Cationic conjugated oligoelectrolytes (COEs) are a class of compounds that can be tailored to achieve relevant in vitro antimicrobial properties with relatively low cytotoxicity against mammalian cells. Three distyrylbenzene-based COEs were designed containing amide functional groups on the side chains. Their properties were compared to two representative COEs with only quaternary ammonium groups. The optimal compound, COE2-3C-C3-Apropyl, has an antimicrobial efficacy against Escherichia coli with an MIC=2 μg mL-1 , even in the presence of human serum albumin low cytotoxicity (IC50 =740 μg mL-1 ) and minimal hemolytic activity. Moreover, we find that amide groups increase interactions between COEs and a bacterial lipid mimic based on calcein leakage assay and allow COEs to readily permeabilize the cytoplasmic membrane of E. coli. These findings suggest that hydrogen bond forming moieties can be further applied in the molecular design of antimicrobial COEs to further improve their selectivity towards bacteria.

A total of eighteen novel 2-(1H-indol-1-yl)-N-(5-phenyl-1,3,4-thiadiazol-2-yl) acetamide derivatives were synthesized through the condensation reaction of 2-(1H-indol-1-yl) acetic acid and 5-phenyl1,3,4-thiadiazol-2-amine via fragment-based design strategy. The target compounds were subjected to screening for antibacterial activity against Xanthomonas oryzae pv. Oryzae, Xanthomonas axonopodis pv. Citri and Pseudomonas syringae pv. Actinidiae. The preliminary antibacterial evaluation of the target compounds showed that some target compounds possessed moderate to good activities against Xoo and Xac. Among them, compound 3p exhibited best inhibition effects against Xoo (77.85% and 62.15%, respectively) and Xac (52.51% and 36.48%, respectively) at 100 lg/mL and 50 lg/mL, which were superior to Thiodiazole-copper and Bismerthiazol. To the best of our knowledge, it is the first report on the design, synthesis, and antibacterial evaluation of novel indole-derived compounds containing 1,3,4-thiadiazole and amide moieties. We have synthesized and characterized a Ru II -OH 2 complex ( 2 ), which has a pentadentate ligand with two pivalamide groups as bulky hydrogen-bonding (HB) moieties in the second coordination sphere (SCS). Complex 2 exhibits a coordination equilibrium through the coordination of one of the pivalamide oxygens to the Ru center in water, affording a η 6 -coordinated complex, 3 . A detailed thermodynamic analysis of the coordination equilibrium revealed that the formation of 3 from 2 is entropy-driven owing to the dissociation of the axial aqua ligand in 2 . Complex 2 was oxidized by a Ce IV salt to produce the corresponding Ru III (OH) complex ( 5 ), which was characterized crystallographically. In the crystal structure of 5 , hydrogen bonds are formed among the NH groups of the pivalamide moieties and the oxygen atom of the hydroxo ligand. Further 1e - -oxidation of 5 yields the corresponding Ru IV (O) complex, 6 , which has intramolecular HB of the oxo ligand with two amide N-H protons. Additionally, the Ru III (OH) complex, 5 , exhibits disproportionation to the corresponding Ru IV (O) complex, 6 , and a mixture of the Ru II complexes, 2 and 3 , in an acidic aqueous solution. We investigated the oxidation of a phenol derivative using complex 6 as the active species and clarified the switch of the reaction mechanism from hydrogen-atom transfer at pH 2.5 to electron transfer, followed by proton transfer at pH 1.0. Additionally, the intramolecular HB in 6 exerts enhancing effects on oxygen-atom transfer reactions from 6 to alkenes such as cyclohexene and its water-soluble derivative to afford the corresponding epoxides, relative to the corresponding Ru IV (O) complex ( 6' ) lacking the HB moieties in the SCS., Competing Interests: The authors declare no competing financial interest., (© 2023 The Authors. A series of novel 2-oxo-2-(arylamino)ethyl 4-formylbenzoates were prepared via the reaction of the potassium salt of p-formylbenzoic acid with 2-chloro-N-aryl-acetamide in DMF at reflux. A new series of 1,4- dihydropyridin-3,5-dicarbonitriles, hexahydroacridine-1,8-diones, decahydropyrimido[4,5-b]quinolines, and hexahydrobenzo[4,5]imidazo[2, 1-b]quinazolines which are linked to N-aryl-benzoyloxyacetamide were obtained through the Hantzsch reaction of various 2-oxo-2-(arylamino)ethyl 4-formylbenzoate with the respective 3-aminocrotononitrile or a mixture of 1,3-dicarbonyl compounds and amine source. Targeting the use of the second harmonic generation (SHG) as a bioimaging technique to unravel the formation of aggregates, the SHG first hyperpolarizabilities (β) of assemblies of benzene-1,3,5-tricarboxamide derivatives have been evaluated at the density functional theory level. Calculations have revealed that i) the assemblies exhibit SHG responses and the total first hyperpolarizability responses of the aggregates are evolving with their size. The largest aggregation effect is a 18-times increase for βHRS of B4 when going from the monomer to the pentamer, that ii) the intrinsic SHG responses described by the hyper-Rayleigh Scattering  are enhanced in presence of iodine atoms on the phenyl core, that iii) the side chains affect the relative orientation of the dipole moment and first hyperpolarizability vectors, which impacts more the EFISHG quantities than their moduli, and that iv) the radial component to β is dominant for the compounds having the largest responses. The transfer model suggests that urea unfolds proteins mainly by increasing the solubility of the amide backbone, probably through urea-induced increase in hydrogen bonding. Other studies suggest that urea addition increases the magnitude of solvent-solute van der Waals interactions, which increases the solubility of nonpolar sidechains. More recent analyses hypothesize that urea has a similar effect in increasing the solubility of backbone and sidechain groups. In this work, we compare the effects of urea addition on the solvation of amides and alkyl groups. At first, we study the effects of urea addition upon solvent hydrogen bonding acidity and basicity through the perturbation in the fluorescence spectrum of probes 1-AN and 1-DMAN. Our results demonstrate that the solvent's hydrogen bonding properties are minimally affected by urea addition. Subsequently, we show that urea addition does not perturb the intra-molecular hydrogen bonding in salicylic acid significantly. Finally, we investigate how urea preferentially interacts with amide and alkyl groups moieties in water by comparing the effects of urea addition upon the solubility of acetaminophen and 4-tertbutylphenol. We show that urea affects amide and t-butyl solubility (lowers the transfer free energy of both amide (backbone) and alkyl (sidechain) groups) in a similar fashion. Flexible displays are a systematic revolution in the field of display, in which high-performance and high-barrier polymer substrates are considered to be one of the most important key materials. In this work, high water vapor barrier polyimides containing amide moieties were synthesized via the ternary polymerization of 4,4′-diaminobenzailide (DABA), 4,4′-diaminodipheny ether (ODA), and 3,3′,4,4′-biphenyl-tetracarboxylic acid dianhydride (BPDA) followed by thermal imidization. The relationship between the content of amide moieties and the water vapor barrier property of the prepared polyimides was studied by means of density test, water absorbing test, water contact angle test, water vapor permeation test, fourier transform infrared spectroscopy (FT-IR), thermogravimetric analysis (TGA), thermogravimetry coupled with fourier transform infrared spectrometry (TG-FTIR), wide-angle X-ray diffraction analysis (WXRD), mechanical performance test, etc. The results show that the introduction of amide groups into polyimide (PI) main chains can improve the water vapor barrier properties of the polyimides effectively. The water vapor transmission rate (WVTR) of the polyimide films can be improved from 8.2365 g·(m2·24 h)−1 to 0.8670 g·(m2·24 h)−1 with the increasing content of amide moieties.

