In 1936, Rusznyak and Szent-Gyorgyi first drew attention to the therapeutically beneficial role of dietary flavonoids, which are the most common group of polyphenols ubiquitously present in plant based food and beverages. Recent years have witnessed a renascence of interest on these nutraceuticals, which, because of their high potency and low systemic toxicity, are gradually emerging as promising alternatives to conventional therapeutic drugs. There is a mounting evidence that various proteins frequently serve as targets for therapeutically important flavonoids. In this article we present perspectives exemplifying the growing potential of fluorescence spectroscopy as an exquisitely sensitive tool for noninvasive sensing of protein-flavonoid interactions at physiologically relevant concentrations, via measurements of steady state emission parameters as well as decay kinetics studies of the intrinsic fluorescence of the target (protein) and/or ligand (flavonoid). Especially, we highlight novel applications of the remarkably environment sensitive ‘two color’ fluorescence exhibited by many important flavonoids, which permits multiparametric and ratiometric measurements. To consolidate findings obtained via fluorescence spectroscopy, use of other relevant experimental biophysical techniques and molecular modeling have proved to be valuable and are also discussed here. Such complementary studies provide additional insights regarding the thermodynamics and conformational aspects of the protein-flavonoid interactions, together with details, at atomistic level, of the dominant noncovalent interactions involved in the docking of different flavonoids to their target proteins.

Flavonols are plant pigments that are ubiquitous in nature. Quercetin (3,3',4',5,7-pentahydroxyflavone) and other related plant flavonols have come into recent prominence because of their usefulness as anticancer, antitumor, anti-AIDS, and other important therapeutic activities of significant potency and low systemic toxicity. Quercetin is intrinsically weakly fluorescent in aqueous solution, showing an emission maximum at ~538 nm. Upon binding to human serum albumin (HSA), quercetin undergoes dramatic enhancement in its fluorescence emission intensity, along with the appearance of dual emission behavior, consisting of normal and excited-state proton transfer (ESPT) fluorescence. In addition, the occurrence of a third emitting species has been noted for the first time. This is attributed to a electronic ground-state complex formed in the protein environment. High values of the fluorescence anisotropy (r) are obtained in the presence of HSA for the ESPT tautomer (r = 0.18), as well as the complex species (r = 0.37) of quercetin, indicating that the precursor ground-state molecules for both these emitting species of quercetin molecules are located in the motionally constrained sites of HSA. The steady-state emission data suggest that quercetin binds to two distinct sites in HSA from which the emissions from the normal tautomer and complex species take place. The preliminary results of studies on emission decay kinetics are also reported herein. Studies by far-UV circular dichroism spectroscopy reveal that binding of quercetin induces no significant perturbation in the secondary structure of HSA. © 2003 Wiley Periodicals, Inc. A number of recent studies have presented perspectives on the hydrophobic fluorescence probe tryptophan octyl ester (TOE). This molecule has attracted notable attention as a suitable model for the natural fluorophore tryptophan, in case of membrane proteins. We report here, for the first time, the fluorescence emission behaviour of TOE in reverse micelles of aerosol-OT (AOT) in n-heptane, containing different amounts of water. Relevant studies in representative homogeneous solvent media are also included for comparison. The fluorescence emission parameters (especially emission maximum, relative intensity, and anisotropy) of TOE are found to exhibit significant variation upon changes in the water/surfactant molar ratio (w0) of the reverse micelles. Fluorescence decay studies on TOE which we have also performed, indicate biexponential decay kinetics in reverse micelles as well as in homogeneous solvent media. The implications of these findings are examined in relation to the potentialities of TOE as a novel fluorescence probe for membrane proteins present in water restricted environments prevailing at the interfaces of biomembranes (for which reverse micelles serve as ideal model systems).

Photoinduced excited state intramolecular proton transfer (ESIPT) reactions comprise an important and extensively explored class of reactions in photochemistry. Till date, plant flavonols are one of the most widely known class of naturally abundant organic molecules exemplifying ESIPT and ‘two color’ fluorescence. From a bio-medical perspective, flavonols and related polyphenols, which are powerful antioxidants, have attracted significant interest as novel drugs (of high potency and low cyto-toxicity) for the prophylaxis and therapy of free radical induced and other important diseases. This article presents perspectives on proton transfer in photoexcited organic molecules from a historical context, emphasizing ESIPT reactions, in particular. Highlights of representative research findings are discussed, exemplifying the promising potential of plant flavonols as their own ESIPT based ‘fluorescence sensors’ for exploring their interactions with proteins, DNA (possessing duplex as well as higher order structures), and biomembranes, which represent the targets underlying the various pharmacological actions of flavonols. The usefulness of such approach for studying the confinement of intrinsically fluorescent flavonols in nano-vehicles for drug encapsulation, is also demonstrated.

A new technique is proposed for the estimation of critical micellar concentrations (cmcs) based on fluorescence polarization anisotropy (r) measurements as a function of surfactant concentration ([surf]). Representative results are presented for Triton X 100 (TX 100), sodium dodecyl sulphate (SDS) and cetyl trimethyl ammonium bromide (CTAB). The first and second cmc values (cmc1 and cmc2) obtained from break points in r vs. [surf] plots are in reasonable agreement with existing literature data.

The yellow-green luminescence of 3-hydroxyflavone and quercetin at room temperature in solution arises from a tautomer of the molecules produced by excited state proton-transfer across a barrier in the double-minimum hydrogen-bonding potential. At 77 K in 2-methylbutane rigid matrix, a normal UV-violet fluorescence is observed in correspondence with the UV absorption. Excitation spectra and deuteration effects confirm the mechanism.

The amino acid analogue 7-azatryptophan has attracted significant recent attention as a novel optical probe for protein structure, function and dynamics. We report here, for the first time, its fluorescence emission behavior in a membrane mimetic model system, namely reverse micelles of aerosol-OT in n-heptane, containing varying amounts of added H2O or D2O. Upon increase in the water/surfactant molar ratio from 0.5 to 50 the emission maximum undergoes a pronounced red shift (by 20 nm), which is accompanied by dramatic quenching of the fluorescence emission and sharp decrease in its average lifetime. These data are used to infer the microenvironments of the fluorophore in the reverse micelles. Furthermore, the highly sensitive dependence of the fluorescence emission parameters of 7-azatrytophan on the water content of the reverse micelles highlights its suitability as a probe for water restricted environments, with possible applications to interfacial regions of biomembranes.
