House mice (Mus musculus) are an invasive species on Auckland Island in the New Zealand subantarctic and planning for their eradication is underway. Mast seeding events cause rodent populations to irrupt, though little is known about this phenomenon in snow tussock grass (Chionochloa spp.) systems on Southern Ocean islands. The aim of this study was to understand population fluctuations of mouse abundance on Auckland Island for the 2 years following a mast event, and with which tools to monitor abundance, to inform planning of bait application for eradication. Mouse populations were studied using kill and live trapping at two sites on Auckland Island, and mouse density was estimated using spatially explicit capture-recapture models. Mouse population density was highest during summer mast seeding of Chionochloa antarctica and then declined the following winter and subsequently remained low for the following year. Breeding remained seasonal, with a pulse in early summer and a very low level continuing through winter in both years, regardless of mast conditions. These results are similar to those from other cool temperate Southern Ocean islands where seasonal resource availability appears to drive breeding. Throughout the study the capture probability of mice was generally higher when population density was lower, which highlights that conclusions about population trends could be misleading if abundance indices are not calibrated to measures of population density. Mouse eradication should preferentially take place outside of a mast event but would likely still succeed during and following a mast event. Our work fills a key knowledge gap about rodent population trends during mast events for Southern Ocean islands, which is particularly important where eradications are planned. Antipodes Island is part of New Zealand’s World Heritage subantarctic region and hosts special biodiversity values and significant species endemism. Invasive house mice were the only introduced mammal and detrimentally impacted invertebrate and native bird communities. Eradication of mice from Antipodes Island was undertaken in 2016 and confirmed in 2018. We present the monitoring used to confirm eradication of mice and the ecological outcomes measured over the 6 years since the eradication. Result monitoring for confirmation applied a simple regime to search for mice following a delay of two mouse breeding seasons since baiting was completed. Outcome monitoring targeted endemic land bird taxa for possible changes due to operational impacts and ecological recovery following eradication of mice. The operation had no longterm negative impacts and endemic land bird taxa have recovered quickly from variable levels of non-target mortality. Estimates of abundance of Antipodes Island snipe, Antipodes Island pipit and Reischek’s parakeet showed strong long-term positive response to mouse eradication. [ABSTRACT FROM AUTHOR]

Auckland Island, the fifth largest island in New Zealand, is the only island in New Zealand’s subantarctic region where introduced mammalian pests remain (pigs, Sus scrofa; mice, Mus musculus; cats, Felis catus). The island has unique biodiversity and is a key site for progressing New Zealand’s goal to be free of several introduced predators by 2050. Recent island eradication successes have rekindled interest in eradicating pests from Auckland Island, and for the first time considering all three pests in one project. Over a 3-year period, we tested the feasibility of eradicating pigs, mice and cats by looking at what it would take to succeed, rather than what we could practically deliver with the tools we currently have. We proposed adaptations to current methods and used an evidence-based approach by undertaking large-scale field trials to test uncertainties and emerging technologies in-situ. We gathered data and evaluated proposed methods against five established principles of eradication while considering the logistics and infrastructure requirements of the project. Eradicating pigs, mice and cats from Auckland Island is worthwhile and feasible but dependent on further development of emerging technologies and capabilities for efficient delivery with an acceptable level of risk. Three eradication operations are required with specific sequencing and timing, supported by initial establishment of infrastructure. The project needs a large investment spread over 8 to 10 years to yield permanent and internationally important benefits with low ongoing cost to sustain. The feasibility study exposed the project’s scale and was used to inform decision makers, who postponed the work in 2020 in response to the economic impacts of COVID-19. The study focusses future preparations on identified planning issues and dependencies to progress project readiness in anticipation of it being launched when economic conditions allow. As a major threat to New Zealand’s biodiversity, feral cats (Felis catus) are the subject of planned eradications on a number of offshore islands, including Rakiura Stewart Island. We used camera traps to estimate population density of feral cats on the north-east coast of Rakiura, and to investigate their movement behaviour and detection probability. We also used camera footage to compare the consumption of two types of non-toxic sausage baits (chicken and rabbit) with a view to future use of toxic baits. Population density of feral cats was likely between 1 and 2 cats per km². Non-target species (rats and possums) removed more than half the baits, greatly reducing bait availability for feral cats. Deer and birds (including kiwi) encountered baits but did not eat them. Cats had an apparent preference for chicken over rabbit baits, although small sample sizes prevent firm conclusions. Both bait types appeared to decline rapidly in palatability, and no baits were consumed by cats more than 5 days after deployment. Future trials and baiting regimes should consider ways to improve bait availability. Increased bait density, exclusion of rats and possums and/or more frequent replacement of baits will likely increase encounter rates by feral cats. [ABSTRACT FROM AUTHOR]

