In the central-eastern Tibetan Plateau (TP) there is abundant organic matter in topsoils, which plays a crucial role in determining soil hydraulic properties that need to be properly described in land surface models. Limited soil parameterizations consider the impacts of soil organic matter (SOM), but they still show poor performance in the TP. A dedicated field campaign is therefore conducted by taking undisturbed soil samples in the central TP to obtain in situ soil hydraulic parameters and to advance SOM parameterizations. The observed findings are twofold: 1) The SOM pore-size distribution parameter, derived from measured soil water retention curves, has been demonstrated to be much underestimated in previous studies. 2) SOM saturated hydraulic conductivity is overestimated. Accordingly, a new soil hydraulic parameterization is established by modifying a commonly used one based on observations, which is then evaluated by incorporating it into Noah-MP. Compared with the original ones, the new parameterization significantly improves surface soil liquid water simulations at stations with high surface SOM content, especially in the warm season. A further application with the revised Noah-MP indicates that SOM can enhance sensible heat flux but decrease evaporation and subsurface soil temperature in the warm season and tends to have a much weak effect in the cold season. This study provides insights into the role of SOM in modulating soil state and surface energy budget. Abstract This work was designed to compare the effectiveness of in-situ coagulation and MIEX as pre-treatments prior to ultrafiltration (UF) to improve organic matter (OM) removal and mitigate membrane fouling. Three kinds of OMs, i.e. salicylic acid (SA), humic acid (HA) and bovine serum albumin (BSA) were employed. The experimental results show that coagulation-UF led to most effective removal of HA (almost 90%), while the SA was uncoagulated and least removable, with the rejection rate of about 55%. Conversely, MIEX present superior ability for removing SA, contributing to additional efficiency of 71.95–77.21% than UF alone. Proper dosage of Al-based coagulants could alleviate flux loss, especially in the cases of HA and BSA. Increasing coagulant dose resulted in continuous decrement of irreversible resistance (R ir), which dominated the membrane fouling development by the SA water. For HA and BSA waters, alternatively, variations of R r determined the flux declines. Floc compact degree was the decisive factor for R r for coagulated SA; while for HA and BSA, R r was most related to the floc size and foulant-foulant interaction. MIEX was most effective for alleviating flux loss when treating the hydrophilic SA with small molecules and for all the cases, MIEX exerted little influence on the R r values. Efficient conversion of cellulosic sugars in cellulosic hydrolysates is important for economically viable production of biofuels from lignocellulosic biomass, but the goal remains a critical challenge. The present study reports a new approach for simultaneous fermentation of cellobiose and xylose by using the co-culture consisting of recombinant Saccharomyces cerevisiae specialist strains. The co-culture system can provide competitive advantage of modularity compared to the single culture system and can be tuned to deal with fluctuations in feedstock composition to achieve robust and cost-effective biofuel production. This study characterized fermentation kinetics of the recombinant cellobiose-consuming S. cerevisiae strain EJ2, xylose-consuming S. cerevisiae strain SR8, and their co-culture. The motivation for kinetic modeling was to provide guidance and prediction of using the co-culture system for simultaneous fermentation of mixed sugars with adjustable biomass of each specialist strain under different substrate concentrations. The kinetic model for the co-culture system was developed based on the pure culture models and incorporated the effects of product inhibition, initial substrate concentration and inoculum size. The model simulations were validated by results from independent fermentation experiments under different substrate conditions, and good agreement was found between model predictions and experimental data from batch fermentation of cellobiose, xylose and their mixtures. Additionally, with the guidance of model prediction, simultaneous co-fermentation of 60 g/L cellobiose and 20 g/L xylose was achieved with the initial cell densities of 0.45 g dry cell weight /L for EJ2 and 0.9 g dry cell weight /L SR8. The results demonstrated that the kinetic modeling could be used to guide the design and optimization of yeast co-culture conditions for achieving simultaneous fermentation of cellobiose and xylose with improved ethanol productivity, which is critically important for robust and efficient renewable biofuel production from lignocellulosic biomass. Cassiae Semen, belonging to the family Leguminosae, is derived from the dry mature seeds of Cassia obtusifolia L. or Cassia