The work presented in this thesis centres on the synthesis and measurements of paramagnetic rotaxane molecules consisting of [Cr₇NiF₈(O₂C^tBu)₁₆]⁻ {Cr₇Ni} octametallic rings. The possibility of using {Cr₇Ni} rings as qubits has triggered interest from research chemists. The aim of the project is to synthesise molecules which connect {Cr₇Ni} rings and dissimilar qubits via an ammonium thread. Paramagnetic NMR is used to investigate the relative solution stabilities for a series of pseudorotaxanes by exchanging the ammonium thread. An understanding of the binding preferences of the octametallic macrocycles is important in the design of more complex molecular assemblies. This work begins by exploring the binding preferences between a [Cr₇CoF₈(O₂CCH₂^tBu)₁₆]⁻ macrocycle and dialkylammonium cations through equilibrium constants, quantified using paramagnetic NMR. The study concludes that the relative stability of a pseudorotaxane is dependent upon the structure of the ammonium cation and solvent used. Knowledge of the molecules' relative stabilities was used to synthesise new pseudorotaxanes via an indirect route in high yields (> 80 %). The observations are the first step in the design of more elaborate [n]rotaxane molecules and dynamic systems. A series of two- and three-spin [2]rotaxanes have been synthesised involving [Cr₇NiF₈(O₂C^tBu)₁₆]⁻ {Cr₇Ni} macrocycles, charge-balanced by ammonium threads terminated with metal salen groups (where M = Cu, Ni, VO). Each compound has been characterised using single crystal X-ray diffraction. One of the complexes synthesised includes a three-hetero-spin [2]rotaxane comprising three spin = ½ centres. Continuous wave and pulsed electron paramagnetic resonance (EPR) experiments were implemented to investigate the structure of the molecules in solution. The inter-spin distances obtained from pulsed EPR experiments are consistent with those observed in the crystal. A twelve-membered nickel cluster has been used to bring multiple {Cr₇Ni} rings together to form a series of [7]rotaxanes. The structure of each molecule differs in the crystal depending on the structure of the coordinated thread. Small angle X-ray scattering supported by molecular dynamic simulations were used to investigate the structure of the molecules in solution. The results indicate that the molecule is stable in solution but adopts conformations which differ to that in the crystal.

Hornworts comprise a bryophyte lineage that diverged from other extant land plants >400 million years ago and bears unique biological features, including a distinct sporophyte architecture, cyanobacterial symbiosis and a pyrenoid-based carbon-concentrating mechanism (CCM). Here, we provide three high-quality genomes of Anthoceros hornworts. Phylogenomic analyses place hornworts as a sister clade to liverworts plus mosses with high support. The Anthoceros genomes lack repeat-dense centromeres as well as whole-genome duplication, and contain a limited transcription factor repertoire. Several genes involved in angiosperm meristem and stomatal function are conserved in Anthoceros and upregulated during sporophyte development, suggesting possible homologies at the genetic level. We identified candidate genes involved in cyanobacterial symbiosis and found that LCIB, a Chlamydomonas CCM gene, is present in hornworts but absent in other plant lineages, implying a possible conserved role in CCM function. We anticipate that these hornwort genomes will serve as essential references for future hornwort research and comparative studies across land plants.

