In this paper, a data-driven framework aimed at investigating how weather factors affect the spatio-temporal patterns of air traffic flow in the terminal maneuvering area (TMA) is presented. The framework mainly consists of three core modules, namely, trajectory structure characterization, flow pattern recognition, and association rule mining. To fully characterize trajectory structure, abnormal trajectories and typical operations are sequentially extracted based on a deep autoencoder network with two specially designed loss functions. Then, using these extracted elements as basic components to further construct and cluster per-hour-level descriptions of airspace structure, the spatio-temporal patterns of air traffic flow can be recognized. Finally, the association rule mining technique is applied to find sets of weather factors that often appear together with each flow pattern. Experimental analysis is demonstrated on two months of arrival flight trajectories at Hong Kong International Airport (HKIA). The results clearly show that the proposed framework effectively captures spatial anomalies, fine-grained trajectory structures, and representative flow patterns. More importantly, it also reveals that those flow patterns with non-conforming behaviors result from complex interactions of various weather factors. Rheumatoid arthritis (RA) is an autoimmune disease that significantly impacts quality of life by disrupting CD4+ T cell immune homeostasis. The identification of a low-side-effect drug for RA treatment is urgently needed. Our previous study suggests that Trichinella spiralis paramyosin (Ts-Pmy) has immunomodulatory effects, but its potential effect on CD4+ T cell response in RA remains unclear. In this study, we used a murine model to investigate the role of rTs-Pmy in regulating CD4+ T cell differentiation in collagen-induced arthritis (CIA). Additionally, we assessed the impact of rTs-Pmy on CD4+ T cell differentiation towards the Th1 and Th17 phenotypes, which are associated with inflammatory responses in arthritis, using in vitro assays. The results demonstrated that rTs-Pmy administration reduced arthritis severity by inhibiting Th1 and Th17 response while enhancing Treg response. Prophylactic administration of Ts-Pmy showed superior efficacy on CIA compared to therapeutic administration. Furthermore, in vitro assays demonstrated that rTs-Pmy could inhibit the differentiation of CD4+ T cells into Th1 and Th17 while inducing the production of Tregs, suggesting a potential mechanism underlying its therapeutic effects. This study suggests that Ts-Pmy may ameliorate CIA by restoring the immune balance of CD4+ T cells and provides new insights into the mechanism through which helminth-derived proteins exert their effects on autoimmune diseases. Simple Summary: The DKK gene family plays an important role in immune mechanisms and embryonic development, but studies on Pelodiscus sinensis have not been reported. In this study, for the first time in the context of P. sinensis, we analyzed the relevant features of the DKK gene family using bioinformatics methods and constructed transcriptional profiles and tissue expression profiles. The DKK1 and DKK3 genes were found to be highly expressed in the ovaries, while DKKL1 and DKK4 were significantly differentiated in the testes. In this study, we preliminarily investigated the function of the DKK gene family in the context of P. sinensis, and this study lays the foundation for further systematic research on the molecular mechanism of sex differentiation in P. sinensis. The DKK family is a canonical small family of WNT antagonists. Though recent studies have suggested that the DKK gene family may be involved in sex differentiation in Pelodiscus sinensis, there are still a lot of things about the DKK gene family that we do not know. In this study, we used bioinformatics methods to identify members of the DKK gene family in P. sinensis and analyzed their phylogeny, covariance, gene structure, structural domains, promoter conserved sites, signal peptides, gonadal transcription factors, transcriptional profiles, and tissue expression profiles. Additionally, qRT-PCR results were utilized for the validation and preliminary investigation of the function of the DKK gene family in P. sinensis. The results showed that the DKK gene family is divided into six subfamilies, distributed on six different chromosomal scaffolds containing different gene structures and conserved motifs with the same structural domains, and all of the members were secreted proteins. Our transcriptional profiling and embryonic expression analysis showed that DKKL1 and DKK4 were significantly expressed in the testes, whereas DKK1 and DKK3 were significantly upregulated in the ovaries. Environmentally friendly powder coatings which have the advantages of being VOC-free, low-cost, and high-efficiency with a high recovery rate have been attracting increasing