Synthesis of robust covalent macrocycles/cages via multiple amide-bond forming reaction is highly challenging and generally it needs multistep reactions. One-pot reaction of appropriate di-/tri-acyl chloride with a diamine generally results polymers or oligomers instead of discrete architectures. To overcome this limitation, a strategy is reported here using dynamic imine chemistry for facile construction of imine-based macrocycle and cage upon treatment of a diamine with di- and tri-aldehydes respectively, followed by post-synthesis one-step conversion of imine bonds to amides to form the desired robust macrocycle and cage containing multiple amide bonds. While the macrocycle was found to form aggregates in DMSO, the cage was intact without any aggregation. Six amide groups in the confined pocket of the cage made it an ideal receptor for selective binding of fluoride with very high selectivity (∼3 × ${\times }$ 10 3 fold) over chloride, and it was silent towards other halides, phosphate, and other oxyanions., (© 2022 Wiley-VCH GmbH.)

We describe the synthesis and characterization of three novel aromatic diamines containing oxyethylene sequences of different lengths. These diamines were polymerized using the low-temperature solution polycondensation method with isophthaloyl chloride (IPC), terepthaloyl chloride (TPC), [1,1’-biphenyl]-4,4’-dicarbonyl dichloride (BDC), and 4,4′-oxybis(benzoyl chloride) (OBE), obtaining twelve poly(ether amide)s with short segments of polydisperse polyethyleneoxide (PEO) sequences in the polymer backbone. These polymers show reasonably high molecular mass materials (Mw > 12,000), and the relationship between their structure and properties has been carefully studied. Compared with conventional polyamides containing monodisperse PEO sequences, the polydispersity of the PEO segments within the structural units exerts a significant influence on the crystallinity, flexibility, solubility, and the thermal properties of the polymers. For instance, the all-para oriented polyamides (TPCP-A), with an average number of 8.2 ethylenoxide units per structural unit can be transformed conventionally (Tm = 259 °C) in comparison with thermally untransformable polymer with 2 ethylenoxide units (Tm = 425 °C).

