The rapid loss of viability, coupled with the difficulties and limitations in conserving coffee seeds, are some of the challenges that nurseries are currently facing. Thus, the objective of this work was to analyze the physiological quality of stored Arabica coffee seeds from cultivars recommended for planting in Brazilian mountainous regions. Seeds from 10 Arabica coffee cultivars were used: Catucaí-785/15, Catucaí-2SL, Catucaí-24/137, Japan, Arara, Acauã, Catuaí IAC-81, Mundo Novo IAC-379/19, Catuaí IAC-62, and Caturra IAC-479. The seeds were collected in the Arabica coffee seed production field, in the municipality of Marechal Floriano-ES. They were subsequently processed and dried in the shade, at room temperature, until reaching a humidity of 35 ± 1%, analyzed and stored in a natural laboratory environment (25 ± 2 °C), for 24 months. The following were analyzed: seed water content, germination, germination speed index, electrical conductivity and potassium leaching. The experimental design used was completely randomized, with ten cultivars, two storage times, with four replications of 25 seeds. Storing arabica coffee seeds for 24 months results in a drop in the germination percentage. Stored seeds of the Catucai 24/137 and Arara cultivars showed germination percentages similar to those of newly harvested seeds. Seeds of the Catuai IAC-62 cultivar maintain vigor during storage. The stored seeds of the Caturra IAC-476 and Japi cultivars showed a reduction in physiological quality. Electrical conductivity and potassium leaching tests are efficient in identifying seeds in an advanced state of deterioration. Seeds of Arabica coffee cultivars stored for 24 months, under the conditions of the present study, produce abnormal seedlings.

Palm seedlings are visually selected from mature fruits in a slow process that leads to nonuniform germination and high embryo mortality. In this study, we determined the levels of monosaccharides, their crystallinity, and their role in the formation of Euterpe edulis endosperm during seed maturation. Seeds harvested from 108 to 262 days after anthesis (DAA) were analyzed morphologically, physiologically, and chemically to measure soluble and insoluble lignins, ashes, structural carbohydrates, degree of crystallinity, and endo-β-mannanase. The seeds achieved maximum germination and vigor at 164 DAA. During the early stages, only compounds with a low structural order were formed. The contents of soluble and insoluble lignins, ashes, glucans, and galactans decreased during maturation. Those of mannans, the main structural carbohydrate in the endosperm, increased along with the degree of crystallinity, as suggested by a mannan-I-type X-ray diffraction pattern. Similarly, endo-β-mannanase activity peaked at 262 DAA. The superior physiological outcome of seeds and seedlings at 164 DAA implies a 98-day shorter harvesting time. The production of seedlings of the passion fruit tree, usually, is sexual, and the seeds are not uniform in the seedling emergence, and soaking treatments of seeds can provide faster and more uniform germination. It was aimed to study the action of plant growth regulators and the mobilization of reserves in the stages of soaking of yellow passion fruit seeds. The seeds were soaked for five hours in solutions containing plant growth regulators, in a completely randomized design, in a factorial 8 x 4, with four replications. The first factor corresponds to eight plant growth regulators: T1 - distilled water (control); T2 - 6-benzylaminepurine ​​500 mg L-1; T3 - 4-(3-indolyl) butyric acid 500 mg L-1; T4 - gibberellic acid 500 mg L-1; T5 - spermine 250 mg L-1; T6 - spermine 750 mg L-1; T7 - spermidine 750 mg L-1; T8 - spermidine 1250 mg L-1; and the second factor, to the four soaking times: zero, four, 72 and 120 hours, corresponding, respectively, to the dry seed, and to phases I, II, and III of the imbibition curve. It was evaluated the biochemical composition of seeds (lipids, soluble sugars and starch). The seeds showed accumulation of lipids in phase III; the content of soluble sugars increased in phase I and decreased in phase II. The starch content increased until the phase II and decreased in phase III. Starch is the main reserve in the seeds and the main source of energy used in phase III; soaking the seeds in polyamines generates an accumulation of lipids in the seeds and soaking in plant growth regulators increases the burning of starch.