5. Creating a Culture: How School Leaders Can Optimise Behaviour

- Abstract
The national picture of school behaviour is complex, but numerous indicators suggest that it can be better in a great number of schools and contexts. Every leader should consciously aspire to the very best behaviour possible in their schools as a matter of priority. There are a number of strategies that schools with outstanding behaviour use frequently, and these should be shared and made available to all school leaders in order for them to decide if they are appropriate for their schools. This report is an attempt to distill the experience and wisdom of practitioners, for the most part still current in their field. As a result, it is authoritative but not definitive or exhaustive. While its conclusions are a distillation of very broad experiential constituencies, it is also a meditative document. Its observations and recommendations are designed to be practical and represent the real issues faced in school leadership, and the very great possibilities offered by improvement in the area of whole school behaviour. Mono‐ and bis‐salen functionalised [2]rotaxanes have been synthesised from the esterification of [2]rotaxanes containing phenol‐terminated threads (salen=N,N′‐bis(salicylidene)ethylenediamine). The [2]rotaxanes have general formula [RH][Cr7NiF8(O2CtBu)16], where [RH]+ is a thread with a central secondary ammonium site that templates a [Cr7NiF8(O2CtBu)16]− ring. The threads are terminated at one or both ends by carboxylic acid functionalised salen groups. The {M(salen)} groups can be free‐base [M=(H+)2] or metallated [M=Cu2+, Ni2+, (VO)2+]. The [2]rotaxanes have been characterised by single crystal XRD and solid‐ and solution‐state EPR spectroscopy. Where two paramagnetic M ions are involved [M=Cu2+ and/or (VO)2+] the [2]rotaxanes contain three electron spin S=1/2 ${{ 1/2 }}$ centres, since the {Cr7Ni} ring has an S=1/2 ${{ 1/2 }}$ ground state which is well isolated at low temperatures. These three‐spin [2]rotaxanes have been characterised in solution by pulsed dipolar EPR spectroscopies (DEER, also known as PELDOR, and RIDME). The M⋅⋅⋅M and M⋅⋅⋅{Cr7Ni} interactions measured are consistent with dipolar interactions and also with the distances from single crystal XRD. The authors declare no competing financial interest. SUMMARY: Strigolactones (SLs) are key regulators of shoot growth and responses to environmental stimuli. Numerous studies have indicated that nitrogen (N) limitation induces SL biosynthesis, suggesting that SLs may play a pivotal role in coordinating systemic responses to N availability, but this idea has not been clearly demonstrated. Here, we generated triple knockout mutants in the SL synthesis gene TaDWARF17 (TaD17) in bread wheat and investigated their phenotypic and transcriptional responses under N limitation, aiming to elucidate the role of SLs in the adaptation to N limitation. Tad17 mutants display typical SL mutant phenotypes, and fail to adapt their shoot growth appropriately to N. Despite exhibiting an increased tillering phenotype, Tad17 mutants continued to respond to N limitation by reducing tiller number, suggesting that SLs are not the sole regulators of tillering in response to N availability. RNA‐seq analysis of basal nodes revealed that the loss of D17 significantly altered the transcriptional response of N‐responsive genes, including changes in the expression profiles of key N response master regulators. Crucially, our findings suggest that SLs are required for the transcriptional downregulation of cytokinin (CK) synthesis and signalling in response to N limitation. Collectively, our results suggest that SLs are essential for the appropriate morphological and transcriptional adaptation to N limitation in wheat, and that the repressive effect of SLs on shoot growth is partly mediated by their repression of CK synthesis. Significance Statement: This research investigates the key role of strigolactones (SLs) in bread wheat tillering and adaptation to nitrogen limitation. Our findings using TaDWARF17 mutants reveal that SLs, while not the sole regulators of tillering under nitrogen limitation, crucially mediate the transcriptional downregulation of cytokinin synthesis and signalling, underscoring the importance of SLs in coordinating morphological and transcriptional responses to nitrogen availability in wheat. 9. The School Research Lead

- Abstract
The researchED movement has generated a new debate about the role of research in schools. Of course there have always been teachers interested in undertaking research and applying the research findings of others. However, involvement in research has tended to be the personal enthusiasm of the individual teacher rather than a coordinated whole school endeavour. One of the many exciting aspects of the current debate that researchED has stimulated is the emergence of the idea of the Research Lead. Many schools now have a key individual who takes a whole school view of the use of evidence. So how should new Research Leads see their role? This report addresses the question in two ways. Tom Bennett, the founder of researchED, has written this thought-provoking essay that sets out some of the ways in which the work of the Research Lead can be conceptualised. He explores a range of different approaches to research leadership. In addition to his tour of the horizon, several pioneering Research Leads have provided fascinating case studies of research leadership in action in their own schools. Both his analysis and the vignettes from the schools collectively make a powerful case for the new role and the potential benefits in terms of better teaching and learning. The different contributions also make it clear that there is no single blueprint that schools should follow as they explore research engagement. The author provides a deliberately diverse set of possible interpretations of research leadership. The unifying factor that is present in all of his approaches is a strong sense of research as a collective and collegiate endeavour. Similarly the case studies, while illustrating different styles of research leadership, are linked by a shared view of the school as a professional learning community. This is an important and timely report. Our understanding of teacher professionalism is in flux and we have a chance to re-define teaching as a research-engaged calling. The new cadre of Research Leads can influence this and make sure that our view of evidence is securely institutional rather than individualistic. [Introduction by Tony McAleavy. 10. Evolution of strigolactone receptors by gradual neo-functionalization of KAI2 paralogues

