Abstract: Considering the high degree of sequence conservation within the Rosaceae family and, in particular, among the Prunus species, we employed the first available peach oligonucleotide microarray (μPEACH 1.0) for studying the transcriptomic profile during apricot (cv. ‘Goldrich’) fruit development. Apricot fruits were harvested at three distinct developmental stages, corresponding to immature green (6 weeks before fully ripe stage), mature firm ripe (change of peel colour, 1 week before fully ripe stage) and fully ripe, namely T1, T2 and T3, respectively. When applied to μPEACH1.0, apricot target cDNAs showed significant hybridization with an average of 43% of spotted probes, validating the use of μPEACH1.0 to profile the transcriptome of apricot fruit. Microarray analyses carried out separately on peach (cv. ‘Fantasia’) and apricot fruit to profile transcriptome changes during fruit development showed that 70% of genes had the same expression pattern in both species. Such data indicate that the transcriptome is quite similar in apricot and peach fruit and also highlight the presence of species-specific transcript changes. In apricot, 400 and 74 differentially expressed genes were found during the transition from T1 to T2 and from T2 to T3, respectively. Among these, a considerable number of genes encoding IAA protein in action regulators (Aux/IAA) and heat shock proteins (HSPs) were highly up-regulated at early and late ripening, respectively. Intriguingly, the expression profiles of all considered HSPs and some of IAA protein /IAA genes showed different patterns between apricot and peach during the last stages of on-tree fruit development, suggesting the presence of diverse mechanism regulating ripening in these two close phylogenetically related species.

Mulberries are members of the genus Morus L., a taxonomic group showing a great genetic variability and adaptability to different environmental conditions. This study deals with the use of AFLP-based fingerprints as a tool for estimating genetic variability within as well as among three different mulberry species (i.e., M. alba L., M. latifolia Poir. and M. bombycis Koidz.). A high level of polymorphism (72.2) was found over all the 48 accessions analyzed. Genetic similarity (GS) within single Morus species ranged from 0.845 (M. bombycis) to 0.884 (M. alba) being intermediate in M. latifolia (0.869). The between-species mean genetic similarity estimates based on pair-wise AFLP marker fingerprint comparison were very similar ranging from 0.861 to 0.874. The partition of the genetic variation over the three Morus species was unexpected a proportion of the among-species genetic diversity as low as GST= 0.084 pointed out that about 92% of the total genetic diversity found among Morus accessions is due to DNA polymorphisms within a species, while only 8% of the total variation was highlighted among species. Our data indicate that some of the introduced accessions showing distinctive phenotypes, clearly differentiated from those revealed in the original habitat where they have been selected and adapted, hide an identical genotype.

The physiological drop of immature fruits was studied in relation to the activation of the abscission zone located between the fruit and the receptacle. Light- and electron-microscopy observations demonstrated that this zone consisted of two types of parenchymatous cells: in the distal region, closer to the fruit, were groups of small thick-walled cells with few intercellular spaces; in the proximal region, closer to the pedicel, the stillgrouped cells were larger, polyphenolic-rich, and thick-walled but with many wide intercellular spaces. Separation of the fruit occurred by dissolution of the middle lamella of the cells of this zone followed by an increase in the size of the intercellular spaces. Lysis of the middle lamella began at the corners of the cells and spread from there across the entire wall surface. Structural changes were paralleled by an increase in soluble proteins, endo-cellulase and exo-polygalacturonase activity. Isoelectric focusing indicated that both enzymes were present as isoenzymes whose patterns were affected by embryoctomy and 2-chloroethylphosphonic acid treatments.
