With the rapid development of sports, sports injuries have become an inevitable problem in sports. Repairing sports injuries has always been the focus of attention in the field of sports and medicine. The wide application of biomedical materials leads to the innovation and development of medical technology. It is of great significance to use them to repair sports injuries, but the current research results are not ideal. In this paper, the biocompatibility of composite biomaterials used to repair sports injuries was studied. First, the rat bone marrow mesenchymal stem cells were cultured to prepare a cell suspension. Then, the tricalcium phosphate-gelatin composite scaffold material and the collagen material were scraped into powder to make the material extract, which are, respectively, the experimental group and the control group. An apoptosis detection kit and a flow cytometer were used to detect apoptosis rate and survival rate of all samples, and a CKK-8 kit was used to detect cytotoxicity and calculate cell proliferation activity. The absorbance value was read with a microplate reader, and the coagulation performance and hemolysis rate of different groups were calculated. The experimental results showed that the cell survival rate of the experimental group was 91.41%, the early apoptosis rate was 2.81%, the stem cell proliferation activity was 0.83, and the anticoagulant performance and hemolysis rate were 64.8% and 0.28%, respectively. This indicates that although the anticoagulant performance of the tricalcium phosphate-gelatin composite scaffold is weak, its cell proliferation activity is strong, which is more suitable for cell adhesion and proliferation, and the cell apoptosis and hemolysis rate are lower. Adhesion to host cells is the first step in the virulence cycle of any pathogen. In Gram‐negative bacteria, adhesion is mediated, among other virulence factors such as the lipopolysaccharides, by specific outer‐membrane proteins generally termed adhesins that belong to a wide variety of families and have different evolutionary origins. In Brucella, a widespread zoonotic pathogen of animal and human health concern, adhesion is central as it may determine the intracellular fate of the bacterium, an essential stage in its pathogenesis. In the present paper, we further characterised a genomic locus that we have previously reported encodes an adhesin (BigA) with a bacterial immunoglobulin‐like domain (BIg‐like). We found that this region encodes a second adhesin, which we have named BigB; and PalA, a periplasmic protein necessary for the proper display in the outer membrane of BigA and BigB. Deletion of bigB or palA diminishes the adhesion of the bacterium and overexpression of BigB dramatically increases it. Incubation of cells with the recombinant BIg‐like domain of BigB induced important cytoskeletal rearrangements and affected the focal adhesion sites indicating that the adhesin targets cell–cell or cell–matrix proteins. We additionally show that PalA has a periplasmic localisation and is completely necessary for the proper display of BigA and BigB, probably avoiding their aggregation and facilitating their transport to the outer membrane. The present study outlines the evaluation of textile materials that are currently in the market for cell culture applications. By using normal LaserJet printing techniques, we created the substrates, which were then characterized physicochemically and biologically. In particular, (i) we found that the weave pattern and (ii) the chemical nature of the textiles significantly influenced the behaviour of the cells. Textiles with closely knitted fibers and cell adhesion motifs, exhibited better cell adhesion and proliferation over a period of 7 days. All the substrates supported good viability of cells (>80%). At present, the healing of osteopathy, especially the healing of cartilage, has been proven to be difficult. Commonly used treatment methods are autogenous bone grafts and allogeneic bone grafts, but grafts cannot fully meet the clinical treatment requirements due to problems related to the source, price, immunity, and other concerns. Thus, the combination of biomaterials and tissue engineering technology has become a new direction in research. Among studies on tissue engineering bone and cartilage materials, hydrogels that show biological activity, absorbability after degradability, plasticity, and easy preparation have become the focus. Hydrogels are used as extracellular matrix mimics. Although various materials are able to form hydrogels, hyaluronic acid and its derivatives are prominently used. Hyaluronic acid hydrogels have many advantages, such as promoting cell adhesion and proliferation and wound healing. They also demonstrate sufficient biological activity for stimulating a microenvironment for cell survival. However, their disadvantages