BackgroundStrigolactones (SLs) are a class of plant hormones that control many aspects of plant growth. The SL signalling mechanism is homologous to that of karrikins (KARs), smoke-derived compounds that stimulate seed germination. In angiosperms, the SL receptor is an α/β-hydrolase known as DWARF14 (D14); its close homologue, KARRIKIN INSENSITIVE2 (KAI2), functions as a KAR receptor and likely recognizes an uncharacterized, endogenous signal ('KL'). Previous phylogenetic analyses have suggested that the KAI2 lineage is ancestral in land plants, and that canonical D14-type SL receptors only arose in seed plants; this is paradoxical, however, as non-vascular plants synthesize and respond to SLs.ResultsWe have used a combination of phylogenetic and structural approaches to re-assess the evolution of the D14/KAI2 family in land plants. We analysed 339 members of the D14/KAI2 family from land plants and charophyte algae. Our phylogenetic analyses show that the divergence between the eu-KAI2 lineage and the DDK (D14/DLK2/KAI2) lineage that includes D14 occurred very early in land plant evolution. We show that eu-KAI2 proteins are highly conserved, and have unique features not found in DDK proteins. Conversely, we show that DDK proteins show considerable sequence and structural variation to each other, and lack clearly definable characteristics. We use homology modelling to show that the earliest members of the DDK lineage structurally resemble KAI2 and that SL receptors in non-seed plants likely do not have D14-like structure. We also show that certain groups of DDK proteins lack the otherwise conserved MORE AXILLARY GROWTH2 (MAX2) interface, and may thus function independently of MAX2, which we show is highly conserved throughout land plant evolution.ConclusionsOur results suggest that D14-like structure is not required for SL perception, and that SL perception has relatively relaxed structural requirements compared to KAI2-mediated signalling. We suggest that SL perception gradually evolved by neo-functionalization within the DDK lineage, and that the transition from KAI2-like to D14-like protein may have been driven by interactions with protein partners, rather than being required for SL perception per se.

13. Group Work for the Good: Unpacking the Research behind One Popular Classroom Strategy

- Abstract
As a philosophy and religious studies high school teacher in the United Kingdom, Tom Bennett discovered that a good deal of what was considered orthodoxy in the profession was unsubstantiated. It was the inspiration for his book, "Teacher Proof: Why Research in Education Doesn't Always Mean What It Claims, and What You Can Do about It," on which this article is based. The book bluntly exposes some of the bigger education myths that still rattle their chains in the classroom. Each chapter is devoted to a questionable educational theory; Bennett examines each claim made for its efficacy by simply following the research crumbs backward. He argues here that one of the most enduring myths he has encountered in education is the subject of this article: group work. Here he presents a discussion of the myth, and everyday realities of group work, and highlights 4 of the usually ignored drawbacks of group work. The article closes with Bennett's recommendation to his colleagues that they use group work when they feel it is appropriate to the task they want their students to achieve and at no other time.

16. What if There Is Another Way? Year 7 Use Diagrammatic Representations of Counterfactuals to Develop Their Causal Reasoning

Tom Bennett begins his article with a tale of a frustrating afternoon with Year 7. We've all been there. In his case, his frustration was caused by his finding a conceptual gap between how well his class wanted to do and the actual quality of their causal thinking. Bennett decided to use counterfactuals to improve their thinking. This article shows how his own thinking changed, too. He analyses why his students had struggled with the Battle of Hastings, and then shows the process by which he decided what to do about it, absorbing and adapting the ideas in the literature. The result is a discussion of how to use counterfactual diagrams and formulae, and improved student understanding of what (and Wat) caused the Peasants' Revolt.

17. Cytokinin and reproductive shoot architecture: bigger and better?

Cytokinin (CK) is a key plant hormone, but one whose effects are often misunderstood, partly due to reliance on older data from before the molecular genetic age of plant science. In this mini-review, we examine the role of CK in controlling the reproductive shoot architecture of flowering plants. We begin with a long overdue re-examination of the role of CK in shoot branching, and discuss the relatively paucity of genetic evidence that CK does play a major role in this process. We then examine the role of CK in determining the number of inflorescences, flowers, fruit and seed that plants initiate during reproductive development, and how these are arranged in space and time. The genetic evidence for a major role of CK in controlling these processes is much clearer, and CK has profound effects in boosting the size and number of most reproductive structures. Conversely, the attenuation of CK levels during the reproductive phase likely contributes to reduced organ size seen later in flowering, and the ultimate arrest of inflorescence meristems during end-of-flowering. We finish by discussing how this information can potentially be used to improve crop yields. [ABSTRACT FROM AUTHOR]

