1. Husk Separation (Kubessa Method) Impacts the Aging Chemistry of Beer

Abstract: The removal of husks before the mashing process, also known as the Kubessa method, is an established brewing practice often positively associated with smoothness and better flavor-stability of beer. Empirical evidence on the effect of the Kubessa method on beer, however, has been lacking. Similarly, our study’s comprehensive analysis of established brewing attributes revealed that traditional methods do not fully capture the impact of husk separation in beer brewing. Conclusive evidence of the Kubessa method’s impact on beer aging chemistry was obtained through ultrahigh resolution mass spectrometry (FT-ICR-MS), revealing intricate molecular details inaccessible to conventional analytical techniques. The compositional information on thousands of molecules in Kubessa beer was resolved and compared to whole malt mashing. Machine learning algorithms applied to aging experiments identified over 500 aging-related compounds inhibited by husk separation. Complementary Time of flight mass spectrometry (ToF-MS) coupled with chromatography further confirmed that the mashing of husks introduces sulfur-containing lipid compounds. These significant differences in the beer composition provide valuable insights for further investigation into the staling protective effect of husk-separation (Kubessa process) during beer production, as empirically demonstrated in this work.

2. Rendezvous mit einer autonom navigierenden Paketstation—Ein innovatives Geschäftsmodell für die letzte Meile Logistik mit Gaia-X

Abstract: Autonome Liefer- und Zustellroboter durchdringen zunehmend das Pakettransportgeschäft, um die letzte Meile Logistik (LML) effizienter und nachhaltiger zu gestalten. Dabei entstehen neue Formen der Mensch-Maschine-Interaktion, die neben den operativen Vorteilen auch das Kundenerlebnis verbessern können. Gleichzeitig wird der Aufbau dezentraler Datenräume mit Gaia‑X in Europa für einen souveränen Datenaustausch und fairen Datenwettbewerb im Mobilitätssektor vorangetrieben. Trotz dieser vielversprechenden Entwicklungen und Möglichkeiten im Mobilitätsbereich werden transferorientierte Geschäftsmodelle bisher unzureichend für die LML thematisiert. Der vorliegende Beitrag widmet sich dem Konsortialforschungsprojekt Gaia-X 4 ROMS und stellt ein innovatives Geschäftsmodell für neuartige autonom navigierende Paketstationen (ANP) auf Basis eines interaktiven Rendezvous-Prinzips vor. Mittels Design Thinking werden die zentralen Bestandteile für ein ANP-spezifisches Geschäftsmodell identifiziert und in einem Business Model Canvas für Flottenbetreiber des Paket- und Expresslogistikmarktes exemplarisch entworfen. Das gestaltete ANP-Geschäftsmodell wird abschließend in Handlungsempfehlungen für LML-Stakeholder überführt, die marktfähige ANP-Innovationen fokussieren und gleichzeitig eine hohe Datensouveränität anstreben. Gaia-X-Kompetenzen, Technologieoffenheit, Vertrauen und Zusammenarbeit sowie Schnelligkeit und Kontinuität stellen entscheidende Aspekte dar, damit die Nutzung zukünftiger föderierter Dateninfrastrukturen und der gezielte ANP-Einsatz im Bereich der LML erfolgreich gelingen können.

3. Kosten der diabetischen Retinopathie und Nephropathie in Deutschland

Abstract: Zusammenfassung: Im Rahmen einer Literaturübersicht wurden die Kosten der diabetischen Retinopathie und diabetischen Nephropathie dargestellt und bewertet. Es wurde eine systematische Literaturrecherche durchgeführt, die insgesamt 441 Publikationen identifizierte, von denen 26 relevante Daten zu den Kosten der genannten Komplikationen enthielten. Der Fokus lag auf der Betrachtung der Kosten für Deutschland, jedoch wurden auch Publikationen anderer westeuropäischer und nordamerikanischer Staaten hinzugezogen. Insgesamt lagen für Deutschland 6 Publikationen vor, von denen 4 Angaben zu den genannten Komplikationen enthielten. Eine Publikation beschäftigte sich ausschließlich mit den Kosten der diabetischen Retinopathie, eine weitere Studie gab die Kosten mikro- und makrovaskulärer Komplikationen an, ohne jedoch diese näher aufzuschlüsseln. Zwei Publikationen für Deutschland zeigten die Kosten einzelner Komplikationen im Vergleich zu den Gesamtkosten des Diabetes mellitus auf. Die diabetische Retinopathie verursachte in Deutschland jährliche Kosten von 745 EUR bis 4347 EUR pro Patient. Für die Behandlung der diabetischen Nephropathie wurden Summen von 4426 EUR und 7548 EUR pro Patient angegeben. Die Behandlungskosten pro Tag betrugen für die diabetische Retinopathie 6,51 EUR und 6,41 EUR für die diabetische Nephropathie.

