The current article is focused on ideal characteristics, patented technologies, significant features and formulating methods including the use of superdisintegrants, which achieve rapid onset of action, better patient compliance and increased bioavailability. Sublingual tablets dissolve instantaneously, releasing the drug, within a few seconds without the need of water and chewing. The objective behind this review was to summaries the benefits of sublingual formulation, mechanism of action, advantages of route of administration, factors affecting permeability of drug, various in vitro evaluation parameters and commercially available sublingual dosage forms. Different sublingual technologies address pharmaceutical industries and patient needs to enhanced lifecycle and appropriate dosing for pediatric, geriatric, psychiatric patients also patients with dysphagia. Consequently, many scientific techniques including freeze drying, molding, spray drying, sublimation, direct compression, mass extrusion, melt granulation method etc. have been employed for development of sublingual tablets. Due to its better stability and bioavailability, the sublingual route could be considered as a promising alternative to oral and parenteral routes.

Like all other fields, healthcare is also fully blessed with Artificial Intelligence (AI) in current era. From managing records and other data, AI has impacted various facets of COVID-19 epidemic in addition to post COVID-19 crisis in positive way. During the scary period of COVID-19, AI has emerged out as blessing for all the patients, healthcare professionals and even for whole world as it aided in rapid diagnosis, treatment, paradoxically in the post COVID-19 crisis too. The AI has emoted a very noteworthy part in early detection of COVID, prediction of morbidity and mortality as well as detection of deterioration of patients in emergency. In nutshell, the current review is an assemblage of the role of AI in COVID-19 along with post COVID-19 in all ways from forecasting of COVID cases to the burial facility in case of any mortality.

Severe acute respiratory syndrome (SARS) coronavirus-2 was declared as world pandemic by WHO in 2019. It has been spreading very rapidly around the world and responsible for various health issues. Different forms of corona viruses are capable of initiating infections in humans ranging from common cold to respiratory disorders including diarrhea. In this paper, we have utilized the drug repurposing approach for identifying the therapeutic potential candidate for the inhibition of the nsp1 protein of a new porcine coronavirus 6lpa. 81 FDA approved antiviral agents were docked against this virus using discovery studio and PyRx software. Analysis of result reveals three compounds on the basis of the best docking score: Abivertinib, Vazegepant and Elbasvir. Pharmacological and toxicological properties of the leads were also determined. Furthermore, Molecular dynamics simulation studies were performed to determine the stability of complexes. RMSD of protein- ligand complex is showing excellent stability with all three drugs. Least conformational changes can be seen within Abivertinib complex which shows its stability during simulation process. These screened drugs obtained through drug repurposing approach can prove to be useful in treating the infections caused by porcine corona virus 6lpa after further studies.

Over the years researchers have been making an attempt to improve the potency of utilization of medicament for treatment of numerous diseases. During this endeavour, drug delivery systems have helped greatly by achieving reduced dose, reduced dosage frequency, improved patient compliance, improved bioavailability and maximum concentration of the drug at the target site. Recent years have seen an unprecedented growth in the use of nanotechnology in designing drug delivery systems. Niosomes are vesicles composed of non-ionic surfactants, which are biodegradable, comparatively non-toxic, stable, economical and effective alternate to liposomes. This review is deepening and widening the interest of Niosomes in several scientific disciplines and notably its application in drug delivery. The traditional medical study is an integral part of the culture and the perception of health by indigenous population in almost world since primordial times. India has used its knowledge of traditional or herbal medicine to prevent and cure diseases. As herbal medicine have fewer side effects in comparison with synthetic medicine they are getting increase patient compliance against allopathic medicines. Oral ulcers are very common ailment of the oral mucosal membrane. An open sore of the skin or mucus membrane lining marked by sloughing of inflamed dead tissue is known as a mouth ulcer. In this article we discuss on the topic “herbal gel for the treatment of mouth ulcer”. There is a list of herbs which contain flavonoids which are most commonly used for the treatment.