18. Studying Cation Exchange in {Cr7Co} Pseudorotaxanes: Preparatory Studies for Making Hybrid Molecular Machines.

In the design of dynamic supramolecular systems used in molecular machines, it is important to understand the binding preferences between the macrocycle and stations along the thread. Here, we apply 1H NMR spectroscopy to investigate the relative stabilities of a series of linear alkylammonium templated pseudorotaxanes with the general formula [H2NRR'][Cr7CoF8(O2CCH2tBu)16] by exchanging the cation in solution. Our results show that the pseudorotaxanes are able to exchange threads via a dissociative mechanism. The position of equilibrium is dependent upon the ammonium cation and solvent used. Short chain primary ammonium cations are shown to be far less favourable macrocycle stations than secondary ammonium cations. Collision‐induced dissociation mass spectrometry (CID‐MS) has been used to look at disassembly of the pseudorotaxanes in a solvent‐free environment and stability trends compared to those in acetone‐d6. The energy needed to induce 50 % of the precursor ion loss (E50) is used and shows a similar trend to the equilibria measured by NMR. The relative stabilities of these hybrid inorganic‐organic pseudo‐rotaxanes are different to those of host‐guest compounds involving crown ethers and this may be valuable for the design of molecular machines. The rhizosphere is a complex physical and chemical interface between plants and their underground environment, both biotic and abiotic. Plants exude a large number of chemicals into the rhizosphere in order to manipulate these biotic and abiotic components. Among such chemicals are strigolactones, ancient signalling molecules that in flowering plants act as both internal hormones and external rhizosphere signals. Plants exude strigolactones to communicate with their preferred symbiotic partners and neighbouring plants, but at least some classes of parasitic organisms are able to 'crack' these private messages and eavesdrop on the signals. In this review, we examine the intentional consequences of strigolactone exudation, and also the unintentional consequences caused by eavesdroppers. We examine the molecular mechanisms by which strigolactones act within the rhizosphere, and attempt to understand the enigma of the strigolactone molecular diversity synthesized and exuded into the rhizosphere by plants. We conclude by looking at the prospects of using improved understanding of strigolactones in agricultural contexts. [ABSTRACT FROM AUTHOR]

The α/β-hydrolase signalling protein KARRIKIN-INSENSITIVE2 (KAI2), a close homologue of the strigolactone receptor DWARF14 (D14), is involved in this process, but it is unclear how the effects of KAI2 on development are mediated. Here, using a combination of physiological, pharmacological, genetic and imaging approaches in Arabidopsis thaliana (Heynh.) we show that kai2 phenotypes arise because of a failure to downregulate auxin transport from the seedling shoot apex towards the root system, rather than a failure to respond to light per se. We demonstrate that KAI2 controls the light-induced remodelling of the PIN-mediated auxin transport system in seedlings, promoting a reduction in PIN7 abundance in older tissues, and an increase of PIN1/PIN2 abundance in the root meristem. We show that removing PIN3, PIN4 and PIN7 from kai2 mutants, or pharmacological inhibition of auxin transport and synthesis, is sufficient to suppress most kai2 seedling phenotypes. We conclude that KAI2 regulates seedling morphogenesis by its effects on the auxin transport system. We propose that KAI2 is not required for the light-mediated changes in PIN gene expression but is required for the appropriate changes in PIN protein abundance within cells.

- Abstract
The increasing deployment of renewable energy sources and greater electrification of demand is requiring more frequent use of load following in pressurised water reactors (PWRs) . A limiting aspect with respect to load following is the thermo-mechanical behaviour of the fuel. Hence a greater use of best-estimate multi-physics tools that can more accurately capture such behaviour during load following manoeuvres – in particular, extended reduced power operation (ERPO) – has become important in recent years. To this end, we have developed a coupled, whole-core analysis framework and demonstrated it with a new generic PWR model. The demonstration is for uncontrolled RCCA bank withdrawal at power (URWAP) faults following a period of ERPO, since such scenarios are often limiting with respect to fuel integrity. The coupled, whole-core analysis framework that was developed consists of the PARCS neutronics code, the RELAP and CTF thermal–hydraulics codes, the ENIGMA fuel performance code, and the NEXUS platform for whole-core fuel performance. The generic PWR model constructed is for a 1240 MWe plant with a modern core design (maximum assembly burnup of 51.0 GWd/tHM, cycle length of 16 months, and an average fuel residence time of 2.3 cycles per assembly). Previous work in the literature on URWAP faults has focused on coupled neutronics and thermal–hydraulics analysis, with fuel performance assessment and the effects of load following neglected. These shortcomings are therefore addressed here. From a fuel performance perspective no fuel was predicted to fail due to pellet-clad interaction (PCI). Of the remaining failure indicators, margin to clad yield stress was most limiting, although no failure was predicted in all cases under the scenarios considered., Peer Reviewed, Postprint (published version)