research attention. The introduction of antibacterial agents into the powder coatings endows them with a capacity to kill bacteria and viruses on the surface of objects; additionally, this enables them to inhibit the indirect transmission of pathogenic microorganisms. Silver, possessing broad-spectrum, strong, and stable antibacterial properties, is considered to be a promising antibacterial material for use in coating applications. Carrier materials for active silver play an important role in its activity and stability. However, there is a lack of systematic studies on the effects of different types of carriers in such coating systems, especially in green powder coating systems. In this paper, we investigated two types of carriers for active silver agents: zeolite, i.e., Linde type A (LTA) zeolite and Y-type zeolite; clay-based materials, i.e., montmorillonite and vermiculite. All the agents showed high antibacterial activity, with antibacterial rates of over 99% as compared to commercial agents. Among the four agents, the Ag-LTA zeolite antimicrobial agent showed a reduction rate of over 99.99%; additionally, it maintained a reduction rate of 99% after seven washing cycles. Thus, this agent was demonstrated to have the highest effectiveness and high durability; these features can be attributed to the high silver content and small particle size. The LTA zeolite also provides a protective effect for silver ions, protecting them from reduction, due to the restriction of elemental silver formation within the confined interior space of the α-cage structure. The Y-type zeolite antimicrobial agent exhibited a slightly lower antimicrobial performance due to its higher silicon-to-aluminum ratio and its lower cation exchange capacity. Comparatively, antimicrobial agents utilizing clay-based carriers have lower cation exchange capacity, resulting in poorer antimicrobial effectiveness than zeolite carriers. In addition, silver loaded on clay-based materials is prone to detach from the carrier and undergo a reduction reaction, making the coating yellowish in color. This study first provides information on the roles of different types of carriers in powder coating systems; then, this information guides the selection of carriers for active silver for the development of efficient antimicrobial agents and coatings. The parent–offspring relationships of all offspring in four consecutive years were confirmed using sixteen microsatellite loci. We also summarised the reproductive results of all females and counted the annual number of offspring and the variation in the number of offspring. The females were then divided into three types (stable, undulating and levelling off) according to the continuity. This study can provide the basis and materials for the creation of a good reproductive group and the study of the reproductive biology of turtles in M. mutica aquaculture. Simple Summary: In aquaculture, the survival and health of aquatic organisms are influenced greatly by external environmental factors. Photoperiod is one of the key environmental factors that affect the early growth, survival, metabolism, and immune system of crustaceans significantly. The aim of the present study was to investigate the impact of photoperiod on the growth, survival, enzyme activity, body color, and expression of growth-related genes in juvenile redclaw crayfish. Five photoperiod experimental groups were established (0L:24D, 6L:18D, 12L:12D, 18L:6D, and 24L:0D), each with three replicates, and reared for 30 days. The group with an 18L:6D photoperiod had higher juvenile crayfish survival rates, better growth performance, stronger antioxidant stress response, and immune defense capabilities. Additionally, artificial photoperiods altered the body color of the redclaw crayfish. The results of the present study lay a foundation for the elucidation of the mechanisms underlying photoperiod regulation of the physiological and biochemical responses of juvenile redclaw crayfish. Through a 30-day experiment, this study investigated the effects of five photoperiods (0L:24D, 6L:18D, 12L:12D, 18L:6D, and 24L:0D) on the survival, enzyme activity, body color, and growth-related gene expression of redclaw crayfish (Cherax quadricarinatus) juveniles. The results showed that C. quadricarinatus juveniles under 18L:6D and 24L:0D photoperiods exhibited the highest survival rate, which was significantly higher than the survival rates of juveniles under the other three photoperiods (p < 0.05). However, the 0L:24D group had the highest final body weight and weight gain rate, significantly surpassing those of the 12L:12D, 18L:6D, and 24L:0D groups (p < 0.05). Regarding enzyme activity and hormone levels, juveniles under the 18L:6D photoperiod exhibited relatively higher activity of superoxide dismutase (SOD), acid phosphatase (ACP), and lysozyme (LZM) enzymes than those under other photoperiods, but their levels of melatonin and