With the continuous evolution of self-powered technology, wearable devices utilizing Triboelectric Nanogenerators (TENG) have garnered extensive attention as a solution to the limitations associated with conventional sensors. In this study, we introduce a non-invasive, sensitive, self-powered triboelectric biosensor (TEBS) designed for simultaneous real-time monitoring of both biochemical and physical aspects of the body. The TEBS features a streamlined design, facilitating an efficient reaction interface between sweat metabolites and enzymes. Demonstrating impressive responsiveness, the TEBS registers glucose, uric acid, and lactate at rates of 101 %, 64.98 %, and 64.80 %, respectively, while also discerning various body movements such as walking, running, squatting, and jumping. To further heighten the reliability and sensitivity of the TENG, we employ Polyvinylidene Fluoride (PVDF) nanofilms modified with Metal-Organic Framework (MOF)-derived materials as the triboelectric layer. This modification proves effective in mitigating charge dissipation on the triboelectric layer's surface, establishing abundant micro capacitors within the nanofiber films. As a result, the TENG exhibits a remarkable six-fold increase in short-circuit current compared to its unmodified PVDF counterpart. Notably, the wearable device showcases the ability to independently power LEDs in external circuits, enabling timely health warnings without reliance on external power sources. The proposed TEBS offers a compelling glimpse into the future of healthcare.

Nerve infiltration into the tumor is a common feature of the tumor microenvironment. The mechanisms of axonogenesis in breast cancer remain unclear. We hypothesized that vascular endothelial growth factor (VEGF), as well as nerve growth factor (NGF), is involved in the axonogenesis of breast cancer. A N-methyl-N-nitrosourea (MNU)-induced rat model of breast cancer was used to explore the presence of axonogenesis in breast tumor and the involvement of VEGF, as well as NGF, in the axonogenesis of breast tumor. Nerve infiltration into the tumor was found in MNU-induced rat model of breast cancer including the sensory and sympathetic nerve fibers. Nerve density was increased following the growth of tumor. The sensory neurons innervating the thoracic and abdominal mammary tumors peaked at T5 to T6 and L1 to L2 dorsal root ganglions, respectively. Either VEGF receptor inhibitor or antibody against VEGF receptor 2, as well as NGF receptor inhibitor, apparently decreased both the nerve density and vascular density of breast tumor. The reduced nerve density was correlated with the decreased vascular density induced by these treatments. In cultured dorsal root ganglion neurons, phosphatidylinositol 3 (PI3K)/Akt, extracellular signal-regulated protein kinase (ERK), and p38 inhibitors significantly attenuated VEGF-induced neurite elongation. These findings provide direct evidence that VEGF, as well as NGF, may control the axonogenesis of breast cancer.

Perfluorooctanesulfonic acid (PFOS) is an emerging environmental organic pollutant that has been widely used in daily life products in the last century. Numerous studies showed that the accumulation of PFOS in human through food chain would lead to various disease. However, there is currently no report about its in situ localization in the tissue. In present study, we aimed to develop a reproductive and less-cost method to quantitatively detect and determine the spatial distribution of PFOS in mouse kidney by matrix-assisted laser desorption/ionization imaging mass spectrometry (MALDI-IMS) with a commercially available matrix. α-Cyano-4-hydroxycinnamic acid (CHCA) matrix was optimized for PFOS detection in MALDI-IMS analysis. Compared to other organic matrices, CHCA used in negative ion mode showed less background interference and enhanced MS signal intensity and high spatial resolution (80 μm) for PFOS analysis. The use of a CHCA matrix with an autospray system led to successful identification of the PFOS ion signals on the perfusion kidney tissue. The detection limit was at the μg/mL level, with direct visualization from a MS image. The developed method with the optimized parameters was successfully employed to obtain the PFOS spatial distribution in the kidney collected from mice after the PFOS exposure for 14 days. PFOS was mainly distributed in the kidney cortex region, which was consistent with the histological analysis results. Taken together, a rapid, economic, and efficient method was developed for PFOS detection by MALDI-IMS using a CHCA matrix. Mapping the distribution of PFOS by MALDI-IMS with a CHCA matrix provides an innovative approach for the analysis of environmental pollutants in animal or human tissues.

