In this paper, a non-intrusive load identification algorithm for residents based on prior knowledge and statistical learning model is proposed to solve the problem of insufficient electric heating subdivision capability in traditional identification technology. In this paper, the electric heating subdivision research is carried out for the auxiliary heating equipment of washing machine, electric kettle, electric rice cooker, electric water heater. The subdivision of auxiliary heating equipment is realized through the equipment operation association algorithm, and the model training of non-auxiliary heating equipment classification is realized based on the limited feedback information of users and expert annotation. The experimental results show that the technical framework proposed in this paper realizes the subdivision of electric heating equipment on the basis of the event detection load identification algorithm and F1 socre above 0.9 is achieved in the decomposition of operation state.

The excavation thickness of the coal seam and the corresponding motion space of the overlying strata are so large in the fully-mechanized top coal caving face, that roof accidents are occur easily. In order to understand the mechanism of roof disasters in fully-mechanized caving working face, some cases of roof disasters in fully-mechanized caving working face are statistically analyzed. According to the roof conditions and disaster characteristics, the roof disasters in fully-mechanized caving working face are divided into two types: large-area roof cutting and support crushing for soft roof, large-area roof collapse with roof cutting and support crushing for hard roof. Taking Cuimu Mine and Caojiatan Mine as examples, the characteristics of two types of roof disasters are analyzed, and the corresponding roof disaster mechanism is presented: The roof in soft roof working face is of weak cementation. If the actual support stiffness is insufficient, the roof will break at the coal wall, and is easy to be unstable after breaking. As the same time, the insufficient support strength eventually leads to the leads to cutting and support crushing disaster of roof. The roof in hard roof working face is of good integrity and is not easy to break. When the suspended roof area is enough, the sudden breaking of roof will cause an impact on the support. If the breaking line is at the coal wall, it will easily leads to cutting and support crushing disaster of roof. Combined with engineering practice, prevention and control measures for two types of roof disaster are presented. For the soft roof working face, the measures such as reasonably improving the support stiffness, the supporting efficiency and the advancing speed of the working face, and the early warning and so on are used to prevent the occurrence of large-scale roof cutting and support crushing. For the hard roof working face, in addition to support management and rational mining design, it is necessary to improving roof conditions. For examples, the regional hydraulic fracturing technology is used to weaken the roof and prevent large-area roof collapse and support crushing disasters.

The application of hydraulic fracturing technology on rock fracturing for hard and complete roof, and destressing for roadways with high stresses is increasingly widespread. Based on a top coal caving working face with extra-thick coal seam and complete roof layers in the Caojiatan coal mine in Shaanxi Province, the roof rock geomechanical measurements, compressibility tests, theory of hydraulic fracture propagation, and 3D numerical simulation were carried out, and the determination approaches for fracturing rock horizon above a working face, fracturing borehole layout and parameters, and fracturing technology were put forward. The underground industrial trial and a comprehensive surface microseismic monitoring with actual time were conducted, and the spatial distribution characteristics of hydraulic fractures in roof were obtained. Meanwhile, shield resistance, periodic weighting step distance and duration distance, weighting dynamic load coefficient, and roof breaking energy were monitored and analyzed, and the hydraulic fracturing effect was synthetically evaluated. A complete set of technology for hydraulic fracturing with directional boreholes in underground coal mines were preliminarily established, which includes determination of fracturing rock horizon and parameter design, fracturing technology and equipment with directional boreholes, monitoring of hydraulic fracture spatial distribution, and comprehensive evaluation of hydraulic fracturing effect. The underground trial pointed out that: on the in-situ stress regime in the Caojiatan coal mine, that is the minimum principal stress is vertical stress, the hydraulic fractures were mainly horizontal fractures with a propagation distance of about 80 m on both sides of boreholes, the hard and complete roofs above the working face were effectively weakened, and regional roof reconstruction was achieved. The intensive ground pressure appearance of the working face was obviously reduced in the fracturing area, and production safety was ensured. Finally, the existing issues associated with hydraulic fracturing were discussed, and technical developments were envisaged.

Natural killer (NK) cells provide important host defense against herpesvirus infections and influence subsequent T cell control of replication and maintenance of latency. NK cells exhibit phases of expansion, contraction and memory formation in response to the natural mouse pathogen murine cytomegalovirus (MCMV). Innate and adaptive immune responses are tightly regulated in mammals to avoid excess tissue damage while preventing acute and chronic viral disease and assuring resistance to reinfection. Caspase (CASP)8 is an autoactivating aspartate-specific cysteine protease that initiates extrinsic apoptosis and prevents receptor interacting protein (RIP) kinase (RIPK)1-RIPK3-driven necroptosis. CASP8 also promotes death-independent signal transduction. All of these activities make contributions to inflammation. Here, we demonstrate that CASP8 restricts NK cell expansion during MCMV infection but does not influence NK memory. Casp8-/-Ripk3-/- mice mount higher NK response levels than Casp8+/-Ripk3-/- littermate controls or WT C57BL/6 J mice, indicating that RIPK3 deficiency alone does not contribute to NK response patterns. MCMV m157-responsive Ly49H+ NK cells support increased expansion of both Ly49H- NK cells and CD8 T cells in Casp8-/-Ripk3-/- mice. Surprisingly, hyperaccumulation of NK cells depends on the pronecrotic kinase RIPK1. Ripk1-/-Casp8-/-Ripk3-/- mice fail to show the enhanced expansion of lymphocytes observed in Casp8-/-Ripk3-/- mice even though development and homeostasis are preserved in uninfected Ripk1-/-Casp8-/-Ripk3-/- mice. Thus, CASP8 naturally regulates the magnitude of NK cell responses in response to infection where strong activation signals depend on another key regulator of death signaling, RIPK1. In addition, the strong NK cell response promotes survival of effector CD8 T cells during their expansion. Thus, hyperaccumulation of NK cells and crosstalk with T cells becomes amplified in the absence of extrinsic cell death machinery.