Abstract: Sparsentan, a novel, non-immunosuppressive, single-molecule, dual endothelin angiotensin receptor antagonist, significantly reduced proteinuria versus irbesartan, an angiotensin II receptor blocker, at 36 weeks (primary endpoint) in patients with immunoglobulin A nephropathy in the phase 3 PROTECT trial's previously reported interim analysis. Here, we report kidney function and outcomes over 110 weeks from the double-blind final analysis.

7. Effect of an Intranasal Corticosteroid on Quality of Life and Local Microbiome in Young Children With Chronic Rhinosinusitis: A Randomized Clinical Trial

Abstract: IMPORTANCE: Intranasal corticosteroids (INCs) remain the first-line treatment of chronic rhinosinusitis (CRS) in both adults and children, despite the lack of evidence regarding their efficacy in the pediatric population. Similarly, their effect on the sinonasal microbiome has not been well documented. OBJECTIVE: To assess the clinical, immunological, and microbiological effects of 12 weeks of an INC in young children with CRS. DESIGN, SETTING, AND PARTICIPANTS: This open-label randomized clinical trial was performed in a pediatric allergy outpatient clinic in 2017 and 2018. Children aged 4 to 8 years with CRS diagnosed by a specialist were included. Data were analyzed from January 2022 to June 2022. INTERVENTIONS: Patients were randomized to receive intranasal mometasone in an atomizer for 12 weeks (1 application per nostril, once per day) and supplemental 3-mL sodium chloride (NaCl), 0.9%, solution in a nasal nebulizer once a day for 12 weeks (INC group) or 3-mL NaCl, 0.9%, solution in a nasal nebulizer once a day for 12 weeks (control group). MAIN OUTCOMES AND MEASURES: Measures taken both before and after treatment included the Sinus and Nasal Quality of Life Survey (SN-5), a nasopharynx swab for microbiome analysis by next-generation sequencing methods, and nasal mucosa sampling for occurrence of innate lymphoid cells (ILCs). RESULTS: Of the 66 children enrolled, 63 completed the study. The mean (SD) age of the cohort was 6.1 (1.3) years; 38 participants (60.3%) were male and 25 (39.7%) were female. The clinical improvement reflected by reduction in SN-5 score was significantly higher in the INC group compared with the control group (INC group score before and after treatment, 3.6 and 3.1, respectively; control group score before and after treatment, 3.4 and 3.8, respectively; mean between-group difference, −0.58; 95% CI, −1.31 to −0.19; P = .009). The INC group had a greater increase in nasopharyngeal microbiome richness and larger decrease in nasal ILC3 abundance compared with the control group. A significant interaction was observed between change in microbiome richness and the INC intervention on the prediction of significant clinical improvement (odds ratio, 1.09; 95% CI, 1.01-1.19; P = .03). CONCLUSIONS AND RELEVANCE: This randomized clinical trial demonstrated that treatment with an INC improved the quality of life of children with CRS and had a significant effect on increasing sinonasal biodiversity. Although further investigation is needed of the long-term efficacy and safety of INCs, these data may reinforce the recommendation of using INCs as a first-line treatment of CRS in children. Abstract: The therapeutic potential of a novel, targeted-release formulation of oral budesonide (Nefecon) for the treatment of IgA nephropathy (IgAN) was first demonstrated by the phase 2b NEFIGAN trial. To verify these findings, the phase 3 NefigArd trial tested the efficacy and safety of nine months of treatment with Nefecon (16 mg/d) versus placebo in adult patients with primary IgAN at risk of progressing to kidney failure (ClinicalTrials.gov: NCT03643965). NefIgArd was a multicenter, randomized, double-blind, placebo-controlled two-part trial. In Part A, 199 patients with IgAN were treated with Nefecon or placebo for nine months and observed for an additional three months. The primary endpoint for Part A was 24-hour urine protein-to-creatinine ratio (UPCR) after nine months. Secondary efficacy outcomes evaluated included estimated glomerular filtration rate (eGFR) at nine and 12 months and the UPCR at 12 months. At nine months, UPCR was 27% lower in the Nefecon group compared with placebo, along with a benefit in eGFR preservation corresponding to a 3.87 ml/min/1.73 m2difference versus placebo (both significant). Nefecon was well-tolerated, and treatment-emergent adverse events were mostly mild to moderate in severity and reversible. Part B is ongoing and will be reported on later. Thus, NefIgArd is the first phase 3 IgA nephropathy trial to show clinically important improvements in UPCR and eGFR and confirms the findings from the phase 2b NEFIGAN study.

