Serine protease inhibitors are a superfamily of proteins that regulate various physiological processes including fibrinolysis, inflammation and immune responses. In parasite systems, serpins are believed to play important roles in parasite colonization, inhibition of host immune serine proteases and penetration of defensive barriers. However, serpins are less well characterized in schistosomes. In this study, a Schistosoma mansoni serpin (Smserpin-p46) containing a 1360 base pair open reading frame, was cloned, expressed and functionally characterized. Bioinformatics analysis revealed that Smserpin-p46 contains the key residues, structural domains and motifs characteristic of inhibitory serpins. Gene expression profiling demonstrated stage-specific expression of Smserpin-p46 with the highest expression in adult male worms. Recombinant Smserpin-p46 (rSmserpin-p46) inhibited both human neutrophil cathepsin G and elastase, key serine proteases involved in NETosis, a program for the formation of neutrophil extracellular traps. Using specific rabbit antiserum, Smserpin-p46 was detected in soluble worm antigen preparation and was localized to the adult worm tegument. Cumulatively, the expression of Smserpin-p46 on the parasite tegument and its ability to inhibit proteases involved in NETosis highlights the importance of this serpin in parasite-host interactions and encourages its further investigation as a candidate vaccine antigen for the control of schistosomiasis. Increased tourism in Belize over the last decade and the growth of the local population have led to coastal development and infrastructure expansion. Land use alteration and anthropogenic activity may change the sediment and nutrient loads in coastal systems, which can negatively affect ecosystems via mechanisms such as reducing photosynthetically active radiation fields, smothering sessile habitats, and stimulating eutrophication events. Accurate monitoring and prediction of water quality parameters such as Total Suspended Sediments (TSS), are essential in order to understand the influence of land-based changes, climate, and human activities on the coastal systems and devise strategies to mitigate negative impacts. This study implements machine learning algorithms such as Random Forests (RF), Extreme Gradient Boosting (XGB), and Deep Neural Networks (DNN) to estimate TSS using Sentinel-2 reflectance data in the Belize Coastal Lagoon (BCL) and validates the results using TSS data collected in situ. DNN performed the best and estimated TSS with a testing R2 of 0.89. Time-series analysis was also performed on the BCL's TSS trends using Bayesian Changepoint Detection (BCD) methods to flag anomalously high TSS spatio-temporally, which may be caused by dredging events. Having such a framework can ease the near-real-time monitoring of water quality in Belize, help track the TSS dynamics for anomalies, and aid in meeting and maintaining the sustainable goals for Belize. The purpose of this paper is to examine how children's art can document emergent sensemaking of spatial reasoning. Spatial reasoning is the understanding of how both people and objects interact with, and relate to, one another. The recent literature has argued for spatial reasoning to be part of multiple domains in STEAM education by highlighting the dynamic nature of spatial thinking relevant in everyday life. The data come from a larger participatory design-based research project that incorporated play, environmental education, and embodiment in a STEAM curriculum. The paper analyzed art created by a focal group of children (6–8 years) as they learned about the kelp forest ecosystem over time. Findings reveal that spatial reasoning is not only an inseparable part of sensemaking in STEAM education, but has implications for environmental education in the elementary curriculum. Computational skills are advantageous for teaching students to investigate environmental change using satellite remote sensing. This focus is especially relevant given the disproportionate underrepresentation of minorities and women in STEM fields. This study quantified the effects in both a STEM and a non-STEM class of Earth science remote sensing modules in Google Earth Engine on students' self-efficacy in coding, understanding remote sensing, and interest in science and a career in environmental research. Additionally, the STEM students engaged in a course-based undergraduate research experience (CURE) on water quality. Satellite imagery was used to visualize water quality changes in coastal areas around the world due to the COVID-19 pandemic shutdown. Pre- and post-surveys reveal statistically significant changes in most students' confidence to apply coding skills to investigate environmental change and understand remote sensing. The intervention was not sufficient to lead to significant changes in interest in science or a career in environmental research. There is great benefit in incorporating remote sensing labs to teach environmental concepts to STEM and non-STEM students and to bolster the confidence of underrepresented minorities and females in STEM. Christian, especially Protestant, identity is often framed through the lens of belief, particularly belief understood as an interior orientation. Through an examination of the non-denominational Protestant group, the 'Little Flock', founded by Watchman Nee in the early 20th century, we trace enduring aspects of Little Flock theology in contemporary Chinese Protestant practice. In particular, we attend to conceptions of and debates surrounding belief and how to determine the quality of one's faith—whether or not one might be considered not just a Christian, but a 'good' one. This paper foregrounds the Turnerian experiences of pilgrims themselves, for whom pilgrimage is perhaps first and foremost the process of experiencing faith with their whole body and mind. At the Chinese Catholic pilgrimage site of Sheshan, located in western Shanghai, multiple meanings and possibilities are written onto the body of the pilgrim as it interacts with sacred materialities. In the process, the pilgrim materially orients themselves towards the transcendent other and to people and events throughout time. The boundary between subject and object is increasingly blurred in the pilgrim's imagination, and pilgrimage becomes a 'porous' mind-body experience