The structural information of offshore oil production equipment is the basis for the functional modification and upgrading of offshore oil drilling platforms. In order to solve the problems of low efficiency in the process of acquiring offshore oil production equipment structure information by traditional measurement methods, we propose a deep learning-based point cloud data processing scheme for offshore oil production equipment. First, a point cloud dataset of offshore oil production equipment for deep learning is constructed, and a deep learning network based on the two-step downsampling method and the local feature aggregation of each point after downsampling is implemented for the semantic segmentation of the dataset. Second, the combined point cloud filtering process based on radius filtering and statistical filtering is implemented on the segmented point cloud data. Third, an implicit surface reconstruction based on contextual prior information is implemented for offshore oil production equipment components. The dataset contains six types of point clouds, including pipelines, flanges, shelves, bends, valves, and oil recovery trees. Based on this dataset for semantic segmentation, the overall segmentation accuracy reaches 98.87% and the mIoU reaches 92.79%. Combined filtering is performed on the segmented offshore oil production equipment data, and the denoising rate can reach 94%. Finally, the denoised point cloud data is utilized for 3-D reconstruction, and the overall consistency accuracy can reach 1.85 mm, which can provide fast, efficient, and reliable data support for the upgrading of offshore oil rigs.

In response to the emergence of SARS-CoV-2 variants of concern, the global scientific community, through unprecedented effort, has sequenced and shared over 11 million genomes through GISAID, as of May 2022. This extraordinarily high sampling rate provides a unique opportunity to track the evolution of the virus in near real-time. Here, we present outbreak.info, a platform that currently tracks over 40 million combinations of Pango lineages and individual mutations, across over 7,000 locations, to provide insights for researchers, public health officials and the general public. We describe the interpretable visualizations available in our web application, the pipelines that enable the scalable ingestion of heterogeneous sources of SARS-CoV-2 variant data and the server infrastructure that enables widespread data dissemination via a high-performance API that can be accessed using an R package. We show how outbreak.infocan be used for genomic surveillance and as a hypothesis-generation tool to understand the ongoing pandemic at varying geographic and temporal scales.

Outbreak.info Research Library is a standardized, searchable interface of coronavirus disease 2019 (COVID-19) and severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) publications, clinical trials, datasets, protocols and other resources, built with a reusable framework. We developed a rigorous schema to enforce consistency across different sources and resource types and linked related resources. Researchers can quickly search the latest research across data repositories, regardless of resource type or repository location, via a search interface, public application programming interface (API) and R package.

Background: STAM-binding protein-like 1 (STAMBPL1) functions as a deubiquitinase to cleave Lys63 ubiquitin linkage, and is associated with cancer dissemination and progression. The role of STAMBPL1 in colorectal cancer (CRC) remains unclear.Methods: STAMBPL1 expression was determined by western blot and qRT-PCR. Cell proliferation was detected by colony formation and MTT assays, and apoptosis was assessed by flow cytometry. The metastasis was evaluated by transwell and wound healing assays. An animal xenograft experiment was used to investigate the effect of STAMBPL1 on tumor growth.Results: The expression of STAMBPL1 was elevated in CRC cells. Knockdown of STAMBPL1 reduced cell viability of CRC and suppressed the proliferation, invasion, and migration. Apoptosis of CRC was induced by silence of STAMBPL1. Tumor growth of CRC was also suppressed by the silence of STAMBPL1. Knockdown of STAMBPL1 increased IκB and decreased phosphorylation of IκB to reduce p65 phosphorylation.Conclusion: Knockdown of STAMBPL1 inhibited cell growth and metastasis of CRC through inactivation of the NF-κB pathway.