Restoration initiatives of ecosystems transformed by human actions require optimisation of eradication measures of introduced species, particularly in fragile insular ecosystems. We studied aspects of the spatial ecology of introduced feral cats (Felis catus) on subantarctic Auckland Island of New Zealand to assist eradication efforts of pests from this remote, biologically rich island. Firstly, we estimated home range sizes and identified core areas of activity based on movement-rooted dynamic Brownian bridge models. Second, we used resource selection functions with generalised linear mixed models to identify seasonal patterns of space use associated to topographic, vegetation and other landscape predictors. Lastly, we quantified cats daily movement rates within home ranges. Average home range size was larger than on other offshore islands and mainland New Zealand, which might relate to lower cat densities and the abundance and predictability of food resources on the island. Cats mostly selected mosaic areas of forest, shrubs and tall tussocks near the coast, and in predominantly flat areas or nearby steep cliffs, which are all typical habitats of seabirds and terrestrial birds. Cats also selected alpine short tussocks during the cold season, likely related to the upsurge of mice (Mus musculus) due to tussock mast seeding and to transiting to steep cliffy areas. Male cats had home ranges that were larger, contained more core areas, and covered longer daily distances in the warm season than females, which might be associated with different breeding and reproductive behaviour. Eradication tools will need to target all habitats on Auckland Island with increased efforts in areas of identified higher use by cats. Understanding aspects of pest species’ spatial ecology on offshore islands worldwide can assist decision-makers in optimising eradication programs such as Predator Free 2050 in New Zealand. Judas pigs (Sus scrofa) will be integral to the success of the proposed Auckland Island (45 891 ha) pig eradication. Judas pigs must be permanently unable to breed and reliably retain a radio-transmitter to enable tracking and retrieval. This study tested the techniques and practical requirements to undertake the procedures required to produce a Judas pig in a remote location. Four adult female wild pigs were captured using a netgun from a helicopter and processed at a base site in the field. They were anaesthetised using intramuscular injection of one of two anaesthetic protocols (Zoletil™, ketamine and xylazine; or medetomidine, ketamine and butorphanol). Surgical sterilisation consisted of tubal ligation and resection via a ventral midline incision. Pigs received either a subcutaneous or intra-abdominal implanted radio-transmitter and had a radio-collar and radioear tag attached. The anaesthetic was reversed prior to release close to the capture location. Radio-tracking at 10 and 104 days showed all pigs had survived and radio-tracking was effective using all the transmitter types. Pigs were humanely dispatched and necropsied on day 104. No adverse events were detected from the surgeries or radio-transmitters. All transmitter types except one subcutaneous transmitter model were shown to be secure and had an effective radio range for a Judas pig programme. Pig growth rates implicate a limited humane collaring period and ear transmitters may be at greater risk of transmitter loss. Implantable intra-abdominal radio transmitters provide the most security, but multiple transmitters are advised. Surgical sterilisation by tubal ligation and resection under general anaesthesia using an intramuscular injection containing medetomidine, butorphanol and ketamine and reversed with atipamezole was shown to be feasible in field conditions. Sourdough technology has been known for its role in the improvement of texture, flavor, and quality of mainly wheat and rye-based breads for decades. However, little is reported about its use in the improvement of whole-grain oat bread, especially concerning flavor formation, which is one major consumer drivers. This study investigated the effects of sourdough obtained by different lactic acid bacteria and yeast starters consortia on the texture and flavor of 100% oat bread. Four different consortia were selected to obtain four oat sourdoughs, which were analyzed to assess the main features due to the different starter fermentation metabolism. Sourdoughs were added to breads as 30% dough weight. Bread quality was technologically monitored via hardness and volume measurements. Sourdough breads were softer and had higher specific volume. The sensory profile of sourdoughs and breads was assessed by a trained panel in sensory laboratory conditions, and the volatile profile was analyzed by HS-SPME-GC-MS. Sourdoughs were rated with higher intensities than untreated control for most of attributes, especially concerning sour aroma and flavor attributes. Sourdough breads were rated with higher intensities than control bread for sour vinegar flavor and total odor intensity, in addition they had richer volatile profile. Our results confirmed that sourdough addition can lead to an enhanced flavor, moreover, it demonstrated that the use of different consortia of lactic acid bacteria and yeast strains leads to the improvement of texture and altered sensory profile of whole-oat bread.