The magnitude of CD8 T cell responses against viruses is checked by the balance of proliferation and death. Caspase-8 (CASP8) has the potential to influence response characteristics through initiation of apoptosis, suppression of necroptosis, and modulation of cell death-independent signal transduction. Mice deficient in CASP8 and RIPK3 (Casp8 -/- Ripk3 -/- ) mount enhanced peak CD8 T cell levels against the natural mouse pathogen murine cytomegalovirus (MCMV) or the human pathogen herpes simplex virus-1 compared with littermate control RIPK3-deficient or WT C57BL/6 mice, suggesting an impact of CASP8 on the magnitude of antiviral CD8 T cell expansion and not on contraction. The higher peak response to MCMV in Casp8 -/- Ripk3 -/- mice resulted from accumulation of greater numbers of terminally differentiated KLRG1hi effector CD8 T cell subsets. Antiviral Casp8 -/- Ripk3 -/- T cells exhibited enhanced proliferation when splenocytes were transferred into WT recipient mice. Thus, cell-autonomous CASP8 normally restricts CD8 T cell proliferation following T cell receptor activation in response to foreign antigen. Memory inflation is a hallmark quality of the T cell response to cytomegalovirus infection. Surprisingly, MCMV-specific memory inflation was not sustained long-term in Casp8 -/- Ripk3 -/- mice even though these mice retained immunity to secondary challenge. In addition, the accumulation of abnormal B220+CD3+ T cells in these viable CASP8-deficient mice was reduced by chronic MCMV infection. Combined, these data brings to light the cell death-independent role of CASP8 during CD8 T cell expansion in mice lacking the confounding impact of RIPK3-mediated necroptosis.

The development of dual linkage covalent organic frameworks (DL-COFs) is fundamentally important for creating diversified structures and functions. Key Clinical Message: This article reports a case of mediastinal lymph node tuberculosis with no obvious symptoms and a concealed focus. This patient, a 33‐year‐old male, suffered from pain behind the sternum after eating. He underwent three gastroscopic examinations and two fine needle punctures guided by ultrasound gastroscopy but was not diagnosed. Chest‐enhanced CT revealed a mediastinal mass compressing the adjacent esophagus, suggesting the possibility of enlarged lymph nodes. Furthermore, T cells from patients infected with tuberculosis tested positive. Ultrasound bronchoscopy revealed enlarged lymph nodes in area 7, and then EBUS‐TBNA was performed in that region. Only a few scattered lymphocytes and necrotic tissue were found under the biopsy microscope. The EBUS‐TBNA biopsy Xpert MTB/RIF showed low positive results, and the EBUS‐TBNA puncture fluid Xpert MTB/RIF was positive. Therefore, he was diagnosed with mediastinal lymph node tuberculosis. After antituberculosis treatment with the 2HREZ/10HRE regimen, the patient's pain behind the sternum gradually alleviated, and the enlarged mediastinal lymph nodes gradually narrowed. The vibration parameters during drilling play a critical role in enhancing drilling speed and ensuring safety. However, traditional downhole vibration sensors face limitations in their power supply methods, hindering widespread adoption. To address this challenge, our research introduces a novel solution: a hybrid downhole vibration sensor (HDV-TENG) utilizing triboelectric nanogenerators. This sensor not only enables the measurement of low- to medium–high-frequency vibrations using self-power but also serves to energize other downhole devices. We utilized a self-constructed vibration simulator to replicate downhole drilling tool vibrations and conducted a comprehensive series of sensor tests. The test results indicate that the frequency measurement bandwidth of the HDV-TENG spans from 0 to 200 kHz. Especially, the measurement errors for vibrations within the low-frequency range of 0 to 10 Hz and the high-frequency range of 10 to 200 k Hz are less than 5% and 8%, respectively. Additionally, the experimental findings regarding load matching demonstrate that the HDV-TENG achieves an output power level in the milliwatt range, representing a significant improvement over the output power of traditional triboelectric nanogenerators. Unlike traditional downhole vibration measurement sensors, HDV-TENG operates without requiring any external power supply, thereby conserving downhole space and significantly enhancing drilling efficiency. Furthermore, HDV-TENG not only offers a broad measurement range but also amplifies output power through the synergy of a triboelectric nanogenerator (TENG), piezoelectric nanogenerator (PENG), and electromagnetic power generator (EMG). This capability enables its utilization as an emergency power source for other micropower equipment downhole.
