Abstract: Boron neutron capture therapy (BNCT) is increasingly being used in the treatment of several aggressive cancers, including cerebral glioblastoma multiforme. The main requirement for this therapy is selective targeting of tumor cells by sufficient quantities of 10B atoms required for their capture/irradiation with low-energy thermal neutrons. The low content of boron targeting species in glioblastoma multiforme accounts for the difficulty in selective targeting of this very malignant cerebral tumor by this radiation modality. In the present study, we have used for the first time boron nitride nanotubes as carriers of boron atoms to overcome this problem and enhance the selective targeting and ablative efficacy of BNCT for these tumors. Following their dispersion in aqueous solution by noncovalent coating with biocompatible poly-l-lysine solutions, boron nitride nanotubes were functionalized with a fluorescent probe (quantum dots) to enable their tracking and with folic acid as selective tumor targeting ligand. Initial in vitro studies have confirmed substantive and selective uptake of these nanovectors by glioblastoma multiforme cells, an observation which confirms their potential clinical application for BNCT therapy for these malignant cerebral tumors.

Investigations over half a century have indicated that mechanical forces induce neurite growth, with neurites elongating at a rate of 0.1–0.3 μm h−1pN−1when mechanical force exceeds a threshold, with this being identified as 400–1000 pN for neurites of PC12 cells. In this article, we demonstrate that neurite elongation of PC12 cells proceeds at the same previously identified rate on application of mechanical tension of ∼1 pN, which is significantly lower than the force generated in vivo by axons and growth cones. This observation raises the possibility that mechanical tension may act as an endogenous signal used by neurons for promoting neurite elongation.

Background: Diabetic retinopathy (DR) is a major complication of diabetes, characterized by extensive vascular pathology leading to vision loss. Neuronal suffering and death are also present in the diabetic retina as a result of different molecular mechanisms that are compromised or modified in response to high glucose. The aim of this paper is to highlight recent data indicating that neurodegeneration is likely to play a primary role in the development of DR and that strategies based on nanomedicine may be exploited to deliver neuroprotection to the retina. Methods: An extensive analysis of the publications dealing with the role of neuroprotection in DR and with nanoparticle-mediated drug delivery to the retina has been conducted using PubMed, with particular attention to the most recent papers. Results: There are important limitations related to possible systemic side effects of neuroprotective substances and to drug bioavailability in the retina such as, for instance, the amount of drug reaching the retina, the need of keeping to a minimum the number of administrations (especially, for example, in the case of intraocular injections) and the need of assuring a long-lasting, graded intraocular drug delivery. In recent years, a variety of investigations have been aimed at the exploitation of approaches of nanomedicine to enhance the pharmacokinetics and pharmacodynamic activity of intraocularly delivered drugs. In particular, we provide some preliminary results that we have obtained about the feasibility of delivering magnetic nanoparticles functionalized with a neuroprotectant to mouse eyes through intraocular injections. Conclusion: We propose that nanoparticles functionalized with neuroprotective substances may be used to protect the diabetic retina, thus causing an impact in the design of future pharmacologic treatments for DR.

New strategies to promote neuronal regeneration should aim to increase the speed of axonal elongation. Biochemical signaling is a key factor in axon growth, but recent discoveries have shown that mechanical force, through a process referred to as stretch growth, can significantly influence the elongation rate. Here, we develop a method to apply forces to primary hippocampal neurons from mice using magnetic microposts that actuate in response to an external magnetic field. Neurons are cultured onto these microposts and subjected to an average displacement of 0.2 μm at a frequency of 5 Hz. We find that the mechanical stimulation promotes an increase in the length of the axons compared to control conditions. In addition, there is an increase in the density of microtubules and in the amount of cisternae of the endoplasmic reticulum, providing evidence that stretch growth is accompanied by a mass addition to the neurite. Together, these results indicate that magnetically actuated microposts can accelerate the rate of axon growth, paving the way for future applications in neuronal regeneration.

Mechanical force is crucial in guiding axon outgrowth before and after synapse formation. This process is referred to as “stretch growth.” However, how neurons transduce mechanical input into signaling pathways remains poorly understood. Another open question is how stretch growth is coupled in time with the intercalated addition of new mass along the entire axon. Here, we demonstrate that active mechanical force generated by magnetic nano-pulling induces remodeling of the axonal cytoskeleton. Specifically, the increase in the axonal density of microtubules induced by nano-pulling leads to an accumulation of organelles and signaling vesicles, which, in turn, promotes local translation by increasing the probability of assembly of the “translation factories.” Modulation of axonal transport and local translation sustains enhanced axon outgrowth and synapse maturation.

In this paper we used Multi Wall Carbon Nanotubes (MWCNTs) containing 3 of residuals and impurities of Fe, Al, and Zn. MWCNTs, by virtue of Fe at their tips, are able to respond to the effects of external magnetic fields. We demonstrated that MWCNTs interact with PC12 cells without compromising their viability and differentiation with outgrowth of neurites. We also document that when exposed to a magnetic field, both undifferentiated and differentiated PC12 cells cultured in CNT-containing medium solution are able to move towards the magnetic source.

Carbon nanotubes are widely used for biomedical applications as intracellular transporters of biomolecules, due to their high propensity to cross cell membranes. In this review we summarize the recent advancement in the functionalization of carbon nanotubes, focusing particular attention on the chemistry involved in the preparation of nanovectors for drug delivery systems, and on the physical and chemical properties that affect cellular up-take and biodistribution phenomena.

Whereas several biomedical applications of carbon nanotubes have been proposed, the use of boron nitride nanotubes (BNNTs) in this field has been largely unexplored despite their unique and potentially useful properties. Our group has recently initiated an experimental program aimed at the exploration of the interactions between BNNTs and living cells. In the present paper, we report on the magnetic properties of BNNTs containing Fe catalysts which confirm the feasibility for their use as nanovectors for targeted drug delivery. The magnetisation curves of BNNTs characterised by the present study are typical of superparamagnetic materials with important parameters, including magnetic permeability and magnetic momentum, derived by employing Langevin theory. In-vitro tests have demonstrated the feasibility for influencing the uptake of BNNTs by living cells by exposure to an external magnetic source. A finite element method analysis devised to predict this effect produced predictive data with close agreement with the experimental observations.

The aim of this paper is to develop highly magnetized, biodegradable and biocompatible, polymeric nanoparticles for drug delivery in cell therapy. Alginate magnetic nanoparticles are realized by an emulsion/reticulation technique, after the dispersion of magnetite in an alginate solution. Such nanoparticles are characterized in terms of external morphology (FIB imaging), microstructure (TEM imaging), size distribution, zeta potential, magnetic properties (SQUID analysis) and drug release behaviour. Magnetization curves show the typical trend of superparamagnetic materials. Important parameters, such as magnetic permeability and magnetic momentum, are derived by employing Langevin theory. Experimental results reveal that a bi-exponential model fully describes the drug release. Finally, in vitro experiments on NIH/3T3 cells are carried out and demonstrate that our magnetic alginate nanoparticles can effectively drive the drug delivery towards an external magnetic field source.