9. Real-Time Monitoring of Miniaturized Thermal Food Processing by Advanced Mass Spectrometric Techniques

Abstract: Mass spectrometry is a popular and powerful analytical tool to study the effects of food processing. Industrial sampling, real-life sampling, or challenging academic research on process-related volatile and aerosol research often demand flexible, time-sensitive data acquisition by state-of-the-art mass analyzers. Here, we show a laboratory-scaled, miniaturized, and highly controllable setup for the online monitoring of aerosols and volatiles from thermal food processing based on dielectric barrier discharge ionization (DBDI) mass spectrometry (MS). We demonstrate the opportunities offered by the setup from a foodomics perspective to study emissions from the thermal processing of wheat bread rolls at 210 °C by Fourier transformation ion cyclotron resonance MS. As DBDI is an emerging technology, we compared its ionization selectivity to established atmospheric pressure ionization tools: we found DBDI preferably ionizes saturated, nitrogenous compounds. We likewise identified a sustainable overlap in the selectivity of detected analytes with APCI and electrospray ionization (ESI). Further, we dynamically recorded chemical fingerprints throughout the thermal process. Unsupervised classification of temporal response patterns was used to describe the dynamic nature of the reaction system. Compared to established tools for real-time MS, our setup permits one to monitor chemical changes during thermal food processing at ultrahigh resolution, establishing an advanced perspective for real-time mass spectrometric analysis of food processing.

10. Differentiation of activity rhythms and space use between two competing water shrew species in a laboratory experiment

Abstract: Hypotheses about the differentiation of activity rhythms and space use between two competing species of water shrews, Neomys anomalus[AA] and N. fodiens[FF], were verified in a laboratory experiment. The animals were kept together for 2 months in an enclosure (195 × 145 cm) and their activity was video recorded using the time-lapse mode. Changes in activity rhythms, distances kept between active animals, and frequency of interspecific conflicts were compared between the early (first 24 h) and late (last 24 h, after 2 months) phases of the animals’ coexistence in the mixed group. In both phases, the activity of both species was relatively evenly distributed over 24 h, with breaks in activity nearly every 1 h. During the early phase, both species were equally and simultaneously active, whereas in the late phase, as predicted, the activity of FF was lower than that of AA, and asynchronous. In the late phase, pairs of different species (F-A) were rarely active simultaneously and kept long inter-individual distances. Consequently, the number of conflicts decreased. The results suggest that, over time, water shrews can develop mechanisms diminishing interference competition, which are based on shifts in activity rhythms, maintaining distance between individuals, and the rare usage of the same shelters and feeding sites.

Abstract: Resistant hypertension is associated with increased cardiovascular risk. The endothelin pathway has been implicated in the pathogenesis of hypertension, but it is currently not targeted therapeutically, thereby leaving this relevant pathophysiological pathway unopposed with currently available drugs. Abstract: Background: Recent data revealed differences in human absorption kinetics and metabolism between food folates and folic acid supplements and fortificant. Objective: The objective was to determine folate bioavailability after ingestion of' breads or a breakfast meal fortified with either 5-CH3-H4 folate or folic acid by using a stable-isotope area under the curve (AUC) and ileostomy model. Design: In a randomized crossover trial, healthy ileostomists (n = 8) ingested single doses of whole-meal bread that contained [approximately equal to] 450 nmol (200 [micro]g) of either (6S)-[[sup.13][C.sub.5]5-C[H.sub.3]-[H.sub.4] folate or [[sup.13][C.sub.5]]folic acid or a breakfast meal that contained [approximately equal to]450 nmol (200 [micro]g) [[sup.13][C.sub.5]]folic acid. We collected blood from the subjects during 12 h postdose for assessment of plasma kinetics. Nonabsorbed folate was assessed from labeled folate contents in stomal effluent 12 and 24 h postdose. Results: The median (range) plasma [AUC.sub.o[right arrow]12] (AUC from 0 to 12 h after ingested dose) of 66 nmol * h/L (34-84 nmol. h/L) after ingestion of bread that contained (6S)-[[sup.13][C.sub.5]]5-C[H.sub.3]-[H.sub.4] folate was significantly greater (P < 0.001) than that after ingestion of [[sup.13][C.sub.5]] folic acid in fortified bread [28 nmol - h/L (15-38 nmol * h/L)] and a fortified breakfast meal [26 nmol * h/L (15-60 nmol * h/L)]. 15. Rhythmic gymnasts’ injuries in a pediatric sports medicine clinic in the United States: a 10-year retrospective chart review

