The threat of microplastics to marine animals and habitats is increasing, which may affect sea turtle nesting grounds. The Qilianyu Islands are the largest remaining green turtle (Chelonia mydas) nesting grounds in China. Despite being far from the mainland, microplastic pollution cannot be ignored. In this study, the level of microplastic pollution in surface sediments from three different zones, namely, the bottom, intertidal, and supratidal zone, was investigated on North Island, Qilianyu Islands. The results showed that the abundance of microplastics in the supratidal zone was significantly higher than that in the bottom zone and intertidal zone (r = 3.65, p = 0.011), with the highest average abundance of microplastics located on the southwest coast of North Island. In the bottom zone, only plastic blocks (88%) and fibers (12%) were found. The main types of microplastics in the intertidal and supratidal zones were plastic blocks (48%) and foam (42%), with polyethylene (PE) (40%) and polystyrene (PS) (34%) being the predominant components. These types and components of microplastics differed from those in the surrounding seawater, but corresponding types and components were found in the plastic debris on the beach. Meanwhile, it was also observed that there were multiple instances of fragmented plastic on the beach. Thus, we suggest that the microplastics on the beach in North Island were mainly derived from the fragmentation of microplastic debris, indicating secondary microplastics. It is recommended to further strengthen the regular cleaning of plastic debris on the beach, especially the removal of small plastic debris, in order to reduce the pollution from secondary microplastics generated by the fragmentation of beach plastic debris and to better protect China's most important sea turtle nesting site in the South China Sea. The purpose of the present study was to investigate the associations of the objectively assessed total sedentary behavior (SB) amount and SB patterns with phase angle (PhA) in older populations and to conduct a comparison analysis for those who are physically active (meet 150 min/week) and inactive (less than 150 min/week). During May to August 2023, a convenience sampling was used to recruit older adults (aged ≥ 65 years) living in a community in Taiwan. The total SB amount (minutes/day), SB patterns (including SB bouts and breaks), and physical activity were assessed by a triaxial accelerometer. A multifrequency bioelectrical impedance analyzer (BIA) was used to measure PhA. Multiple linear regression analysis was employed to examine the associations between SB and PhA in the total number of participants and stratified in the physically active and inactive groups. For the whole sample (n = 166; mean age: 72.1 ± 5.5 years), the total SB amount and patterns of SB were not associated with PhA. For those physically active, the total SB amount and SB patterns were not associated with PhA. Among those physically inactive, the total SB amount was negatively related to PhA (B: −0.059, 95% CI: −0.118, 0.000). This study underscores the importance of minimizing the total SB amount in physically inactive older adults, providing evidence for future interventions targeting SB and PhA in this population. Fusarium wilt is a severe and worldwide disease in potato cultivation. In this study, Fusarium foetens was first identified as the pathogen of potato wilt. Bacillus subtilis SF1 has the potential for controlling potato wilt induced by F. foetens, resulting in a mycelium growth inhibition of 52.50 ± 2.59% in vitro and a significant decrease in incidence rate by 45.56% in vivo. This research highlighted the antifungal activity of surfactin from B. subtilis SF1 and attempted to reveal the unknown antifungal mechanisms. Surfactin inhibited F. foetens mycelium growth beyond the concentration of 20 µg/µL. Surfactin-treated mycelium appeared to have morphological malformation. Surfactin enhanced reduced glutathione production and caused the increase in values of the extracellular fluids in OD260 and OD280. Surfactin induced differential protein expression and changed the genes' transcription levels. Surfactin binds to fungal DNA via groove-binding mode, with a binding constant of Kb 2.97 x 104 M-1. Moreover, B. subtilis SF1 harbored genes encoding plant-promoting determinants, making potato seedlings grow vigorously. The results will help provide a comprehensive understanding of the mechanisms of surfactin against filamentous fungi and the application of surfactin-producing microbial in the biocontrol of plant pathogenic fungi. The typical two-dimensional layered structure materials, MXenes, are widely used in energy conversion and storage due to their high conductivity, ion transport ability, and rich surface structures. Recently, MXenes and their composites have been widely employed in secondary batteries, especially sodium-ion batteries (SIBs), with obvious performance improvement. As anodic materials, MXenes, metal oxides, metal sulfides, and other materials contain certain advantages in Na+ storage, but they individually also suffer from some issues and challenges, such as low conductivity and serious volume change, as well as the associated low capacity and poor cyclability. By virtue of the advantages of MXenes, with their high conductivity and ultrathin two-dimensional structures, the construction of surface-functionalized MXenes and MXene-based composites could effectively improve the conductivity and mass-transport properties of composites, alleviate volume expansion, and, thus, enhance the capacity properties, rate performances, and cycle stability of SIBs. Herein, we review the latest research status of the structural design of MXenes and Mxene-based materials, as well as their applications in SIBs. We briefly introduce the research background and introduce MXenes and SIBs, and focus on their structural designs and corresponding applications in SIBs. Finally, the important challenges of MXene-based materials applied to SIBs are discussed, and the future prospects of MXene-based composite developments in SIBs are presented. [ABSTRACT FROM AUTHOR]