Appropriately substituted 2-amino-4-phenylthiazole derivatives were designed and synthesized according to the structural characteristics of crizotinib. The obtained compounds were characterized using 1H NMR, 13C NMR, and HRMS. The target compounds 5a–o were evaluated for their in vitro antiproliferative activity against A549, HeLa, HT29, and Karpas299 human cancer cell lines. Based on results of biological studies, some of these compounds exhibited significant antiproliferative activity. Compound 5b possessed outstanding growth inhibitory effects on the four cell lines, especially for HT29 cell with IC50 value of 2.01 µM. Along with the biological assay data, a molecular docking study suggests that the target compounds were a potential inhibitor.

Two families of organic molecules with different backbones have been considered. The first family is based on a macrolactam-like unit that is constrained in a particular conformation. The second family is composed by a substituted central phenyl that allows a larger mobility for its substituents. They have however a common feature, three amide moieties (within the cycle for the macrolactam-like molecule and as substituents for the phenyl) that permit hydrogen bonding when molecules are stacked. In this study we propose a computational protocol to unravel the ability of the different families to self-assemble into organic nanotubes. Starting from the monomer and going towards larger assemblies like dimers, trimers, and pentamers we applied the different protocols to rationalize the behavior of the different assemblies. Both structures and thermodynamics were investigated to give a complete picture of the process. Thanks to the combination of a quantum mechanics approach and molecular dynamics simulations along with the use of tailored tools (non covalent interaction visualization) and techniques (umbrella sampling), we have been able to differentiate the two families and highlight the best candidate for self-assembling purposes.

The control of the localization of the macrocycle, which surrounds a molecular axle, along the thread leads to translational isomers of very different properties. Although many moieties have been used as sites of interactions for crown ethers, the very straightforwardly obtained amide motif has more rarely been envisaged as molecular station. In this article, we report the use of secondary and tertiary amide moieties as efficient secondary molecular station in pH-sensitive molecular shuttles. Depending on the N-substitution of the amide station, and on deprotonation or deprotonation-carbamoylation, the actuation of the molecular machinery differs accordingly to very distinct interactions between the axle and the DB24C8.

Dipeptidyl peptidase-IV (DPP-4) is a validated target for T2DM treatment. We previously reported a novel series of triazole-based uracil derivatives bearing aliphatic carboxylic acids with potent DPP-4 inhibitory activities in vitro, but these compounds showed poor hypoglycemic effects in vivo. Herein we further optimized the triazole moiety by amidation of the carboxylic acid to improve in vivo activities. Two series of compounds 3a-f and 4a-g were designed and synthesized. By screening in DPP-4, compound 4c was identified as a potent DPP-4 inhibitor with the IC 50 value of 28.62 nM. Docking study revealed compound 4c has a favorable binding mode and interpreted the SAR of these analogs. DPP-8 and DPP-9 tests indicated compound 4c had excellent selectivity over DPP-8 and DPP-9. Further in vivo evaluations revealed that compound 4c showed more potent hypoglycemic activity than its corresponding carboxylic acid in ICR mice and dose-dependently reduced glucose levels in type 2 diabetic C57BL/6 mice. Dipyrrolyldiketone BF 2 complexes with amide units at pyrrole α-positions were synthesized in modest yields and showed extremely high anion-binding affinities because of multiple hydrogen-bonding interactions. The phenyl-amide-substituted derivative formed solid-state chiral helical structures, wherein π planes were oriented parallel to the helical directions. Examination of anion-binding behavior using 1 H NMR spectroscopy combined with theoretical studies suggested fast pyrrole inversions, particularly for the phenyl-amide-substituted derivative. The previous studies in a series of 8-methoxy-1,3-dimethyl-2,6-dioxo-purin-7-yl derivatives revealed their analgesic properties. We extended the study with these compounds in aim to assess their impact on inflammatory process. For this purpose we used: the zymosan-induced peritonitis and the carrageenan-induced edema model. Furthermore, the antioxidant activity of the investigated compounds by the FRAP assay was determined. For the most active derivatives from evaluated series their influence on plasma TNF-α level was also tested in vivo. All investigated purine-2,6-dione derivatives 1-11 decreased neutrophils count and inhibited intensity of early vascular permeability. Furthermore, all evaluated compounds reduced the volume of edema caused by subcutaneous injection of carrageenan. Derivatives 1 (with ester moiety), 3 and 4 (with carboxylic group) showed the highest activity in the zymosan-induced peritonitis. In addition, a significant inhibition of plasma TNF-α level in rats with endotoxemia was observed following intraperitoneal administration of these compounds. In turn, compounds 6 and 8-11 containing amide moiety showed the greatest anti-inflammatory (antiedematous) effect in the carrageenan-induced paw edema model. All compounds did not show significant antioxidant properties. The present studies revealed that the presented purine-2,6-dione derivatives exhibit a significant anti-inflammatory activity and this effect may result from their ability to lower TNF-α level.
