Abstract

The Large Hadron electron Collider (LHeC) is designed to move the field of deep inelastic scattering (DIS) to the energy and intensity frontier of particle physics. Exploiting energy recovery technology, it collides a novel, intense electron beam with a proton or ion beam from the High Luminosity--Large Hadron Collider (HL-LHC). The accelerator and interaction region are designed for concurrent electron-proton and proton-proton operation. This report represents an update of the Conceptual Design Report (CDR) of the LHeC, published in 2012. It comprises new results on parton structure of the proton and heavier nuclei, QCD dynamics, electroweak and top-quark physics. It is shown how the LHeC will open a new chapter of nuclear particle physics in extending the accessible kinematic range in lepton-nucleus scattering by several orders of magnitude. Due to enhanced luminosity, large energy and the cleanliness of the hadronic final states, the LHeC has a strong Higgs physics programme and its own discovery potential for new physics. Building on the 2012 CDR, the report represents a detailed updated design of the energy recovery electron linac (ERL) including new lattice, magnet, superconducting radio frequency technology and further components. Challenges of energy recovery are described and the lower energy, high current, 3-turn ERL facility, PERLE at Orsay, is presented which uses the LHeC characteristics serving as a development facility for the design and operation of the LHeC. An updated detector design is presented corresponding to the acceptance, resolution and calibration goals which arise from the Higgs and parton density function physics programmes. The paper also presents novel results on the Future Circular Collider in electron-hadron mode, FCC-eh, which utilises the same ERL technology to further extend the reach of DIS to even higher centre-of-mass energies., Comment: 373 pages, many figures, to be published by J. Abstract

In this paper, we propose a real-time free-running time scale based on four remote hydrogen masers. The clocks in the ensemble were scattered around Beijing, connected by urban fiber links using a novel frequency synchronization system. The remote clock ensemble prevents the time scale from potential problems caused by correlation among co-located clocks. Insofar as it is real-time, it fulfills the requirements for applications such as navigation, telecommunications and so on. The free-running time scale is updated every 1200 s, and a disturbance-resistant algorithm makes it robust to fiber link disturbances and clock malfunctions. The results of a continuous experiment over 224 days are reported. The stability of the time scale outperformed any clock in the ensemble for averaging times of more than approximately 10000 s.

Abstract

The correlation of co-located hydrogen masers (H-masers) is difficult to measure because their common-mode noise induced by the environment will be cancelled out during the comparison measurement. With the development of fibre-based high-precision time and frequency transfer technique, the correlation of co-located hydrogen masers can be directly measured with the help of remote H-masers. Recently, a fiber-based frequency synchronization network was constructed in the Beijing region by connecting 5 H-masers from 4 institutions. The correlation coefficient of atomic clocks is defined and the correlation between two co-located H-masers is measured using both experimental and simulative methods. The results show that the correlation is not prominent until the averaging time is larger than $\sim10^3$s; then, the coefficient grows rapidly for averaging times ranging from $\sim10^3$s to $\sim10^5$s and decreases beyond $\sim10^5$s up to 5 days.

6. The elastic anisotropy, thermodynamic and tensile properties of Ti2AX (A = Al and Ga, X = C and N) MAX phases: first-principles calculations.

Abstract

Although Ti2AX has excellent mechanical properties of both metals and ceramics, there are few papers exploring the correlation between mechanics and thermodynamics simultaneously. In this paper, the structure properties, elastic anisotropies, thermodynamic and tensile properties of Ti2AX (A = Al and Ga, X = C and N) MAX phases are studied by first-principles calculations. The results show that these MAX phases have thermodynamical, dynamical, and mechanical stabilities. The calculated anisotropy indexes indicate that the MAX phases are anisotropic in elastic modulus. Ti-X chemical bonds possess covalent-ionic mixed nature. Moreover, the sound velocities and thermal conductivities of these Ti2AX MAX phases are also anisotropic. According to the tensile calculation, the tensile strength along the [110] direction is greater than that along the [001] direction, and fracture failure will occur in the [110] direction. 9. Highly accurate fiber transfer delay measurement with large dynamic range

Abstract

A novel and efficient method for fiber transfer delay measurement is demonstrated. Fiber transfer delay measurement in time domain is converted into the frequency measurement of the modulation signal in frequency domain, accompany with a coarse and easy ambiguity resolving process. This method achieves a sub-picosecond resolution, with an accuracy of 1 picosecond, and a large dynamic range up to 50 km as well as no measurement dead zone.