cortisol were relatively low. In addition, the 24L:0D group showed the highest malondialdehyde (MDA) content. Analysis of gene expression levels revealed that retinoid X receptor (RXR) and α-amylase (α-AMY) genes in C. quadricarinatus juveniles exhibited significantly higher expression levels under the 18L:6D photoperiod than those under the other four photoperiods (p < 0.05). With increasing daylight exposure, the body color of C. quadricarinatus changed from pale blue to yellow–brown. In summary, C. quadricarinatus juveniles achieved high survival rates, good growth performance, strong antioxidant stress response, and immune defense capabilities under an 18 h photoperiod. Therefore, in the industrial seedling cultivation of redclaw crayfish, it is recommended to provide 18 h of daily light. Further, the study demonstrated the ability to manipulate the body color of C. quadricarinatus through controlled artificial photoperiods. Male and female Chinese soft-shelled turtles (Pelodiscus sinensis) have sex-dimorphic growth patterns, and males have higher commercial value because of their larger size and thicker calipash. Thus, developing sex-specific markers is beneficial to studies on all-male breeding in P. sinensis. Here, we developed an accurate and efficient workflow for the screening of sex-specific sequences with ZW or XY sex determination systems. Based on this workflow, female and male P. sinensis reference genomes of 2.23 Gb and 2.26 Gb were obtained using de novo assembly. After aligning and filtering, 4.01 Mb female-specific sequences were finally identified. Subsequently, the seven developed sex-specific primer pairs were 100% accurate in preliminary, population, and embryonic validation. The presence and absence of bands for the primers of P44, P45, P66, P67, P68, and P69, as well as two and one bands for the PB1 primer, indicate that the embryos are genetically female and male, respectively. NR and functional annotations identified several sex-determining candidate genes and related pathways, including Ran, Eif4et, and Crkl genes, and the insulin signaling pathway and the cAMP signaling pathway, respectively. Collectively, our results reveal that a ZW-type sex-determination system is present in P. sinensis and provide novel insights for the screening of sex-specific markers, sex-control breeding, and the studies of the sex determination mechanism of P. sinensis. Salinity stress can trigger a series of physiological changes. However, the mechanism underlying the response to acute salinity stress in Macrobrachium rosenbergii remains poorly understood. In this study, osmoregulation, physiological metabolism, antioxidant capacity, and apoptosis were examined over 96 h of acute salinity stress. Hemolymph osmolality increased with increasing salinity. After 48 h of salinity exposure, the glucose, triglycerides, total protein, and total cholesterol contents in two salinity stress groups (13 and 26‰ salinity) were significantly lower than those in the 0‰ salinity group. The highest levels of these parameters were detected at 6 h; however, superoxide dismutase (SOD), total antioxidant capacity (T-AOC), and malondialdehyde (MDA) were the lowest at 96 h in the 13‰ salinity group. The activity of immunity-related enzyme alkaline phosphatase (AKP) showed a decreasing trend with increasing salinity and remained at a low level in the 26‰ salinity group throughout the experiment. No significant differences were observed in aspartate aminotransferase (AST), alanine aminotransferase (ALT), or lysozyme (LZM) among the three treatments at 96 h. After 96 h of salinity treatments, the gill filament diameter significantly decreased, and a more pronounced terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL)-positive signal was detected in the 13‰ and 26‰ groups compared to that in the 0‰ group. Expression levels of apoptosis-related genes, including Cysteine-aspartic acid protease 3 (Caspase 3), Cysteine-aspartic acid protease 8 (Caspase 8), Cytochrome c (Cyt-c), tumor suppressor gene (P53), Nuclear factor kappa-B (NF-κB), and B cell lymphoma 2 ovarian killer (Bok) were significantly higher in the 26‰ salinity group than in the other groups at 24 h, but lower than those in the 0‰ salinity group at 96 h. Cyt-c and P53 levels exhibited a significantly positive relationship with MDA, AST, and LZM activity during salinity stress. In the 13‰ salinity group, Bok expression was significantly correlated with SOD, T-AOC, AKP, acid phosphatase, and LZM activity, whereas in the 26‰ group, the AST content was positively correlated with Caspase 8, Cyt-c, and P53 expression. A significant negative relationship was observed between Caspase 3 expression and catalase (CAT) activity. These findings provide insight into the mechanisms underlying the response to acute salinity stress and will contribute to improving M. rosenbergii aquaculture and management practices. Simple Summary: We