A precisely designed dual-color biosensor has realized a visual assessment of thymidine kinase 1 (TK1) mRNA in both living cells and cell lysates. The oligonucleotide probe is constructed by hybridizing the antisense strand of the target and two recognition sequences, in which FAM serves as the donor and TAMRA as the acceptor. Once interacting with the target, two recognition strands are replaced, and then the antisense complementary sequence forms a more stable double-stranded structure. Due to the increasing spatial distance between two dyes, the FRET is attenuated, leading to a rapid recovery of FAM fluorescence and a reduction of TAMRA fluorescence. A discernible color response from orange to green could be observed by the naked eye, with a limit of detection (LOD) of 0.38 nM and 5.22 nM for spectrometer- and smartphone-based assays, respectively. The proposed ratiometric method transcends previous reports in its capacities in visualizing TK1 expression toward reliable nucleic acid biomarker analysis, which might establish a general strategy for ratiometric biosensing via strand displacement. Multi-behavior recommendation systems exploit multi-type user–item interactions (e.g., clicking, adding to cart and collecting) as auxiliary behaviors for user modeling, which can alleviate the problem of data sparsity faced by traditional recommendation systems. The key point of multi-behavior recommendation systems is to make full use of the auxiliary behavior information for the learning of user preferences. However, there are two challenges in existing methods that need to be explored: (1) capturing personalized user preferences based on multiple auxiliary behaviors, especially for negative feedback signals; and (2) explicitly modeling the semantics between auxiliary and target behaviors, and learning the explicit interactions between multiple behaviors. To tackle the two problems described above, we propose a novel model, called explicit behavior interaction with heterogeneous graph for multi-behavior recommendation (MB-EBIH). In particular, we first construct a heterogeneous behavior graph, including both positive and negative behaviors. A pre-trained model based on graph neural network (GNN) is then used to generate explicit behavior interaction values as the edge weights for the heterogeneous behavior graph. These weights reflect the importance of each of the auxiliary behaviors in an explicit manner. Finally, the extracted explicit behavior interaction information is incorporated into the multi-behavior user–item bipartite graphs to learn better representations. Experimental results on four real-world datasets demonstrate the effectiveness of our model in terms of exploring multi-behavioral data; and ablation and analysis experiments further demonstrate the effectiveness of explicit behavior interaction information. Mitophagy has a critical role in maintaining cellular homeostasis through acidic lysosomes engulfing excess or impaired mitochondria, thereby pH fluctuation is one of the most significant indicators for tracking mitophagy. Then such precise pH tracking demands the fluorogenic probe that has tailored contemporaneous features, including mitochondrial-specificity, excellent biocompatibility, wide pH-sensitive range of 8.0–4.0, and especially quantitative ability. However, available molecular probes cannot simultaneously meet all the requirements since it is extremely difficult to integrate multiple functionalities into a single molecule. To fully address this issue, we herein integrate two fluorogenic pH sensitive units, a mitochondria-specific block, cell-penetrating facilitator, and biocompatible segments into an elegant silica nano scaffold, which greatly ensures the applicability for real-time tracking of pH fluctuations in mitophagy. Most significantly, at a single wavelength excitation, the integrated pH-sensitive units have spectra-distinguishable fluorescence towards alkaline and acidic pH in a broad range that covers mitochondrial and lysosomal pH, thus enabling a ratiometric analysis of pH variations during the whole mitophagy. Single-cell nucleic acid analysis aims at discovering the genetic differences between individual cells which is well known as the cellular heterogeneity. This technology facilitates cancer diagnosis, stem cell research, immune system analysis, and other life science applications. The conventional platforms for single-cell nucleic acid analysis more rely on manual operation or bulky devices. Recently, the emerging