Recently, interferometric altimeters (IAs), such as the surface water and ocean topography (SWOT) satellite, have been launched and will improve the observation of ocean dynamics. The validation of altimeters is a complex and important process that can effectively improve their observation accuracy. For the IA validation of sea surface height (SSH) in 2-D, it is normal to use the steric height (SH) to validate the SSH. However, the validation method is affected by many factors. In order to improve the validation accuracy, the South China Sea (SCS), which is a suitable area for validation of China’s offshore, was selected in this study to analyze the impact of wind on the validation procedures. Through the analysis of the spatial–temporal relationships between wind speed and SH and SSH (SH-SSH), we found correlations between wind speed and validation results. The high wind speeds can lead to a certain deviation in the relationship between SH and SSH, and the special weather conditions make it unsuitable for validation. Meanwhile, in terms of time, May is the time when the wind speed is relatively low, making it more suitable for validation. Also, it is better when the wind speed is less than 12 m/s and the range of U wind and V wind should be −4 to 2 m/s. These analyses are useful for the validation procedures in SSH.

With their unique structures and functionalities, rotating machineries are widely employed in various high-end industries. However, eccentric faults of rotors during continuous high-speed operation represent a critical factor in causing safety accidents. Therefore, real-time monitoring of rotor health is crucial for promptly identifying potential safety hazards and fault indications, enabling preventative maintenance, and enhancing safety performance. Nonetheless, traditional monitoring methods rely on sensors installed on rotating components, which are prone to interference in rotational environments, involve complex installation and maintenance procedures, and pose significant challenges to rotor health monitoring. In this study, a self-powered rotor eccentricity monitoring sensor (RE-TENGs) is designed and fabricated, integrating triboelectric nanogenerator technology. This sensor utilizes the rotor as the self-sensing entity, requiring no external power supply and featuring simplified installation, enabling real-time monitoring of rotor health during rotational machinery operation. A linear relationship between the RE-TENG's eccentric response and electrical characteristics was established through dynamic testing. Furthermore, the actual eccentricity orientation of the rotor was successfully determined by employing vector analysis and synthesis methods with multiple RE-TENGs coupled. To further validate the practicality and accuracy of RE-TENGs, a monitoring testbed for eccentric faults of the spindle rotor in a CNC machine tool was constructed, with the spindle rotor's eccentric fault simulated through misaligned installation. Experimental results demonstrate that RE-TENGs can effectively detect rotor faults induced by eccentric forces during the actual operation of machine tools, accurately determining the eccentric direction and displacement. With an actual angle recognition error of less than ±0.824° and an eccentric distance recognition accuracy exceeding 98%, the exceptional performance of RE-TENGs in monitoring and precise identification of eccentric faults in rotating machinery rotors is unequivocally verified. By integrating triboelectric nanogenerator (TENG) technology, this study realizes real-time monitoring and precise identification of eccentric faults in rotating machinery rotors. It provides a novel solution for health condition monitoring of rotating machinery and offers a reference for overcoming the cumbersome installation and complex wiring of traditional sensors in practical applications.

Bisphenol A (BPA) is a typical endocrine disruptor. Bisphenol S (BPS) has been widely used as a substitute for various plastic materials due to the limited application of BPA. However, it does not mean that BPS is a safe substitute due to the lack of effective evaluation of BPS. In this study, the clinical model of Caenorhabditis elegans(C. elegans) was used to study the effects of BPS on the locomotion behavior, growth, reproduction, lifespan and antioxidant system. Our study found that C. elegansexposed to 0.01 μM BPS could have significantly inhibited locomotion behavior and growth, as well as damaged reproductive and antioxidant systems and lifespan. It is interesting to note that in multi-generational exposure studies, we found that BPS exhibits complex genotoxicity. With the transmission to the offspring, BPS showed more significant inhibition of the head thrashes of the nematode, while the effect on the body bends and body length was gradually weakened. The effect of BPS on the brood size shows different rules according to different concentrations and offsprings. Therefore, the safety of BPS still needs further evaluation, especially the multi-generational genotoxicity.