Due to the advantages of rich resources, low cost, high energy conversion efficiency, long cycle life, and low maintenance fee, sodium–ion batteries have been regarded as a promising energy storage technology. However, their relatively low energy density compared with the commercialized lithium–ion batteries still impedes their application for power systems. Anode–free rechargeable sodium–metal batteries (AFSMBs) pose a solution to boost energy density and tackle the safety problems of metal batteries. At present, researchers still lack a comprehensive understanding of the anode-free cells in terms of electrolytes, solid–electrolyte interphase (SEI), and current collectors. This review is devoted to the field of AFSMBs, and outlines the breakthroughs that have been accomplished along with our perspective on the direction of future development for AFSMBs and the areas that warrant further investigation. Objective: A comparative analysis was performed to investigate the potential risk factors of Adverse Events Following Immunization (AEFI) after receiving different booster vaccines. Methods: From 18 January 2021 to 21 January 2022, the Health Care Workers (HCWs) of Guizhou Provincial Staff Hospital (Guizhou Province, China) who received a third Booster vaccine, that was either homologous (i.e., (i) a total of three doses of Vero cell vaccine or (ii) three doses of CHO cell vaccine) or (iii) heterologous with two first doses of Vero cell vaccine, being either CHO cell vaccine or adenovirus type-5 (Ad5) vectored COVID-19 vaccine, were asked to complete a self-report questionnaire form to provide information on any AEFI that may have occurred in the first 3 days after vaccination with the booster. The frequency of AEFI corresponding to the three different booster vaccines was compared, and the risk factors for predicting AEFI were determined by multivariate logistic regression analysis. Results: Of the 904 HCWs who completed the survey, 792 met the inclusion criteria. The rates of AEFI were 9.8% (62/635) in the homologous Vero cell booster group, 17.3% (13/75) in the homologous CHO cell booster group, and 20.7% (17/82) in the heterologous mixed vaccines booster group, and the rates were significantly different (c2 = 11.5, p = 0.004) between the three groups of vaccines. Multivariate logistic regression analysis showed that: (1) compared to the homologous Vero cell booster group, the risk of AEFI was about 2.1 times higher (OR = 2.095, 95% CI: 1.056-4.157, p = 0.034) in the CHO cell booster group and 2.5 times higher (OR = 2.476, 95% CI: 1.352-4.533, p = 0.003) in the mixed vaccines group; (2) the odds for women experiencing AEFI were about 2.8 times higher (OR = 2.792, 95% CI: 1.407-5.543, p = 0.003) than men; and (3) compared to the non-frontline HCWs, the risk of AEFI was about 2.6 times higher (OR = 2.648, 95% CI: 1.473-4.760, p = 0.001) in the doctors. Conclusion: The AEFI in all three booster groups are acceptable, and serious adverse events are rare. The risk of AEFI was higher in doctors, which may be related to the high stress during the COVID-19 epidemic. Uncoordinated carbon-nitrogen ratio in raw materials will lead to excessive contents of higher alcohols in alcoholic beverages. The effect of GAT1 gene, the GATA transcription activator, on higher alcohol biosynthesis was investigated to clarify the mechanism of Saccharomyces cerevisiae regulating higher alcohol metabolism under high concentrations of free amino nitrogen (FAN). The availability of FAN by strain SDT1K with a GAT1 double-copy deletion was 28.31% lower than that of parent strain S17, and the yield of higher alcohols was 33.91% lower. The transcript levels of the downstream target genes of GAT1 and higher alcohol production in the double-copy deletion mutant suggested that a part of the effect of GAT1 deletion on higher alcohol production was the downregulation of GAP1, ARO9, and ARO10. This study shows that GATA factors can effectively regulate the metabolism of higher alcohols in S. cerevisiae and provides valuable insights into higher alcohol biosynthesis, showing great significance for the wheat beer industry. Consumption of a high-fat diet (HFD) promotes the development of obesity, a disease resulting from an imbalance between energy intake and energy expenditure. Brown adipose tissue (BAT) has thermogenic capacity that burns calories to produce heat, and it is a potential target for the treatment and prevention of obesity. There is limited information regarding the impact of HFD on the BAT transcriptome. We hypothesized that HFD-induced obesity would lead to transcriptional regulation of BAT genes. RNA sequencing was used to generate global transcriptome profiles from BAT of lean mice fed with a low-fat diet (LFD) and obese mice fed with a HFD. Gene Ontology (GO) analysis identified increased expression of genes involved in biological processes (BP) related to immune responses, which enhanced molecular function (MF) in chemokine activity; decreased expression of genes involved in BP related to ion transport and muscle structure development, which reduced MF in channel and transporter activity and structural binding. Kyoto Encyclopedia of Genes and Genomes (KEGG) functional pathway analysis indicated that pathways associated with innate immunity were enhanced by HFD, while pathways associated with muscle contraction and calcium signaling were suppressed by HFD. Collectively, these results suggest that diet-induced obesity changes transcriptomic signatures of BAT, leading to dysfunction involving inflammation, calcium signaling, ion transport, and cell structural development. Compared to traditional DNA markers, genome-scale datasets can provide mass information to effectively address historically difficult phylogenies. Primula is the largest genus in the family Primulaceae, with members distributed mainly throughout temperate and arctic areas of the Northern Hemisphere. The phylogenetic relationships among Primula taxa still maintain unresolved, mainly due to intra- and interspecific morphological variation, which was caused by frequent hybridization and introgression. In this study, we sequenced and assembled four