microfluidic technology has provided a perfect platform for single-cell nucleic acid analysis with the characteristic of accurate and automatic single-cell manipulation. In this review, we briefly summarized the procedure of single-cell nucleic acid analysis including single-cell isolation, single-cell lysis, nucleic acid amplification, and genetic analysis. And then, three representative microfluidic platforms for single-cell nucleic acid analysis are concluded as valve-, microwell-, and droplet-based platforms. Furthermore, we described the state-of-the-art integrated single-cell nucleic acid analysis systems based on the three platforms. Effects of the number of welding passes on microstructure and properties of 2219-C10S aluminum alloy tungsten inert gas (TIG) welded joints were analyzed in detail. The first capping weld had a solid solution effect on the backing weld. The second capping weld had a solid solution effect on the backing weld close to the capping weld and had an aging effect on the backing weld away from the capping weld. The sizes and quantities of the second phases in partially melted zone (PMZ) and over-aged zone (OAZ) increased slightly with the number of welding passes increased. The hardness of OAZ decreased slightly and its width increased slightly. The larger strain distribution range before tensile fractured of the joints was narrower and the range of plastic deformation of the joints was not significantly improved. The eutectic structure and θ phase at weld toes were not conducive to the tensile properties of joints. The tensile properties of double-pass welded joints were relatively ideal. Metal–organic framework (MOF) supported nanocatalysts are attracting great attentions in heterogeneous catalysis. However, they are still less explored and poorly understood in catalyzing solid–gas reactions. In current work, two MOF supported Pt nanocatalysts were prepared by immersion method coupled with the most commonly used NaBH4 (Pt/UiO-66-NaBH4) and H2 (Pt/UiO-66-H2) reduction, and their catalytic CO oxidation performances were investigated. NaBH4 and H2 reduction both provide highly dispersed Pt/UiO-66 nanocatalysts, and the two prepared catalysts display excellent CO oxidation activities. Moreover, CO oxidation durability tests reveal that Pt/UiO-66-NaBH4 shows superior durability and structure stability, which are much better than those of Pt/UiO-66-H2. The high structure stability of Pt/UiO-66-NaBH4 also endows it with good H2O and CO2 tolerance. This work attempts to elucidate the influence of reduction methods on the structures and catalytic CO oxidation performances of Pt/UiO-66 catalysts by employing XRD, TEM, N2-sorption, XPS and in situ DRIFTS techniques. Furthermore, the mechanism of CO oxidation was studied by in situ DRIFTS, and the result reveals that the CO oxidation on the two catalysts follows different mechanism. [ABSTRACT FROM AUTHOR]

Purpose: To determine the diagnostic performance of intravoxel incoherent motion (IVIM) parameters to predict tumor recurrence after hepatectomy in patients with hepatitis B virus (HBV)-related hepatocellular carcinoma (HCC).Materials and Methods: One hundred and fifty-seven patients (mean age 52.54 ± 11.32 years, 87% male) with surgically and pathologically confirmed HCC were included. Regions of interests were drawn including the tumors by two independent radiologists. ADC and IVIM-derived parameters (true diffusion coefficient [D]; pseudodiffusion coefficient [D*]; pseudodiffusion fraction [f]) were obtained preoperatively. The Cox proportional hazards model was used to analyze the predictors associated with tumor recurrence after hepatectomy.Results: Forty-seven of 157 (29.9%) patients experienced tumor recurrence. The multivariate Cox proportional hazards model revealed that a D value < 0.985 × 10-3 mm2/s (hazard ratio (HR), 0.190; p = 0.023) was a risk factor for tumor recurrence. Additional risk factors included younger age (HR, 0.328; p = 0.034) and higher serum alpha-fetoprotein (AFP) level (HR, 2.079; p = 0.013). Further, receiver operating characteristic (ROC) analysis showed that the area under the curve (AUC) of the obtained Cox regression model improved from 0.68 for the combination of AFP and age alone to 0.724 for the combination of D value, AFP, and age.Conclusion: The D value derived from the IVIM model is a potential biomarker for the preoperative prediction of recurrence after hepatectomy in patients with HCC. Purpose: To evaluate histologic differentiation of