A 150‐d feeding experiment was conducted to assess the effect of the dietary energy to protein ratio (E/P ratio) on growth, biological indices, proximate composition, and amino acid profiles in the muscle of Varicorhinus macrolepis. Six experimental diets were formulated with different E/P ratios (30.51, 33.90, 37.82, 42.34, 47.70, and 54.09 kJ/g). In the experiment, 360 fish with a mean weight of 8.75 g (SD, 0.59) were randomly divided into six groups (three replicates per group, 20 fish per replicate) and fed three times daily with the experimental diet. Results showed that when the dietary E/P ratio was 37.82 kJ/g, Varicorhinus macrolepistended to gain the highest specific growth rate. With a change in the increment of dietary E/P ratio, the condition factor, intestosomatic index, hepatosomatic index, and viscerosomatic index all changed in the quadric polynomial regression model. Muscle crude lipid content showed a tendency of leveling‐off first and then increased with increasing dietary E/P ratio. Varicorhinus macrolepisgained the highest concentration of total amino acids and essential amino acids when the dietary E/P ratio was 37.82 kJ/g. Except for methionine + cysteine, phenylalanine + tyrosine, and the amino acid score of lysine, the amino acid scores and chemical scores of the other essential amino acids were highest when the dietary E/P ratio was 37.82 kJ/g. These results suggest that Varicorhinus macrolepisobtained optimal growth and muscle amino acid composition when the dietary E/P ratio was 37.82 kJ/g.

Gravity anomalies (GAs) and vertical gravity gradient anomalies (VGGs) are regularly used to predict bathymetry. However, few studies have explored the combination of the GAs and VGGs to predict bathymetry. We introduce the back propagation (BP) neural network into bathymetric prediction field and propose a method to predict depth from GAs and VGGs. The method was tested in the Mariana Trench region, and a neural network bathymetry model was constructed using feature data obtained from GAs and VGGs as the input of the BP neural network. Additionally, single‐beam sounding data were used as label data. By comparing the neural network and the gravity‐geologic method (GGM), the neural network was found to provide better performance with an accuracy improvement of 19%. The root‐mean‐square of the absolute difference between the neural network bathymetry model and the single‐beam sounding data was 72.40 m, with a relative accuracy of 1.71%. Approximately 50% of the differences were distributed within ±20 m, and 90% were distributed within ±100 m. The neural network bathymetry model was also compared with the GGM bathymetry model for different depths and topographies, and the results verified the feasibility and effectiveness of the BP neural network method. The gravity data currently used to predict bathymetry include gravity anomalies and vertical gravity gradient anomalies. We propose a new method that introduces the back propagation neural network into bathymetric prediction, based on two kinds of gravity data, which are used to predict depth. The results show that the depths predicted by the neural network are closer to the shipborne depths than are those estimated with the current method. We also test the adaptability of the neural network method for different depths and topographies (seamount, trench, and plain). Finally, we find that the BP neural network method is competent for various inversion tasks. Barley is rich in nutritionally positive compounds, but the quality of bread made of wheat–barley composite flours is impaired when a high percentage of barley is used in the mixture. A number of enzymes have been reported to be useful additives in breadmaking. However, the effect of ß-glucanase on breadmaking has scarcely been investigated. In this paper, the influence of different levels (0.02, 0.04, 0.06, and 0.08%, based on composite flour) of ß-glucanase (100,000 U/g) on the properties of dough and bread from 70% wheat, 30% barley composite flour were studied. Although dough development time, dough stability, and protein weakening value decreased after ß-glucanase addition, dough properties such as softness and elasticity as well as bread microstructure were improved compared with the control dough. ß-Glucanase also significantly improved the volume, texture, and shelf life of wheat–barley composite breads. The use of an optimal enzyme concentration (0.04%) increased specific volume (57.5%) and springiness (21%), and it reduced crumb firmness (74%) and staling rate. Bread with added ß-glucanase had a better taste, softness, and overall acceptability of sensory characteristics compared with the control bread. Moreover, the quality of wheat–barley composite bread after addition of 0.04% ß-glucanase was nearly equal to the quality of pure wheat bread. These results indicate that dough rheological characteristics and bread quality of wheat–barley composite flour can be improved by adding a distinct level of ß-glucanase.