complete plastid genomes (Primula handeliana, Primula woodwardii, Primula knuthiana, and Androsace laxa) by Illumina paired-end sequencing. A total of 10 Primula species (including 7 published plastid genomes) were analyzed to investigate the plastid genome sequence divergence and their inferences for the phylogeny of Primula. The 10 Primula plastid genomes were similar in terms of their gene content and order, GC content, and codon usage, but slightly different in the number of the repeat. Moderate sequence divergence was observed among Primula plastid genomes. Phylogenetic analysis strongly supported that Primula was monophyletic and more closely related to Androsace in the Primulaceae family. The phylogenetic relationships among the 10 Primula species showed that the placement of P. knuthiana-P. veris clade was uncertain in the phylogenetic tree. Magnetic molecularly imprinted polymers (MMIPs), combination of outstanding magnetism with specific selective binding capability for target molecules, have proven to be attractive in separation science and bio-applications. Herein, we proposed the core-shell magnetic molecularly imprinted polymers for food analysis, employing the Fe3O4 particles prepared by co-precipitation protocol as the magnetic core and MMIP film onto the silica layer as the recognition and adsorption of target analytes. The obtained MMIPs materials have been fully characterized by scanning electron microscope (SEM), Fourier transform infrared spectrometer (FT-IR), vibrating sample magnetometer (VSM), and re-binding experiments. Under the optimal conditions, the fabricated Fe3O4@MIPs demonstrated fast adsorption equilibrium, a highly improved imprinting capacity, and excellent specificity to target sterigmatocystin (ST), which have been successfully applied as highly efficient solid-phase extraction materials followed by high-performance liquid chromatography (HPLC) analysis. The MMIP-based solid phase extraction (SPE) method gave linear response in the range of 0.05-5.0 mg⋅L-1 with a detection limit of 9.1 µg⋅L-1. Finally, the proposed method was used for the selective isolation and enrichment of ST in food samples with recoveries in the range 80.6-88.7% and the relative standard deviation (RSD) <5.6%. The situation of stable and slightly advancing glaciers in the Karakoram is called the “Karakoram anomaly”. Glacier surface velocity is one of the key parameters of glacier dynamics and mass balance, however, the response of glacier motion to this regional anomaly is not fully understood. Here, we characterize the spatial-temporal variations in glacier velocity over the Central Karakoram from 1999–2003. The inter-annual glacier velocity fields were retrieved using a cross-correlation-based algorithm applied to four Landsat-7 Enhanced Thematic Mapper Plus (ETM+) panchromatic image pairs. We find that most of the glaciers on the southern slope flowed faster than those on the northern slope, which might be attributed to the differences in glacier sizes. Furthermore, ice motion observations over four years reveal that most of the glaciers were quasi-stable or experienced small fluctuations of flow velocity during our study period. We identify a new surging event for the South Skamri Glacier in the study period by investigating the glacier frontal changes and the longer-term time series of surface velocities between 1996 and 2006. From the transverse velocity profiles of seven typical glaciers, we infer that basal sliding is the predominant motion mechanism of the middle and upper glaciers, whereas internal deformation dominates closest to the glacier terminus. To accurately estimate leaf area index (LAI) in mangrove areas, the selection of appropriate models and predictor variables is critical. However, there is a major challenge in quantifying and mapping LAI using multi-spectral sensors due to the saturation effects of traditional vegetation indices (VIs) for mangrove forests. World View-2 (WV2) imagery has proven to be effective to estimate LAI of grasslands and forests, but the sensitivity of its vegetation indices (VIs) has been uncertain for mangrove forests. Furthermore, the single model may exhibit certain randomness and instability in model calibration and estimation accuracy. Therefore, this study aims to explore the sensitivity of WV2 VIs for estimating mangrove LAI by comparing artificial neural network regression (ANNR), support vector regression (SVR) and random forest regression (RFR). The results suggest that the RFR algorithm yields the best results (RMSE = 0.45, 14.55% of the average LAI), followed by ANNR (RMSE = 0.49, 16.04% of the average LAI), and then SVR (RMSE = 0.51, 16.56% of the average LAI) algorithms using 5-fold cross validation (CV) using all VIs. Quantification of the variable importance shows that the VIs derived from the red-edge band consistently remain the most important contributor to LAI estimation. When the red-edge band-derived VIs are removed from the models, estimation accuracies measured in relative RMSE (RMSEr) decrease by 3.79%, 2.70% and 4.47% for ANNR, SVR and RFR models respectively. VIs derived from red-edge band also yield better accuracy compared with other traditional bands of WV2, such as near-infrared-1 and near-infrared-2 band. Furthermore, the estimated LAI values vary significantly across different mangrove species. The study demonstrates the utility of VIs of WV2 imagery and the selected machine-learning algorithms in developing LAI models in mangrove forests. With the rapid development of China's economy, the demand for labor in the coastal cities continues to grow. Due to restrictions imposed by China's household registration system, a large number of floating populations have subsequently appeared. The relationship between floating populations and crime, however, is not well understood. This paper investigates the impact of a floating population on residential burglary on a fine spatial scale. The floating population was divided into the floating population from other provinces (FPFOP) and the floating population from the same province as ZG city (FPFSP), because of the high heterogeneity. Univariate spatial