ABSTRACTFocussed searches were made across New Zealand between 2013 and 2016, for endemic aphids from the Schizaphis(Rhopalosiphina) genus, which is currently represented by two putative, undescribed species from the endemic host plants Aciphyllaand Dracophyllum. Cytochrome c oxidase I (COI) gene sequences (48 in total) from the Schizaphiswere analysed together with those from a broader collection of New Zealand endemic aphids that has been assembled since the year 2000. The bulk of the Schizaphisbelonged to two clusters corresponding to the host plant genera. Two aphids from central North Island Dracophyllumrepresented a much diverged lineage without clear affiliations to other New Zealand Schizaphis. Inter-population variation in the New Zealand Schizaphiswas high compared with that seen in international studies of Aphidinae and among populations of other endemic New Zealand Aphidina. Within Schizaphisfrom Dracophyllum, geography played an apparent role in genetic structuring, with populations from Taranaki (North Island) and especially Mt Lyford (South Island) being divergent from those on the South Island main divide. Two distinct lineages of Schizaphis, which co-occurred at some sites, were found on Aciphylla. Our sequence comparisons, including GMYC analyses, indicated up to five New Zealand Schizaphislineages, and two newly discovered endemic Aphisspecies from the host plants Clematisand Hebe.

We present a mitochondrial gene tree for representative species of all the genera in the subfamily Myobatrachinae, with special emphasis on Crinia and Geocrinia. This group has been the subject of a number of long-standing taxonomic and phylogenetic debates. Our phylogeny is based on data from approximately 780 bp of 12S rRNA and 676 bp of ND2, and resolves a number of these problems. We confirm that the morphologically highly derived monotypic genera Metacrinia, Myobatrachus, and Arenophryne are closely related, and that Pseudophryne forms the sister group to these genera. Uperoleia and the recently described genus Spicospina are also part of this clade. Our data show that Assa and Geocrinia are reciprocally monophyletic and together they form a well-supported clade. Geocrinia is monophyletic and the phylogenetic relationships with the genus are fully resolved with two major species groups identified: G. leai, G. victoriana, and G. laevis; and G. rosea, G. alba, and G. vitellina (we were unable to sample G. lutea). We confirm that Taudactylus forms the sister group to the other myobatrachine genera, but our data are equivocal on the phylogenetic position of Paracrinia. The phylogenetic relationships among Crinia species are well resolved with strong support for a number of distinct monophyletic clades, but more data are required to resolve relationships among these major Crinia clades. Crinia tasmaniensis and Bryobatrachus nimbus form the sister clade to the rest of Crinia. Due to the lack of generic level synapomorphies for a Bryobatrachus that includes C. tasmaniensis, we synonymize Bryobatrachus with Crinia. Crinia georgiana does not form a clade distinct from other Crinia species and so our data do not support recognition of the genus Ranidella for other Crinia species. Crinia subinsignifera, C. pseudinsignifera, and C. insignifera are extremely closely related despite differences in male advertisement call. A preliminary investigation of phylogeographic substructure within C. signifera revealed significant divergence between samples from across the range of this species.

The rare Australian venomous elapid snake ‘Echiopsis’ atriceps has been the subject of considerable taxonomic instability with the five known specimens assigned to four genera by various authorities. Phylogenetic affinities of the rare Elapognathus minor also are poorly understood and have been the subject of some disagreement. To examine the phylogenetic affinities of these two rare taxa, a molecular data set comprising 1680 base pairs of mtDNA was assembled from a representative of each of the terrestrial Australian viviparous elapid genera and two species of Drysdalia, a genus about which there also has been phylogenetic controversy. A total of 936 base pairs of 12S rRNA, 454 base pairs of 16S rRNA and 290 base pairs of cytochrome b mtDNA were sequenced for 15 species. The Asian elapid Naja naja was used as the outgroup. These mtDNA regions provided 195, 38 and 72 parsimony informative sites, respectively, for a total of 315 parsimony informative characters. Unweighted phylogenetic analyses were performed under both parsimony and neighbour-joining criteria. Parsimony analyses of the unweighted, combined data set resulted in a single fully resolved most parsimonious tree 1225 steps long. The neighbour-joining tree differed by only a single weakly supported branch. These data strongly support a sister group relationship between ‘Echiopsis’ atriceps and the Australian broadheaded snakes of the genus Hoplocephalus with a bootstrap value of 99%. Templeton tests soundly reject all previous taxonomic arrangements for this species. Our data also strongly support a sister group relationship between Elapognathus minor and Drysdalia coronata with a bootstrap value of 98%. Importantly, Drysdalia coronata and Drysdalia coronoides do not form a monophyletic group, supporting some previous studies. Based on our results, we allocate ‘Echiopsis’ atriceps to a new monotypic genus and re-describe Elapognathus to include ‘Drysdalia’ coronata.

We provide details of seven microsatellite loci (out of 20 primer pairs designed) that exhibited the cleanest and strongest banding patterns for the Eastern tiger snake Notechis scutatus. These loci were used to screen 76 individuals from across the geographical range of N. scutatusand a further 14 individuals of the closely related and endangered broadheaded snake Hoplocephalus bungaroides. We observed large numbers of alleles per locus (14–52) and relatively high levels of heterozygosity (0.270–0.696) within the N. scutatussample. These markers are also likely to be informative for work on H. bungaroides.

A panel of six samples from three Eulamprus species was used to screen 24 primer pairs developed from the Australian water skink Eulamprus kosciuskoi. We provide details of 10 microsatellite loci that exhibited the cleanest and strongest banding patterns. We also screened six of these microsatellite loci against 118 individuals from a population of E. heatwolei. These microsatellites exhibited large numbers of alleles per locus (4–19) and high heterozygosity (0.271–0.898) in this population. This suggests that they will be extremely useful for investigating mating systems and other facets of population biology in these lizards.
