gene RAG-1 by in situ hybridization to thymi from mice bearing transgenes for the T-cell receptor (TCR) alpha chain, TCR beta chain, or both TCR alpha and beta chains. RAG-1 transcription was found in the thymic cortex of transgenic mice carrying a single TCR alpha- or TCR beta-chain transgene, comparable to normal mice. However, RAG-1 transcription was strikingly reduced in the thymic cortex from transgenic mice carrying both TCR alpha- and beta-chain genes and expressing major histocompatibility complex (MHC) class I (H-2b) molecules necessary for positive selection of the transgenic TCR. In contrast, thymi of transgenic mice also carrying both TCR alpha- and beta-chain genes but expressing MHC molecules (H-2d) that did not positively select the transgenic TCR displayed high levels of RAG-1 transcription. The low thymic RAG-1 expression coincided with high transgenic TCR alpha-chain surface expression and with inhibition of endogenous TCR alpha-chain rearrangement. Our findings suggest that binding of the TCR to self MHC molecules during positive selection down-regulates RAG-1 transcription in cortical thymocytes and thereby prevents further TCR alpha-chain rearrangements.

Gene disruptions and deletions of up to 20kb have been generated by homologous recombination with appropriate targeting vectors in murine embryonic stem (ES) cells. Because we could not obtain a deletion of about 200 kb in the mouse amyloid precursor protein gene by the classical technique, we employed strategies involving the insertion of loxP sites upstream and downstream of the region to be deleted by homologous recombination and elicited excision of the loxP-flanked region by introduction of a Cre expression vector into the ES cells. In the first approach, the loxP sequences were inserted in two successive steps and after each step, ES cell clones were isolated and characterized. Deletion of the loxP-flanked sequence was accomplished by introducing the cre gene in a third step. In the second approach, ES cells containing the upstream loxP cassette were electroporated simultaneously with the downstream loxP targeting vector and the Cre expression plasmid. ES cells were obtained that gave rise to chimeric mice capable of germ-line transmission of the deleted amyloid precursor protein allele.