Triclosan (TCS), an extensively used antimicrobial agent, has raised considerable concern due to its hepatocarcinogenic potential. However, previous hepatotoxicity studies primarily focused on the activation of specific intracellular receptors, the underlying mechanisms still warrant further investigation at the metabolic level. Herein, we applied metabolomics in combination with lipidomics to unveil TCS-related metabolic responses in human normal and cancerous hepatocytes. Endogenous and exogenous metabolites were analyzed for the identification of metabolic biomarkers and biotransformation products. In L02 normal cells, TCS exposure induced the up-regulation of purine metabolism and amino acid metabolism, caused lipid accumulation, and disturbed energy metabolism. These metabolic disorders in turn enhanced the overproduction of reactive oxygen species (ROS), leading to the alteration of antioxidant enzyme activities, down-regulation of endogenous antioxidants, and peroxidation of lipids. TCS-induced oxidative stress is thus considered to be one crucial factor for hepatotoxicity. However, in HepG2 cancer cells, TCS underwent fast detoxification through phase II metabolism, accompanied by the enhancement of energy metabolism and elevation of antioxidant defense system, which contributed to the potential effects of TCS on human hepatocellular carcinoma development. These different responses of metabolism between normal and cancerous hepatocytes provide novel and robust perspectives for revealing the mechanisms of TCS-triggered hepatotoxicity.

AbstractRapid hydrolysis of waste activated sludge (WAS) can be achieved by improving activities of extracellular enzymes under proper temperature conditions. As short-chain fatty acids (SCFAs) accumulation is always consumed by methanogens under anaerobic conditions, and considering that the micro-aerobic condition can inhibit the activities of methanogens, in this study, effects of temperature (55–75°C) on thermophilic micro-aerobic fermentation of WAS were investigated. Results showed that the highest soluble chemical oxygen demand (SCOD) yield was obtained at 60°C (4,407 ± 80 mg/L, 36 h), 2.0 times higher than that obtained at 75°C (2,180 ± 40 mg/L, 36 h), the corresponding hydrolysis rate was 0.6689 d−1. The highest SCFAs yield was 2,928 ± 12 mg COD/L at 60°C and 36 h, 4.9 times higher than that obtained at 75°C (594 ± 10 mg COD/L, 36 h). The analysis of SCFAs composition showed that acetic acid (HAc) accounted for the most percentage (>40%), followed by n-valeric (n-HVa) (20–25%), and propionic acids (HPr) (10–15%). Total suspended solids removal efficiency reached 18.7% after 192-h fermentation at 60°C. These results suggested that the optimal thermophilic micro-aerobic conditions for WAS hydrolysis and SCFAs accumulation from WAS were 60°C and 36-h fermentation time.

The global ocean heat content evolution is a key component of the Earth's energy budget and can be consistently determined by ocean reanalyses that assimilate hydrographic profiles. This work investigates the impact of the atmospheric reanalysis forcing through a multiforcing ensemble ocean reanalysis, where the ensemble members are forced by five state‐of‐the‐art atmospheric reanalyses during the meteorological satellite era (1979–2013). Data assimilation leads the ensemble to converge toward robust estimates of ocean warming rates and significantly reduces the spread (1.48 ± 0.18 W/m2, per unit area of the World Ocean); hence, the impact of the atmospheric forcing appears only marginal for the global heat content estimates in both upper and deeper oceans. A sensitivity assessment performed through realistic perturbation of the main sources of uncertainty in ocean reanalyses highlights that bias correction and preprocessing of in situ observations represent the most crucial component of the reanalysis, whose perturbation accounts for up to 60% of the ocean heat content anomaly variability in the pre‐Argo period. Although these results may depend on the single reanalysis system used, they reveal useful information for the ocean observation community and for the optimal generation of perturbations in ocean ensemble systems. The epicuticular waxy layer of plant leaves enhances the extreme environmental stress tolerance. However, the relationship between waxy layer and saline tolerance was not established well. The epicuticular waxy layer of rice (Oryza sativa L.) was studied under the NaHCO3stresses. In addition, strong saline tolerance Puccinellia tenuiflorawas chosen for comparative studies. Scanning electron microscope (SEM) images showed that there were significant changes in waxy morphologies of the rice epicuticular surfaces, while no remarkable changes in those of P. tenuifloraepicuticular surfaces. The NaHCO3-induced morphological changes of the rice epicuticular surfaces appeared as enlarged silica cells, swollen corns-shapes and leaked salt columns under high stress. Energy dispersive X-ray (EDX) spectroscopic profiles supported that the changes were caused by significant increment and localization of [Na+] and [Cl−] in the shoot. Atomic absorption spectra showed that [Na+]shoot/[Na+]rootfor P. tenuifloramaintained stable as the saline stress increased, but that for rice increased significantly. In rice, NaHCO3stress induced localization and accumulation of [Na+] and [Cl−] appeared as the enlarged silica cells (MSC), the swollen corns (S-C), and the leaked columns (C), while no significant changes in P. tenuiflora.