require further modification and include a poor degradation rate and insufficient mechanical performance. In this paper, hyaluronic acid-based hydrogels, their modifications, applications, and mechanisms, as well as new techniques for processing hyaluronic acid hydrogels in bone and cartilage tissue engineering, are briefly reviewed, and their future prospects and directions for future work are discussed. Objective: To investigate the effects of a drug‐coated balloon (DCB) on inflammatory cytokines in patients with coronary artery calcification (CAC) after interventional therapy. Methods: This study included 58 patients with coronary heart disease who underwent coronary angiography (CAG) from October 2020 to September 2021. Patients were divided into CAC and non‐CAC groups, and a DCB was used to intervene in the target lesions. Ten‐milliliter preoperative and postoperative blood samples were drawn from the coronary lesions in both groups to detect the expression of serum interleukin‐6 (IL‐6), tumor necrosis factor‐alpha (TNF‐α), and intercellular adhesion molecule‐1 (ICAM‐1). All patients were subjected to a 6‐month follow‐up to observe the incidence of major adverse cardiac events (MACEs). Results: No significant differences in baseline clinical data were found between the groups. Serum IL‐6, TNF‐α, and ICAM‐1 expressions in coronary blood samples immediately before DCB were not significantly different from those after DCB in all patients. After DCB, serum TNF‐α expression in the CAC group was significantly lower than that in the non‐CAC group (p < 0.05). In contrast, no significant difference in serum IL‐6 and ICAM‐1 expression was found between the groups. During the 6‐month follow‐up, no significant difference in the incidence of MACE was found between both groups. Conclusions: DCB reduced the expression of inflammatory cytokine TNF‐α in CAC, which may be one of the key mechanisms underlying the treatment of CAC by DCB. [ABSTRACT FROM AUTHOR]

Bone tissue engineering necessitates the development of scaffolds with optimal properties to provide a suitable microenvironment for cell adhesion, proliferation, and osteogenic differentiation. The selection of appropriate scaffold materials remains a critical challenge in this field. In this study, we aimed to address this challenge by evaluating and comparing the performance of hydrogel scaffolds reinforced with β‐tricalcium phosphate (β‐TCP), allograft, and a combination of allograft and strontium hydroxyapatite (SrHA). In this study, scaffolds containing the following compounds with a weight ratio of 75 : 25 : 50 were made using a 3D printer: group (1) alginate + gelatin + β‐TCP (TCP), group (2) alginate + gelatin + allograft (Allo), and group (3) alginate + gelatin + allograft + strontium hydroxyapatite (Str). Stem cells extracted from rat bone marrow (rBMSCs) were cultured on scaffolds, and cell proliferation and differentiation tests were performed. Also, the physical and chemical properties of the scaffolds were investigated. The two/one‐way analysis of variance (ANOVA) by Tukey's post hoc test was performed. There was no significant difference between scaffolds with pore size and porosity. TCP scaffolds' mechanical strength and degradation rate were significantly lower than the other two groups (P < 0.05). Also, the swelling ratio of Allo scaffolds was higher than in other samples. The amount of cell proliferation in the samples of the TCP group was lower than the other two, and the Allo samples had the best results in this concern (P < 0.01). However, the scaffolds containing strontium hydroxyapatite had significantly higher bone differentiation compared to the other two groups, and the lowest results were related to the scaffolds containing β‐TCP. Hydrogel scaffolds reinforced with allograft or its combination with strontium showed better physicochemical and biological behavior compared to those reinforced with β‐TCP. Besides, adding strontium had a limited impact on the physicochemical features of allograft‐containing scaffolds while improving their potential to induce osteogenic differentiation. There is an increasing concern that the term adipose tissue-derived stem cell (ASC) is inappropriately used to refer to the adipose stromal vascular fraction (SVF). To evaluate the accuracy and quality of reporting, 116 manuscripts on the application of ASC in humans and animals were examined based on the 2013 published International Federation for Adipose Therapeutics and Science (IFATS)/ International Society for Cellular Therapy (ISCT) joint statement and in reference to current guidelines for clinical trials and preclinical studies. It is disconcerting that 4 among the 47 papers or 8.51% (CI 2.37–20.38) surveyed after publication of IFATS/ISCT statement reported using ASCs but in fact they used unexpanded cells. 