patterns in residential burglary and the floating population in ZG were explored using Moran's I and LISA (local indicators of spatial association) models. Furthermore, a geographically weighted Poisson regression model, which addressed the spatial effects in the data, was employed to explore the relationship between the floating population and residential burglary. The results revealed that the impact of the floating population on residential burglary is complex. The floating population from the same province did not have a significant impact on residential burglary in most parts of the city, while the floating population from other provinces had a significantly positive impact on residential burglary in most of the study areas and the magnitude of this impact varied across the study area. DL0410, containing biphenyl and piperidine skeletons, was identified as an acetylcholinesterase (AChE) and butyrylcholinesterase (BuChE) inhibitor through high-throughput screening assays, and further studies affirmed its efficacy and safety for Alzheimer's disease treatment. In our study, a series of novel DL0410 derivatives were evaluated for inhibitory activities towards AChE and BuChE. Among these derivatives, compounds 6-1 and 7-6 showed stronger AChE and BuChE inhibitory activities than DL0410. Then, pharmacophore modeling and three-dimensional quantitative structure activity relationship (3D-QSAR) models were performed. The R² of AChE and BuChE 3D-QSAR models for training set were found to be 0.925 and 0.883, while that of the test set were 0.850 and 0.881, respectively. Next, molecular docking methods were utilized to explore the putative binding modes. Compounds 6-1 and 7-6 could interact with the amino acid residues in the catalytic anionic site (CAS) and peripheral anionic site (PAS) of AChE/BuChE, which was similar with DL0410. Kinetics studies also suggested that the three compounds were all mixed-types of inhibitors. In addition, compound 6-1 showed better absorption and blood brain barrier permeability. These studies provide better insight into the inhibitory behaviors of DL0410 derivatives, which is beneficial for rational design of AChE and BuChE inhibitors in the future. [ABSTRACT FROM AUTHOR]

In our study, we attempted to investigate the influences of P-glycoprotein (P-gp) on DL0410, a novel synthetic molecule for Alzheimer's disease (AD) treatment, for intestinal absorption and blood-brain barrier permeability in vitro and related binding mechanisms in silico. Caco-2, MDCK, and MDCK-MDR1 cells were utilized for transport studies, and homology modelling of human P-gp was built for further docking study to uncover the binding mode of DL0410. The results showed that the apparent permeability (Papp) value of DL0410 was approximately 1 × 10-6 cm/s, indicating the low permeability of DL0410. With the presence of verapamil, the directional transport of DL0410 disappeared in Caco-2 and MDCK-MDR1 cells, suggesting that DL0410 should be a substrate of P-gp, which was also confirmed by P-gp ATPase assay. In addition, DL0410 could competitively inhibit the transport of Rho123, a P-gp known substrate. According to molecular docking, we also found that DL0410 could bind to the drug binding pocket (DBP), but not the nucleotide binding domain (NBD). In conclusion, DL0410 was a substrate as well as a competitive inhibitor of P-gp, and P-gp had a remarkable impact on the intestine and brain permeability of DL0410, which is of significance for drug research and development. Automatic extraction of power lines using airborne LiDAR (Light Detection and Ranging) data has been one of the most important topics for electric power management. However, this is very challenging over complex urban areas, where power lines are in close proximity to buildings and trees. In this paper, we presented a new, semi-automated and versatile framework that consists of four steps: (i) power line candidate point filtering, (ii) local neighborhood selection, (iii) spatial structural feature extraction, and (iv) SVM classification. We introduced the power line corridor direction for candidate point filtering and multi-scale slant cylindrical neighborhood for spatial structural features extraction. In a detailed evaluation involving seven scales and four types for local neighborhood selection, 26 structural features, and two datasets, we demonstrated that the use of multi-scale slant cylindrical neighborhood for individual 3D points significantly improved the power line classification. The experiments indicated that precision, recall and quality rate of power line classification is more than 98%, 98% and 97%, respectively. The storm hyetograph is critical in drainage design since it determines the peak flooding volume in a catchment and the corresponding drainage capacity demand for a return period. This study firstly compares the common design storms such as the Chicago, Huff, and Triangular curves employed to represent the storm hyetographs in the metropolitan area of Guangzhou using minute-interval rainfall data during 2008-2012. These common design storms cannot satisfactorily represent the storm hyetographs in sub-tropic areas of Guangzhou. The normalized time of peak rainfall is at 33 ± 5% for all storms in the Tianhe and Panyu districts, and most storms (84%) are in the 1st and 2nd quartiles. The Huff curves are further improved by separately describing the rising and falling limbs instead of classifying all storms into four quartiles. The optimal time intervals are 1-5 min for deriving a practical urban design storm, especially for short-duration and intense storms in Guangzhou. Compared to the 71 observed storm hyetographs, the Improved Huff curves have smaller RMSE and higher NSE values (6.43, 0.66) than those of the original Huff (6.62, 0.63), Triangular (7.38, 0.55), and Chicago (7.57, 0.54) curves. The mean relative difference of peak flooding volume simulated with SWMM using the Improved Huff curve as the input is only 2%, -6%, and 8% of those simulated by observed rainfall at the three catchments, respectively. In contrast, those simulated by the original Huff (-12%, -43%, -16%), Triangular (-22%, -62%, -38%), and Chicago curves (-17%, -19%, -21%) are much smaller and greatly underestimate the peak flooding volume. The relationship between burglary and socio-demographic factors has long been a hot topic in crime research. Spatial dependence and spatial heterogeneity are two issues to be addressed in modeling geographic data. When these two issues arise at the same time, it is difficult to model them simultaneously. A cross-comparison of three models is presented in this study to identify which spatial effect should be addressed first in crime analysis. The negative binominal model (NB), Bayesian hierarchical model (BHM) and the geographically weighted Poisson regression model (GWPR) were implemented based on a three-year residential burglary data set from ZG, China. The modeling result shows that both BHM and GWPR outperform NB as they capture either of the spatial effects. Compared to the NB model, the mean absolute deviation (MAD) of BHM and GWPR was decreased by 83.71% and 49.39%, the mean squared error (MSE) of BHM and GWPR was decreased by 97.88% and 77.15% and the R2d of BHM and GWPR was improved by 26.7% and 19.1%, respectively. In comparison with BHM and GWPR, BHM fits the data better with lower MAD, MSE and higher R2d. The empirical analysis indicates that the percentage of renter population, percentage of people from other provinces, bus line density and bus stop density have a significantly positive impact on the number of residential burglaries. Uveitis, defined as inflammation of the uveal tract, may cause blindness in both young and middle-aged people. Approximately 10–15% of blindness in the West is caused by uveitis. Therefore, a comprehensive investigation to determine the disease pathogenesis is urgent, as it will thus be possible to design effective treatments. Identification of the disease genes that cause uveitis is an important requirement to achieve this goal. To begin to answer this question, in this study, a computational method was proposed to identify novel uveitis-related genes. This method was executed on a large protein–protein interaction network and employed a popular ranking algorithm, the RandomWalk with Restart (RWR) algorithm. To improve the utility of the method, a permutation test and a procedure for selecting core genes were added, which helped to exclude false discoveries and select the most important candidate genes. The five-fold cross-validation was adopted to evaluate the method, yielding the average F1-measure of 0.189. In addition, we compared our method with a classic GBA-based method to further indicate its utility. Based on our method, 56 putative genes were chosen for further assessment. We have determined that several of these genes (e.g., CCL4, Jun, and MMP9) are likely to be important for the pathogenesis of uveitis. [ABSTRACT FROM AUTHOR]

To elucidate the chemical compositions of the aerial parts of Chrysanthemum coronarium L., the ethanol extracts of Ch. coronarium L. were firstly isolated by the MCI-gel resin column. The caffeoylquinic acid-rich fractions were further purified by various chromatographic columns including silica gel, Sephadex LH-20, and semi-preparative HPLC to yield the compounds. The purified compounds were characterized by 1H-Nuclear Magnetic Resonance (1H-NMR), 13C-NMR, and high resolution electrospray ionisation mass spectral (HR-ESI-MS) spectroscopy. Seven caffeoylquinic acid (CQA) compounds were isolated from this plant. Their structures were clarified by spectrometric methods and identified as 3-O-caffeoylquinic acid (1), 5-O-caffeoylquinic acid (2), 4-O-caffeoylquinic acid (3), 3,4-di-O-caffeoylquinic acid (4), 1,5-di-O-caffeoylquinic acid (5), 3,5-di-O-caffeoylquinic acid (6), and 4,5-di-O-caffeoylquinic acid (7). Caffeoylquinic acids were the major constituents present in the aerial parts of Ch. coronarium L. All of the isolates except for compounds 2 and 6 were reported for the first time from this species. Deficiency of the cholinergic system is thought to play a vital role in cognitive impairment of dementia. DL0410 was discovered as a dual inhibitor of acetylcholinesterase (AChE) and butyrylcholinestease (BuChE), with potent efficiency in in-vitro experiments, but its in vivo effect on the cholinergic model has not been evaluated, and its action mechanism has also not been illustrated. In the present study, the capability of DL0410 in ameliorating the amnesia induced by scopolamine was investigated, and its effect on the cholinergic system in the hippocampus and its binding mode in the active site of AChE was also explored. Mice were administrated DL0410 (3 mg/kg, 10 mg/kg, and 30 mg/kg), and mice treated with donepezil were used as a positive control. The Morris water maze, escape learning task, and passive avoidance task were used as behavioral tests. The test results indicated that DL0410 could significantly improve the learning and memory impairments induced by scopolamine, with 10 mg/kg performing best. Further, DL0410 inhibited the AChE activity and increased acetylcholine (ACh) levels in a dose-dependent manner, and interacted with the active site of AChE in a similar manner as donepezil. However, no difference in the activity of BuChE was found in this study. All of the evidence indicated that its AChE inhibition is an important mechanism in the anti-amnesia effect. Rehmannia is a non-parasitic genus in Orobanchaceae including six species mainly distributed in central and north China. Its phylogenetic position and infrageneric relationships remain uncertain due to potential hybridization and polyploidization. In this study, we sequenced and compared the complete chloroplast genomes of six Rehmannia species using Illumina sequencing technology to elucidate the interspecific variations. Rehmannia plastomes exhibited typical quadripartite and circular structures with good synteny of gene order. The complete genomes ranged from 153,622 bp to 154,055 bp in length, including 133 genes encoding 88 proteins, 37 tRNAs, and 8 rRNAs. Three genes (rpoA, rpoC2, accD) have potentially experienced positive selection. Plastome size variation of Rehmannia was mainly ascribed to the expansion and