Abstract: ABSTRACTObjectivesRhythmic gymnastics injuries have not been studied thoroughly especially in the United States. Existing research studies are predominantly from Europe or Canada or from more than 15 years ago. The purpose of our study was to provide an updated description of injury patterns among rhythmic gymnasts in the United States.MethodsA retrospective chart review was conducted of 193 rhythmic gymnastics injuries in 79 females, ages 6–20. Patients were seen between January 2010 and March 2020 in a hospital-based pediatric sports medicine clinic. Gymnast demographics, injury locations, and injury types were collected as available. Descriptive and bivariate statistical analysis was performed using general linear mixed models.ResultsOur cohort had a mean age of 14.61 2.61 years. Overuse injuries (76.7%) were more common than acute injuries (23.3%). The most common injury types were strain (20.7%), nonspecific pain (15.5%), and tendinitis/tenosynovitis (10.36%). The most frequently injured body regions were lower extremity (75.1%), followed by trunk/back (19.2%), upper extremity (4.7%), and head/neck (1.0%). The most common injured body parts were foot (24.9%), ankle (15.5%), knee (15.0%), lower back (14.0%), and hip (13.0%). General linear mixed models revealed that older age (p = 0.001) and higher competitive level (p = 0.016) were associated with a greater number of diagnoses. Gymnasts with foot injuries were older than gymnasts with ankle (p = 0.026), hip (p < 0.0001), and knee (p = 0.002) injuries. Gymnasts with higher BMI-for-age percentile were more likely to have acute injuries than overuse (p = 0.035).ConclusionOur data showed that injuries among rhythmic gymnasts were most frequently located in the lower extremities, specifically the foot, followed by trunk/back. Additionally, the most frequent injury types were strains and nonspecific pain, and overuse was the most prevalent mechanism. Gymnasts with foot injuries were older than gymnasts with ankle, hip, and knee injuries. Higher BMI is a predictor of acute injuries.

Abstract: The risk of diabetic nephropathy is partially genetically determined. Diabetic nephropathy is linked to a gene locus on chromosome 18q22.3-q23. We aimed to identify the causative gene on chromosome 18 and to study the mechanism by which the product of this gene could be involved in the development of diabetic nephropathy. DNA polymorphisms were determined in 135 case (diabetic nephropathy) and 107 control (diabetes without nephropathy) subjects. The effect of carnosine on the production of extracellular matrix components and transforming growth factor-β (TGF-β) after exposure to 5 and 25 mmol/l D-glucose was studied in cultured human podocytes and mesangial cells, respectively. A trinucleotide repeat in exon 2 of the CNDP1 gene, coding for a leucine repeat in the leader peptide of the carnosinase-1 precursor, was associated with nephropathy. The shortest allelic form (CNDP1 Mannheim) was more common in the absence of nephropathy (P = 0.0028, odds ratio 2.56 [95% CI 1.36-4.84]) and was associated with lower serum carnosinase levels. Carnosine inhibited the increased production of fibronectin and collagen type VI in podocytes and the increased production of TGF-β in mesangial cells induced by 25 mmol/l glucose. Diabetic patients with the CNDP1 Mannheim variant are less susceptible for nephropathy. Abstract: Introduction. Bullous pemphigoid (BP) is one of the most common bullous diseases with an autoimmune background. The etiology and pathogenesis of BP are believed to be influenced not only by environmental, genetic, and immunological factors as well as by oxidative stress. BP is observed more frequently in elderly patients. Additionally, more potent oxidative stress is observed just in old age. Glutathione S-transferases (GSTs) play key roles in the detoxification of xenobiotics, metabolism of endogenous substrates, and the defense against oxidative stress. The present study examines whether polymorphism of genes encoding three selected GSTs (GSTM1, GSTT1, and GSTP1) might be associated with a higher risk for BP.

Abstract: The study concerns a comprehensive analysis of a multistage hot-die forging on hammers, in order to produce a yoke-type forging, used as a component of excavator power transmission systems. The investigations were conducted with the aim to analyze and identify the sensitive areas in the process and then improve the currently implemented forging technology by using finite element (FE) simulation. QuantorForm (the developer of the QForm program) has developed a thermomechanical numerical model for the production of forked forging. The software Computer-Aided Three-Dimensional Interactive Application (CATIA) was used to develop and build Computer-Aided Design (CAD) models of forging tools. As a result of the numerical simulations, the plastic deformations and temperature distributions for the forgings and tools were obtained, and the force courses during the forging process were analyzed. The obtained results enabled a thorough analysis of the forging process, including identification of potential forging defects (laps) as well as those tool areas that are the most loaded and exposed to damage. On this basis, changes were implemented in the production process, which allowed for the improvement of the currently implemented technology and obtaining the corrected forgings.

Abstract: Ispinesib is a potent inhibitor of kinesin spindle protein (KSP), which has been identified as a promising target for antimitotic anticancer drugs. Herein, we report the synthesis of half-sandwich complexes of Ru, Os, Rh, and Ir bearing the ispinesib-derived N,N-bidentate ligands (R)- and (S)-2-(1-amino-2-methylpropyl)-3-benzyl-7-chloroquinazolin-4(3H)-one and studies on their chemical and biological properties. Using the enantiomerically pure (R)- and (S)-forms of the ligand, depending on the organometallic moiety, either the SM,Ror RM,Sdiastereomers, respectively, were observed in the molecular structures of the Ru- and Os(cym) (cym = η6-p-cymene) compounds, whereas the RM,Ror SM,Sdiastereomers were found for the Rh- and Ir(Cp*) (Cp* = η5-pentamethylcyclopentadienyl) derivatives. However, density functional theory (DFT) calculations suggest that the energy difference between the diastereomers is very small, and therefore a mixture of both will be present in solution. The organometallics exhibited varying antiproliferative activity in a series of human cancer cell lines, with the complexes featuring the (R)-enantiomer of the ligand being more potent than the (S)-configured counterparts. Notably, the Rh and Ir complexes demonstrated high KSP inhibitory activity, even at 1 nM concentration, which was independent of the chirality of the ligand, whereas the Ru and especially the Os derivatives were much less active.