explored the effect of maternal size on offspring size and number using a 4-year dataset based on paternity analyses in Asian yellow pond turtle, Mauremys mutica, and found that the maternal size significantly affected the offspring number but not offspring size. The offspring size was significantly correlated with maternal age. Our results suggest that the offspring size-number trade-off theory does not apply to cultured M. mutica and also provides basic data for the efficient artificial breeding management of M. mutica in captivity. Offspring size-number trade-off is a critical component of life-history theory and is important for further understanding the reproductive strategies of animals. The relationship between this trade-off and maternal size has been explored in several turtle species, except for the Asian yellow pond turtle, Mauremys mutica. To investigate how the maternal condition affects offspring size and number, we explored the relationships among the maternal body size and the number and size of cultured M. mutica hatchlings using a 4-year dataset. Our results showed that different females not only produced different sizes of offspring but also produced different numbers of offspring. No trade-off in egg size number was detected. According to regression analysis, we did not find that the maternal body size significantly influenced the offspring mass; however, we detected that the offspring size was significantly correlated with the clutch size and maternal age. The mean body mass of offspring increased with maternal age, and the clutch size varied significantly over four years, which was correlated with offspring size, maternal body size and age. However, the number of offspring per female increased with the maternal plastron length rather than age. Our results were inconsistent with the optimal offspring size theory in that females did not increase their offspring size but rather increased the offspring number to increase their fitness, which will also provide a basis for the efficient cultivation management of turtles. Different weakening buffer operators in a time-series model analysis usually result in different model sensitivities, which sometimes affect the effectiveness of relevant operator-based methods. In this paper, the stability of two classic fractional-order weakening buffer operator-based series models is studied; then, a new data preprocessing method based on a novel fractional-order bidirectional weakening buffer operator is provided, whose effect in improving the model's stability is tested and utilized in prediction problems. Practical examples are employed to demonstrate the efficiency of the proposed method in improving the model's stability in noise scenarios. The comparison indicates that the proposed method overcomes the disadvantage of many weakening buffer operators in the subjectively biased weighting of the new or old information in forecasting. Simple Summary: Hibernation in turtle species is an adaptive survival strategy to colder winter conditions or food restrictions. However, the mechanisms underlying seasonal adaptions remain unclear. In this study, we isolated and performed scRNA-seq analysis on P. cantorii hemocytes in the active state and during hibernation to classify the hemocyte types and obtain the relevant cell maps. We identified the marker genes of each cell population and the differentiation pathways associated with their maturation. We also discovered different immune-related genes that might play crucial roles in hemocyte differentiation before and after hibernation. Our study aims to provide a reference dataset for the unified classification of P. cantorii hemocytes and improve our understanding of the immune system of P. cantorii. Moreover, several heat shock genes, including the Hsp40 family chaperone gene (DNAJ) and HSP temperature-responsive genes (HSPs), were upregulated during the hibernation period, which predicted these genes may play crucial roles in the stress response during hibernation. Additionally, compared to hemocytes in the active state, several upregulated differentially expressed immune-related genes, such as stat1, traf3, and socs6, were identified in hemocytes during the hibernation period, thus indicating the important immune function of hemocytes. Therefore, our findings provide a unified classification of P. cantorii hemocytes and identify the genes related to the stress response, thereby providing a better understanding of the adaptive mechanisms of hibernation. Simple Summary: The Asian giant soft-shelled turtle (Pelochelys cantorii), belonging to the order Testudines (family Trionychidae, genus Pelochelys), is one of the largest inland aquatic turtle species. However, due to excessive economic development, Pc. cantorii is critically endangered and rarely seen in the wild. As early as 1989, China listed the turtle as a key aquatic wildlife protection animal at the national