Abstract

Rigorous external validation is crucial for assessing the generalizability of prediction models, particularly by evaluating their discrimination (AUROC) on new data. This often involves comparing a new model's AUROC to that of an established reference model. However, many studies rely on arbitrary rules of thumb for sample size calculations, often resulting in underpowered analyses and unreliable conclusions. This paper reviews crucial concepts for accurate sample size determination in AUROC-based external validation studies, making the theory and practice more accessible to researchers and clinicians. We introduce powerROC, an open-source web tool designed to simplify these calculations, enabling both the evaluation of a single model and the comparison of two models. The tool offers guidance on selecting target precision levels and employs flexible approaches, leveraging either pilot data or user-defined probability distributions. We illustrate powerROC's utility through a case study on hospital mortality prediction using the MIMIC database. more...

Abstract

PLATO (PLAnetary Transits and Oscillations of stars) is ESA's M3 mission designed to detect and characterise extrasolar planets and perform asteroseismic monitoring of a large number of stars. PLATO will detect small planets (down to <2 R_(Earth)) around bright stars (<11 mag), including terrestrial planets in the habitable zone of solar-like stars. With the complement of radial velocity observations from the ground, planets will be characterised for their radius, mass, and age with high accuracy (5 %, 10 %, 10 % for an Earth-Sun combination respectively). PLATO will provide us with a large-scale catalogue of well-characterised small planets up to intermediate orbital periods, relevant for a meaningful comparison to planet formation theories and to better understand planet evolution. It will make possible comparative exoplanetology to place our Solar System planets in a broader context. In parallel, PLATO will study (host) stars using asteroseismology, allowing us to determine the stellar properties with high accuracy, substantially enhancing our knowledge of stellar structure and evolution. The payload instrument consists of 26 cameras with 12cm aperture each. For at least four years, the mission will perform high-precision photometric measurements. Here we review the science objectives, present PLATO's target samples and fields, provide an overview of expected core science performance as well as a description of the instrument and the mission profile at the beginning of the serial production of the flight cameras. Abstract

Understanding the causal impact of medical interventions is essential in healthcare research, especially through randomized controlled trials (RCTs). Despite their prominence, challenges arise due to discrepancies between treatment allocation and actual intake, influenced by various factors like patient non-adherence or procedural errors. This paper focuses on the Complier Average Causal Effect (CACE), crucial for evaluating treatment efficacy among compliant patients. Existing methodologies often rely on assumptions such as exclusion restriction and monotonicity, which can be problematic in practice. We propose a novel approach, leveraging supervised learning architectures, to estimate CACE without depending on these assumptions. Our method involves a two-step process: first estimating compliance probabilities for patients, then using these probabilities to estimate two nuisance components relevant to CACE calculation. Building upon the principal ignorability assumption, we introduce four root-n consistent, asymptotically normal, CACE estimators, and prove that the underlying mixtures of experts' nuisance components are identifiable. Our causal framework allows our estimation procedures to enjoy reduced mean squared errors when exclusion restriction or monotonicity assumptions hold. Abstract

Typing mathematics is sometimes difficult with text editor functions for students with motor impairment and other associated impairments (visual, cognitive). Based on the HandiMathKey software keyboard, a user-centred design method involving the ecosytem of disabled students was applied to design the HMK-D physical keyboard for mathematical input. We opted for the Stream Deck device because of its multimedia features and its appeal to young students to the HMK-D. Preliminary tests with 8 students (5 in secondary school and 3 in high school) shows that HMK-D is highly accepted, accessible and fun for mathematical input by students with impairments. Abstract

Self-supervised monocular depth estimation methods aim to be used in critical applications such as autonomous vehicles for environment analysis. To circumvent the potential imperfections of these approaches, a quantification of the prediction confidence is crucial to guide decision-making systems that rely on depth estimation. In this paper, we propose MonoProb, a new unsupervised monocular depth estimation method that returns an interpretable uncertainty, which means that the uncertainty reflects the expected error of the network in its depth predictions. We rethink the stereo or the structure-from-motion paradigms used to train unsupervised monocular depth models as a probabilistic problem. Within a single forward pass inference, this model provides a depth prediction and a measure of its confidence, without increasing the inference time. We then improve the performance on depth and uncertainty with a novel self-distillation loss for which a student is supervised by a pseudo ground truth that is a probability distribution on depth output by a teacher. To quantify the performance of our models we design new metrics that, unlike traditional ones, measure the absolute performance of uncertainty predictions. Abstract