29. Impact of wound duration on diabetic foot ulcer healing: evaluation of a new sucrose octasulfate wound dressing

Abstract: Objective:A common and frequent complication of diabetes is diabetic foot ulcers (DFU), which can have high treatment costs and severe adverse events. This study aims to evaluate the effects of wound duration on wound healing and the impact on costs, including treatment with a new sucrose octasulfate dressing compared with a control dressing.Method:Based on the Explorer study (a two-armed randomised double-blind clinical trial), a cost-effectiveness analysis compared four different patient groups distinguished by their wound duration and additionally two DFU treatment options: a sucrose octasulfate dressing and a neutral dressing (as control). Clinical outcomes and total direct costs of wound dressings were evaluated over 20 weeks from the perspective of the Social Health Insurance in Germany. Simulation of long-term outcomes and costs were demonstrated by a five cycle Markov model.Results:The results show total wound healing rates between 71% and 14.8%, and direct treatment costs for DFU in the range of €2482–3278 (sucrose octasulfate dressing) and €2768–3194 (control dressing). Patients with a wound duration of ≤2 months revealed the highest wound healing rates for both the sucrose octasulfate dressing and control dressing (71% and 41%, respectively) and had the lowest direct treatment costs of €2482 and €2768, respectively. The 100-week Markov model amplified the results. Patients with ≤2 months' wound duration achieved wound healing rates of 98% and 88%, respectively and costs of €3450 and €6054, respectively (CE=€3520, €6864). Sensitivity analysis revealed that the dressing changes per week were the most significant uncertainty factor.Conclusion:Based on the findings of this study, early treatment of DFU with a sucrose octasulfate dressing is recommended from a health economic view due to lower treatment costs, greater cost-effectiveness and higher wound healing rates.

Abstract: Les recommandations de l’European Society of Gynaecologic Oncology (ESGO) couvrent désormais la totalité des situations cliniques courantes en cancérologie gynécologique. Elles sont élaborées selon une méthodologie stricte comportant une revue systématique de littérature, la constitution d’un groupe d’experts sur la base de l’activité scientifique, de la répartition géographique et de la multidisciplinarité, et la revue externe par des utilisateurs et des représentants de patientes. Les recommandations pour la prise en charge du cancer vulvaire ont été élaborées dans l’année 2015 publiées en 2017. Elles sont disponibles en accès libre sur le site internet de l’ESGO. Une application smartphone « ESGO guidelines » permet de les implémenter en pratique quotidienne. Cet article reprend les éléments de stratégie sur le diagnostic et la prise en charge de l’atteinte ganglionnaire dans les cancers vulvaires. Une revue complémentaire de bibliographie depuis 2015 a été réalisée pour la rédaction de cet article afin de compléter la documentation. La stratégie ganglionnaire dans les cancers vulvaires comporte un volet diagnostique, où la clinique et l’imagerie jouent encore un rôle essentiel, mais où la méthode du ganglion sentinelle a permis une avancée majeure dans les cancers précoces. Elle comporte un volet thérapeutique, où le curage ganglionnaire et la radio(chimio)thérapie ont une place codifiée.

Abstract: Acute-on-chronic liver failure (ACLF) is well-studied in adults and characterized by decompensated cirrhosis, multi-organ failure, and early mortality. Studies of ACLF in children are limited. We sought to characterize the prevalence and clinical factors associated with pediatric ACLF (PACLF). A retrospective review of children 3 months to 18 years listed for liver transplantation and hospitalized for decompensated cirrhosis between January 2007 and December 2017 at a single pediatric hospital. Primary outcome was the development of PACLF, characterized as failure of at least 1 extrahepatic organ (mechanical ventilation, renal replacement therapy, vasoactive medications, grade III/IV hepatic encephalopathy). Characteristics were recorded for each hospitalization. Sixty-six patients had 186 hospitalizations with mean age at admission 4.0?±?5.6 years and diagnosis of biliary atresia (BA) in 65%. In line, there is an increasing need to describe and comprehend the potential immunomodulatory effects of toxins that can be produced from fungi proliferating even in a domestic environment like, for instance, Alternaria alternata. Taking this as a starting point, we investigated the effects of one of the most potent genotoxic compounds produced by this fungi type, namely altertoxin II (ATXII) on THP-1 macrophages. In noncytotoxic concentrations (0.1–1 μM), ATXII inhibited the activation of the transcription factor NF-κB, and this event was accompanied by significant mitochondrial superoxide production (1 μM ATXII). Both responses seemed dependent on membrane structure and morphology since they were modulated by the coincubation with the cholesterol complexing agent methyl-β-cyclodextrin (MβCD, 10–50 μM). Moreover, toxicity of ATXII was enhanced by cholesterol load (cholesterol-MβCD). The mycotoxin induced also lipid peroxidation (1–10 μM, ATXII) possibly streaming down at the mitochondrial level and suppressing NF-κB activation in THP-1 macrophages.