tora L. and has long been used as a laxative, hepatoprotective, improve eyesight, and antidiabetic complications medicine or functional food in Asia. This review summarizes the integrated research progress of botany, traditional uses, phytochemistry, pharmacology, toxicity, and quality control of Cassiae Semen. Additionally, the emerging challenges and possible developing directions are discussed as well. The information on Cassiae Semen was collected from published scientific materials, including ancient books of traditional Chinese Medicine; Ph.D. and M. Sc. dissertations; monographs on medicinal plants; pharmacopoeia of various countries and electronic databases, such as PubMed, Web of Science, ACS, Science Direct, J-STAGE, Springer link, Taylor, CNKI and Google Scholar, etc. First, the traditional uses and plant origins of Cassiae Semen are outlined. Secondly, approximately 137 compounds, including anthraquinones, naphthopyranones, naphthalenes, flavones, polysaccharides and other compounds, have been isolated and identified from Cassia obtusifolia L. and Cassia tora L. Third, the pharmacological activities and mechanisms of crude extract of Cassiae Semen and its main bioactive compounds are summarized. Moreover, the processing, toxicity, and quality control are introduced briefly. Cassiae Semen is a frequently used Chinese Materia Medica with pharmacological effects that mainly affect the digestive system, cardiovascular systems and nervous system. This review summarized its botany, traditional uses, phytochemistry, and pharmacology, it also exhibited recent scientific research advances and gaps, which provide a deeper insight into the understanding and application of Cassiae Semen. In future research on Cassiae Semen, more attention should be given to the pharmacological activities of naphthopyranones and polysaccharides and the mechanism of action for improving eye diseases. Meanwhile, it is essential to focus on strengthening the study on the pharmacokinetics research and the safety evaluation of related health products research. The fermentation inhibitors from the pretreatment of lignocellulosic materials, e.g., acetic acid and furfural, are notorious due to their negative effects on the cell growth and chemical production. However, the metabolic reprogramming of the cells under these stress conditions, especially metabolic response for resistance to mixed inhibitors, has not been systematically investigated and remains mysterious. Therefore, in this study, 13C metabolic flux analysis (13C-MFA), a powerful tool to elucidate the intracellular carbon flux distributions, has been applied to two Saccharomyces cerevisiae strains with different tolerances to the inhibitors under acetic acid, furfural, and mixed (i.e., acetic acid and furfural) stress conditions to unravel the key metabolic responses. By analyzing the intracellular carbon fluxes as well as the energy and cofactor utilization under different conditions, we uncovered varied metabolic responses to different inhibitors. Under acetate stress, ATP and NADH production was slightly impaired, while NADPH tended towards overproduction. Under furfural stress, ATP and cofactors (including both NADH and NADPH) tended to be overproduced. However, under dual-stress condition, production of ATP and cofactors was severely impaired due to synergistic stress caused by the simultaneous addition of two fermentation inhibitors. Such phenomenon indicated the pivotal role of the energy and cofactor utilization in resisting the mixed inhibitors of acetic acid and furfural. Pharbitidis Semen (the seeds of Ipomoea nil (L.) Roth or Ipomoea purpurea (L.) Roth), a popular traditional Chinese medicine, is also known as "Heichou" or "Baichou" (Chinese: 黑丑, 白丑). It can purge the bowels, promote diuresis, remove stagnated accumulation, and kill worms. It can be used for treating anasarca with constipation and oliguria; dyspnea and cough caused by retained fluid; abdominal pain because of intestinal parasitosis; ascariasis; and taeniasis. This review discusses the botany, ethnopharmacology, phytochemistry, pharmacological activities, toxicology, and quality control of Pharbitidis Semen, to obtain a complete understanding of its effects and provide a basis for further research and the development of new drugs. The literature on Pharbitidis Semen is mainly obtained from pharmacopoeias of different countries, masterpieces of traditional Chinese medicine, Master's and Ph.D. theses, and published articles obtained from literature retrieval websites, such as CNKI, PubMed, SciFinder, WanFang data, Web of Science, Springer, ScienceDirect, Wiley, ACS Publications, Taylor & Francis, J-STAGE, and Google Scholar. Its botany, ethnopharmacology, phytochemistry, pharmacological activities, toxicology, and quality control are discussed to understand its effects and provide a basis for further research. Pharbitidis semen has been used ethnomedically in many tropical and subtropical countries as deobstruents, diuretics, and anthelmintics. About 