28/47 or 59.57% (CI 44.27–73.63) explicitly reported that adherent cells were used, 35/47 or 74.47% (CI 59.65–86.06) identified expression of surface markers, and 25/47 or 53.19% (CI 14.72–30.65) verified the multilineage potential of the cells. While there are a number of papers examined in this survey that were not able to provide adequate information on the characteristics of ASCs used with some erroneously referring to the SVF as stem cells, there are more room for improvement in the quality of reporting in the application of ASCs in humans and animals. Purpose. The crystal adhesion caused by the damage of renal tubular epithelial cells (HK-2) is the key to the formation of kidney stones. However, no effective preventive drug has been found. This study aims to explore the recovery effects of four Laminaria polysaccharides (SLPs) with different sulfate (–OSO3–) contents on damaged HK-2 cells and the difference in the adhesion of damaged cells to nanometer calcium oxalate monohydrate (COM) and calcium oxalate dihydrate (COD) before and after recovery. Methods. Sodium oxalate (2.6 mmol/L) was used to damage HK-2 cells to establish a damaged model. SLPs (LP0, SLP1, SLP2, and SLP3) with –OSO3– contents of 0.73%, 15.1%, 22.8%, and 31.3%, respectively, were used to restore the damaged cells, and the effects of SLPs on the adhesion of COM and COD, with a size of about 100 nm before and after recovery, were measured. Results. The following results were observed after SLPs recovered the damaged HK-2 cells: increased cell viability, restored cell morphology, decreased reactive oxygen levels, increased mitochondrial membrane potential, decreased phosphatidylserine eversion ratio, increased cell migration ability, reduced expression of annexin A1, transmembrane protein, and heat shock protein 90 on the cell surface, and reduced adhesion amount of cells to COM and COD. Under the same conditions, the adhesion ability of cells to COD crystals was weaker than that to COM crystals. Conclusions. As the sulfate content in SLPs increases, the ability of SLPs to recover damaged HK-2 cells and inhibit crystal adhesion increases. A novel nanointerface of graphene oxide-doped polypyrrole (GO/PPy) is prepared on the surface of an indium tin oxide (ITO) electrode for electrochemical impedance detection of cell adhesion and proliferation through a facile one-step electropolymerization. The prepared GO/PPy nanocomposite had a robust surface and provided a biocompatible substrate for A549 cells adhesion and proliferation. The adhesion and proliferation of A549 cells on the surface of the GO/PPy modified ITO electrode directly increased the electron transfer resistance of [Fe(CN)6]3−/4− redox probe and influenced the impedance properties of the GO/PPy modified ITO electrode system. Based on these results, the adhesion and proliferation of A549 cells could be detected by electrochemical impedance technology using two approaches. Therefore, the present paper confirms that the GO/PPy nanocomposite film provides an excellent biological-electrical interface for cell immobilization and offers advantages of simple, low-cost fabrication and multiparameter detection and possesses potential application in cytological studies. Summary: Bioaugmentation is considered a potential tool to boost the performance of microbial fuel cells (MFCs) by modifying electrodes with biopolymers. However, research on biopolymers in MFCs to treat real wastewater is very limited. Polylactic acid (PLA) is a biopolymer obtained from renewable feedstock, extensively being used in the form of nanofibers due to its biocompatible nature, while polyaniline (PANI) is a favorite candidate for electrode modification in MFC under its good electrical conductivity and stability. Interestingly, PANI‐based degradable nanofibers are known to support cell adhesion and proliferation. The current work is, therefore, aimed at finding a new environmentally benign composite material using electrospun PLA nanofiber decorated with in situ polymerized PANI. The resultant PANI/PLA electrode was tested for its efficacy as anode with domestic wastewater‐fed MFCs against conventional graphite felt. The flexible anode outperformed the flat graphite felt anode displaying a 92.32% decrement in charge transfer impedance and 8‐fold increase in maximum power density with excellent cell adherence under which simultaneous COD removal of 91.7% was also achieved. The present study has successfully added a new dimension to the design of sustainable anode material to enhance microbial binding and achieve high power output, revealing the enormous potential of biopolymer‐based MFCs in the future. By switching to the biopolymer flexible electrode that rivals its traditional polymers, the MFCs demonstrate promise to be a vital tool accessible in the remotest part of the world lacking electrical infrastructure for