level, but the conservation biology of Pc. cantorii has not been fully elucidated due to a lack of reference genomes. Here, based on a high-quality chromosome-level genome for Pc. cantorii, acquired by a combination of Illumina short-read, PacBio long-read and Hi-C scaffolding technologies in a previous study, we analyzed the evolutionary state of Pc. cantorii. Moreover, we found that several candidate genes associated with tumor suppression, growth and age were expanded, implicating their potential roles in the exceptional longevity of turtles. These findings will be an enabling resource for genetic and genomic studies to support fundamental insights into Pc. cantorii conservation. The Asian giant soft-shelled turtle, Pelochelys cantorii (Trionychidae), is one of the largest aquatic turtles in China and was designated as a First-Grade Protected Animal in China in 1989. Previous investigation based on a combination of Illumina short-read, PacBio long-read and Hi-C scaffolding technologies acquired a high-quality chromosome-level genome of Pc. cantorii. In this study, comparative genomic analysis between Pc. cantorii and 16 other vertebrate genomes indicated that turtles separated from the ancestor of archosaurians approximately 256.6 (95% highest posterior density interval, 263.6–251.9) million years ago (Mya) (Upper Permian to Triassic) and that Pc. cantorii separated from the ancestor of Pd. sinensis and R. swinhoei approximately 59.3 (95% highest posterior density interval, 64.3–54.3) Mya. Moreover, several candidate genes, such as VWA5A, ABCG2, A2M and IGSF1, associated with tumor suppression, growth and age were expanded, implicating their potential roles in the exceptional longevity of turtles. This new chromosome-level assembly has important scientific value in the study of conservation of Pc. cantorii and also enriches the evolutionary investigation of turtle species. Simple Summary: Pelochelys cantorii is critically endangered and rarely seen in the wild, and only 13 adults are being kept in captivity in China. For the purpose of reinforcing the conservation and management of P. cantorii, the Pearl River Fisheries Research Institute (Chinese Fishery Academy of Sciences) successfully bred more than 800 turtles from 2015 to 2020. In this study, we developed and characterized 10 simple sequence repeat markers from the RNA transcriptome of P. cantorii, established two multiplex PCR systems, and obtained the genetic structure and genetic diversity of the artificially conserved population. The aim was to obtain viable second-generation P. cantorii with the highest genetic diversity to implement population recovery plans for this species. To understand the genetic structure of the protected turtle species Pelochelys cantorii we used transcriptome data to design more than 30,000 tri- and tetranucleotide repeat microsatellite primer pairs, of which 230 pairs were used for laboratory experiments. After two screenings, only 10 microsatellite markers with good specificity, high amplification efficiency, and polymorphisms were obtained. Using the selected primers, two multiplex PCR systems were established to compare and analyze the genetic diversity of artificially assisted breeding generations from four parents (two females and two males) continuously bred over two years. A total of 25 alleles were detected among the 10 microsatellite loci of the offspring. The polymorphism information content (PIC) was 0.313–0.674, with an average of 0.401, among which two loci were highly polymorphic (PIC ≥ 0.5). The number of alleles was 2–5 and the number of effective alleles was 1.635–3.614. The observed heterozygosity was 0.341–0.813, with an average of 0.582, whereas the average expected heterozygosity was 0.389–0.725, with an average of 0.493. Moreover, on the basis of Nei's genetic distance and the Bayesian clustering algorithm, the 182 offspring were divided into two subgroups, and the results corresponded to the two maternal lines. This is the first study to investigate the molecular markers of P. cantorii, and the results obtained can be used to protect genetic resources and provide a genetic basis for the design of population recovery plans. Although some significant achievements have been obtained in the gonadal cells' culture or manipulation across multiple phyla including teleost and mammals, there is limited study on gonadal cell manipulation in turtles. In this study, we isolated an ovarian cell line (fGSCs), from the juvenile Asian yellow pond turtle in vitro. The cultured fGSCs were characterized with germline stem cell specific markers' expression using transcriptome sequencing on Illumina platform, reverse transcription-polymerase chain reaction (RT-PCR) and immunofluorescence (IF). Our study aims to provide a valuable tool to elaborate the molecular mechanisms behind germ cells development, differentiation and oogenesis in the turtle even in reptiles, and also, the