Fisheries harvest strategies are formal frameworks that represent a best-practice approach for sustainable fisheries management. A key component of a harvest strategy is a ’pre-agreed rule’, known as a harvest control rule (HCR), that sets fishing opportunities, e.g. catch limits, based on an estimate of fish stock status, e.g. estimated stock biomass. The harvest strategy development process is driven by stakeholders who are required to make a range of informed decisions, including on the selection of the preferred HCR. Capacity building may be required to facilitate the stakeholder engagement, particularly regarding the technical components of harvest strategies, including HCRs. The AMPLE package for R provides three interactive apps that support capacity building and stakeholder engagement on HCRs. These apps have been used during in-country national workshops around the western and central Pacific Ocean (WCPO) to support the development of harvest strategies for the Western and Central Pacific Fisheries Commission. These apps include several novel features: they take users from a gentle introduction to how HCRs work, to using methods for testing, comparing and selecting a preferred HCR from a suite of candidates. They include an introduction to the impact of uncertainty on the performance of an HCR, introduce performance indicators and discuss methods for selecting the preferred HCR based on management objectives. As such they provide a more detailed overview of HCRs than currently existing alternatives. These apps provide an effective platform for hands-on learning and have proven to be successful at supporting capacity building on HCRs in the WCPO. For example, using them for group activities and competitions stimulated productive discussions and increased understanding. As the model fishery in AMPLE is generic and not based on a real example, the apps will also be of interest to scientists, managers and stakeholders developing harvest strategies in other regions.

Micropropagation techniques offer opportunity to proliferate, maintain, and study dynamic plant responses in highly controlled environments without confounding external influences, forming the basis for many biotechnological applications. With medicinal and recreational interests for Cannabis sativa L. growing, research related to the optimization of in vitro practices is needed to improve current methods while boosting our understanding of the underlying physiological processes. Unfortunately, due to the exorbitantly large array of factors influencing tissue culture, existing approaches to optimize in vitro methods are tedious and time-consuming. Therefore, there is great potential to use new computational methodologies for analyzing data to develop improved protocols more efficiently. Here, we first tested the effects of light qualities using assorted combinations of Red, Blue, Far Red, and White spanning 0–100 μmol/m2/s in combination with sucrose concentrations ranging from 1 to 6% (w/v), totaling 66 treatments, on in vitro shoot growth, root development, number of nodes, shoot emergence, and canopy surface area. Collected data were then assessed using multilayer perceptron (MLP), generalized regression neural network (GRNN), and adaptive neuro-fuzzy inference system (ANFIS) to model and predict in vitro Cannabis growth and development. Based on the results, GRNN had better performance than MLP or ANFIS and was consequently selected to link different optimization algorithms [genetic algorithm (GA), biogeography-based optimization (BBO), interior search algorithm (ISA), and symbiotic organisms search (SOS)] for prediction of optimal light levels (quality/intensity) and sucrose concentration for various applications. Predictions of in vitro conditions to refine growth responses were subsequently tested in a validation experiment and data showed no significant differences between predicted optimized values and observed data. Thus, this study demonstrates the potential of machine learning and optimization algorithms to predict the most favorable light combinations and sucrose levels to elicit specific developmental responses. Based on these, recommendations of light and carbohydrate levels to promote specific developmental outcomes for in vitro Cannabis are suggested. Ultimately, this work showcases the importance of light quality and carbohydrate supply in directing plant development as well as the power of machine learning approaches to investigate complex interactions in plant tissue culture.

