- In this study we used simulation of genetic parameters based on microsatellite data to investigate the reproductive system of three Hymenaea courbaril L. species populations. Different selfing, migration and clonal reproduction rates were tested using the Easypop software. The results suggest the presence of both sexual and clonal reproduction in these populations. The simulated populations showed gradual decrease of the genetic diversity due the low gene flow caused by fragmentation. Due to connectivity importance, it is suggested expanding the area and replacing the explored trees using high genetic diversity germplasm in order to minimize the negative effects of fragmentation.

O objetivo do presente trabalho foi avaliar a influência da temperatura e do tamanho da semente na germinação de Plinia peruviana. As sementes foram classificadas em três tamanhos: pequenas (≤ 0,6 cm de diâmetro), médias (0,6 - 1,0 cm) e grandes (≥ 1,5 cm), sendo semeadas em caixas plásticas contendo como substrato vermiculita e Plantimax® (1:1). As caixas foram distribuídas em duas condições de temperatura: ambiente (22 ºC ± 2) e estufa (30 ºC ± 2). O experimento foi conduzido em delineamento inteiramente casualizado. Após 45 dias, avaliou-se a germinação (%), índice de velocidade de germinação (IVG), tempo médio de germinação (TMG), velocidade média de germinação (VMG), número de folhas por plântula (NF), altura da parte aérea das plântulas (APA) e comprimento da maior raiz (CMR). As maiores médias em todas as variáveis foram obtidas com sementes grandes, exceto para TMG. A temperatura de 30 °C ± 2 ocasionou maior IVG, TMG e VMG. Sementes grandes resultaram em mudas de melhor qualidade e quando colocadas para germinar em estufa originaram plântulas com maior NF, APA e CMR. As temperaturas mais elevadas de germinação promovem o desenvolvimento mais rápido de Plinia peruviana.

ABSTRACT Eugenia involucrata is an important Brazilian forest species, which it can be used for timber, such as firewood and charcoal, and non-timber purposes, for culinary and medicinal use. Considering the limitations of its seminal reproduction, vegetative propagation, particularly by micropropagation, becomes an alternative for plantlets production. In this study we evaluated the effect of the phytoregulators 6-benzylaminopurine (BAP) and 3-indolebutyric acid (IBA) on the in vitro multiplication stage of nodal segments. The experiment was carried out in a completely randomized design, in a 3 x 4 factorial array, the main factors were BAP (0; 0.1 or 0.2 µM) and IBA (0; 10; 20 or 30 µM). After 60 days of in vitro inoculation, evaluations on the the number of buds, number of shoots, number of leaves, callus formation (%) and number of roots per explant were carried out. BAP had not significant effect, however, the use of IBA alone favours the callogenesis and negatively affected leaves, shoots and roots formation. A moderate multiplication rate was obtained when considering in vitro bud formation. BAP and IBA are dispensable in in vitro multiplication of E. involucrata.

ABSTRACT Gene flow allows connectivity of geographically isolated populations and which genetic diversity is reduced. Understanding how gene flow occurs is essential to conservation and selection of priority populations for intervention. In this study, the rates of migration and selfing of Copaifera langsdorffii Desf. sampled in Cerrado fragments in the State of São Paulo, Brazil, were estimated through computer simulations. Different selfing and migration rates were tested and the combination that showed values of observed and expected heterozygosity closest to those previously obtained with microsatellite markers was selected. According to the simulations, selfing and migration were low. Due to the high geographical distances among the fragments, gene flow was limited and may have led to low genetic diversity in the fragments. It is recommend enriching the area with high genetic diversity germplasm and reducing disturbance to promote the occurrence of pollinators, in order to increase the genetic diversity.

A descontaminação dos explantes é requisito para a eficiência da micropropagação. O trabalho teve como objetivo avaliar o modo de utilização do antibiótico sulfato de estreptomicina (SE) no controle de bactérias endógenas em segmentos nodais micropropagados de Eugenia involucrata. Os tratamentos foram: T1 (explantes sem contaminação prévia); T2 (explantes com contaminação prévia); T3 (explantes sem contaminação e imersos em solução de SE a 100 mg L-1 durante 5 min); T4 (explantes com contaminação e imersos em solução de SE a 100 mg L-1 durante 5 min); T5 (explantes sem contaminação e inoculados em meio contendo 100 mg L-1 de SE); e T6 (explantes com contaminação e inoculados em meio contendo 100 mg L-1 de SE). Decorridos 30 dias de cultivo in vitro não houve efeito significativo sobre as variáveis sobrevivência (média de 94,44%) e contaminação fúngica (média 6,94%). As maiores contaminações bacterianas foram observadas em T3 e T4 (100%), já em T5 (16,67%) observou-se a menor média. Ocorreu maior número de folhas em T5 (3,83), que diferiu estatisticamente de T2 e T4, os quais apresentaram a menor média. A imersão não controla a proliferação de bactérias. A inoculação na ausência de contaminação prévia controla a proliferação, mas não reduz as bactérias quando há contaminação. O desenvolvimento das folhas é prejudicado pela presença de bactérias.