hepatitis B virus (HBV)-related hepatocellular carcinomas (HCCs) using apparent diffusion coefficient (ADC) and intravoxel incoherent motion (IVIM)-derived metrics and to compare findings with alpha-fetoprotein (AFP) levels alone and in combination. Materials and method: One hundred and six chronic HBV-related HCC patients who underwent IVIM diffusion-weighted magnetic resonance imaging with eleven b values were enrolled. Mean ADC, diffusion coefficient ( D), pseudodiffusion coefficient ( D*), and perfusion fraction ( f) values were determined for all detected lesions. The metrics and AFP levels of different histologically differentiated groups were compared. Spearman's rank correlation was used to assess the statistical dependence among the histologically differentiated HCCs. Receiver operating characteristic (ROC) analysis was performed to evaluate diagnostic performance of these metrics and AFP levels alone and in combination. Results: ADC, D, and f values and AFP levels were significantly different among well-, moderately, and poorly differentiated HCCs. The four metrics were significantly correlated with histologic differentiation. The area under the ROC curve (AUC-ROC) of ADC, D, f, and AFP for diagnosing well-differentiated HCCs was 0.903, 0.84, 0.782, and 0.806, respectively, and the AUC-ROC of above metrics for diagnosing poorly differentiated HCCs was 0.787, 0.726, 0.624, and 0.633, respectively. The combination of ADC and AFP provided an AUC-ROC of 0.945 for well-differentiated HCC. However, this did not provide better performance for diagnosing poorly differentiated HCC. Conclusion: ADC, IVIM metrics, and AFP levels may be useful for evaluating histologic differentiation of HBV-related HCCs, and the combination of ADC and AFP provides better diagnostic performance for well-differentiated HCC. Recent studies in subcortical ischemic vascular disease (SIVD) suggest the involvement of white matter (WM) abnormalities underlying the pathogenesis of cognitive function impairment. Here, we performed magnetic resonance diffusion tensor imaging (DTI) on detecting WM damage and to investigate the correlations between DTI measures and cognitive dysfunction in SIVD patients. Fifty right-handed SIVD patients were recruited and divided into vascular cognitive impairment on dementia (VCIND) group and normal cognition (NC) group. Twenty-two VCIND patients and 28 NC patients underwent DTI scanning and neuropsychological assessment. Atlas-based analysis (ABA) was performed on each subject for extracting FA and MD measures from supratentorial tracts. Among VCIND, as compared to NC patients, decreased FA and increased MD were observed in all projection fibers (bilateral anterior, posterior limb, and retrolenticular part of internal capsule, anterior, superior, and posterior corona radiata and posterior thalamic radiation), association fibers (bilateral sagittal stratum, external capsule, cingulum, fornix, and stria terminalis, superior longitudinal fasciculus, superior fronto-occipital fasciculus, and uncinate fasciculus), and commissural fibers (genu, body, splenium, and bilateral tapetum of corpus callosum). Furthermore, we also found that MoCA scores correlated with DTI values in all supratentorial WM tracts. The results suggested that SIVD patients demonstrated abnormal WM connectivity in all supratentorial regions. Moreover, the severity of damage in WM tracts correlated with cognitive dysfunction. Multi-center studies are advantageous for enrolling participants of varying pathological and demographical conditions, and especially in neurological studies. Hence stability of the obtained quantitative R2* and susceptibility in multicenter studies is a key issue for their widespread applications. In this work, the stabilities of simultaneously obtained R2* and susceptibility are investigated and compared across 10 sites that are equipped with the same scanner and receiver coil, the same post-processing process was used to achieve consistent experiment setup. Two healthy adult volunteers (one male and female) participated in this study. High intraclass correlation coefficient was obtained for both susceptibility (0.94) and R2* (0.96). The coefficients of variance for all measurements obtained were smaller than 0.1, the largest variations of measurements in all the chosen ROIs fall within ±20% from the median value. Higher level of stability was obtained in R2* as compared to susceptibility at 1 mm resolution (P < 0.05) and at 1.5 mm (P < 0.01). [ABSTRACT FROM AUTHOR]