Abstract

Functional characterization of disease-causing variants at risk loci has been a significant challenge. Here we report a high-throughput single-nucleotide polymorphisms sequencing (SNPs-seq) technology to simultaneously screen hundreds to thousands of SNPs for their allele-dependent protein-binding differences. This technology takes advantage of higher retention rate of protein-bound DNA oligos in protein purification column to quantitatively sequence these SNP-containing oligos. We apply this technology to test prostate cancer-risk loci and observe differential allelic protein binding in a significant number of selected SNPs. We also test a unique application of self-transcribing active regulatory region sequencing (STARR-seq) in characterizing allele-dependent transcriptional regulation and provide detailed functional analysis at two risk loci (RGS17 and ASCL2). Together, we introduce a powerful high-throughput pipeline for large-scale screening of functional SNPs at disease risk loci.

20. MiR-186 promotes the apoptosis of glioma U87 cells by down-regulating the expression of Smad6.

Abstract

OBJECTIVE: MiRNA family gene is an evolutionarily conserved non-coding small RNA that directly participates in a variety of physiological processes and cancer development via regulating gene expression in the biological level of transcription. To research the specific mechanism by which miR-186 regulates apoptosis within gliomas. PATIENTS AND METHODS: RT-qPCR was performed to verify the transcriptional level of miR-186 within glioma tissues and glioma cells. miRanda and Dual-Luciferase assay were performed to predict and confirm that Smad6 gene is an effective target of miR-186 within glioma. The expression of Smad6 protein was tested by Western blot following cell effective transfection. Apoptosis of gliomas was analyzed by inverted fluorescence microscopy and flow cytometry. RESULTS: The mRNA level of miR-186 was suppressed within glioma tissues and glioma U87 cells. MiR-186 is associated with apoptosis in glioma. Overexpression of miR-186 promoted U87 cell apoptosis, whereas suppression of miR-186 had the opposite effect. Besides, miR-186 directly targeted Smad6 and suppress its expression in glioma. The expression of Smad6 affected the regulation of miR-186 on glioma cell apoptosis, restoration of Smad6 rescued apoptosis of glioma U87 cells induced by miR-186 mimics, whereas inhibition of Smad6 promoted apoptosis. CONCLUSIONS: As noted above, miR-186 exerts a tumor-suppressing effect by targeting Smad6. We propose that miR-186 can be used as a novel biomarker for glioma diagnosis in the future, or as a new pharmacy target in the cure of gliomas. [ABSTRACT FROM AUTHOR]

21. Antiplatelet agents for the secondary prevention of ischaemic stroke in patients with or without renal dysfunction.

Abstract

Background and purpose: Studies on using antiplatelet agents for secondary prevention in ischaemic stroke patients with renal dysfunction are limited. The Taiwan Stroke Registry database was used to compare the efficacy of antiplatelet agents. Methods: From the Taiwan Stroke Registry data, 39 174 acute ischaemic stroke patients were identified and were classified into three groups by antiplatelet agent: aspirin, clopidogrel and dual antiplatelet therapy (DAPT) with a combination of aspirin and clopidogrel. The re‐stroke incidence and 1‐year mortality were stratified by estimated glomerular filtration rate (eGFR) levels at admission: ≥90, 60–89 and <60 ml/min/1.73 m2 or on dialysis. Results: Compared to the aspirin group, the re‐stroke differences were not statistically significant for the clopidogrel group [adjusted subhazard ratio 0.95, 95% confidence interval (CI) 0.84–1.08] and the DAPT group (adjusted subhazard ratio 1.03, 95% CI 0.77–1.39) after controlling for the competing risk of death. The mortality rate increased as the eGFR level declined. In addition, compared to patients taking aspirin, there was no statistically significant difference in overall 1‐year mortality for the clopidogrel group (adjusted hazard ratio 1.11, 95% CI 0.95–1.29) and for the DAPT group (adjusted hazard ratio 1.01, 95% CI 0.67–1.54). The results were consistent in different subgroups stratified by eGFR levels. Conclusions: There was no difference in the risks of recurrent stroke and 1‐year mortality amongst ischaemic stroke patients with or without renal dysfunction receiving antiplatelet agents with aspirin, clopidogrel or dual agents with a combination of aspirin and clopidogrel, regardless of their renal dysfunction status. 30. Zyxin: a mechanotransductor to regulate gene expression.

