OBJECTIVES: Traditional treatment methods are becoming popular and commonly used in many societies and have become the first treatment option for most people. While some of these methods are helpful, they can interact with medications the patient is taking for another disease and cause a variety of life-threatening risks. Valerian (catweed) plant is used in traditional medicine as a sleep aid due to its sedative effects. Valerian may also exert anticancer effect in vitro. MATERIALS AND METHODS: In this study, the cytotoxicty and oxidative stress effects of valerian root extract were evaluated in human liver hepatocellular carcinoma (Hepg2) and human colorectal adenocarcinoma (Caco2) cell lines. The cytotoxicity was evaluated via the 3-(4,5-dimethythiazol-2-yl)-2,5-diphenyl tetrazolium bromide test. Total reactive oxygen species analysis was performed via a 2’,7’-dichlorodihydrofluorescein diacetate assay in flow cytometry. RESULTS: Inhibition concentration 50 values were calculated as 936.6 and 1097.5 µg/mL in the Hepg2 and Caco2 cell lines, respectively. It was observed that valerian root extract did not induce oxidative stress in HepG2 and Caco2 cell lines. CONCLUSION: These results indicate that the use of valerian root extract as an alternative method in cancer treatment may not be effective and may cause a risk for public health. On the other hand, it may be safe at recommended tolerated concentrations since it does not cause oxidative stress.

Diabetes mellitus insülin sekresyonunun, insülin etkisinin veya her ikisinin de bozulması sonucu oluşan hiperglisemiyle karakterize metabolik bir hastalıktır. Gerek endüstrileşme, kentleşme ve yaşam alışkanlıkların değişmesi ile artan görülme sıklığı gerekse sosyo-ekonomik etkileri nedeniyle 21. yüzyılın en ciddi sorunlarından biridir. Diyabetin neden olduğu kardiyovasküler hastalıklardan korunma ve tedavide yaşam tarzı değişiklikleri ve glisemik kontrol için oral antidiyabetik kullanımı ve dislipidemi tedavisinde statin kullanımı önemlidir. Yapılan çalışmalarda diyabetin erkek üreme sistemine hasar verdiği gösterilirken atorvastatin ve gliklazid için sonuçlar çelişkilidir.Bu çalışmada 250-300 g ağırlığındaki sağlıklı genç erişkin erkek Sprague-Dawley sıçanlarda streptozotosin ile diyabet indüklendi. Diyabetik sıçanlara 28 gün boyunca oral gavaj yöntemiyle atorvastatin, gliklazid ve atorvastatin/gliklazid kombinasyonu uygulandı. Deney hayvanları 28. günde sakrifiye edilerek kalp kanlarında hormon analizleri, spermde morfolojik, testislerde ışık mikroskobik ve ultrastrüktürel yöntemlerle histolojik incelemeler yapıldı. Elde edilen veriler tek yönlü varyans analizi ile değerlendirildi.Bu çalışmada (i) diyabetin ve tek tek ilaçların hormon seviyelerini değiştirmediği ancak atorvastatin/gliklazid kombinasyonunun testosteron seviyesini azalttığı, (ii) diyabetin sperm sayısını azalttığı, morfolojisini bozduğu, testiste hasar oluşturduğu ve spermatogenezi bozduğu, (iii) atorvastatinin diyabetik sıçanlarda sperm sayısını etkilemediği, testis yapısında iyileşmeye neden olmamamakla birlikte sperm morfolojisini düzelttiği, (iv) gliklazidin diyabetik sıçanlarda sperm sayısını arttırdığı, morfolojisini iyileştirdiği, testis yapısında iyileşme sağladığı ve spermatogenezi düzelttiği, (v) atorvastatin/gliklazid kombinasyonunun ise diyabetik sıçanlarda sperm sayısını etkilemediği, morfolojisini düzelttiği, testis yapısını iyileştirdiği ancak iyileşmenin kontrol düzeyinde olmadığı saptandı. Diyabetin erkek üreme sistemindeki olumsuz etkileri üzerine atorvastatin ve gliklazid kombinasyonunun etkisinin değerlendirildiği bu çalışmadan elde edilen verilerin literatüre katkı sağlayacağına inanılmaktadır. Diabetes mellitus is characterized by hyperglycemia resulting from abnormalities in insulin secretion, insulin action, or both. It is one of the most serious challanges of 21th century because of both increased prevalence due to industrialization, urbanization and changed lifestyle habits, and socio-economic impacts. Lifestyle reforms, treatment of oral antidiabetics for glycemic control and of statins to improve the dyslipidemia are crucial in the prevention and treatment of cardiovascular diseases caused by diabetes. Numerous studies proved that diabetes has severe impacts on male reproductive system, however, data obtained about atorvastatin and gliclazide are conflicted.In this study, diabetes was induced in 250-300 g young adult male Sprague-Dawley rats by streptozotocin. Diabetic rats were given atorvastatin, gliclazide and atorvastatin/gliclazide combination by oral gavage for 28 days. Rats were sacrificed on day 28 and hormone analysis in heart blood, morphological evaluations in sperms, light microscopic and ultrastructural examinations in testes were conducted. The obtained data were analyzed by using one-way ANOVA.The following results were obtained: (i) diabetes and individually the drugs did not change the hormone levels, while atorvastatin/gliclazide combination reduced the testosterone level, (ii) diabetes decreased sperm count, altered sperm morphology, caused damage in testes and impaired spermatogenesis, (iii) in diabetic rats, atorvastatin did not effect sperm count and not improve testes structure, yet ameliorated sperm morphology, (iv) in diabetic rats, gliclazide increased sperm count, ameliorated sperm morphology, improved testes structure and spermatogenesis, (v) in diabetic rats, atorvastatin/gliclazide combination did not effect sperm count, ameliorated sperm morphology, improved testes structure, however, the improvement has not reached to control level. In conclusion, data obtained from the study, which the effects of atorvastatin and gliclazide combination in diabetes-induced damage on male reproductive system were evaluated, is believed to contribute to the literature. 135