established cell line has potential applications to enrich the conservation strategy of aquatic animal germplasm resources. Although some significant achievements have been obtained in the gonadal cells' culture or manipulation across multiple phyla including teleost and mammals, there is limited study on gonadal cell manipulation in turtles. In this study, we established a new ovarian cell line from the young Asian yellow pond turtle (Mauremys mutica), which exhibited a normal diploid karyotype with high alkaline phosphatase activity. The cell line, designated as YTO2, was then characterized through the analysis of gene expression profiles. The transcriptome analysis and the reverse transcription polymerase chain reaction (RT-PCR) showed that the cells expressed germline genes such as tdrd7, nanos1, klf5, igtb1, hsd17b4 and rad51. Moreover, the immunostaining showed that the germ cell markers, Tdrd7 and Rad51 proteins, were detected predominant in cytoplasm of perinuclear region, while proliferation marker, PCNA, was dominantly observed in the nuclei of cultured cells. Intriguingly, the cells could respond to the retinoic acid induction with significantly increasing the expression level of some meiosis genes, including vasa, dazl, figla, and dmc1. Furthermore, YTO2 cells could be efficiently transfected with the pHBAd-BHG-EGFP adenovirus and properly expressed the exogenous genes. To sum up, an ovarian cell line of the Asian yellow pond turtle had been established and could be stably propagated under in vitro culture condition, as well as being capable of efficiently expressing the exogenous gene tdrd7. This cell line would provide a valuable tool to elaborate the molecular mechanisms behind germ cells development, differentiation and oogenesis in the turtle, even in reptiles. Simple Summary: Turtles have been on Earth for 300 million years, but their survival has been challenged as human activity has expanded. The results of a census and statistical analysis of artificially domesticated aquatic turtles in 15 provinces of China showed that 29 species were aquatic turtles native to China and a large number of exotic aquatic turtles are domesticated in China. This paper presents the current situation and problems of aquaculture and conservation of aquatic turtles in major provinces of China, and puts forward some suggestions for the conservation and management of aquatic turtles. China is a major country in turtle cultivation and has a long history of artificial breeding of turtles. In this study, a census and statistical analysis of artificially domesticated aquatic turtles in 15 provinces of China were conducted. The results showed that 29 species were aquatic turtles native to China, accounting for approximately 9% of the world's total, and a large number of exotic aquatic turtles are also domesticated in China. The present situation of artificial breeding and protection of aquatic turtles in major provinces of China is shown, and existing problems are also put forward, with suggestions for the protection and management of aquatic turtles. Simple Summary: Chinese soft-shell turtle (Pelidiscus sinensis) is a widely farmed aquatic reptile in China. It has of high nutritional, pharmaceutical, and economic value, but wild Chinese soft-shell turtle has been faced with an extreme decline of populations. Thus, it is of great significance to study the germ cells biology and develop the techniques for turtle breeding and genetic resource preserving. Nanog is a homeodomain-containing transcription factor, and it plays a vital role in maintaining the pluripotency of embryonic stem cells; there is a lack of reporting on the Nanog gene in reptile. In this study, the cDNA sequence and promoter of a Nanog gene was identified in P. sinensis, designed as PsNanog. PsNanog gene products were mainly expressed in gonads and were especially rich in ovary. Particularly, PsNanog mRNA and protein was exclusively expressed in germ cells in ovary and testis. Collectively, this is the first report on the gene structure and expression of Nanog in turtle, which may lay a foundation for further functional analysis of the Nanog in turtle, even in the reptiles. Further, PsNanog probably has the conserved and divergent function in regulating cell pluripotency and germ cell development in turtle, compared with other species, including teleosts and mammals. PsNanog has a highly conserved HD domain and shares a high identity with that of Chelonia mydas and the lowest identity with Oryzias latipes. Similarly, PsNanog presented a tight cluster with C. mydas Nanog, but was far from those of teleosts. Additionally, we cloned a length of 1870 bp PsNanog promoter. Dual luciferase assay showed that the DNA fragment of −1560 to +1 exhibited a high promoter activity. The RT-PCR and RT-qPCR results showed that PsNanog was predominantly expressed in ovary, and then in testis. The in situ hybridization and