• A parametrized nonlinear aeroelastic model of the folding wing with bilinear-stiffness hinges has been established. • The nonlinear aeroelastic responses of the folding wing at variable structural parameters can be calculated efficiently by the parameterized model. • Harmful subcritical or beneficial supercritical LCOs will occur at the cases with different hinge stiffness or different folding angles. • For a folding wing designed with specific stiffness values, if there is nonlinearity in the folding mechanism, then supercritical behavior appears and helps to suppress the flutter. A morphing wing with structural nonlinearities may encounter complex nonlinear oscillations during the in-flight morphing process. This paper investigates the nonlinear aeroelastic vibrations of a folding wing with bilinear stiffness in its hinge joints. The parameterized-fictitious-mode method is used to compute the nonlinear aeroelastic response of the morphing wing [as a subcritical or supercritical limit-cycle oscillation (LCO)] efficiently at different folding angles without having to perform repeated finite-element modeling. A low-aspect-ratio morphing wing is selected as a test case to investigate the nonlinear oscillations, and how the hinge stiffness and folding angle affect the nonlinear aeroelastic behavior is also investigated efficiently. The numerical results show that the nonlinear aeroelastic behavior of the folding wing is very complex. For a folding wing with sufficiently strong nonlinearity, the flight region of nonlinear oscillations is above the flutter boundary, and benign LCOs occur at flow speeds higher than the flutter speed. As the hinge stiffness is decreased, the region moves downward and is eventually entirely below the flutter boundary, whereupon detrimental LCOs occur at flow speeds lower than the flutter speed. Atherosclerosis is an inflammatory response to accumulation of cholesterol in the artery wall. Low-density lipoproteins (LDL) accumulate and are oxidized to proinflammatory compounds in the arterial intima during hypercholesterolemia, leading to activation of endothelial cells, macrophages, and T cells. We sought to define the role of oxidized LDL (oxLDL) in the poloxamer 407 (P-407)-induced mouse model of dose-dependent hyperlipidemia and atherosclerosis developed in our laboratory. The hyperlipidemic agent P-407 was evaluated for its ability to oxidize native LDL in vitro as determined by measuring the rate of formation of conjugated dienes, as well as malondialdehyde (MDA) production using the thiobarbituric acid reactive substances assay. Additionally, plasma obtained from C57BL/6 mice treated with P-407 for 100 days and maintained on either a normal diet or a diet supplemented with 0.5% w/w cholic acid was assayed for both MDA and lipid hydroperoxide content. Lastly, plasma from these same groups of mice was analyzed for the presence of immunoglobulin (Ig) G and IgM autoantibodies against oxLDL. Our results indicate that P-407 is unable to directly oxidize native LDL in vitro. However, plasma obtained from P-407-treated mice demonstrated a significant (P< 0.05) increase in the content of oxidized lipids, but showed a significant (P< 0.05) decrease in the concentration of lipid hydroperoxides when compared to controls. Both plasma IgG and IgM antibodies to MDA-modified LDL (MDA-LDL) were significantly (P< 0.05) elevated in P-407-treated mice, as was IgG1 anti-MDA-LDL, whereas the titer of IgG2a anti-MDA-LDL was significantly (P< 0.05) reduced. We suggest that P-407 causes oxidation of LDL in vivo by an indirect mechanism and we further conclude that P-407-induced hypercholesterolemia in C57BL/6 mice is associated with T cell-dependent (IgG) and T cell-independent (IgM) B-cell responses to MDA-LDL.

During the last few decades, the pathogenesis of atherosclerosis has been related not only to cholesterol deposition and cell proliferation in the lesions, but also to infiltration of immune cells, which are involved in both systemic and local, innate as well as adaptive, immune responses. A number of antigen candidates, such as oxidised low-density lipoprotein and heat-shock proteins, have been associated with the disease process. As some inflammatory and autoimmune diseases could be treated by immunologically based therapy, it is of particular interest whether such principles can also be applied to prevent or treat atherosclerosis. Indeed, modification of immune reactions in animal models can greatly affect the development and progression of atherosclerosis. This review provides an overview of our current understanding of how immunomodulation changes the course of atherosclerosis and how vaccination may be used for preventing the disease.