35. Zinc-mediated Neurotransmission in Alzheimer's Disease: A Potential Role of the GPR39 in Dementia

Abstract: With more people reaching an advanced age in modern society, there is a growing need for strategies to slow down age-related neuropathology and loss of cognitive functions, which are a hallmark of Alzheimer's disease. Neuroprotective drugs and candidate drug compounds target one or more processes involved in the neurodegenerative cascade, such as excitotoxicity, oxidative stress, misfolded protein aggregation and/or ion dyshomeostasis. A growing body of research shows that a G-protein coupled zinc (Zn2+) receptor (GPR39) can modulate the abovementioned processes. Zn2+itself has a diverse activity profile at the synapse, and by binding to numerous receptors, it plays an important role in neurotransmission. However, Zn2+ is also necessary for the formation of toxic oligomeric forms of amyloid beta, which underlie the pathology of Alzheimer’s disease. Furthermore, the binding of Zn2+ by amyloid beta causes a disruption of zincergic signaling, and recent studies point to GPR39 and its intracellular targets being affected by amyloid pathology. In this review, we present neurobiological findings related to Zn2+ and GPR39, focusing on its signaling pathways, neural plasticity, interactions with other neurotransmission systems, as well as on the effects of pathophysiological changes observed in Alzheimer's disease on GPR39 function. Direct targeting of the GPR39 might be a promising strategy for the pharmacotherapy of zincergic dyshomeostasis observed in Alzheimer’s disease. The information presented in this article will hopefully fuel further research into the role of GPR39 in neurodegeneration and help in identifying novel therapeutic targets for dementia.

38. Contact with adult hens affects the composition of skin and respiratory tract microbiota in newly hatched chicks

Abstract: Chickens in commercial production are hatched in hatcheries without any contact with their parents and colonization of their skin and respiratory tract is therefore dependent on environmental sources only. However, since chickens evolved to be hatched in nests, in this study we evaluated the importance of contact between hens and chicks for the development of chicken skin and tracheal microbiota. Sequencing of PCR amplified V3/V4 variable regions of the 16S rRNA gene showed that contact with adult hens decreased the abundance of E. coli, Proteus mirabilisand Clostridium perfringensboth in skin and the trachea, and Acinetobacter johnsoniiand Cutibacterium acnesin skin microbiota only. These species were replaced by Lactobacillus gallinarum, Lactobacillus aviarius, Limosilactobacillus reuteri, and Streptococcus pasterianusin the skin and tracheal microbiota of contact chicks. Lactobacillican be therefore investigated for their probiotic effect in respiratory tract in the future. Skin and respiratory microbiota of contact chickens was also enriched for Phascolarctobacterium, Succinatimonas, Flavonifractor, Blautia, and [Ruminococcus] torquethough, since these are strict anaerobes from the intestinal tract, it is likely that only DNA from nonviable cells was detected for these taxa.

39. Molecular Characterization of Cooking Processes: A Metabolomics Decoding of Vaporous Emissions for Food Markers and Thermal Reaction Indicators

Abstract: Thermal processing of food plays a fundamental role in everyday life. Whereas most researchers study thermal processes directly in the matrix, molecular information in the form of non- and semivolatile compounds conveyed by vaporous emissions is often neglected. We performed a metabolomics study of processing emissions from 96 different food items to define the interaction between the processed matrix and released metabolites. Untargeted profiling of vapor samples revealed matrix-dependent molecular spaces that were characterized by Fourier-transform ion cyclotron resonance–mass spectrometry and ultra-performance liquid chromatography–mass spectrometry. Thermal degradation products of peptides and amino acids can be used for the differentiation of animal-based food from plant-based food, which generally is characterized by secondary plant metabolites or carbohydrates. Further, heat-sensitive processing indicators were characterized and discussed in the background of the Maillard reaction. These reveal that processing emissions contain a dense layer of information suitable for deep insights into food composition and control of cooking processes based on processing emissions.