Sygygium cumini, commonly known as Malabar plum, Java plum, black plum, jamun or jambolan, is an evergreen tropical tree in the flowering plant family Myrtaceae. The present study the antioxidant effects of microparticles of S. cumini plant extract were evaluated using in vitro, 2, 2-diphenylpicrylhydrazine photometric assay, total antioxidant activity and super oxide free radical scavenging activity. The microparticles showed different levels of radicals scavenging activity in a dose-dependent manner over the range of 50–150 µg/mL concentration, indicating the high antioxidative capacity of the extract. In all the experimental screening models microparticles of aqueous extract show the maximum inhibition of free radicals comparison of other experimental model at different concentrations. These findings suggest that the seed of Syginium cumini has potent antioxidant activity which may be responsible for some of its reported pharmacological activities and can be used as antioxidant supplement.

The main objective of present study was to prepare a herbo-mineral facial scrub. Majorly facial skin comes in direct contact of dirt, pollution, dust particles and having large number of dead cells. In order to remove the dead cells and make the skin healthy, cleaned and nourished, some facial preparations required. The prepared scrub contains various natural ingredients which are safer for use and having fewer side effects and also they possess antiseptic, anti-infective, antioxidant, anti-aging and humectant properties. The scrub was prepared by using simple mixing method using various ingredients such as poppy seeds, neem extract, tulsi extract, aloe vera gel, almond oil, mixed in carbopol 934, rest of ingredients such as glycerin, triethanolamine, preservatives and perfuming agent were also added to this preparation with homogeneous mixing. The formulated scrub was evaluated for various parameters such as physical appearance, color, texture, odor, pH, viscosity, irritability, washability, homogeneity, extrudability, spreadability and found fruitful results for all the parameter tested. Thus the prepared formulation can be used effectively as it shows good scrubbing properties and it can be used to make a healthy, clean and glowing skin. This research work was aimed to enhance the oral bioavailability and provide faster onset of action of Prochlorperazine maleate (used for the treatment nausea and vomiting) by formulating its mouth dissolving film (MDF). Prochlorperazine belongs to BCS II and oral bioavailability of it’s about 11-15%. The MDF of Prochlorperazine maleate was prepared by solvent casting method using HPMC (film forming agent),Glycerol (plasticizer), Betacyclodextrin (solubilizing agent), Citric acid (saliva stimulating agent), Mannitol (sweetening agent). The formulation was optimized by two factors, three levels (32) was used for the formulation optimization of fast dissolving film of Prochlorperazine maleate and experimental trials are performed on all 9 formulation. In which the amount of HPMC, Glycerol were selected as independent variables (factor) varied at three different level: low (-1), medium (0), and high (+1) levels. The drug release and disintegration time used as dependent variables (response). and formulation was evaluated for weight variation, thickness, folding endurance, drug content, in- vitro disintegration, in vitro dissolution study and stability study. Mouth dissolving film is the most advanced oral solid dosage form due to its flexibility and comfort in use. Mouth dissolving films are oral solid dosage form that disintegrate and dissolve within a minute when placed in mouth without taking water or chewing. This dosage form allows the medication to bypass the first pass metabolism so bioavailability of medication may be improved .Mouth dissolving film has potential to improve onset of action lower the dosing and eliminate the fear of chocking. Formulation of mouth dissolving films involves both the visual and performance characteristics as plasticized hydrocolloids, API taste masking agents are being laminated by solvent casting and semisolid casting method. Solvent casting method being the most preferred method over other methods because it offers great uniformity of thickness and films prepared having fine glossy look and better physical properties. Mouth dissolving films are evaluated for its various parameters like thickness, physical property like folding endurance, disintegration and dissolution time. This review gives an idea about formulation techniques, evaluation parameters, overview on packaging and some available marketed products of mouth dissolving films. Orodispersible tablets (ODTs), also known as fast melt, quick melts, fast disintegrating have the unique property of disintegrating in the mouth in seconds without chewing and the need of water. Oral bioavailability of Atorvastatin Calcium is low (14%) and shows extensive intestinal clearance and first-pass metabolism, which is the main cause for the low systemic availability. In the present work, orodispersible tablets of Atorvastatin calcium were prepared by direct compression method using Hibiscus rosa sinesis mucilage as natural superdisintegrant with a view to enhance patient compliance and to avoid hepatic first pass metabolism and to improve its bioavailability. The prepared batches of tablets were evaluated for hardness, friability, drug content uniformity, wetting time, water-absorption ratio and in-vitro dispersion time. Short-term stability studies on the promising formulation indicated that there are no significant changes in drug content and in vitro dispersion time. Objective: The objective of this study was to formulate orodispersible tablets containing empagliflozin by direct compression method with sufficient hardness and rapid disintegration time and to study the effect of functionality differences of super-disintegrants on the tablet properties. Methods: A two factor three level factorial design (32) was used for the formulation optimization of orodispersible tablets of Empagliflozin and experimental trials were performed on all possible formulations, in which the amount of β-cyclodextrin, crospovidone and croscarmellose sodium were selected as independent variables (factor) varied at three different levels: low (-1), medium (0), and high (+1) levels. The drug release and disintegration time were used as dependent variables (response). All formulations were characterized for parameters such as diameter, hardness, weight, thickness, friability, disintegration time, drug release. Results: Formulation FD6 having 30 sec disintegration time, 98.84% drug release after 30 min, 2.8 kg/cm2 hardness and 0.292% friability was found best among all formulations and selected as an optimized formulation with rapid onset of action and enhanced bioavailability (more than 98% drug release within 30 min.) as compared to the oral empagliflozin tablet. Conclusion: Empagliflozin orodispersible tablets with different superdisintegrants were successfully prepared and formulation containing highest percentage of crospovidone was found best among all other formulations in terms of bioavailability and rapid onset of action.