PLAnetary Transits and Oscillations of stars (PLATO) is the ESA M3 space mission dedicated to detect and characterise transiting exoplanets including information from the asteroseismic properties of their stellar hosts. The uninterrupted and high-precision photometry provided by space-borne instruments such as PLATO require long preparatory phases. An exhaustive list of tests are paramount to design a mission that meets the performance requirements, and as such, simulations are an indispensable tool in the mission preparation. To accommodate PLATO's need of versatile simulations prior to mission launch - that at the same time describe accurately the innovative but complex multi-telescope design - we here present the end-to-end PLATO simulator specifically developed for the purpose, namely PlatoSim. We show step-by-step the algorithms embedded into the software architecture of PlatoSim that allow the user to simulate photometric time series of CCD images and light curves in accordance to the expected observations of PLATO. In the context of the PLATO payload, a general formalism of modelling, end-to-end, incoming photons from the sky to the final measurement in digital units is discussed. We show the strong predictive power of PlatoSim through its diverse applicability and contribution to numerous working groups within the PLATO Mission Consortium. This involves the on-going mechanical integration and alignment, performance studies of the payload, the pipeline development and assessments of the scientific goals. PlatoSim is a state-of-the-art simulator that is able to produce the expected photometric observations of PLATO to a high level of accuracy. Abstract

Background: Bone metastasis is a significant concern in advanced solid tumors, contributing to diminished patient survival and quality of life due to skeletal-related events (SREs). Denosumab (DMAB), a monoclonal antibody targeting the receptor activator of nuclear factor kappa-B ligand (RANKL), is used to prevent SREs in such cases. The RANK/RANKL axis, crucial in immunological processes, has garnered attention, especially with the expanding use of immune checkpoint inhibitors (ICI) in modern oncology. Objective: Our study aims to explore the potential synergistic antitumor effects of combining immunotherapy with denosumab, as suggested by anecdotal evidence, small cohort studies, and preclinical research. Methods: We conducted a retrospective analysis using the IMMUCARE database, encompassing patients receiving ICI treatment since 2014 and diagnosed with bone metastases. We examined overall survival (OS), progression-free survival (PFS) and switch of treatment line based on denosumab usage. Patients were stratified into groups: without denosumab, ICI followed by denosumab, and denosumab followed by ICI. Survival curves and multivariate Cox regression analyses were performed. Results: Among the 268 patients with bone metastases, 154 received treatment with ICI alone, while 114 received ICI in combination with denosumab at some point during their oncological history. No significant differences were observed in overall survival (OS) or progression-free survival (PFS) between patients receiving ICI monotherapy and those receiving ICI with denosumab (p = 0.29 and p = 0.79, respectively). However, upon analyzing patients who received denosumab following ICI initiation (17 patients), a notable difference emerged. The group receiving ICI followed by denosumab exhibited a significant advantage compared to those without denosumab (154 patients) or those receiving denosumab before ICI initiation (72 patients) (p = 0.022). Conclusion: This retrospective investigation supports the notion of potential benefits associated with sequential administration of ICI and denosumab, although statistical significance was not achieved. Future studies, including prospective trials or updated retrospective analyses, focusing on cancers treated with first-line immunotherapy, could provide further insights into this therapeutic approach. more...

Abstract

Individualized treatment rules (ITRs) are deterministic decision rules that recommend treatments to individuals based on their characteristics. Though ubiquitous in medicine, ITRs are hardly ever evaluated in randomized controlled trials. To evaluate ITRs from observational data, we introduce a new probabilistic model and distinguish two situations: i) the situation of a newly developed ITR, where data are from a population where no patient implements the ITR, and ii) the situation of a partially implemented ITR, where data are from a population where the ITR is implemented in some unidentified patients. In the former situation, we propose a procedure to explore the impact of an ITR under various implementation schemes. In the latter situation, on top of the fundamental problem of causal inference, we need to handle an additional latent variable denoting implementation. To evaluate ITRs in this situation, we propose an estimation procedure that relies on an expectation-maximization algorithm. In Monte Carlo simulations our estimators appear unbiased with confidence intervals achieving nominal coverage. Academic rankings, despite their pitfalls, play a major role in the "business of business schools." The claim that "rankings are here to stay" continues to be frequently accepted at face value. This essay challenges this situation, arguing that while many business schools perceive rankings as a source of legitimacy, their participation in rankings can actually undermine the very legitimacy they seek. By adopting an effectual stance—which focuses on problem-solving and innovation using available resources—we theorize about a phased approach to steer business schools (and other higher education institutions) away from reliance on "ranking-based legitimacy" toward "research-based legitimacy." In the first phase, deans and academics can consider short-term bridging measures that do not fundamentally change the ranking game but influence it in self-serving directions. Rather than being powerless "ranking takers," some proactive approaches can allow business schools to change the zero-sum game nature of rankings and become "ranking makers." In the second phase, deans and academics can challenge and reject the ranking game by reaffirming the true purpose of business education.
