[Display omitted] Structural information of grassland changes on the Tibetan Plateau is essential for understanding alterations in critical ecosystem functioning and their underlying drivers that may reflect environmental changes. However, such information at the regional scale is still lacking due to methodological limitations. Beyond remote sensing indicators only recognizing vegetation productivity, we utilized multivariate data fusion and deep learning to characterize formation-based plant community structure in alpine grasslands at the regional scale of the Tibetan Plateau for the first time and compared it with the earlier version of Vegetation Map of China for historical changes. Over the past 40 years, we revealed that (1) the proportion of alpine meadows in alpine grasslands increased from 50% to 69%, well-reflecting the warming and wetting trend; (2) dominances of Kobresia pygmaea and Stipa purpurea formations in alpine meadows and steppes were strengthened to 76% and 92%, respectively; (3) the climate factor mainly drove the distribution of Stipa purpurea formation, but not the recent distribution of Kobresia pygmaea formation that was likely shaped by human activities. Therefore, the underlying mechanisms of grassland changes over the past 40 years were considered to be formation dependent. Overall, the first exploration for structural information of plant community changes in this study not only provides a new perspective to understand drivers of grassland changes and their spatial heterogeneity at the regional scale of the Tibetan Plateau, but also innovates large-scale vegetation study paradigm. Despite microbes' key roles in driving biogeochemical cycles, the mechanism of microbe-mediated feedbacks to global changes remains elusive. Recently, soil transplant has been successfully established as a proxy to simulate climate changes, as the current trend of global warming coherently causes range shifts toward higher latitudes. Four years after southward soil transplant over large transects in China, we found that microbial functional diversity was increased, in addition to concurrent changes in microbial biomass, soil nutrient content and functional processes involved in the nitrogen cycle. However, soil transplant effects could be overridden by maize cropping, which was attributed to a negative interaction. Strikingly, abundances of nitrogen and carbon cycle genes were increased by these field experiments simulating global change, coinciding with higher soil nitrification potential and carbon dioxide (CO2) efflux. Further investigation revealed strong correlations between carbon cycle genes and CO2 efflux in bare soil but not cropped soil, and between nitrogen cycle genes and nitrification. These findings suggest that changes of soil carbon and nitrogen cycles by soil transplant and cropping were predictable by measuring microbial functional potentials, contributing to a better mechanistic understanding of these soil functional processes and suggesting a potential to incorporate microbial communities in greenhouse gas emission modeling. Aim: The formation of thermokarst lakes could make a large amount of carbon accessible to microbial degradation, potentially intensifying the permafrost carbon‐climate feedback via carbon dioxide/methane emissions. Because of their diverse functional roles, prokaryotes could strongly mediate biogeochemical cycles in thermokarst lakes. However, little is known about the large‐scale patterns and drivers of these communities. Location: Permafrost regions in the Northern Hemisphere. Time period: Present day. Major taxa studied: Prokaryotes. Methods: Based on a combination of large‐scale measurements on the Tibetan Plateau and data syntheses in pan‐Arctic regions, we constructed a comprehensive dataset of 16S rRNA sequences from 258 thermokarst lakes across Northern Hemisphere permafrost regions. We also used the local contributions to beta diversity (LCBD) to characterize the variance of prokaryotic species composition and screened underlying drivers by conducting a random forest modelling analysis. Results: Prokaryotes in thermokarst lake water were dominated by the orders Burkholderiales, Micrococcales, Flavobacteriales and Frankiales. The relative abundance of dominant taxa was positively associated with dissolved organic matter (DOM) properties, especially for the chromophoric/aromatic compounds. Microbial structure differed between high‐altitude and high‐latitude thermokarst lakes, with the dominance of Flavobacterium in high‐altitude lakes, and the enrichment of Polynucleobacter in high‐latitude lakes. More importantly, climatic variables were among the main drivers shaping the large‐scale variation of prokaryotic communities. Specifically, mean annual precipitation was the best predictor for prokaryotic beta diversity across the Northern Hemisphere, as well as in the high‐altitude permafrost regions, while mean annual air temperature played a key role in the high‐latitude thermokarst lakes. Main conclusions: Our findings demonstrate significant associations between dominant taxa and DOM properties, as well as the important role of climatic factors in affecting prokaryotic communities. Permafrost soil in high latitude tundra is