Tackling smoking is an integral component of efforts to improve health outcomes in Aboriginal communities. Social marketing is an effective strategy for promoting healthy attitudes and influencing behaviours; however, there is little evidence for its success in reducing smoking rates in Aboriginal communities. This paper outlines the development, implementation and evaluation of Kick the Habit Phase 2, an innovative tobacco control social marketing campaign in Aboriginal communities in New South Wales (NSW). The Aboriginal Health & Medical Research Council worked with three Aboriginal communities and a creative agency to develop locally tailored, culturally relevant social marketing campaigns. Each community determined the target audience and main messages, and identified appropriate local champions and marketing tools. Mixed methods were used to evaluate the campaign, including surveys and interviews with community members and Aboriginal Community Controlled Health Service staff. Community survey participants demonstrated high recall of smoking cessation messages, particularly for messages and images specific to the Kick the Habit campaign. Staff participating in interviews reported an increased level of interest from community members in smoking cessation programs, as well as increased confidence and skills in developing further social marketing campaigns. Aboriginal community-driven social marketing campaigns in tobacco control can build capacity, are culturally relevant and lead to high rates of recall in Aboriginal communities. This paper describes a methodology that combines meta-population theory and stock assessment models to gain insights about spatial heterogeneity of the meta-population in an operational time frame. The methodology was tested with stochastic simulations for different degrees of connectivity between sub-populations and applied to two case studies, North Sea cod (Gadus morua) and Northeast Atlantic sardine (Sardina pilchardus). Considering that the biological components of a population can be partitioned into discrete spatial units, we extended this idea into a property of additivity of sub-population abundances. If the additivity results hold true for putative sub-populations, then assessment results based on sub-populations will provide information to develop and monitor the implementation of finer scale/local management. The simulation study confirmed that when sub-populations are independent and not too heterogeneous with regards to productivity, the sum of stock assessment model estimates of sub-populations' SSB is similar to the SSB estimates of the meta-population. It also showed that a strong diffusion process can be detected and that the stronger the connection between SSB and recruitment, the better the diffusion process will be detected. On the other hand it showed that weak to moderate diffusion processes are not easy to identify and large differences between sub-populations productivities may be confounded with weak diffusion processes. The application to North Sea cod and Atlantic sardine exemplified how much insight can be gained. In both cases the results obtained were sufficiently robust to support the regional analysis.

Objective: Player availability post-surgery can be challenging, however, implementing a GPS guided load management program can maximise readiness. Methods: In Fall 2017, Patient A (20 yrs, Centre Back) underwent patellofemoral realignment and Patient B (21 yrs, Centre Midfielder) underwent ACL reconstruction. Both players were cleared medically to play in the 2018 season and seventy-six sessions were monitored during the season. Modification of total distance (TD) and high-speed running (HSR) quantified by GPS was implemented with the medical staff to manage ongoing injury site symptoms. Training loads were based on absolute and relative values of individual match TD and HSR. TD and HSR (velocity >14.4km/h) were measured in metres (m). Results: Patient A's average match duration was 84±23 minutes (min) and Patient B's average match duration was 90±12min. Overall team match duration was 95±8min. Patient A match loads were TD 8763±3826m and HSR 1039±509m. Patient B match loads were TD 8652±2281m and HSR 981±307m. Team match loads were TD 7431±1430m and HSR 1090±235m. Patient A training loads were TD 5201±1048m and HSR 463±352m; which was 59.3% of TD and 44.5% of HSR match load. Athlete B training loads were TD 4415±1650m and HSR 333±209m, which was 51% of TD and 33.9% of HSD match load. Team training loads were TD 4612±1296m and HSR 474±301m. This was 62% of TD and 43.4% of HSR match load. Conclusion: As a result of implementing a GPS guided load management program, both athletes were available for starter selection for all matches throughout the whole competitive season and were able to play the majority of match duration. Clinical implications: Successful collaboration between the medical and sport science teams can ensure injured player readiness and availability for matches. Estimating fish stock status is very challenging given the many sources and high levels of