contraction of the border regions between the inverted repeat (IR) region and the single-copy (SC) regions. Despite of the conserved structure in Rehmannia plastomes, sequence variations provide useful phylogenetic information. Phylogenetic trees of 23 Lamiales species reconstructed with the complete plastomes suggested that Rehmannia was monophyletic and sister to the clade of Lindenbergia and the parasitic taxa in Orobanchaceae. The interspecific relationships within Rehmannia were completely different with the previous studies. Metal nanoparticles (NPs) possess unique physicochemical attributes for creating effective recognition and transduction processes in chem/bio-sensing. In this work, we suggested that citrate-capped Au/Ag NPs could be used as the reporters for the design of hydrogen peroxide (H2O2) sensors with a simple manipulation principle and an easy detection procedure. Specifically, p-benzenediboronic acid (BDBA) induced the aggregation of citrate-capped Au NPs through the cross-linking reaction between citrate and boronic acid of BDBA in solution. By modifying the electrode with a boronic acid derivative, the BDBA-induced assembly of Au NPs was achieved on the electrode surface. This led to a significant decrease in the electron transfer resistance due to the unique conductive ability of Au NPs. However, when the boronate group on the electrode surface was oxidized into its phenol format, the assembly of Au NPs on the electrode surface was not achieved. As a result, a higher electron transfer resistance was observed. The process could be monitored by electrochemical impedance technique. Furthermore, when Ag NPs were used instead of Au NPs in this design, the H2O2 concentration could be determined by measuring the linear-sweep voltammetry (LSV) current through the solid-state Ag/AgCl reaction of Ag NPs. Alzheimer’s disease (AD), the most common form of dementia in adults, is a progressive neurodegenerative disorder of the brain characterized by loss of memory and steady deterioration of cognition. Here, a series of symmetrical molecules containing biphenyl/bibenzyl scaffolds (12–36) were designed, synthesized, and evaluated for their ability to inhibit both acetylcholinesterase (AChE) and butyrylcholinesterase (BuChE). A biological evaluation showed that most of these biphenyl derivatives were potent AChE and BuChE inhibitors. Among them, compound 15 displayed the greatest ability to inhibit BuChE (IC50 = 0.74 µM) and was also a good AChE inhibitor (IC50 = 1.18 µM). Compound 19 was not only a potent AChE inhibitor (IC50 = 0.096 µM), but also a mild BuChE inhibitor (IC50 =1.25 µM). Evaluating the impacts of climatic changes and morphometric features on glacier mass balance is crucial to providing insight into glacier changes and their effects on regional water resources and ecosystems. Here, we presented an evaluation of morphometric effects on the glacier mass balances of the Puruogangri ice field (PIF) on the Tibetan Plateau. A clear spatial variability of glacier mass balances, ranging from -0.035 to +0.019 m·w.e.·year-1, was estimated by comparing the TanDEM-X DEM (2012) with the SRTM-X DEM (2000). In general, the observed glacier mass changes were consistent with our fieldwork investigations. Furthermore, by applying the method of linear regression analysis, we found that the mass changes of individual glaciers on the PIF were mainly dominated by the mean altitude (R = 0.84, p < 0.001), however, they were statistically independent of glacier size, aspect, and surface velocity. Chili pepper is used as a food, seasoning and has been revered for its medicinal and health claims. It is very popular and is the most common spice worldwide. Capsaicin (CAP) is a major pungent and bioactive phytochemical in chili peppers. CAP has been shown to improve mitochondrial biogenesis and adenosine triphosphate (ATP) production. However, there is limited evidence around the effects of CAP on physical fatigue and exercise performance. The purpose of this study was to evaluate the potential beneficial effects of CAP on anti-fatigue and ergogenic functions following physiological challenge. Female Institute of Cancer Research (ICR) mice from four groups (n = 8 per group) were orally administered CAP for 4 weeks at 0, 205, 410, and 1025 mg/kg/day, which were respectively designated the vehicle, CAP-1X, CAP-2X, and CAP-5X groups. The anti-fatigue activity and exercise performance was evaluated using forelimb grip strength, exhaustive swimming time, and levels of serum lactate, ammonia, glucose, BUN (blood urea nitrogen) and creatine kinase (CK) after a 15-min swimming exercise. The grip strength and exhaustive swimming time of the CAP-5X group were significantly higher than other groups. CAP supplementation dose-dependently reduced serum lactate, ammonia, BUN and CK levels, and increased glucose concentration after the 15-min swimming test. In addition, CAP also increased hepatic glycogen content, an important energy source for exercise. The possible mechanism was relevant to energy homeostasis and the physiological modulations by CAP supplementation. Human chorionic gonadotropin (hCG) has been regarded as a biomarker for the diagnosis of pregnancy and some cancers. Because the currently used methods (e.g., disposable Point of Care Testing (POCT) device) for hCG detection require the use of many less stable antibodies, simple and cost-effective methods for the sensitive and selective detection of hCG have always been desired. In this work, we have developed a graphene oxide (GO)-based fluorescent platform for the detection of hCG using a fluorescein isothiocyanate (FITC)-labeled hCG-specific binding peptide aptamer (denoted as FITC-PPLRINRHILTR) as the probe, which can be manufactured cheaply and consistently. Specifically, FITC-PPLRINRHILTR adsorbed onto the surface of GO via electrostatic interaction showed a poor fluorescence signal. The specific binding of hCG to FITC-PPLRINRHILTR resulted in the release of the peptide from the GO surface. As a result, an enhanced fluorescence signal was observed. The fluorescence intensity was directly proportional to the hCG concentration in the