for them. In the process, as pilgrims repeatedly physically enact doctrine and doctrinal texts in the course of pilgrimage—while simultaneously rooting them in their own personal lives—Sheshan is, through the concrete actions of worshippers layered up over time, continually being re-made as sacred. Architected cellular materials encompass a wide range of design and performance possibilities. While there has been significant interest in periodic cellular materials, recent emphasis has included consideration of aperiodicity, most commonly in studies of stochastic and graded cellular materials. This study proposes a classification scheme for aperiodic cellular materials, by first dividing the design domain into three main types: gradation, perturbation, and hybridization. For each of these types, two design decisions are identified: (i) the feature that is to be modified and (ii) the method of its modification. Considerations such as combining different types of aperiodic design methods, and modulating the degree of aperiodicity are also discussed, along with a review of the literature that places each aperiodic design within the classification developed here, as well as summarizing the performance benefits attributed to aperiodic cellular materials over their periodic counterparts. Featured Application: With its term mapping capability, MARIE can be used to improve data interoperability between different biomedical institutions. It can also be applied to text data pre-processing or normalization in non-biomedical domains. With growing interest in machine learning, text standardization is becoming an increasingly important aspect of data pre-processing within biomedical communities. As performances of machine learning algorithms are affected by both the amount and the quality of their training data, effective data standardization is needed to guarantee consistent data integrity. Furthermore, biomedical organizations, depending on their geographical locations or affiliations, rely on different sets of text standardization in practice. To facilitate easier machine learning-related collaborations between these organizations, an effective yet practical text data standardization method is needed. In this paper, we introduce MARIE (a context-aware term mapping method with string matching and embedding vectors), an unsupervised learning-based tool, to find standardized clinical terminologies for queries, such as a hospital's own codes. By incorporating both string matching methods and term embedding vectors generated by BioBERT (bidirectional encoder representations from transformers for biomedical text mining), it utilizes both structural and contextual information to calculate similarity measures between source and target terms. Compared to previous term mapping methods, MARIE shows improved mapping accuracy. Furthermore, it can be easily expanded to incorporate any string matching or term embedding methods. Clostridium difficile infection (CDI) is a potentially fatal illness, especially in the elderly and hospitalized individuals. The recurrence and rates of CDI are increasing. In addition, some cases of CDI are refractory to the currently available antibiotics. The search for improved modalities for the management of primary and recurrent CDI is underway. This review discusses the current antibiotics, fecal microbiota transplantation (FMT) and other options such as immunotherapy and administration of non-toxigenic Clostridium difficile (CD) for the management of both primary and recurrent CDI. In the colonial era, many Spanish missionaries in the Andes sought a total temporal and cultural break between the pagan past and desired a Christian future of indigenous Andeans. Discussions of Christian conversion in the modern-day Andes have often echoed this line of thinking, portraying conversion—whether to Protestantism or to Roman Catholicism—as an event of radical discontinuity, and mapping the rupture of conversion onto the rupture of the Spanish invasion and subsequent evangelisation of the Americas. In doing so, however, scholars have often portrayed Catholicism as a veneer over the 'authentic' Andes—which was assumed to not be Catholic, and indeed could never be. Recently, however, in the south-central Peruvian Andean parish of Talavera—under the guidance of a first generation of an indigenous Catholic priesthood, made up entirely of men born and raised in the local area—discourses surrounding conversion portray the past as a source of continuity rather than discontinuity with Catholicism. Drawing from historical and ethnographic sources, this article demonstrates that although conversion has been and continues to be an important point of reference in contemporary Roman Catholicism in the Andes, the question of what people convert from has shifted. Today, the Andes are spoken of as already inherently and profoundly Catholic; conversion, in the sense of the need to make the Andes 'really' Catholic, is considered long accomplished. As the article discusses, in a national context where Catholicism is dominant and ubiquitous to the point of hegemony, this is an inherently political stance which runs counter to longstanding harmful stereotypes of indigenous Andeans as not 'real' Catholics and thus unable to be 'real' Peruvians. In the years directly following the Second Vatican Council under the guidance of its second bishop Mons. Enrique Pelach i Feliu, the Andean diocese of Abancay—founded in 1959 in one of the most rural and most indigenous areas of Peru—experienced the founding of a new seminary intended to train a new generation of native clergy, and a concerted clerical effort to revive and promote the Marian pilgrimage of the Virgin of Cocharcas. The former meant the advent of a generation of native clergy made up of men born and raised in rural farming families in Abancay and native speakers of Quechua, the local indigenous language, which transformed the relationship between the institutional Church and indigenous Catholics from one rooted in antipathy and hostility to one based in a shared cultural background and language. The latter meant the elevation of the indigenous figure of Sebastian Quimichu as exemplar of both Andean Catholic faith and practice for his role in founding the Marian shrine of Cocharcas, and the legitimisation of popular Andean Catholic practices