immunohistochemical analysis showed that PsNanog was expressed in the early primary oocytes and the cytoplasm of the cortical region of stage VIII oocytes in ovary, and distributed in most stages of germ cells in testis. Collectively, the results imply that PsNanog probably has the conserved function in regulating germ cell development across phyla and is also a pluripotent cell gene and expressed in germ cells, which is similar to that in teleosts and mammals. Simple Summary: Spermatogenesis in Pelodiscus sinensis has distinct seasonal characteristics. Spats1 acts a key function in germ cell differentiation in mammals, yet its study in P. sinensis has not been reported. Here, we cloned the coding sequences (CDS) of Spats1 for the first time and characterized its expression in different tissues. In addition, the expression characteristics of Spats1 in the testis during different seasons and the expression pattern in embryos and adults after steroid hormone treatment were also investigated. The present research provides a preliminary study on the physiological regulation of Spats1 in P. sinensis, and lays the foundation for further systematic investigation of the molecular mechanisms of spermatogenesis and release in P. sinensis. Spats1 (spermatogenesis-associated, serinerich 1) has been characterized as a male-biased gene which acts an important role in the germ cell differentiation of mammals. Nevertheless, the function of Spats1 in the Chinese soft-shelled turtle (P. sinensis) has not yet been reported. To initially explore the expression of Spats1 in P. sinensis and its response to sex steroid treatment, we cloned the CDS of Spats1 for the first time and analyzed its expression profile in different tissues, including the testes in different seasons. The Spats1 cDNA fragment is 1201 base pairs (bp) in length and contains an open reading frame (ORF) of 849 bp, which codes for 283 amino acids. Spats1 mRNA was highly expressed in the testes (p < 0.01) and barely detectable in other tissues. In P. sinensis, the relative expression of Spats1 also responsive to seasonal changes in testis development. In summer (July) and autumn (October), Spats1 gene expression was significantly higher in the testes than in other seasons (p < 0.05). Spats1 mRNA was found to be specifically expressed in germ cells by chemical in situ hybridization (CISH), and it was mainly located in primary spermatocytes (Sc1), secondary spermatocytes (Sc2) and spermatozoa (St). Spats1 expression in embryos was not significantly changed after 17α-methyltestosterone (MT)and 17β-estradiol (E2) treatment. In adults, MT significantly induced Spats1 expression in male P. sinensis. However, the expression of Spats1 in testes was not responsive to E2 treatment. In addition, the expression of Spats1 in females was not affected by either MT or E2 treatment. Additionally, some potential genes linked to sex differentiation, such as bmp2, ran, and sox3, were also isolated in P. sinensis. Functional analysis showed that the DE miRNAs and DE ncRNAs were enriched in the pathways related to sex differentiation, including ovarian steroidogenesis, the hippo signaling pathway, and the calcium signaling pathway. Remarkably, a lncRNA/circRNA–miRNA–mRNA interaction network was constructed, containing the key genes associated with sex differentiation, including fgf9, foxl3, and dmrta2. Collectively, we constructed a gender dimorphism profile of the female and male gonads of P. sinensis, profoundly contributing to the exploration of the major genes and potential ncRNAs involved in the sex differentiation of P. sinensis. Trichinella spiralis maintains chronic infections within its host, involving a variety of immunomodulatory properties, the mechanisms of which have not been completely elucidated. In this study, we found that T. spiralis infection induced strong regulatory T cell responses through parasite excretory–secretory (ES) products, characterized by increase of CD4+CD25+Foxp3+ and CD4+CD25−Foxp3+ Treg cells accompanied by high levels of IL-10 and TGF-β. T. spiralis adult worm excretory–secretory products (AES) and muscle larvae excretory–secretory products (MES) were both able to activate BMDCs in vitro to facilitate their maturation and to create regulatory cytokines IL-10 and TGF-β. The T. spiralis AES- and MES-pulsed dendritic cells (DCs) possessed abilities not only to present antigens to sensitized CD4+ T cell to stimulate their proliferation but also to induce naive CD4+ T cells to differentiate to Treg cells secreting IL-10 and TGF-β. The passive transfer of T. spiralis AES- and MES-pulsed bone marrow-derived dendritic cells (BMDCs) conferred the naive mice to acquire the differentiation of Treg cells. T. spiralis AES possesses a better ability to induce Treg cells than did MES, although the latter has the ability to induce CD4+CD25−Foxp3+ Treg cells. The results obtained in this study suggested that T. spiralis