Parenteral are sterile dosage form of therapeutic agent that are free of viable micro- organisms and they are injected through the skin or mucous membranes into the internal body compartments. Ciprofloxacin is a classic drug (Model drug) which is used in treating different kinds of severe bacterial infections. It is weakly basic and shows solubility only in acidic medium but it leads to irritation when injected. This problem can be solved by adding chemical modifiers like PEG-400 and PG at low concentration which would give 100% bioavailability with no toxicity and stable in pH 7.4. This study is performed to enhance solubility of Ciprofloxacin IV infusion by adding PEG-400 and PG have 75.33±0.57 and 98.66±0.52 mg/mL respectively. All formulations (F1-F9) were found to be sterile, clear, isotonic with blood plasma. Stability study showed that all formulations remains stable at temperature 40±2°C and 75±5% RH. Among all formulations F3 is best suited and passed all evaluation parameters.

Nowadays, novel fast dissolving oral films (FDF) have come in existence as an alternative dosage form in comparison with tablet, capsules, syrup and other oral dosage forms with respect to patient convenience and compliance. Fast dissolving oral films are helpful to paediatric and geriatric patients who experience difficulties in swallowing traditional oral solid-dosage forms. The FDF drug delivery systems are solid dosage form which disintegrate or dissolve within seconds when placed in the mouth cavity without need of water or chewing. FDF provide better drug dissolution, faster onset of action, bypassing the first pass metabolism of drugs and thus enhance their oral bioavailability with reduced dosing frequency. These formulations are suitable for cough, cold, sore throat, allergenic conditions, nausea, pain, hypertension and CNS disorders. The present review provides the details about the recent advancement in design and development of oral fast dissolving film.