one of the largest terrestrial carbon (C) stocks and is highly sensitive to climate warming. Understanding microbial responses to warming-induced environmental changes is critical to evaluating their influences on soil biogeochemical cycles. In this study, a functional gene array (i.e., geochip 4.2) was used to analyze the functional capacities of soil microbial communities collected from a naturally degrading permafrost region in Central Alaska. Varied thaw history was reported to be the main driver of soil and plant differences across a gradient of minimally, moderately, and extensively thawed sites. Compared with the minimally thawed site, the number of detected functional gene probes across the 15-65 cm depth profile at the moderately and extensively thawed sites decreased by 25% and 5%, while the community functional gene β-diversity increased by 34% and 45%, respectively, revealing decreased functional gene richness but increased community heterogeneity along the thaw progression. Particularly, the moderately thawed site contained microbial communities with the highest abundances of many genes involved in prokaryotic C degradation, ammonification, and nitrification processes, but lower abundances of fungal C decomposition and anaerobic-related genes. Significant correlations were observed between functional gene abundance and vascular plant primary productivity, suggesting that plant growth and species composition could be co-evolving traits together with microbial community composition. Altogether, this study reveals the complex responses of microbial functional potentials to thaw-related soil and plant changes and provides information on potential microbially mediated biogeochemical cycles in tundra ecosystems. Summary: Despite the important roles of soil microbes, especially the most diverse rare taxa in maintaining community diversity and multifunctionality, how different climate regimes alter the stability and functions of the rare microbial biosphere remains unknown. We reciprocally transplanted field soils across a latitudinal gradient to simulate climate change and sampled the soils annually after harvesting the maize over the following 6 years (from 2005 to 2011). By sequencing microbial 16S ribosomal RNA gene amplicons, we found that changing climate regimes significantly altered the composition and dynamics of soil microbial communities. A continuous succession of the rare and abundant communities was observed. Rare microbial communities were more stable under changing climatic regimes, with lower variations in temporal dynamics, and higher stability and constancy of diversity. More nitrogen cycling genes were detected in the rare members than in the abundant members, including amoA, napA, nifH, nirK, nirS, norB and nrfA. Random forest analysis and receiver operating characteristics analysis showed that rare taxa may act as potential contributors to maize yield under changing climatics. The study indicates that the taxonomically and functionally diverse rare biosphere has the potential to increase functional redundancy and enhance the ability of soil communities to counteract environmental disturbances. With ongoing global climate change, exploring the succession process and functional changes of rare taxa may be important in elucidating the ecosystem stability and multifunctionality that are mediated by microbial communities. Abstract Global climate change is projected to intensify soil drying-rewetting (DRW) events with extended drought, especially in arid and semiarid ecosystems. However, the extent to which the soil DRW with intensified drought can alter soil respiration (R s) in forests is still under debate, and subsequent legacy effects on R s are not well understood. Here, we conducted a 180-d soil incubation experiment to investigate how soil DRW with different drought intensities alter the R s in poplar (Populus simonii) and Mongolian pine (Pinus sylvestris var. mongolica) plantations. The incubation experiment included four 30-d cycles of 1) constant moisture treatment (control), 2) DRW with 10-d drying and 20-d rewetting (DRW 10-20) or 3) DRW with 20-d drying and 10-d rewetting (DRW 20-10), and then an extended 60-d incubation under constant moisture. During the four DRW cycles, the direct C release with respiration of Mongolian pine soils (27 g C·m−2 in DRW 10-20 and 140 g C·m−2 in DRW 20-10 , respectively) decreased to a much lower extent than that of poplar soils (228 g C·m−2 in DRW 10-20 and 498 g C·m−2 in DRW 20-10 , respectively). R s did not significantly change during the extended 60-d incubation in the DRW 10-20 treatment compared to control treatment. However, the respired CO 2 were increased by 68 g C·m−2 in the poplar soils and 19 g C·m−2 in the Mongolian pine soils in the DRW 20-10 treatment, which approximately compensated for 14% of the decreases of total respiration during four DRW cycles. This legacy effect induced by the DRW with intensified drought was attributed to the higher amount of remaining substrates and soil microbial biomass. Our study highlights that DRW can cause both direct and legacy effects on R s , but the effects vary with drought intensity and forest type. Abstract Climate models predict a substantial increase in