range of 0.05-20 IU/mL. The detection limit was found to be 20 mIU/mL. In order to improve the corrosion and wear resistance of biomedical Ti-6Al-4V implants, a Ta2N nanoceramic coating was synthesized on a Ti-6Al-4V substrate by the double glow discharge plasma process. The Ta2N coating, composed of fine nanocrystals, with an average grain size of 12.8 nm, improved the surface hardness of Ti-6Al-4V and showed good contact damage tolerance and good adhesion strength to the substrate. The corrosion resistance of the Ta2N coating in Ringer’s physiological solution at 37 °C was evaluated by different electrochemical techniques: potentiodynamic polarization, electrochemical impedance spectroscopy (EIS), potentiostatic polarization and capacitance measurements (Mott-Schottky approach). The evolution of the surface composition of the passive films at different applied potentials was determined by X-ray photoelectron spectroscopy (XPS). The results indicated that the Ta2N coating showed higher corrosion resistance than both commercially pure Ta and uncoated Ti-6Al-4V in this solution, because of the formed oxide film on the Ta2N coating having a smaller carrier density (Nd) and diffusivity (Do) of point defects. The composition of the surface passive film formed on the Ta2N coating changed with the applied potential. At low applied potentials, the oxidation of the Ta2N coating led to the formation of tantalum oxynitride (TaOxNy) but, subsequently, the tantalum oxynitride (TaOxNy) could be chemically converted to Ta2O5 at higher applied potentials. The nontoxic heat-labile toxin (LT) B subunit (LTB) was used as mucosal adjuvant experimentally. However, the mechanism of LTB adjuvant was still unclear. The LTB and enterovirus 71 (EV71) VP1 subunit (EVP1) were constructed in pET32 and expressed in E. coli BL21, respectively. The immunogenicity of purified EVP1 and the adjuvanticity of LTB were evaluated via intranasal immunization EVP1 plus LTB in Balb/c mice. In order to elucidate the proteome change triggered by the adjuvant of LTB, the proteomic profiles of LTB, EVP1, and LTB plus EVP1 were quantitatively analyzed by iTRAQ-LC-MS/MS (isobaric tags for relative and absolute quantitation; liquid chromatography-tandem mass spectrometry) in murine macrophage RAW264.7. The proteomic data were analyzed by bioinformatics and validated by western blot analysis. The predicted protein interactions were confirmed using LTB pull-down and the LTB processing pathway was validated by confocal microscopy. The results showed that LTB significantly boosted EVP1 specific systematic and mucosal antibodies. A total of 3666 differential proteins were identified in the three groups. Pathway enrichment of proteomic data predicted that LTB upregulated the specific and dominant MAPK (mitogen-activated protein kinase) signaling pathway and the protein processing in endoplasmic reticulum (PPER) pathway, whereas LTB or EVP1 did not significantly upregulate these two signaling pathways. Objective: To screen for influenza virus neuraminidase inhibition and to provide a reference for the clinical treatment of influenza using traditional Chinese medicines (TCM). In this study, 421 crude extracts (solubilized with petroleum ether, ethanol, ethyl acetate, and aqueous solvents) were obtained from 113 TCM. The medicine extracts were then reacted with oseltamivir, using 2'-(4-methylumbelliferyl)- -D-N-acetylneuraminic acid (MUNANA) as the substrate, to determine influenza virus neuraminidase activity using a standard fluorimetric assay. It was found that Chinese medicine extracts from Pyrola calliantha, Cynanchum wilfordii, Balanophora involucrata and Paeonia delavayi significantly inhibited neuraminidase activity at a concentration of 40 μg/mL. Dose-dependent inhibitory assays also revealed significant inhibition. The IC50 range of the TCM extracts for influenza virus neuraminidase was approximately 12.66-34.85 μg/mL, respectively. Some Chinese medicines have clear anti-influenza viral effects that may play an important role in the treatment of influenza through the inhibition of viral neuraminidase. The results of this study demonstrated that plant medicines can serve as a useful source of neuraminidase (NA) inhibitors and further investigation into the pharmacologic activities of these extracts is warranted. Recently, sparse representation has yielded successful results in hyperspectral image (HSI) classification. In the sparse representation-based classifiers (SRCs), a more discriminative representation that preserves the spectral-spatial information can be exploited by treating the HSI as a whole entity. Based on this observation, a tensor block-sparsity based representation method is proposed for spectral-spatial classification of HSI in this paper. Unlike traditional vector/matrix-based SRCs, the proposed method consists of tensor block-sparsity based dictionary learning and class-dependent block sparse representation. By naturally regarding the HSI cube as a third-order tensor, small local patches centered at the training samples are extracted from the HSI to maintain the structural information. All the patches are then partitioned into a number of groups, on which a dictionary learning model is constructed with a tensor block-sparsity constraint. A test sample is also expressed as a small local patch and the block sparse representation is then performed in a class-wise manner to take advantage of the class label information. Finally, the category of the test sample is determined by using the minimal residual. The herpes simplex virus thymidine kinase/ganciclovir (HSV TK/GCV) system is one of the best studied cancer suicide gene therapy systems. Our previous study showed that caspase 3 expression was upregulated and bladder tumor growth was significantly reduced in rats treated with a combination of Bifidobacterium (BF) and HSV TK/GCV (BF-rTK/GCV). However, it was raised whether the BF-mediated recombinant thymidine kinase combined with ganciclovir (BF-rTK/GCV) was safe to administer via venous for cancer gene therapy. To answer this question, the antitumor effects of BF-rTK/GCV were mainly evaluated in a xenograft