170 chemical compounds, including terpenoids, phenylpropanoids, resin glycosides, fatty acids and other compounds, have been isolated. It has been reported to have different effects, including laxative, renal-protective, neuroprotective, insecticidal, antitumor, anti-inflammatory, and antioxidant. Moreover, a brief introduction to processing, toxicity, and quality control is provided. The traditional efficacy of Pharbitidis Semen in diarrhea has been confirmed, but its bioactive and toxic ingredients are not entirely clear. It is necessary to strengthen the research and identification of effective parts or natural active components of Pharbitidis Semen, clarify the molecular mechanism of its toxicity and change rule of endogenous substances to make Pharbitidis Semen better used in clinical practice. Additionally, the imperfect quality standard is also a challenge that must be solved urgently. The purpose of this study was to investigate the effects of leaf extract from Lithocarpus polystachyus Rehd. on type II diabetes mellitus (T2DM) and the active ingredients of this effect. In addition, this study determined, for the first time, the underlying molecular and pharmacological mechanisms of the extracts on hyperglycemia using long-term double high diet-fed and streptozotocin (STZ) induced type II diabetic mice. In the present study, leaf extract, phloridzin and trilobatin were assessed in vivo (gavage) and in vitro (non-invasive micro-test technique, NMT) in experimental T2DM mice. The biochemical parameters were measured including blood glucose and blood lipid level, liver biochemical indexes, and hepatic glycogen. The relative expression of glycometabolism-related genes was detected. The effect of leaf extracts on physiological glucose flux in liver tissue from control and T2DM mice was also investigated. Body weight of experimental T2DM mice increased significantly after the first week, but stabilized over the subsequent three weeks; body weight of all other groups did not change during the four weeks’ study. After four weeks, all treatment groups decreased blood glucose, and treatment with leaf extract had numerous positive effects: a) promoted in glucose uptake in liver, b) increased synthesis of liver glycogen, c) reduced oxidative stress, d) up-regulation of glucokinase (GK), glucose transporter 2 (GLUT2), insulin receptor (IR) and insulin receptor substrate (IRS) expression in liver, e) down-regulation of glucose-6-phosphatase (G-6-P) expression, and f) ameliorated blood lipid levels. Both treatment with trilobatin or phloridzin accelerated liver glycogen synthesis, decreased oxidative stress and increased expression of GK. IRS and phosphoenolpyruvate carboxykinase (PEPCK) were both up-regulated after treatment with trilobatin. Expression of GLUT2, PEPCK and G-6-P were also increased in liver tissue after treatment with phloridzin. Our data indicate that leaf extract from L. polystachyus Rehd. has a preferable hypoglycemic effects than trilobatin or phloridzin alone. Leaf extract significantly increased glucose uptake and hepatic glycogen synthesis while also inducing a decline of hepatic gluconeogenesis and oxidative stress in T2DM mice. From this study, we draw conclusions that L. polystachyus promoted glycogen synthesis in T2DM mice, and that the active compounds were not only the trilobatin or phloridzin. [ABSTRACT FROM AUTHOR]

Lacustrine rift basins in Eastern China possess large amounts of petroleum, especially the shale gas and tight sand gas in deep formations, where "sweet spots" are closely related to organic-rich shale intervals. However, the OM enrichment of deep formation shale in lacustrine rift basins and the main controlling factors have not been well-researched in previous studies. The organic matter (OM) enrichment is mainly controlled by the OM input, preservation, and/or dilution. The shales deposited in deep formations of Songliao Basin, including the Jurassic Huoshiling (J 3 h), Cretaceous Shahezi (K 1 sh), and Yingcheng (K 1 yc) formations, are evaluated as fair to excellent source rocks. Taking the shales as an example, the characters and impact of input, preservation, and dilution on OM enrichment in shales are discussed by utilizing the parameters of element concentrations and biomarkers. Results show that the OM sources are mixed sources of aquatic microorganisms and higher plant materials, and the higher plant materials are dominant. The bio-productivity were moderate to high. The paleoclimate was mainly the cold and semi-humid to semi-arid. The lake salinity was fresh-to brackish-water. The redox conditions were sub-reducing to sub-oxidizing. The dilution degree in Yingcheng was the strongest, followed by the Shahezi and Huoshiling. Consequently, the OM enrichment in the J 3 h shale was called moderate bio-productivity and preservation model; the OM enrichment in the K 1 sh shale was called high bio-productivity and preservation model; the OM enrichment in the K 1 yc shale was named high dilution model.