The use of residues from coffee production to obtain biochar is a sustainable approach, which aims to minimize the environmental impact of these materials. In this study, the effect of adding coffee straw biochar on the physiological quality of lettuce and sorghum seeds was investigated. Thus, the objective of this work was to study the effect of adding different concentrations of coffee biochar in the substrate composition on the physiological quality of lettuce (Lactuca sativa) and sorghum (Sorghum bicolor) seeds. The experimental design used was completely randomized, with five concentrations of biochar (0; 7.5; 15; 30 and 60%), conducted with four replications of 25 seeds. The use of biochar in the concentrations studied does not provide an increase in the average germination percentage and vigor of lettuce and sorghum seeds. The increase in the concentration of biochar caused less seed vigor, suggesting a toxic effect. For seed germination, there was no significant difference between lettuce and sorghum species, regardless of treatment. For the germination speed index, sorghum seeds have higher means, except for the treatment with the addition of 15% coffee straw biochar. Lettuce seeds have higher shoot length averages, except for treatment with 100% commercial substrate. The sorghum seeds have higher mean root length and dry mass than lettuce, regardless of the treatment.

The application of biochar in soil provides various benefits that can vary in intensity as the pyrolysis temperature increases. However, its low density makes this material easily transportable and prone to being removed from the system. The objective of this study was to investigate the pyrolysis temperatures and compression pressure of densified biochar carrier capsules on the physiological quality of Schizolobium parahyba var. amazonicum seeds. Produced at three final pyrolysis temperatures (300, 600, and 900 °C), the biochar was characterized through bulk and true density analyses, immediate composition, pH, electrical conductivity, cation exchange capacity, water-soluble carbon, characterization of organic structures by FTIR, and PAH analysis. Subsequently, the biochar was compacted by briquetting at two compression pressures (50 and 200 psi) with one seed per capsule, and germination, emergence, and quality of generated seedlings were evaluated. After verifying residue normality and variance homogeneity, analysis of variance was conducted following a completely randomized design in a 3 × 2 factorial arrangement, with four replications per treatment and two additional control treatments. Upon identifying significant differences, regression model adjustments were performed. Cluster-based multivariate analysis was used to identify similarities among the studied treatments, both for capsules and controls. Pyrolysis temperature and compression pressure influenced seed germination, emergence, and initial seedling growth. Lower pressure favored shoot development, while higher pressure favored root development and generated seedlings of higher quality. Ateleia glazioveana Baill. is a pioneer, rustic and can be used for forest recovery. This work aimed to study the process of physiological maturation of this species. The research was carried out in the city of Alegre - ES, the trees were identified in the floral anthesis and accompanied during the filling of the fruits and development of the seeds until the complete maturation. The fruits were harvested at the following stages 7, 14, 21, 28, 35 and 42 days after anthesis, and characterized according to: morphometry, moisture, fresh and dry mass of fruits and seeds, germination, germination speed index, shoot and root length and dry mass of seedlings. The regression equations were adjusted for the main characteristics analyzed as a function of the harvest period. The point of physiological maturity of timbó occurred at 42 days after anthesis.

Simultaneous analysis studies of several agronomic traits in cultivated plants make it possible to identify phenotypic and genotypic differences due to environmental variations, such as altitude. Therefore, the objective was to evaluate, through multivariate analysis of agronomic, physicochemical and physiological characters, passion fruit hybrids cultivated in different environments. The hybrids used were Gigante Amarelo, Rubi do Cerrado and Sol do Cerrado, cultivated in the southern region of Espírito Santo in four municipalities/environments: Marataízes (41 m), Jerônimo Monteiro (104 m), Alegre (711 m), and Ibitirama (1016 m). The agronomic characters of the plants, the physical-chemical characteristics of the fruits and the physical, biochemical and physiological qualities of the seeds were analyzed. The Singh method was used to determine the most important differentiating characters between hybrids growing in different environments. Based on these characters, a dissimilarity matrix was generated and a principal coordinate analysis was performed. It was observed that the pulp yield was influenced by altitude. The three hybrids showed greater performance in terms of agronomic characters at altitude (41 m) than at altitude (104 m). The Sol do Cerrado hybrid showed high performance in the physical-chemical characteristics of the fruits at altitude (104 m).