nude mouse model bearing MKN-45 gastric tumor cells. The immune response, including analysis of cytokine profiles, was analyzed to evaluate the safety of intramuscular and intravenous injection of BF-rTK in BALB/c mice. The results suggested that gastric tumor growth was significantly inhibited in vivo by BF-rTK/GCV. However, the BF-rTK/GCV had no effect on mouse body weight, indicating that the treatment was safe for the host. The results of cytokine profile analysis indicated that intravenous injection of a low dose of BF-rTK resulted in a weaker cytokine response than that obtained with intramuscular injection. Furthermore, immunohistochemical analysis showed that intravenous administration did not affect the expression of immune-associated TLR2 and TLR4. Finally, the BF-rTK/GCV inhibited vascular endothelial growth factor (VEGF) expression in mouse model, which is helpful for inhibiting of tumor angiogenesis. Thermokarst is the process of ground subsidence caused by either the thawing of ice-rich permafrost or the melting of massive ground ice. The consequences of permafrost degradation associated with thermokarst for surface ecology, landscape evolution, and hydrological processes have been of great scientific interest and social concern. Part of a tundra patch affected by wildfire in northern Alaska (27.5 km²) was investigated here, using remote sensing and in situ surveys to quantify and understand permafrost thaw dynamics after surface disturbances. A two-pass differential InSAR technique using L-band ALOS-PALSAR has been shown capable of capturing thermokarst subsidence triggered by a tundra fire at a spatial resolution of tens of meters, with supporting evidence from field data and optical satellite images. We have introduced a calibration procedure, comparing burned and unburned areas for InSAR subsidence signals, to remove the noise due to seasonal surface movement. In the first year after the fire, an average subsidence rate of 6.2 cm/year (vertical) was measured. Subsidence in the burned area continued over the following two years, with decreased rates. The mean rate of subsidence observed in our interferograms (from 24 July 2008 to 14 September 2010) was 3.3 cm/year, a value comparable to that estimated from field surveys at two plots on average (2.2 cm/year) for the six years after the fire. These results suggest that this InSAR-measured ground subsidence is caused by the development of thermokarst, a thawing process supported by surface change observations from high-resolution optical images and in situ ground level surveys. A series of novel 2-phenyl-4-trifluoromethyl thiazole-5-carboxamide derivatives have been synthesized and evaluated for their anticancer activity against A-549, Bel7402, and HCT-8 cell lines. Among the tested compounds, highest activity (48%) was achieved with the 4-chloro-2-methylphenyl amido substituted thiazole containing the 2-chlorophenyl group on the two position of the heterocyclic ring. We presented a strategy for fabricating graphene oxide (GO)-based fluorescent biosensors to monitor the change of phosphorylation state and detect phosphatase activity. By regulating the interaction between the negatively charged phosphate group and the positively charged amino residue, we found that GO showed different quenching efficiency toward the phosphorylated and dephosphorylated dye-labeled peptides. To demonstrate the application of our method, alkaline phosphatase (ALP) was tested as a model enzyme with phosphorylated fluorescein isothiocyanate (FITC)-labeled short peptide FITC-Gly-Gly-Gly-Tyr(PO32-)-Arg as the probe. When the negatively charged phosphate group in the Tyr residue was removed from the peptide substrate by enzymatic hydrolysis, the resulting FITC-Gly-Gly-Gly-Tyr-Arg was readily adsorbed onto the GO surface through electrostatic interaction. Asthma is the result of chronic inflammation of the airways which subsequently results in airway hyper-responsiveness and airflow obstruction. It has been shown that an elicited expression of acidic mammalian chitinase (AMCase) may be involved in the pathogenesis of asthma. Our recent study has demonstrated that the specific suppression of elevated AMCase leads to reduced eosinophilia and Th2-mediated immune responses in an ovalbumin (OVA)-sensitized mouse model of allergic asthma. In the current study, we show that the elicited expression of AMCase in the lung tissues of both ovalbumin-and Der P2-induced allergic asthma mouse models. The effects of allergic mediated molecules on AMCase expression were evaluated by utilizing promoter assay in the lung cells. In fact, the exposure of chitin, a polymerized sugar and the fundamental component of the major allergen mite and several of the inflammatory mediators, showed significant enhancement on AMCase expression. The rapid evolution of influenza virus makes antiviral drugs less effective, which is considered to be a major bottleneck in antiviral therapy. The key proteins in the host cells, which are related with the replication cycle of influenza virus, are regarded as potential drug targets due to their distinct advantage of lack of evolution and drug resistance. Cdc2-like kinase 1 (CLK1) in the host cells is responsible for alternative splicing of the M2 gene of influenza virus during influenza infection and replication. In this study, we carried out baculovirus-mediated expression and purification of CLK1 and established a reliable screening assay for CLK1 inhibitors. After a virtual screening of CLK1 inhibitors was performed, the activities of the selected compounds were evaluated. Finally, several compounds with strong inhibitory activity against CLK1 were discovered and their in vitro anti-influenza virus activities were validated using a cytopathic effect (CPE) reduction assay. The assay results showed that clypearin, corilagin, and pinosylvine were the most potential anti-influenza virus compounds as CLK1 inhibitors among the compounds tested. These findings will provide important information for new drug design and development in influenza treatment, and CLK1 may be a potent drug target for anti-influenza drug screening and discovery.