simultaneous clean water and electricity generation. Revascularization of the pulp tissue is one of the fundamental processes and challenges in regenerative endodontic procedures (REPs). In this regard, the current study is aimed at synthesizing the mineral trioxide aggregate- (MTA-) based scaffolds as a biomaterial for REPs. Poly (ε-caprolactone) (PCL)/chitosan (CS)/MTA scaffolds were constructed and evaluated by FTIR, SEM, XRD, and TGA analyses. Proliferation and adhesion of human dental pulp stem cells (hDPSCs) were assessed on these scaffolds by scanning electron microscopy (SEM) and MTT assays, respectively. The expression of angiogenic markers was investigated in gene and protein levels by real-time PCR and western blotting tests. Our results indicated that the obtained appropriate physicochemical characteristics of scaffolds could be suitable for REPs. The adhesion and proliferation level of hDPSCs were significantly increased after seeding on PCL/CS/MTA scaffolds. The expression levels of VEGFR-2, Tie2, and Angiopoietin-1 genes were statistically increased on the PCL/CS/MTA scaffold. In support of these findings, western blotting results showed the upregulation of these markers at protein levels in PCL/CS/MTA scaffold (P < 0.05). The current study results suggested that PCL/CS/MTA scaffolds provide appropriate structures for the adhesion and proliferation of hDPSCs besides induction of the angiogenesis process in these cells. Melioidosis is a serious infectious disease caused by the environmental Gram-negative bacillus Burkholderia pseudomallei. It has been shown that the host immune system, mainly comprising various types of immune cells, fights against the disease. The present study was to specify correlation between septicemic melioidosis and the levels of multiple immune cells. First, the genes with differential expression patterns between patients with septicemic melioidosis (B. pseudomallei) and health donors (control/healthy) were identified. These genes being related to cytokine binding, cell adhesion molecule binding, and MHC relevant proteins may influence immune response. The Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis revealed 23 enriched immune response pathways. We further leveraged the microarray data to investigate the relationship between immune response and septicemic melioidosis, using the CIBERSORT analysis. Comparison of the percentages of 22 immune cell types in B. pseudomallei vs. control/healthy revealed that those of CD4 memory resting cells, CD8+ T cells, B memory cells, and CD4 memory activated cells were low, whereas those of M0 macrophages, neutrophils, and gamma delta T cells were high. The multivariate logistic regression analysis further revealed that CD8+ T cells, M0 macrophages, neutrophils, and naive CD4+ cells were strongly associated with the onset of septicemic melioidosis, and M2 macrophages and neutrophils were associated with the survival in septicemic melioidosis. Objectives: Both thymic stromal lymphopoietin (TSLP) and dendritic cells (DCs) are involved in many autoimmune diseases, but the potential roles of TSLP and DCs in bullous pemphigoid (BP) have not been clarified. We sought to explore the contributions of TSLP and DCs in patients with BP.Methods: TSLP levels in sera and blister fluids were measured by enzyme-linked immunosorbent assay. The TSLP expression in the BP lesional skin was detected by immunohistochemical staining. Infiltration of DCs, marked by DC-specific intercellular adhesion molecule-3-grabbing nonintegrin (DC-SIGN), and its relationship with TSLP and TSLP receptors was evaluated by immunofluorescence staining.Results: We found that TSLP levels in sera and in blister fluids of patients with BP were higher compared to the control groups. In patients with BP, TSLP levels in sera correlated with TSLP levels in blisters. The expression of TSLP in the BP lesional skin was higher compared to the healthy controls' skin. Greater numbers of TSLP-positive cells were observed in the epidermis of patients with BP compared to the healthy controls. Greater numbers of DC-SIGN-positive cells were present in the BP lesional skin compared to the skin of controls. The expression of TSLP was highly upregulated in DC-SIGN-positive cells, and most DC-SIGN-positive cells expressed TSLP receptors.Conclusions: We conclude that TSLP may activate DC-SIGN-positive DCs directly, which may be involved in the pathogenesis of BP. The use of biomaterials capable of achieving controlled release of drugs or chemical and physical interactions with living organisms causes these new materials to be studied in depth to better target their future applications. An example of this is the use of materials that have in their physical structure elements with chelating effects, capable of interacting with the ions and cations present in the cell culture