Atherosclerosis is an inflammatory disease induced by a lipid metabolic disturbance at sites of hemodynamic strain in the vasculature. Studies in both man and experimental animal models show an involvement of innate and adaptive immune mechanisms in the disease process. Our recent studies in apoE-knockout mice show that the level of hypercholesterolemia affects the functional properties of the immune response. Modulating immune activity by injections of polyclonal immunoglobulins inhibits disease progression, suggesting that immunomodulation may be useful to treat atherosclerosis. Analysis of T cell receptor (TCR) mRNA in atherosclerotic lesions shows expansions of T cells expressing TCR-V?6, a receptor type that is also expressed by T cells recognizing oxidized low density lipoprotein (oxLDL). This suggests that oxLDL is an autoantigen that induces strong, local T cell responses in the plaque. Further characterization of this and other candidate antigens, such as heat shock proteins and macromolecular components of Chlamydia pneumoniae, may provide important information on which specific interference with the disease process could be based.

Background: In the course of atherosclerosis, humans and apolipoprotein (apoE) Knockout (KO) mice exhibit an active cell-mediated and humoral immune process, both at the systemic level and within atheromata. Low density lipoproteins (LDL) infiltrate the vascular wall, where they are oxidatively modified. This oxidative modification may generate new epitopes for which tolerance is not achieved during ontogenesis. Such epitopes could constitute new targets for autoreactive immune responses that may have a physiopathological role in disease development. Materials and Methods: Exposing mice to high dose of antigens during thymic T-cell education induces immunological tolerance to the administered antigens. We injected newborn apoE KO mice with oxidized LDL. They were fed a cholesterol-rich diet and aortic atherosclerosis, cell-mediated immune response, and T-cell repertoire were analyzed after 5 months. Results: Injection of oxidized LDL at birth reduced not only the immune response to oxidized LDL, but also susceptibility to atherosclerosis in apoE mice. Injection of oxidized LDL induced T-cell tolerance due to clonal deletion, rather than anergy of the reactive T cells. The T-cell repertoire of apoE KO mice was affected by the development of the disease, whereas tolerization normalized it. Conclusions: This study demonstrates that the immune response against oxidized LDL has a deleterious role in atherogenesis and that a fine-tuning of this response could modify the course of the disease.

•ERCP may be inappropriate for some patients with choledocholithiasis.•One-step PTCSL is beneficial in the treatment of patients with choledocholithiasis.•One-step PTCSL might be a reasonable alternative in the treatment of choledocholithiasis patients, especially when ERCP is not feasible.

Conventional and nanostructured ceramic coatings were prepared with conventional and nanostructured aggregate Al2O3-13wt.%TiO2ceramic powders through plasma spraying and plasma spraying-laser remelting combined technique. Influences of feedstocks and laser remelting on erosion resistance of the prepared coatings were discussed, and the erosion failure mechanisms of four types of coatings were discussed. Results demonstrate that compared with coatings prepared by plasma spraying, the coating prepared by laser remelting has better erosion resistance. Given the equal conditions, nanostructured coating shows stronger erosion resistance than conventional coating. Conventional ceramic coating shows typical brittle erosion characteristics and nanostructured ceramic coating also shows brittle erosion characteristics, accompanied with certain degree of plastic erosion characteristics. Erosive wearing of plasma coating is mainly lamellar peeling, accompanied with certain degree of brittle ceramic particle crushing. Coating prepared by laser remelting present crack initiation and propagation on near surface, finally leading to crystal crushing and peeling of remelting layers.

Due to the geographical peculiarities of the sea islands, we often encounter the difficulty that some coastlines are hard to reach or to identify. Thus a specific measurement method cannot be implemented effectively, which affects the integrity and precision of island coastline surveying. In this paper, we use the digital elevation Model (DEM) by LIDAR date. However, The elevations acquired by an Airborne LIDAR system are ellipsoid heights, which is different from the National Vertical Datum 1985 in existing island coastline. For this reason, a regional quasi-geoid model is constructed with the geometric method, using the EGM2008 gravity field model and GPS/Leveling data. We manage to transform ellipsoid heights to normal heights from observations, and construct the DEM which is based on National Vertical Datum 1985. Finally island coastlines are extracted from the reference DEM and the associated tidal model and compared with that from GPS-RTK measurements.