The physical and chemical characteristics of the soil can influence plant growth. When sewage sludge (SS) is applied as a soil fertilizer, the accumulation of non-essential elements contained in it can be toxic for plants. The aim of this study was to understand the effect of SS dosage on the cell cycle of Lactuca sativa L. meristematic cells and on the initial growth of L. sativa and Passiflora alata Curtis. Nine concentrations of SS + distilled water (mg dm -3 ) corresponding to 0, 20, 40, 60, 80, 120, 160, 320, and 520 t ha -1 were tested in four replicates of 25 seeds. Chemical analysis showed an increase in pH of the sludge from 0 to 80 t ha -1 SS followed by its stabilization thereafter. The highest electrical conductivity was observed at 520 t ha -1 SS. SS negatively affected the germination and initial growth of seedlings from P. alata and L. sativa. Cytogenetic analysis on 6000 L. sativa meristematic cells for each treatment revealed that SS could adversely affect the genetic stability of this species. SS concentrations above 120 t ha -1 adversely affected the germination and early seedling growth of L. sativa and P. alata. Sapucaia or Lecythis pisonis Cambess. is an ornamental tree that produces edible, tasty, and nutritious nuts, and can be used for timber production. Sapucaia has potential as a functional food, owing to optimal nutritional and particularly Se levels. The present study sought to characterize the seeds of 21 sapucaia matrices from forest remnants of a neotropical hotspot (Atlantic Forest) and farms in two Brazilian states. Biometrics, germination, vigor, lignin content in the teguments, minerals content, and phenotypic and molecular diversity were analyzed. The seeds of matrices 16 and 21 were the most vigorous. Nuts from matrices 21 and 17 contained the highest amounts of Se. Matrices 5, 8, and 21 were the most phenotypically distant; whereas matrix 21 was the most genetically distant. Importantly, we validated a new non-destructive and efficient X-ray-based methodology for internal and densitometric analysis of sapucaia seeds, and demonstrated a high genetic divergence among matrices., (Copyright © 2020 Elsevier Ltd. The purpose of this study is to evaluate temporal trends in changes in vegetation patterns within the Sooretama Biological Reserve and its surroundings, located in Espirito Santo State, Brazil. The evaluation will be performed using the EVI and NDVI index of the MODIS sensor, the Mann-Kendall monotonic trend, Seasonal Trend Analysis methods, and monitoring drought events through the VCI drought index for the years 2007 through 2015. The tools utilized were the EVI and NDVI indexes of the MOD13Q1 product and LST from the MOD11A2 product. These indices were used in order to represent the dynamics of the study area biomass and then to analyze the drought occurrence using the index best-suited to the area of study, identified as VCI. The temporal trends in the data set were examined, pixel by pixel, by application of the Mann-Kendall monotonic technique, treating each pixel in space as a one-dimensional temporal series of 16-day cycles. To evaluate the seasonal trend, the analysis used the STA technique (Seasonal Trend Analysis) implemented in the ETM module. The characterization and spatial distribution of drought events were performed through the Vegetation Condition Index (VCI). The use of (a) images and seasonal curves produced by the monotonic trend of Mann-Kendall and (b) analysis of seasonal trends generated the response of the vegetation to climate variations. The VCI indicated a potential for drought occurrence analysis in regions and areas with different vegetation densities. So, the VCI can be used as a powerful tool to compose a comprehensive and early system alert of drought that can accompany the changes in spatial coverage of vegetation and severity of change. Lastly, the analysis of the data from the MODIS NDVI, EVI, and TST images indicated that the data is suitable to a space-time analysis of drought occurrences and vegetation trends., (Copyright © 2019 Elsevier Ltd. Somatic embryogenesis from explants from hermaphrodite papaya mother plants is an alternative for the production of true-to-type plants without the need for sexing. This study aimed to analyze hormonal and osmotic inducers in different somatic embryogenesis stages in the commercial hermaphrodite hybrid papaya UENF/Caliman 01. Leaf disks from in vitro shoots originated from ex vitro hermaphrodite plants were cultured in induction medium supplemented with different concentrations of 2,4-D (6, 9, 12, 15, and 18 μM) and 4-CPA (19, 22, 25, 28, and 31 μM). After 90 days, the formation of somatic embryos was verified. The 2,4-D induced the formation of light brown calli with low frequency (20%) of somatic embryogenesis. However, 4-CPA (25 μM) induced 96% of embryogenic calli, which were transferred to maturation medium (MM) and cultured for 30 days. The MM contained ABA (0.5 μM) and AC (15 g L-1) and produced 36.6 somatic embryos callus-1, mainly on cotyledonary stage. Cotyledonary embryos were transferred to germination medium supplemented with gibberellic acid (GA3) (0.0, 1.44, 2.88, and 4.32 µM), and the conversion into plantlets was enhanced with GA3 at 2.88 µM.