media, drugs, and other elements capable of interacting with the human body. In this work, cellulose acetate hydrogel (HAC) crosslinked with EDTAD (ethylenediaminetetraacetic acid dianhydride) showed a significant chelating effect capable of altering the adhesion of mesenchymal cells in the first days of in vitro tests. This result became our main question in this work, and by using new cell viability assays, it was verified that the hydrogel interacted with the culture medium, removed the salts present, such as Ca2+ and Mg2+, and promoted a decrease in the amount of cells adhered to the material but not cell death. After saturation of Ca2+ and Mg2+, we obtained an improvement in cell adhesion. Electrochemical impedance spectroscopy (EIS) results also verified the dynamics of linked or mobile loads in the volume and interface regions of HAC-EDTA and the presence of characteristic bands related to the interaction between calcium and magnesium present in the culture medium with HAC-EDTA. The DMA traction test showed that there was no dynamic-mechanical change in the structure of the material at temperatures between 20 and 60°C, which is ideal for this studied biomaterial. The purpose of these tests is to aid in the characterization of future biomaterials, where the physicochemical interaction of some of them is often confused with a false cytotoxicity, which in fact may only be an easily identifiable and adaptable structural feature. Titanium (Ti) is widely used as a biomaterial for endosseous implants due to its relatively inert surface oxide layer that enables implanted devices the ability of assembling tissue reparative components that culminate in osseointegration. Topographic modifications in the form of micro- and nanoscaled structures significantly promote osseointegration and enhance the osteogenic differentiation of adult mesenchymal stromal cells (MSCs). While the biological mechanisms central to the differential responses of tissues and cells to Ti surface modifications remain unknown, adhesion and morphological adaptation are amongst the earliest events at the cell-biomaterial interface that are highly influenced by surface topography and profoundly impact the regulation of stem cell fate determination. This study correlated the effects of Ti topographic modifications on adhesion and morphological adaptation of human MSCs with phenotypic change. The results showed that modified Ti topographies precluded the adhesion of a subset of MSCs while incurring distinct morphological constraints on adherent cells. These effects anomalously corresponded with a differential expression of stem cell pluripotency and Wnt signalling-associated markers on both modified surfaces while additionally differing between hydrophobic and hydrophilic surface modifications—though extent of osteogenic differentiation induced by both modified topographies yielded similarly significant higher levels of cellular mineralisation in contrast to polished Ti. These results suggest that in the absence of deposited proteins and soluble factors, both modified topographies incur the selective adhesion of a subpopulation of progenitors with relatively higher cytoskeletal plasticity. While the presence of deposited proteins and soluble factors does not significantly affect adherence of cells, nanotopographic modifications enhance expression of pluripotency markers in proliferative conditions, which are conversely overridden by both modified topographies in osteogenic inductive conditions. Further deciphering the mechanisms underlying cellular selectivity and Ti topographic responsiveness will improve our understanding of stem cell heterogeneity and advance the potential of MSCs in regenerative medicine. We have recently demonstrated that a small molecular weight amino-terminal peptide of L-plastin (10 amino acids; "MARGSVSDEE") suppressed the phosphorylation of endogenous L-plastin. Therefore, the formation of nascent sealing zones (NSZs) and bone resorption are reduced. The aim of this study was to develop a biodegradable and biocompatible PLGA nanocarrier that could be loaded with the L-plastin peptide of interest and determine the efficacy in vitro in osteoclast cultures. L-plastin MARGSVSDEE (P1) and scrambled control (P3) peptide-loaded PLGA-PEG nanoparticles (NP1 and NP3, respectively) were synthesized by double emulsion technique. The biological effect of nanoparticles on osteoclasts was evaluated by immunoprecipitation, immunoblotting, rhodamine-phalloidin staining of actin filaments, and pit forming assays. Physical characterization of well-dispersed NP1 and NP3 demonstrated ~130-150 nm size, < 0.07 polydispersity index, ~-3 mV ζ-potential, and a sustained release of the peptide for three weeks. Biological characterization in osteoclast cultures demonstrated the following: NP1 significantly reduced (a) endogenous