the frequency of extreme drought, suggesting subsequent impacts on the carbon (C) and water cycles. Although many studies have investigated the impacts of extreme drought on ecosystem functioning, it remains unknown how the timing of extreme drought within a growing season may affect carbon and water cycling. Here we conducted a 3-year field experiment to investigate the influence of seasonal drought timing on ecosystem carbon and water exchange by excluding rainfall (for consecutive 30 days) during three periods of the growing season (May–June, July–August and August–September) in fenced and grazed sites of a semiarid temperate steppe in Inner Mongolia, China. In the fenced steppe, extreme drought reduced growing-season net CO 2 uptake regardless of drought timing, while in the grazed steppe, early-growing season drought caused relatively larger reductions to net CO 2 uptake than drought imposed later in the season. The effect of extreme drought on evapotranspiration (ET) was similar to that of CO 2 exchange at the fenced site, with consistent reductions of seasonally-integrated ET for all treatments compared with the ambient condition. In contrast, at the grazed site, the response of ET to extreme drought was more variable, possibly due to the absence of litter and greater bare ground. Surprisingly, both gross and net carbon uptake declined with increasing ET at the grazed site, while the fenced site showed the positive water-carbon linkage typically seen in semiarid ecosystems. The different responses of CO 2 and water exchanges for the fenced and grazed sites were regulated predominately by soil temperature and soil water content. Together, our results show that drought timing within the growing season can significantly alter drought impacts on ecosystem water and CO 2 exchanges, and that grazing management may further mediate the response. To understand soil microbial community stability and temporal turnover in response to climate change, a long-term soil transplant experiment was conducted in three agricultural experiment stations over large transects from a warm temperate zone (Fengqiu station in central China) to a subtropical zone (Yingtan station in southern China) and a cold temperate zone (Hailun station in northern China). Annual soil samples were collected from these three stations from 2005 to 2011, and microbial communities were analyzed by sequencing microbial 16S ribosomal RNA gene amplicons using Illumina MiSeq technology. Our results revealed a distinctly differential pattern of microbial communities in both northward and southward transplantations, along with an increase in microbial richness with climate cooling and a corresponding decrease with climate warming. The microbial succession rate was estimated by the slope (w value) of linear regression of a log-transformed microbial community similarity with time (time-decay relationship). Compared with the low turnover rate of microbial communities in situ (w=0.046, P<0.001), the succession rate at the community level was significantly higher in the northward transplant (w=0.058, P<0.001) and highest in the southward transplant (w=0.094, P<0.001). Climate warming lead to a faster succession rate of microbial communities as well as lower species richness and compositional changes compared with in situ and climate cooling, which may be related to the high metabolic rates and intense competition under higher temperature. Regulatory mechanisms of soil respiratory carbon (C) release induced by substrates (i.e., plant derived substrates) are critical for predicting ecosystem responses to climate change, but the mechanisms are not well understood. In this study, we sampled soils from a long-term field manipulative experiment and conducted a laboratory incubation to explore the role of substrate supply in regulating the differences in soil C release among the experimental treatments, including control, warming, clipping, and warming plus clipping. Three types of substrates (glucose, C3 and C4 plant materials) were added with an amount equal to 1% of soil dry weight under the four treatments. We found that the addition of all three substrates significantly stimulated soil respiratory C release in all four warming and clipping treatments. In soils without substrate addition, warming significantly stimulated soil C release but clipping decreased it. However, additions of glucose and C3 plant materials (C3 addition) offset the warming effects, whereas C4 addition still showed the warming-induced stimulation of soil C release. Our results suggest that long-term warming may inhibit microbial capacity for decomposition of C3 litter but may enhance it for decomposition of C4 litter. Such warming-induced adaptation of microbial communities may weaken the positive C-cycle feedback to warming due to increased proportion of C4 species in plant community and decreased litter quality. In contrast, clipping may weaken microbial capacity for warming-induced decomposition of C4 litter but may enhance it for C3 litter. Warming- and clipping-induced shifts in microbial metabolic capacity may be strongly associated with changes in plant species composition and could substantially influence soil C dynamics in response to global change.
