Abstract Background Human cryptosporidiosis is distributed worldwide, and it is recognised as a leading cause of acute diarrhoea and death in infants in low- and middle-income countries. Besides immune status, the higher incidence and severity of this gastrointestinal disease in young children could also be attributed to the digestive environment. For instance, human gastrointestinal physiology undergoes significant changes with age, however the role this variability plays in Cryptosporidium parvum pathogenesis is not known. In this study, we analysed for the first time the impact of digestive physicochemical parameters on C. parvum infection in a human and age-dependent context using a dynamic in vitro gastrointestinal model. Results Our results showed that the parasite excystation, releasing sporozoites from oocysts, occurs in the duodenum compartment after one hour of digestion in both child (from 6 months to 2 years) and adult experimental conditions. In the child small intestine, slightly less sporozoites were released from excystation compared to adult, however they exhibited a higher luciferase activity, suggesting a better physiological state. Sporozoites collected from the child jejunum compartment also showed a higher ability to invade human intestinal epithelial cells compared to the adult condition. Global analysis of the parasite transcriptome through RNA-sequencing demonstrated a more pronounced modulation in ileal effluents compared to gastric ones, albeit showing less susceptibility to age-related digestive condition. Further analysis of gene expression and enriched pathways showed that oocysts are highly active in protein synthesis in the stomach compartment, whereas sporozoites released in the ileum showed downregulation of glycolysis as well as strong modulation of genes potentially related to gliding motility and secreted effectors. Conclusions Digestion in a sophisticated in vitro gastrointestinal model revealed that invasive sporozoite stages are released in the small intestine, and are highly abundant and active in the ileum compartment, supporting reported C. parvum tissue tropism. Our comparative analysis suggests that physicochemical parameters encountered in the child digestive environment can influence the amount, physiological state and possibly invasiveness of sporozoites released in the small intestine, thus potentially contributing to the higher susceptibility of young individuals to cryptosporidiosis.

Historical cars are an important part of the cultural heritage of the last 150 years. Their preservation in technical museums raises the question of how to preserve their primary functionality, namely their mobility. This implies being able to reactivate and to maintain their thermal engines, which are the source of their motion. However, the diversity and complexity of these engines generally require the presence of highly qualified personnel as well as detailed condition reports to assist the conservators. This study proposes to use acoustic emission techniques to facilitate these conservation procedures by objectifying the evaluation of the state of the engines and by providing systematic quantitative indicators for their health monitoring. To illustrate the implementation and the potentialities of this approach, different tests have been carried out at the National Automobile Museum of Mulhouse, on a Renault Type AG1. A dedicated experimental setup and the associated measurement protocol are presented in this paper. The derived results show the ability of this method to detect specific types of engine malfunctions, both during bench test and in situ measurement conditions. A critical discussion is finally proposed to highlight the feasibility and the possibilities of such laboratory techniques in the context of conservation assistance. [ABSTRACT FROM AUTHOR]

It has been claimed that Nigella sativa seeds (NSS), also known as black cumin, have antidiabetic and lipid‐lowering properties. Our pilot study investigated the effects of powdered NSS on insulin secretion and lipid profile in healthy male volunteers. We conducted a double‐blind, randomized, placebo‐controlled 4‐week trial in 30 subjects, receiving NSS powder (1 g/day) or placebo orally (15 subjects/group). Insulin secretion as determined by the hyperglycaemic clamp technique, insulin sensitivity as well as cholesterol and triglycerides serum concentrations, were measured before and after treatment. NSS powder administration was clinically well tolerated. It did not modify fasting glycaemia and insulinaemia, and was ineffective on glucose‐induced insulin secretion and insulin sensitivity. No significant changes on serum lipids were observed after treatment in any treatment groups, nor between the two treatment groups. However, in the treated group only, there was a significant correlation between total cholesterol change after treatment and its baseline level (r = −0.71, P = 0.006, n = 13), and between low‐density lipoprotein (LDL) cholesterol change after treatment and its baseline level (r = −0.74, P = 0.004, n = 13). No such correlations were found for high‐density lipoprotein (HDL) cholesterol, and for triglycerides. These results do not confirm any NSS effect on glucose regulation; however, they suggest that NSS powder may be of interest in lowering lipid concentrations in hyperlipidaemic subjects.