41. Cost-effectiveness of TLC-sucrose octasulfate versus control dressings in the treatment of diabetic foot ulcers

Abstract: Objective:Diabetes is one of the most widespread diseases in Germany. Common complications are diabetic foot ulcers (DFU), which are associated with a cost-intensive treatment and serious adverse events, such as infections, amputations. This cost-effectiveness analysis compares two treatment options for patients with DFU: a TLC-NOSF dressing versus a neutral dressing, assessed through a European double-blind randomised controlled trial (RCT), Explorer.Methods:The evaluation of the clinical outcomes was associated to direct costs (costs for dressings, nursing time, hospitalisation etc.) of both dressings, from the perspective of the statutory health insurance in Germany. Due to the long mean healing time of a DFU, the observation period was extended from 20 to 100 weeks in a Markov model.Results:After 20 weeks, and with complete closure as a primary endpoint, the model revealed direct treatment costs for DFU of €2,864.21 when treated with a TLC-NOSF dressing compared with €2,958.69 with the neutral control dressing (cost-effectiveness: €6,017.25 versus €9,928.49). In the Markov model (100 weeks) the costs for the TLC-NOSF dressing were €5,882.87 compared with €8,449.39 with the neutral dressing (cost-effectiveness: €6,277.58 versus €10,375.56). The robustness of results was underlined by several sensitivity analyses for varying assumptions. The frequency of weekly dressing changes had the most significant influence in terms of parameter uncertainty.Conclusion:Overall, the treatment of DFU with a TLC-NOSF dressing is supported from a health economic perspective, because both the treatment costs and the cost-effectiveness were superior compared with the neutral wound dressing.

42. Donor site morbidity following radial forearm free flap reconstruction with split thickness skin grafts using negative pressure wound therapy

Abstract: Donor site complications secondary to radial forearm free flap (RFFF) reconstruction can limit recovery. Optimizing hand and wrist function in the post-operative period may allow more efficient self-care and return to activities of daily living. Negative pressure wound dressings (NPD) may increase blood flow and perfusion as compared to static pressure dressings (SPD) designed to minimize shear forces during the healing period. This study aims to compare subjective and objective hand and wrist functional outcomes following RFFF reconstruction with split thickness skin grafts (STSG) in patients treated with NPD and SPD. Adult patients undergoing RFFF with STSG were identified preoperatively and randomized to receive NPD or SPD following their RFFF reconstruction. NPD involved a single-use, portable device capable of applying 80 mmHg of negative pressure to the forearm donor site. SPD involved a volar splint. Dressings were left in place for seven days with subjective and objective function assessed at seven days, one month and three months postoperatively. The primary outcome was self-reported hand function as measured with the function subscale of the Michigan Hand Questionnaire (MHQ). Secondary outcomes included hand and wrist strength, range of motion, sensation, scar aesthetics, and skin graft complications. Twenty-four patients undergoing RFFF were randomized to NPD or SPD. Patients treated with NPD had improved MHQ self-reported functional scores as compared to those treated with SPD at seven days postoperatively (P= 0.016). Flexion at seven days was improved in NPD group (P= 0.031); however, all other strength and range of motion outcomes were similar between groups. There were no differences in rates of graft complications, scar aesthetics, or sensation. In the immediate post-operative period, NPD was associated with improved patient-reported hand and wrist function. Wound care to optimize hand and wrist function could allow for improved patient outcomes in the immediate postoperative period.

Abstract: A turn-on fluorescent probe, HOCD-RB, for monitoring singlet oxygen (1O2) was developed by linking rhodamine B as fluorophore with dimethylhomoocoerdianthrone (HOCD) as 1O2reaction site and fluorescence quencher due to the intramolecular energy transfer (ET) between rhodamine B and HOCD moieties. Upon exposure to 1O2it rapidly forms endoperoxide with HOCD and turns on the fluorescence of rhodamine B by 18-fold. Taking advantage of the HOCD-RB probe that shows fast response, high sensitivity, and selectivity for 1O2, it is applied for imaging of endogenous 1O2in living cells and the fluorometric assay for evaluating 1O2quenching activity of selected common flavonoids found in our daily diets. The results show that the 1O2scavenging activity of flavonoids depends on not only the structure of individual flavonoid but also the competitive interactions between mixed flavonoids. The best antioxidant capacity for individual and mixed flavonoids is epigallocatechin gallate and the mixture of catechin gallate with kaempferol, respectively. Overall, this work provided a new tool for detection and imaging of singlet oxygen activity in a biological system as well as an efficient fluorometric assay of 1O2scavenging activity.

46. Retrospective analysis of coronary interventions in a single centre and comparison of specific differences between radial and femoral access

Abstract: AbstractObjectives:The aim of this study was to analyse the consequence of radial or femoral access during coronary interventions to radiation dose, fluorography time and a number of pseudoaneurysms following each type of intervention.Background:According to the results of many recent studies (RIVAL, RIFLE-STEACS, STEMI RADIAL), current guidelines favour radial over femoral access during coronary intervention for safety, especially in cases of acute coronary syndromes. However, several papers have referred to problems in the design of these studies and the management of antithrombotic therapy. The influence of access site on radiation dose and fluorography time is also still unclear.Methods:We retrospectively analysed 4522 patients who underwent coronary angiography in 2012 and 2016 in a single centre in the Czech Republic. We compared the access site with the average radiation dose and mean effective dose that each patient received in Gy/cm2or mSv, respectively. We also compared average fluorography time in minutes and the incidence of pseudoaneurysms.Results:The radiation dose was a body mass index (BMI)-dependent parameter since each five points of body mass index increased radiation dose approximately by 23%. Use of femoral access resulted in lower fluorography time in all subgroups (p< .001) and decreased radiation dose in patients with coronary artery bypass grafts (CABGs) by 26% (p = .044). On the other hand, there has been 16 times lower frequency (p< .001) of post-catheterisation pseudoaneurysms after radial access than after femoral access.Conclusions:Both the radiation dose and fluorography time were lower after femoral access compared to radial access in patients with CABGs. On the other hand, radial access led to significantly fewer periprocedural pseudoaneurysms. Radial access therefore should be considered as the preferred access site during coronary intervention in patients with a high risk of pseudoaneurysm development, and femoral access should be considered for patients with a high risk of contrast-induced nephropathy.