Weather phenomena El Niño and La Niña are observed by meteorological variables, which allows you to track climate change and its possible effects in certain regions. The objective of this study was to analyze the behavior of rainfall, temperature and evapotranspiration in the Amazon river basin (Latitudes 5° N to 20° S and Longitudes 50° W to 80° W), comparing them with the occurrence of El Niño and La Niña phenomena, from January 2000 to December 2016. The values referring to the meteorological variables were obtained from the TRMM and MODIS orbital sensors. After data pre-processing, the data were separated into monthly and annual scales and per period according to the presence or absence of El Niño and La Niña phenomena. Based on the results obtained, it was observed that the studied variables were affected by modification of both phenomena. The modifications are more noticeable in the distinction between the more and less rainy periods. Among the variables studied, the evapotranspiration was severely affected in the rainiest months, the La Niña phenomenon, and the least rainy months, El Niño. Thus, it was possible to conclude that, in general, the presence of La Niña increased precipitation values in comparison to the Neutral period, but the inverse occurs in the presence of El Niño. The methodology applied in the present study was adequate for the analysis of the modifications of the meteorological variables coming from the El Niño and La Niña phenomena, being able to be adapted to other variables and regions., (Copyright © 2018 Elsevier B.V. This study evaluated the effect of osmoregulators and carbohydrates on the maturation and germination of somatic embryos of papaya 'Golden THB'. Cotyledon explants from papaya seedlings germinated in vitro on basal MS medium were cultured on somatic embryogenesis induction medium (IM) containing MS salts, myo-inositol, sucrose, agar and p-chlorophenoxyacetic acid. After 50 days, embryogenic calli were transferred onto maturation media (MM) for 45 additional days. For experiment 1, a MS-based medium supplemented with abscisic acid, activated charcoal and concentrations of PEG 6000 (0; 40; 50; 60 and 70 g L-1) was used, whereas for experiment 2 malt extract concentrations (0; 0.1; 0.2; 0.3 and 0.4 g L-1) were assessed. The normal cotyledonary somatic embryos produced in experiment 2 were transferred to the germination medium (GM). The GM consisted of full-strength MS medium, sucrose, agar and was supplemented with myo-inositol at varying concentrations (0; 0.275; 0.55 and 0.825 mM). The PEG concentrations tested impaired the maturation of 'Golden THB' papaya somatic embryos. The MM, supplemented with malt extract at 0.153 g L-1, promoted the greatest development of normal somatic embryos (18.28 SE calli-1), that is, two cotyledonary leaves produced 36.56 SE calli-1. The supplementation with 0.45 mM myo-inositol provided the highest germination percentage (47.42%) and conversion to emblings.

Genetic diversity allows identification of potential intraspecific genotypes in the genus Passiflora. The objective of this study was to examine the morphological and genetic diversity of auxin-induced Passiflora mucronata. The experiments were arranged in a complete randomized block design, with a 9 x 2 factorial arrangement (nine genotypes x presence and absence of auxin, indole-3-butyric acid (IBA)), with four replicates of 16 cuttings. The rooting and vegetative growth responses were variable. Genotype 5 was more responsive in the absence of IBA and genotypes 3, 8 and 9 were more responsive in the presence of IBA. Auxin increased rooting rate and percentage, reducing the average time of root protrusion in eight days. IBA also contributed to increase photosynthesis and dry root and shoot mass in 55.55 and 44.44% of the genotypes, respectively. The highest relative contribution to phenotypic diversity in the absence of auxin was rate (38.75%) and percentage (20.27%) of rooting, whereas in the presence of auxin was stomatal conductance (23.19%) and root dry mass (20.91%). Similarity was found for phenotypic and molecular divergence in the presence of IBA, in which genotypes 1 and 6; genotypes 5, 8 and 9; and genotype 3 were clustered in distinct groups.

The aim of this study was to evaluate somatic embryogenesis in juvenile explants of the THB papaya cultivar. Apical shoots and cotyledonary leaves were inoculated in an induction medium composed of different concentrations of 2,4-D (6, 9, 12, 15 and 18 µM) or 4-CPA (19, 22, 25, 28 and 31 µM). The embryogenic calluses were transferred to a maturation medium for 30 days. Histological analysis were done during the induction and scanning electron microscopy after maturing. For both types of auxin, embryogenesis was achieved at higher frequencies with cotyledonary leaves incubated in induction medium than with apical shoots; except for callogenesis. The early-stage embryos (e.g., globular or heart-shape) predominated. Among the auxins, best results were observed in cotyledonary leaves induced with 4-CPA (25 µM). Histological analyses of the cotyledonary leaf-derived calluses confirmed that the somatic embryos (SEs) formed from parenchyma cells, predominantly differentiated via indirect and multicellular origin and infrequently via synchronized embryogenesis. The secondary embryogenesis was observed during induction and maturation phases in papaya THB cultivar. The combination of ABA (0.5 µM) and AC (15 g L-1) in maturation medium resulted in the highest somatic embryogenesis induction frequency (70 SEs callus-1) and the lowest percentage of early germination (4%).