ES products stimulate the differentiation of host Treg cells possibly through activating dendritic cells to create a regulatory environment that benefits the survival of the parasite in the host. Ultrafine powder coatings are one of the development directions in the powder coating industry, as they can achieve thin coatings with good leveling and high surface smoothness comparable to liquid coatings. Compared to regular coatings, they experience a higher sensitivity to any incompatibilities, e.g., filler from coating components. The properties of fillers play a great role in the performance of coating films. Aluminum trihydrate (Al(OH)3) is a well-known filler in solvent-based coatings and other polymer industries. To study and evaluate the performances of Al(OH)3 in ultrafine powder coatings, a popular filler, barium sulfate (BaSO4) is used for comparison. Both fillers are added in ultrafine powder coatings based on two of the most commonly used resin systems (polyester-epoxy and polyester). The differences of physical and chemical properties between both fillers have significant influences on several properties of powder paints and coating films. The polar groups (hydrogen bond) in Al(OH)3 result in the strong interaction between inorganic filler and organic polymer matrix, thus decreasing the molecular network mobility and influencing the chain formation, which is verified by differential scanning calorimetric (DSC). The bed expansion ratio (BERs) of powder paints incorporated with Al(OH)3 are much higher than those with BaSO4, which indicate more uniform gas-solid contact during the spraying process. Samples with Al(OH)3 exhibit much lower specular gloss at 60°, which are expected to achieve remarkable matting effects. Superior corrosion resistances can be observed for almost all the coated panels incorporated with Al(OH)3 in contrast to those with BaSO4. Other aspects are slightly influenced by the difference between the two fillers, such as the angle of repose values (AORs) of powder paints, the impact resistance and flexibility of coating films. Aluminum trihydrate (ATH) is one of the most widely used fillers in solvent-borne coatings and polymer industries. To investigate its performance in powder coatings, four different pigment to binder ratios (P/B) were used in respect to different ATH concentrations in four most widely used resin systems (epoxy, epoxy-polyester, polyurethane and polyester). The angle of repose (AOR) and specific gravity of powder paints increase accordingly with ATH concentration. The mechanical properties, corrosion and UV resistance performances of coating films are enhanced or only slightly decreased with the addition of ATH. Pencil scratch hardness increase significantly with the increase of ATH content. Impact resistance decreases within 5% of the initial resistance at the maximum ATH loading. There is a slightly decrease in corrosion resistance performances of less than 2 mm of failure at scribe in all formulations. The incorporation of ATH has slight effect on the UV resistance performance of all the samples. Moreover, the matting effect of ATH is observed for all the resin systems, especially epoxy, with the specular gloss decreasing more than 70% at P/B of 0.75. [ABSTRACT FROM AUTHOR]

The rapid advancement in Unpiloted Robotic Vehicle technology has significantly influenced ground support operations at airports, marking a critical shift towards future development. This study presents a novel Unpiloted Ground Support Equipment (GSE) detection and control framework, comprising virtual channel delineation, boundary line detection, object detection, and navigation and docking control, to facilitate automated aircraft docking within the aircraft stand. Firstly, we developed a bespoke virtual channel layout for Unpiloted GSE, aligning with operational regulations and accommodating a wide spectrum of aircraft types. This layout employs turning induction markers to define essential navigation points, thereby streamlining GSE movement. Secondly, we integrated cameras and Lidar sensors to enable rapid and precise pose adjustments during docking. The introduction of a boundary line detection system, along with an optimized, lightweight YOLO algorithm, ensures swift and accurate identification of boundaries, obstacles, and docking sites. Finally, we formulated a unique control algorithm for effective obstacle avoidance and docking in varied apron conditions, guaranteeing meticulous management of vehicle pose and speed. Our experimental findings reveal an 89% detection accuracy for the virtual channel boundary line, a 95% accuracy for guiding markers, and an F1-Score of 0.845 for the YOLO object detection algorithm. The GSE achieved an average docking error of less than 3 cm and an angular deviation under 5 degrees, corroborating the efficacy and advanced nature of our proposed approach in Unpiloted GSE detection and aircraft docking.