Tadalafil is used for the treatment of the erectile dysfunction (ED) and pulmonary arterial hypertension. It is having low aqueous solubility thus it shows poor bioavailability of about 28% by after oral administration. To improve its solubility and dissolution profile solid dispersions (SDPs) of Tadalafil was prepared by physical mixing and solvent evaporation method using polyvinyl pyrollidone-K30 (PVP-30) as a hydrophilic polymeric carrier in different proportions with respect to drug (drug to polymer ratio 1:1 to 1:5). Drug and polymer compatibility studies were performed using FTIR study. The best suitable ratio and method was selected on the basis of enhanced aqueous solubility of drugs. Further selected SDPs were evaluated for various parameters like DSC analysis, percentage yield, percent drug content, saturation solubility, percent drug dissolution and stability studies. FTIR study indicated no incompatibility between Tadalafil and PVP-K30. SDPs prepared with drug to polymer ratio 1:3 and solvent evaporation method was found to be best as they shown significant increased (up to 10 fold) in aqueous solubility in comparison with that of others. DSC study also suggested the depression in the crystalline nature of Tadalafil. Selected SDPs exhibited good stability up to 3 months at 25 ± 2°C /60 ± 5% RH. Based on the results it can be concluded that, SDPs shown remarkable increase in the aqueous solubility and dissolution of Tadalafil and it may improve oral bioavailability of drug as compared with plain drug.

The aim of the present research is to formulate a sublingual tablet of antiemetic drug. Doxylamine succinate is an antihistaminic commonly used for the prevention and treatment of nausea and vomiting. Oral bioavailability of doxylamine succinate is low and shows extensive hepatic metabolism. The Objective of the present research is to formulate doxylamine succinate sublingual tablet to avoid hepatic first-pass metabolism and to improve its bioavailability. Nuclear factor-kB (NF-kB) is a crucial transcription factor in the signal transduction cascade of the inflammatory signaling. Activation of NF-κB depends on the phosphorylation of IκBα by IκB kinase (IKKβ) followed by subsequent ubiquitination and degradation. This leads to the nuclear translocation of the p50- p65 subunits of NF-κB, and further triggers pro-inflammatory cytokine gene expression. Thus, in the need of a more effective therapy for the treatment of inflammatory diseases, specific inhibition of IKKβ represents a rational alternative strategy to the current therapies. A computer-aided drug identification protocol was followed to identify novel IKKβ inhibitors from a database of over 1500 Food and Drug Administration (FDA) drugs. The best scoring compounds were compared with the already known high-potency IKKβ inhibitors for their ability to bind and inhibit IKKβ by evaluating their docking energy. Finally, Thioridazinehydrochloride (TDZ), a potent antipsychotic drug against Schizophrenia was selected and its efficiency in inhibiting IκBα protein degradation and NF-κB activation was experimentally validated. Our study has demonstrated that TDZ blocks IκBα protein degradation and subsequent NF-κB activation to inhibit inflammation. Thus, it is a potential repurposed drug against inflammation.

Plasma medicine can be subdivided into three main fields; Non-thermal atmospheric-pressure direct plasma for medical therapy; Plasma-assisted modification of bio-relevant surfaces and Plasma-based bio-decontamination and sterilization. The basis of the research is that as it has free carrier molecules, it has the ability to target specific cells and regulate functions like wound healing. Plasma does not harm healthy human cells but can kill bacteria and possibly even cancer cells to help treat various diseases. Nosocomial infection control, prevention and containment of contagious diseases, disinfection of medical devices, surface treatment (heat and UV sensitive surfaces) are research of interest. Recent success in generating plasma at very low temperature ie. Cold plasma makes the therapy painless. It has the ability to activate cellular responses and important mechanisms in the body. They target specific molecules such as prothrombin for blood coagulation, cytokines for killing bacteria, and angiogenesis for tissue regeneration. Plasma has bactericidal, fungicidal and virucidal properties. Plasma technology has flourishing future in diverse fields like Textiles, Nanofabrication, Automotives, Waste management, Microbiology, Food Hygiene, Medical Science like Skin treatments, sterilisation of wounds, Hand disinfection, Dental treatments etc. Food hygiene using plasma can be achieved in disinfection of food containers, food surface disinfection, hygiene in food handling, preparation and packaging.