Eugenia involucrata é uma espécie florestal nativa do Brasil, com grande potencial para uso madeireiro, frutícola e medicinal. Esse trabalho teve como objetivo avaliar o efeito de ácido naftalenoacético (ANA) e cinetina (CIN) na sua multiplicação in vitro. Foram testadas diferentes combinações de ANA e CIN. Após 90 dias de cultivo in vitro foi observada uma média geral de 1,19 gemas por explante, sendo que o maior número de brotos (1,33) e folhas (3,38) foram obtidos a 2 μM de ANA. Essa concentração de ANA é importante na multiplicação in vitro de E. involucrata, sendo dispensável sua associação com cinetina.

ABSTRACT: Mangaba tree is a fruit tree species whose natural populations are fragmented by anthropic actions. For this reason, studies assessing the impact of fragmentation on the diversity and genetic structure of these populations are required in order to establish suitable conservation strategies. In our study, we used data from analyzes through microsatellite markers in computer simulations to estimate the rates of migration and selfing of six mangaba populations. The studied populations are located in the northeastern states of Ceará, Pernambuco and Sergipe. We tested different selfing and migration rates and selected the combination that showed values of observed and expected heterozygosity closest to those previously obtained with microsatellite markers. According to our simulations, selfing and migration were moderate. This may have led to an increase in inbreeding and genetic drift, resulting in low genetic diversity. We recommend expanding the area and reducing disturbance to promote the occurrence of pollinators, which play an important role in increasing genetic diversity.

O objetivo desse trabalho foi avaliar o efeito do sorbitol e do período de cultivo in vitro sobre o crescimento mínimo e a subsequente retomada do crescimento de brotações de Luehea divaricata. Foram realizados dois ensaios, um de crescimento mínimo e o outro de retomada de crescimento. No crescimento mínimo, observou-se redução nas médias de sobrevivência, estabelecimento, número de folhas e de raízes primárias e secundárias na presença de sorbitol. Quando se considerou o período, observou-se redução aos 30 dias seguida de aumento e estabilização nas médias de raízes primárias e secundárias e do número de folhas e de brotos. Na retomada de crescimento, a presença de sorbitol promoveu uma redução significativa nas médias de sobrevivência, estabelecimento, número de folhas e de brotos. Somente a partir dos 120 dias observou-se um decréscimo das médias na sobrevivência e estabelecimento. A presença de sorbitol é importante para realizar a conservação in vitro de L. divaricata via crescimento mínimo, o qual pode ser realizado por um período de até 120 dias. A subsequente retomada de crescimento das brotações conservadas na presença de sorbitol é limitada a 90 dias.

O objetivo do presente trabalho foi avaliar o efeito do período de cultivo na micropropagação de brotações de Luehea divaricata. Os tratamentos foram compostos pelos períodos de cultivo in vitro: 14; 21; 28; 40; 56; 70; 84; 102; 116; 130; 154, 186 ou 200 dias, sem realizar subcultivo ou transferência para outro meio nutritivo fresco. A sobrevivência e o estabelecimento ficaram em torno de 90% até 84 dias de cultivo. A partir deste período, a mortalidade foi mais acentuada. Para as variáveis número total de folhas e folhas com senescência, o período de cultivo de 56 dias foi o que mostrou uma proporção mais adequada, em que para cada nove folhas formadas apenas uma estava senescente. Em relação à formação de raízes primárias, médias superiores a 50% foram observadas após 70 dias de cultivo in vitro, chegando aos melhores resultados (60%) aos 200 dias. Para raízes secundárias, as maiores médias foram observadas após 102 dias (41,5%), alcançando 45,3% aos 200 dias. A micropropagação de Luehea divaricata é afetada significativamente pelo período de cultivo in vitro, que pode ser estendido até aproximadamente 60 dias. A partir desse período, são registradas altas mortalidades.

ABSCTRACT: Eugenia involucrata DC. is a forest species with high environmental and economic potential. The objective of this study was to quantify the genetic variability and analyzed the genetic structure of three natural fragments located in the central region of the Rio Grande do Sul state, Brazil. We used four microsatellite loci developed for the congener species Eugenia uniflora and using GenAlEx 6.5 software, parameters of genetic variability and its partition among and within fragments were estimated for each locus. We observed high levels of genetic variability (3.67 alleles per locus; HO = 0.815; HE = 0.625; FIS = −0.294), most of which (93%) were distributed within the fragments, suggesting that these individuals came from a single original population. Gene flow between fragments was high (2.35 to 4.56 migrants per generation), resulting in low genetic differentiation indexes (FST values ranging from 0.052 to 0.096). The fragments showed high genetic variability, distributed within the remnants themselves, and low genetic differentiation. Our results have repercussions for planning locally adapted germplasm collections for forest restoration programs, thereby avoiding the implantation of populations with an exogamous depression.