uncertainty surrounding the biological processes (e.g. natural variability in the demographic rates), model selection (e.g. choosing growth or stock assessment models) and parameter estimation. Incorporating multiple sources of uncertainty in a stock assessment allows advice to better account for the risks associated with proposed management options, promoting decisions that are more robust to such uncertainty. However, a typical assessment only reports the model fit and variance of estimated parameters, thereby underreporting the overall uncertainty. Additionally, although multiple candidate models may be considered, only one is selected as the 'best' result, effectively rejecting the plausible assumptions behind the other models. We present an applied framework to integrate multiple sources of uncertainty in the stock assessment process. The first step is the generation and conditioning of a suite of stock assessment models that contain different assumptions about the stock and the fishery. The second step is the estimation of parameters, including fitting of the stock assessment models. The final step integrates across all of the results to reconcile the multi-model outcome. The framework is flexible enough to be tailored to particular stocks and fisheries and can draw on information from multiple sources to implement a broad variety of assumptions, making it applicable to stocks with varying levels of data availability The Iberian hake stock in International Council for the Exploration of the Sea (ICES) Divisions VIIIc and IXa is used to demonstrate the framework, starting from length-based stock and indices data. Process and model uncertainty are considered through the growth, natural mortality, fishing mortality, survey catchability and stock-recruitment relationship. Estimation uncertainty is included as part of the fitting process. Simple model averaging is used to integrate across the results and produce a single assessment that considers the multiple sources of uncertainty.

Purpose: This study aims to create a stigma reduction framework for stuttering in the local context of Québec, Canada using the Participative Concept Mapping Approach (PCMA), focusing on both self and societal stigma., Method: Employing a mixed-methods approach, this study engaged 17 experts-people who stutter, clinicians and health innovation specialists-in PCMA workshops. Via diverse steps, including generation, sorting and rating of ideas in response to the focus prompt, "To effectively address stuttering (self-)stigma, an intervention should…" these sessions led to a framework depicted in visual maps, then refined into actionable principles through qualitative analysis. Mixed-methods data analysis used the open-source R-CMap software to generate visual maps illustrating the relationships among ideas as well as importance and feasibility ratings., Results: The collaborative workshops identified 95 ideas in response to the focus prompt, reunited in 7 clusters, evolving into 16 principles to mitigate stuttering stigma and self-stigma. At the therapy level, these principles emphasize personalized therapy, thorough assessments, stigma-free therapeutic environment, empowerment, and the importance of group inclusivity and educating the relational circles. Societally, they advocate for initiatives such as improved educational outreach, empathy enhancement, and better representation. This dual approach targets individual experiences and societal views on stuttering, stressing the need for an all-encompassing intervention framework., Conclusion: The findings demonstrate PCMA's usefulness in crafting local, culturally sensitive, tailored interventions for stigma reduction. Background: Schizophrenia, schizoaffective disorder, and bipolar affective disorder are debilitating psychiatric conditions characterized by a chronic pattern of emotional, behavioral, and cognitive disturbances. Shared psychopathology includes the pre-eminence of altered affective states, disorders of thoughts, and behavioral control. Additionally, those conditions share epidemiological traits, including significant cardiovascular, metabolic, infectious, and respiratory co-morbidities, resulting in reduced life expectancy of up to 25 years. Nutritional ketosis has been successfully used to treat a range of neurological disorders and preclinical data have convincingly shown potential for its use in animal models of psychotic disorders. More recent data from open clinical trials have pointed toward a dramatic reduction in psychotic, affective, and metabolic symptoms in both schizophrenia and bipolar affective disorder., Objectives: to investigate the effects of nutritional ketosis via a modified ketogenic diet (MKD) over 14 weeks in stable community patients with bipolar disorder, schizoaffective disorder, or schizophrenia., Design: A randomized placebo-controlled clinical trial of 100 non-hospitalized adult participants with a diagnosis of bipolar disorder, schizoaffective disorder, or schizophrenia who are capable of consenting and willing to change their diets., Intervention: Dietitian-led