Abstract: Short-term treatment for people with type 2 diabetes using a low dose of the selective endothelin A receptor antagonist atrasentan reduces albuminuria without causing significant sodium retention. We report the long-term effects of treatment with atrasentan on major renal outcomes.

48. Burden or Opportunity? Parent Experiences When Approached for Research in a Pediatric Intensive Care Unit

Abstract: Despite an ongoing need for pediatric research, low study enrollment may impede study completion, particularly in critical care. We conducted a prospective cross‐sectional survey and chart review study to assess parent experiences with research in a pediatric intensive care unit (PICU). Of the 80 parents who completed the study survey, 54% were approached to participate in a research study in the PICU, and 93% agreed to participate. Motivators included altruism, low burden, low risk, and research that would benefit the child. Barriers included risks to the child, the child's being too sick to participate, feeling overwhelmed, not having enough time to participate, the research's being burdensome, and the research's not being explained well. PICU parents had mostly favorable attitudes toward research participation. Compared with non‐Hispanic survey respondents, respondents of Hispanic ethnicity less often reported having been approached for research, which further evidences the need to track research recruitment processes to help avoid underinclusion of members of minority populations.

49. Acid strength measurements of Amberlyst 15 resin, p-xylene-2-sulfonic acid and chlorosulfonic and sulfuric acid treated SiO2, Al2O3, TiO2and MgO

Abstract: The measurements of the minimum acid strength (HOmin) required for the initiation of cis-stilbene at 303 K and tert-butylbenzene at 313 K, 323 K and 333 K have enabled the establishment of a much more precise acid strength scale based on test reactions. HOmin values obtained were: − 9.0 (cis-stilbene, 303 K), − 9.3, − 9.5 and − 9.9 (tert-butylbenzene, 313 K, 323 K, 333 K). The updated method was used for the determination of the acid strength of different catalysts obtained in the reaction of chlorosulfonic and sulfuric acid on different oxide carriers. It was found that the acid strength values depended on the basicity of the studied supports. For the supports treated with chlorosulfonic acid the values of acid strength were: − 11.5 < HO≤ − 10.8 in the case of the less basic SiO2and − 9.3 < HO≤ − 9.0 for the more basic TiO2, Al2O3and MgO. For the supports treated with sulfuric acid the acid strength was: -9.3 < HO≤ -9.0 for the less basic SiO2and TiO2, − 9.0 < HO≤ − 7.9 for the more basic Al2O3and − 7.9 < HO≤ − 6.9 for the most basic MgO. Silica treated with chlorosulfonic acid possesses higher acid strength than HZSM-5 and amorphous silicaalumina. The catalyst obtained in the modification of TiO2, Al2O3and MgO with chlorosulfonic acid were of lower acid strength comparable to Amberlyst 15 and HZSM-5 (− 9.3 < HO≤ − 9.0), but higher than p-xylene-2-sulfonic acid dihydrate (− 7.9 < HO≤ − 6.9).

50. The Competition Between Immatures of Ixodes ricinusand Dermacentor reticulatus(Ixodida: Ixodidae) Ticks for Rodent Hosts

Abstract: The numbers of Ixodes ricinus(L.) and Dermacentor reticulatus(Fabricius) larvae and nymphs attached to small mammals are influenced by interspecific competition. The present study analyses data collected over several years in two study areas: Kosewo Górne (Mazurian District, N Poland; between July 1997 and July 2009) and Białowieża Primeval Forest (E Poland; in July 2007). In total, 975 ticks were collected from striped field mice (Apodemus agrarius), yellow-necked mice (A. flavicollis), bank voles (Myodes glareolus), and root voles (Microtus oeconomus). In total, of the 203 investigated rodents, 137 were infested with ticks and 39 demonstrated mixed infection. The numbers of the two tick species found on Apodemusmice were significantly negatively correlated with those on root voles; similarly, although bank voles were significantly more frequently infested by I. ricinusthan by D. reticulatus, the reverse was observed in root voles. In addition, among the voles, each tick species was found in different locations on the host body, which could also result from competitive interactions; furthermore, competitive release regarding microhabitat selection was observed on hosts infested with one tick species. This competition may be driven by the limited area of host body available for foraging by ticks, i.e., safe areas of high vascularization covered by thin skin. However, the mechanisms of such competition require further investigation.
