Structural equation model was used to analyze the effects of resilience.Results: The score of FCR in glioma patients was 29.52± 8.30. A total of 47 patients had FCR (total score ≥ 34), with an incidence of 36.7%. There was a correlation between FCR, resilience and social support (P< 0.01). The resilience between perceived social support and FCR in glioma patients had good fitting with the structural equation model. Resilience played a mediating role between perceived social support and FCR, with a mediating effect of 48.4%.Conclusion: The level of resilience can be improved by improving the perceived social support in patients with glioma to reduce the FCR of patients.Keywords: glioma, fear of cancer recurrence, resilience, perceived social support, mediating effect

27. DBX2 Promotes Glioblastoma Cell Proliferation by Regulating RESTExpression

Abstract

Background: Glioblastoma (GBM) is the most common but lethal brain cancer withpoor prognosis. The developing brain homeobox 2 (DBX2) has been reported to play importantroles in tumor growth. However, the mechanisms of DBX2 in GBM are still unknown. Objectives: This study aims to investigate the function and mechanisms of DBX2 in GBM. Methods: The expressions of DBX2 and REST in GBM were measured by analyzing data fromdatabases, and the results were checked by qPCR and/or western blot of GBM cell lines. Cellproliferation was determined by CCK8 assay, immunohistochemistry and colony formationassay. ChIP-qPCR was used to determine the binding sites of DBX2 on REST. Results: In this study, we found that the expression of DBX2 was upregulated in the GBM celllines. The cell proliferation was damaged after blocking DBX2 expression in U87 and U251 GBMcell lines. The expression level of DBX2 had a positive relationship with that of REST. Our ChIPqPCRresults showed that DBX2 is directly bound to the promoter region of REST. Additionally,the increased GBM cell proliferation caused by DBX2 overexpression can be rescued by RESTloss of function. Conclusion: DBX2 could promote cell proliferation of GBM by binding to the promoter regionof REST gene and increasing REST expression.

Abstract

The abnormal metabolic processes following traumatic brain injury (TBI) have been proposed to contribute to secondary injuries after TBI. Therefore, enteral nutrition (EN) support for TBI patients has received more attention. This study aimed to evaluate the complimentary effects of enteral nutrition with glutamine and hyperbaric oxygen (HBO) on the recovery of TBI. TBI model was established in SD rats, which were randomly divided into four groups: TBI, TBI + HBO, TBI + GLN, and TBI + HBO + GLN. Neuronal apoptosis in penumbra area was detected by TUNEL. Serum prealbumin level was detected by ELISA. Motor function was evaluated by beam-balance test. We found that the body weight of the rats had no significant differences in different groups before and after injury. Among the four groups, beam-balance test score was the lowest, serum prealbumin level was the highest, and neuronal apoptosis rate was the lowest in TBI + HBO + GLN group on day 3 and 7 after TBI. In conclusion, our data suggest that hyperbaric oxygen combined with enteral nutrition support with glutamine is effective in reducing neuronal apoptosis, increasing serum prealbumin concentration and improving neurological function after TBI injury.