and medically supervised ketogenic diet compared to a diet following the Australian Guide to Healthy Eating for 14 weeks., Outcomes: The primary outcomes include psychiatric and cognitive measures, reported as symptom improvement and functional changes in the Positive and Negative Symptoms Scale (PANSS), Young Mania Rating Scale (YMS), Beck Depression Inventory (BDI), WHO Disability Schedule, Affect Lability Scale and the Cambridge Cognitive Battery. The secondary metabolic outcomes include changes in body weight, blood pressure, liver and kidney function tests, lipid profiles, and markers of insulin resistance. Ketone and glucose levels will be used to study the correlation between primary and secondary outcomes. Optional hair cortisol analysis will assess long-term stress and variations in fecal microbiome composition. Autonomic nervous system activity will be measured via wearable devices (OURA ring and EMBRACE wristband) in the form of skin conductance, oximetry, continuous pulse monitoring, respiratory rate, movement tracking, and sleep quality. Based on the encouraging results from established preclinical research, clinical data from other neurodevelopment disorders, and open trials in bipolar disorder and schizophrenia, we predict that the ketogenic metabolic therapy will be well tolerated and result in improved psychiatric and metabolic outcomes as well as global measures of social and community functioning. Background: Suicide is the leading cause of death among Australians. One commonly cited explanation is the impact of social media, in particular, the ways in which young people use social media to communicate about their own experiences and their exposure to suicide-related content posted by others. Guidelines designed to assist mainstream media to safely report about suicide are widespread. Until recently, no guidelines existed that targeted social media or young people. In response, we developed the #chatsafe guidelines and a supporting social media campaign, which together make up the #chatsafe intervention. The intervention was tested in a pilot study with positive results. However, the study was limited by the lack of a control group., Objective: The aim of this study is to assess the impact of the #chatsafe social media intervention on young people's safety and confidence when communicating on the web about suicide., Methods: The study employs a pragmatic, parallel, superiority randomized controlled design. It will be conducted in accordance with the Consolidated Standards of Reporting Trials statement over 18 months. Participants will be 400 young people aged 16-25 years (200 per arm). Participants will be recruited via social media advertising and assessed at 3 time points: time 1-baseline; time 2-8-week postintervention commencement; and time 3-4-week postintervention. They will be asked to complete a weekly survey to monitor safety and evaluate each piece of social media content. The intervention comprises an 8-week social media campaign including social media posts shared on public Instagram profiles. The intervention group will receive the #chatsafe suicide prevention content and the control group will receive sexual health content. Both groups will receive 24 pieces of content delivered to their mobile phones via text message. The primary outcome is safety when communicating on the web about suicide, as measured via the purpose-designed #chatsafe online safety questionnaire. Additional outcomes include willingness to intervene against suicide, internet self-efficacy, safety, and acceptability., Results: The study was funded in November 2020, approved by the University of Melbourne Human Research Ethics Committee on October 7, 2022, and prospectively registered with the Australian New Zealand Clinical Trials registry. Trial recruitment began in November 2022 and study completion is anticipated by June 2024., Conclusions: This will be the first randomized controlled trial internationally to test the impact of a social media intervention designed to equip young people to communicate safely on the web about suicide. Given the rising rates of youth suicide in Australia and the acceptability of social media among young people, incorporating social media-based interventions into the suicide prevention landscape is an obvious next step. Introduction: The cumulative burden of chronic stress and life events has been termed allostatic load. Elevated allostatic load indices are associated with different mental health conditions in adulthood. To date, however, the association between elevated allostatic load in childhood and later development of mental health conditions has not been investigated., Methods: Using data from the Avon Longitudinal Study of Parents and Children (ALSPAC), we will calculate allostatic load indices using biomarkers representing the cardiovascular, metabolic, immune, and neuroendocrine systems, at the ages of 9 and 17 years. Bivariate and multivariable logistic regression models will be used to investigate the association between allostatic load and psychiatric disorders in adulthood.
