Genetic variation among 378 melon (Cucumis melo L.) germplasm accessions collected in India in 1992 and 26 accessions in China in 1994 was evaluated with 19 isozyme loci. ‘Top Mark’ and ‘Green Flesh Honeydew’, which represented two distinct C. melo ssp. melo L. groups, Cantalupensis and Inodorus, respectively, were used as reference cultivars. Genetic distances among accessions were calculated, and an initial cluster analysis using these distances resulted in 148 groups of varying size, ranging from two to 47 accessions. One accession from each of the 148 groups was chosen at random and used in a second cluster analysis that identified 11 accession groups. Group 1 was unique and consisted of only two C. melo ssp. agrestis (Naudin) Pangalo accessions. Two large branches were detected at cluster node 2. One branch was comprised of three groups of 3, 12, and 34 accessions, while the other branch contained seven groups of 2, 3, 14, 16, and 47 accessions, and the reference cultivars. Of the 148 accessions, 132 were from 41 sites in Rajasthan and Madhya Pradesh, India, which were distributed unequally across the 11 groups. The 14 Chinese ac- cessions originating from seven provinces were also dispersed unequally in the four major cluster groups. ‘Top Mark’ and ‘Green Flesh Honeydew’ were genetically distinct and uniquely clustered in the same group. Analyses of complete 1143-base pair sequence of the mitochondrial cytochrome b gene demonstrated a sister relationship between Japanese R . nigromaculata and Korean R . plancyi chosenica , but not with Korean R . nigromaculata , while the allozyme data strongly supported the monophyly of the Korean and Japanese populations of R . nigromaculata . We surmise this discordance to be the result of the inheritance of introduced mtDNA and the dilution of introduced nuclear DNA in mixed lineages after past hybridization and genome introgression between the two species, although the direction of introgression is unknown. I consider how the study of genetic variation has influenced efforts to conserve natural populations over the last 50 years. Studies with allozymes in the 1970s provided the first estimates of the amount of genetic variation within and between natural populations at multiple loci. These early studies played an important role in developing plans to conserve species. The description of genetic variation in mitochondrial DNA in the early 1980s laid the foundation for the field of phylogeography, which provided a deeper look in time of the relationships and connectivity among populations. The development of microsatellites in the 1990s provided much more powerful means to describe genetic variation at nuclear loci, including the ability to detect past bottlenecks and estimate current effective population size with a single temporal sample. In the 2000s, single nucleotide polymorphisms presented a cornucopia of loci that has greatly improved power to estimate genetic and population demographic parameters important for conservation. Today, population genomics presents the ability to detect regions of the genome that are affected by natural selection (e.g. local adaptation or inbreeding depression). In addition, the ability to genotype historical samples has provided power to understand how climate change and other anthropogenic phenomena have affected populations. Modern molecular techniques provide unprecedented power to understand genetic variation in natural populations. Nevertheless, application of this information requires sound understanding of population genetics theory. I believe that current training in conservation genetics focuses too much on the latest techniques and too little on understanding the conceptual basis which is needed to interpret these data and ask good questions., (© 2016 John Wiley & Sons Ltd.)