Esse trabalho objetivou avaliar o efeito do tipo de luz no cultivo in vitro de Eugenia involucrata. Os tratamentos consistiram do cultivo sob diodos emissores de luz (LEDs) de cor vermelho, azul, verde ou branco e lâmpadas fluorescentes. Nos cultivos sob LEDs branco e azul foram observadas as maiores médias de estabelecimento; sob o azul e lâmpadas fluorescentes, o maior número de folhas; sob o azul, o maior número de brotos; sob o branco, menor contaminação bacteriana; e sob o verde os piores resultados. LEDs azul, vermelho, branco e lâmpadas fluorescentes influenciam favoravelmente a propagação in vitro de E. involucrata.

Este trabalho objetivou avaliar o efeito de fitorreguladores na calogênese e rizogênese em discos foliares de Eugenia involucrata. Foram testadas, em dois experimentos, combinações de ácido naftalenoacético (ANA) e 6-benzilaminopurina (BAP) e de 2,4-diclorofenoxiacético (2,4-D) e BAP. Aos 45 dias de cultivo todos os tratamentos formaram calos, principalmente naqueles em que foram empregados ANA isolado e, também, combinado com BAP, bem como 2,4-D e BAP (5:5 e 10:10 µM). Apenas ANA isolado (10 µM) induziu rizogênese. Aos 90 dias de cultivo pôde-se perceber melhor o efeito dos fitorreguladores, observando-se que 2,4-D e BAP (5:5 e 10:10 µM) favoreceram a calogênese, enquanto para rizogênese o melhor tratamento foi com 10 µM de ANA. É possível mediar processos de desdiferenciação e rediferenciação celular a partir de discos foliares de Eugenia involucrata.

Abstract The objective of this study was to evaluate the effect of sucrose and substrates on the acclimatization of micropropagated Luehea divaricata plants. First, the effect of sucrose on in vitro growth and development of nodal segments in Woody Plant Medium reduced to its half concentration was tested. After that, these shoots were then acclimatized and the combined effect of sucrose, as previously described, plus the commercial substrates vermiculite, MecPlant® or H-decker® were tested. In the in vitro culture, root and leaf formation were superior (44%, 8.44 leaves) with 20 g L-1 sucrose, and then the presence of sucrose continued to be more efficient in the acclimatization with the MecPlant® or H-decker® commercial substrates. Sucrose at 20 g L-1 is more efficient for in vitro leaf and root formation, and the MecPlant® or H-decker® substrates may be used in acclimatization.

ABSTRACT: The objective of the present research was to evaluate the effect of different concentrations of indolebutyric acid (IBA) on the rooting of Luehea divaricata minicuttings under two different cultivation periods in a greenhouse environment. The minicuttings were immersed in a hydroethanolic solution containing IBA. All minicuttings survived after 30 and 60 days of cultivation. Root formation at 30 days of cultivation with 1000 mg L-1 of IBA was 91.7%, the same rate as that observed with 4000 mg L-1. At the 1000 mg L-1 concentration, the number of roots was the greatest among all treatments, with 8.42 roots per minicutting after 30 days of cultivation. After 30 days of cultivation, the formation and number of roots with the use of 1000 mg L-1 of IBA in the “pulse” treatment produced the best outcome results, allowing the plants to spend less time in a greenhouse at a reduced concentration of auxin when compared with other treatments. However, to obtain higher averages of secondary root formation, root length, and number of leaves, it is necessary to increase the cultivation period to 60 days.

ABSTRACT Eugenia involucrata DC. (Myrtaceae), an economically important forest tree species, is prized for its timber and fruits, and is also an important ornamental plant. This study aimed to evaluate the effect of Thidiazuron (TDZ) and α-Naphthaleneacetic acetic acid (NAA) on in vitro multiplication of nodal segments of E. involucrata. We tested the effect of the absence and presence of NAA (0.5 µM), combined with TDZ at concentrations of 0, 2, 4, 8, 16, or 32 µM, on the in vitro multiplication of E. involucrata. The use of TDZ combined with NAA (0.5 µM) favored the formation of shoots and buds in the explants, especially at 32 µM TDZ concentration. Intermediate concentrations of TDZ also promoted shoot formation but induced hyperhydricity in the explants. It is possible to induce organogenesis leading to the multiplication of E. involucrata nodal segments using TDZ, preferably combined with NAA.