L-plastin phosphorylation; (b) formation of NSZs and sealing rings; (c) resorption. However, the assembly of podosomes which are critical for cell adhesion was not affected. L-plastin peptide-loaded PLGA-PEG nanocarriers have promising potential for the treatment of diseases associated with bone loss. Future studies will use this sustained release of peptide strategy to systematically suppress osteoclast bone resorption activity in vivo in mouse models demonstrating bone loss. Background. Middle turbinate (MT) lateralisation with adhesion formation (MiTLAF) is a common complication following endoscopic sinus surgery, frequently resulting in surgical failure, persistence of preoperative symptoms, and delayed secondary complications. Packing materials, splints, or spacers reduce the risk of MiTLAF but often result in postoperative nasal obstruction and discomfort, along with reduced access for irrigation. Temporary suture medialisation of the MTs reduces the risk of MiTLAF and prevents the problems encountered with packing, splints, or spacers. However, the techniques described in the literature are technically challenging and often ineffectual. Methods. We describe a method of suture placement that provides a secure temporary MT medialisation, without the technical challenges of traditional techniques, using a 4-0 Monocryl (Poliglecaprone 25, Ethicon, Somerville, NJ, USA) suture on a 19 mm precision point reverse cutting PS-2 curved needle. We review 25 consecutive patients undergoing sinonasal procedures with our new technique and assess for MiTLAF. Results. In our cohort, only one patient experienced MiTLAF which was not clinically significant. Conclusions. A variety of genetic approaches, including twin studies, linkage studies, and candidate gene studies, has established a firm genetic basis for addiction. However, there has been difficulty identifying the precise genes that underlie addiction liability using these approaches. This situation became especially clear in genome-wide association studies (GWAS) of addiction. Moreover, the results of GWAS brought into clarity many of the shortcomings of those early genetic approaches. GWAS studies stripped away those preconceived notions, examining genes that would not previously have been considered in the study of addiction, consequently creating a shift in our understanding. Most importantly, those studies implicated a class of genes that had not previously been considered in the study of addiction genetics: cell adhesion molecules (CAMs). Considering the well-documented evidence supporting a role for various CAMs in synaptic plasticity, axonal growth, and regeneration, it is not surprising that allelic variation in CAM genes might also play a role in addiction liability. This review focuses on the role of various cell adhesion molecules in neuroplasticity that might contribute to addictive processes and emphasizes the importance of ongoing research on CAM genes that have been implicated in addiction by GWAS. The importance of the bone marrow microenvironment forming the so-called niche in physiologic hemopoiesis is largely known, and recent evidences support the presence of stromal alterations from the molecular to the cytoarchitectural level in hematologic malignancies. Various alterations in cell adhesion, metabolism, cytokine signaling, autophagy, and methylation patterns of tumor-derived mesenchymal stem cells have been demonstrated, contributing to the genesis of a leukemic permissive niche. This niche allows both the ineffective haematopoiesis typical of myelodysplastic syndromes and the differentiation arrest, proliferation advantage, and clone selection which is the hallmark of acute myeloid leukemia. After peripheral nerve injury, immune/inflammatory responses are triggered, which are critical for nerve regeneration. Despite their importance, the underlying molecular changes in immune/inflammatory responses remain largely unknown. In this study, we systematically analyzed differentially expressed genes in immune/inflammatory-related pathways at high temporal resolution and experimentally validated gene expression changes with RT-PCR following sciatic nerve crush in rats. We found that immune/inflammatory reactions not only occur in the acute injury but also remained activated over two weeks after injury. Detailed bioinformatic studies suggested that multiple immune/inflammatory pathways, including agranulocyte adhesion and diapedesis, granulocyte adhesion and diapedesis, IL-6 signaling, and IL-10 signaling, were sustained activated during nerve degeneration and regeneration. Our current study expands our understanding of the molecular basis of altered immune/inflammatory-related pathways following injury and thus might offer the possibility of targeting related molecules as therapeutic intervention for peripheral nerve regeneration.