that had previously been stigmatised. This article provides a dual historical and ethnographic account of these events, and in doing so demonstrates the profound transformation of rural Andean lived religion and practice in the years following Vatican II. The ECOsystem Spaceborne Thermal Radiometer Experiment on Space Station (ECOSTRESS) provides remotely-sensed estimates of evapotranspiration at 70 m spatial resolution every 1–5 days, sampling across the diurnal cycle. This study, in partnership with an operational water management organization, the Eastern Municipal Water District (EMWD) in Southern California, was conducted to evaluate estimates of evapotranspiration under ideal conditions where water is not limited. EMWD regularly uses a ground-based network of reference evapotranspiration (ETo) from the California Irrigation Management Information System (CIMIS); yet, there are gaps in spatial coverage and questions of spatial representativeness and consistency. Space-based potential evapotranspiration (PET) estimates, such as those from ECOSTRESS, provide consistent spatial coverage. We compared ECOSTRESS ETo (estimated from PET) to CIMIS ETo at five CIMIS sites in Riverside County, California from July 2018–June 2020. We found strong correlations between CIMIS ETo and ECOSTRESS ETo across all five sites (R2 = 0.89, root mean square error (RMSE) = 0.11 mm hr−1). Both CIMIS and ECOSTRESS ETo captured similar seasonal patterns through the study period as well as diurnal variability. There were site-specific differences in the relationship between ECOSTRESS AND CIMIS, in part due to spatial heterogeneity around the station site. Consequently, careful examination of landscapes surrounding CIMIS sites must be considered in future comparisons. These results indicate that ECOSTRESS successfully retrieves PET that is comparable to ground-based reference ET, highlighting the potential for providing observation-driven guidance for irrigation management across spatial scales. Genome doubling is an underlying cause of cancer cell aneuploidy and genomic instability, but few drivers have been identified for this process. Due to their physiological roles in the genome reduplication of normal cells, we hypothesised that the oncogenes cyclins E1 and E2 may be drivers of genome doubling in cancer. We show that both cyclin E1 (CCNE1) and cyclin E2 (CCNE2) mRNA are significantly associated with high genome ploidy in breast cancers. By live cell imaging and flow cytometry, we show that cyclin E2 overexpression promotes aberrant mitosis without causing mitotic slippage, and it increases ploidy with negative feedback on the replication licensing protein, Cdt1. We demonstrate that cyclin E2 localises with core preRC (pre-replication complex) proteins (MCM2, MCM7) on the chromatin of cancer cells. Low CCNE2 is associated with improved overall survival in breast cancers, and we demonstrate that low cyclin E2 protects from excess genome rereplication. This occurs regardless of p53 status, consistent with the association of high cyclin E2 with genome doubling in both p53 null/mutant and p53 wildtype cancers. In contrast, while cyclin E1 can localise to the preRC, its downregulation does not prevent rereplication, and overexpression promotes polyploidy via mitotic slippage. Thus, in breast cancer, cyclin E2 has a strong association with genome doubling, and likely contributes to highly proliferative and genomically unstable breast cancers. [ABSTRACT FROM AUTHOR]

The development of a native clergy in the Andes has long been called for but only recently achieved. Drawing from archival and ethnographic data, this article sets out how intercultural prejudice and discrimination have served to prevent the ordination of native Andeans. In the early colonial period, doubts about the authenticity of Andean conversion to Catholicism were rooted in mainstream Spanish skepticism of and disdain for Andean culture; in the modern day, these same prejudices continue, meaning it is only within the last fifty years that a native clergy has developed in the southern Andes, in the Peruvian diocese of Abancay, as the result of the concerted efforts of its second bishop. Today, Abancay boasts its first generation of native clergy, made up entirely of men who were born and raised in the diocese in which they now serve, and which promises a new, more empathetic institutional relationship between what it has historically meant to be Andean and what it has meant to be Catholic. Background: Clostridium difficile infection (CDI) is one of the most common healthcare-associated infections (HAI) in the United States and Canada, and incidence rates have increased worldwide in recent decades. Currently, antibiotics are the mainstay treatments for both primary and recurrent CDI, but their efficacy is limited, prompting further therapies to be developed. Aim: This review summarizes current and emerging therapies in CDI management including antibiotics, fecal microbiota transplantation, monoclonal antibodies, spore-based therapies, and vaccinations. Microcystins are toxic chemicals generated by certain freshwater cyanobacteria. These chemicals can accumulate to dangerous levels during harmful algal blooms. When exposed to microcystins, humans are at risk of hepatic injury, including liver failure. Here, we describe a method to detect microcystins in human plasma by using immunocapture followed by a protein phosphatase inhibition assay. At least 279 microcystins have been identified, and most of these compounds share a common amino acid, the Adda side chain. We targeted this Adda side chain using a commercial antibody and extracted microcystins from human samples for screening and analysis. To quantitate the extracted microcystins, we fortified plasma with microcystin-LR, one of the most well-studied, commonly detected, and toxic microcystin congeners. The quantitation range for the detection of microcystin in human plasma using this method is 0.030-0.50 ng/mL microcystin-LR equivalents. This method detects unconjugated and conjugated forms (cysteine and glutathione) of microcystins. Quality control sample accuracies varied between 98.9% and 114%, with a precision of 7.18-15.8%. Finally, we evaluated plasma samples from a community health surveillance project of Florida residents living or working near harmful algae blooms.