A solid-core microstructured optical fiber is infiltrated with the Dispersed Yellow 7 and chloroform mixture solution, resulting in the appearance of several transmission dips with different temperature and axial load responses. The temperature- and load-dependent spectral characteristics of these dips have been investigated and the experimental results indicate that they are pretty temperature-sensitive, but insensitive to the variation of applied axial load. Such a compact fiber-optic device with electric perturbation immunity could be employed for load-insensitive temperature sensing with high sensitivity.

Cancer cells adapt to hypoxia through HIFs (hypoxia-inducible factors), which initiate the transcription of numerous genes for cancer cell survival in the hypoxia microenvironment. In this study, we find that the FACT (facilitates chromatin transcription) complex works cooperatively with HIFs to facilitate the expeditious expression of HIF targets for hypoxia adaptation. Knockout (KO) of the FACT complex abolishes HIF-mediated transcription by impeding transcription elongation in hypoxic cancer cells. Interestingly, the FACT complex is post-translationally regulated by PHD/VHL-mediated hydroxylation and proteasomal degradation, in similar fashion to HIF-1/2α. Metabolic tracing confirms that FACT KO suppresses glycolytic flux and impairs lactate extrusion, leading to intracellular acidification and apoptosis in cancer cells. Therapeutically, hepatic artery ligation and anti-angiogenic inhibitors adversely induce intratumoral hypoxia, while co-treatment with FACT inhibitor curaxin remarkably hinders the growth of hypoxic tumors. In summary, our findings suggest that the FACT complex is a critical component of hypoxia adaptation and a therapeutic target for hypoxic tumors.

A fiber in-line Mach–Zehnder interferometer is fabricated by selectively filling liquid into one air hole of the innermost layer of a photonic crystal fiber (PCF). The refractive index of the liquid is so close to that of the background silica in the wavelength range of 1300–1600 nm that the two-mode PCF evolves into multimode PCF with an elliptically shaped core. Due to the different propagation constants, interference can occur between the fundamental mode and higher-order modes of the liquid-filled PCF. Such a device is applied in temperature and strain measurements with high sensitivities of 16.49  nm/°C and −14.595  nm/N, respectively.

There have been no multicity studies on the acute effects of air pollution on stroke mortality in China. This study was undertaken to examine the associations between daily stroke mortality and outdoor air pollution (particulate matter <10 m in aerodynamic diameter, sulfur dioxide, and nitrogen dioxide) in 8 Chinese cities.

Hypoxia, low oxygen (O2), is a key feature of all solid cancers, including hepatocellular carcinoma (HCC). Genome-wide CRISPR-Cas9 knockout library screening is used to identify reliable therapeutic targets responsible for hypoxic survival in HCC. We find that protein-tyrosine phosphatase mitochondrial 1 (PTPMT1), an important enzyme for cardiolipin (CL) synthesis, is the most significant gene and ranks just after hypoxia-inducible factor (HIF)-1α and HIF-1β as crucial to hypoxic survival. CL constitutes the mitochondrial membrane and ensures the proper assembly of electron transport chain (ETC) complexes for efficient electron transfer in respiration. ETC becomes highly unstable during hypoxia. Knockout of PTPMT1 stops the maturation of CL and impairs the assembly of ETC complexes, leading to further electron leakage and ROS accumulation at ETC in hypoxia. Excitingly, HCC cells, especially under hypoxic conditions, show great sensitivity toward PTPMT1 inhibitor alexidine dihydrochloride (AD). This study unravels the protective roles of PTPMT1 in hypoxic survival and cancer development.

The Earth's energy imbalance (EEI) is stored in the oceans for the most part. Thus, estimates of its variability can be ingested in ocean retrospective analyses to constrain the global ocean heat budget. Here we propose a scheme to assimilate top of the atmosphere global radiation imbalance estimates from Clouds and the Earth's Radiant Energy System (CERES) in a coarse‐resolution variational ocean reanalysis system (2000–2014). The methodology proves able to shape the heat content tendencies according to the EEI estimates, without compromising the reanalysis accuracy. Spurious variability and underestimation (overestimation) present in experiments with in situ (no) data assimilation disappear when EEI data are assimilated. The warming hiatus present without the assimilation of EEI data is mitigated, inducing ocean warming at depths below 1,500 m and slightly larger in the Southern Hemisphere, in accordance with recent studies. Furthermore, the methodology may be applied to Earth System reanalyses and climate simulations to realistically constrain the global energy budget. Ocean reanalyses show global heat content tendencies whose variability barely matches that from radiation imbalance measurements at the TOAA scheme is introduced to assimilate energy imbalance estimates into ocean reanalyses for constraining the global heat budgetThe new assimilation successfully corrects the heat content tendency variability without any loss of accuracy in the reanalysis