Abstract

OBJECTIVE: Cells answer to biochemical, electrical and mechanical signals in the environment, which regulate their behavior. Mechanical signals can propagate through mechanically stiff structures like focal adhesions (FAs). Zyxin, a LIM domain protein, is localized primarily at focal adhesion plaques. Growing evidence suggests that zyxin is a vital mechanotransductor to regulate the gene expression. In this review, we summarize the features of zyxin and the molecular mechanism of how zyxin participate in the cellular activity. MATERIALS AND METHODS: An English-language literature search is based on a keyword- based query of multiple databases (MEDLINE, Embase) and bibliographies from identified publications. The references in the selected paper are also considered as an additional source of data. The search was last updated in April 2018; no limitations are applied. RESULTS: Zyxin enhances actin polymerization with the aid of Enabled (Ena)/vasodilator- stimulated phosphoprotein (VASP) proteins in response to mechanical tension, to perform its role in stress fibers (SFs) remodeling and repair. Zyxin can translocate from focal adhesions (FAs) to the nucleus responds to stretch, and regulate gene transcription by interaction with transcription factors like nuclear matrix protein 4 (NMP4). Misregulation of nuclear functions of zyxin appears to be associated with pathogenic effects and diseases, such as prostate cancer and non-small-cell lung cancer. CONCLUSIONS: Zyxin is a crucial ingredient of the cellular mechanotransducing system and can modulate the gene expression. Given its clinical relevance, zyxin is also a promising target for the diagnosis and treatment of certain diseases. Understanding the role of zyxin in force sensing and gene expression regulating provides a compelling challenge for future biomechanics studies, and offers attractive evidence for zyxin as a potential diagnostic marker and therapeutic target for clinical diseases. Abstract: A 10‐week growth trial was run to evaluate effects of myo‐inositol (MI) on growth performance, haematological parameters, antioxidative capacity and salinity stress tolerance of Litopenaeus vannamei. Six practical diets supplemented with graded levels of MI (designated as MI0, MI600, MI1200, MI2400, MI 3600 and MI4800 for 448.8, 974.2, 1568.0, 2810.6, 3835.5 and 4893.6 mg/kg diet, respectively) were fed to six replicate groups of L. vannamei (mean initial body weight 0.63 ± 0.00 g). The results showed that significant increment of growth performance was observed in shrimp fed MI600 diet than those fed MI1200 diet. Lipid concentration in whole body of the shrimp fed MI600 diet was significantly increased. Shrimp fed MI0 diet had lower total protein (TP) as compared to shrimp fed the MI‐supplemented diets (except MI4800 diet). In general, lower activities of antioxidant enzymes and higher malondialdehyde (MDA) content in haemolymph and hepatopancreas were recorded in shrimp fed MI0 diet, compared to those fed the MI‐supplemented diets. Reduced survival after 7‐h salinity stress was present in shrimp fed MI0 diet as compared to those fed MI4800 diet. Cast iron with good mechanical and thermal combination property is preferred material for the cylinder head of high power density diesel engine. Interface between graphite and matrix should play a key role in thermal conductivity of cast iron. However, it is difficult to measure the interfacial contact thermal conductance (ICTC) between graphite inclusion and matrix in cast iron. In this paper, a modeling experimental sample containing a macroscopic interface is designed and ICTC can be estimated with the aid of finite element (FE) calculation. At last the effect of ICTC on the thermal conductivity of cast iron is assessed according to the change of metallographic FE simulation results before and after considering ICTC. Results show that the magnitude of ICTC is between 1.0 × 105 and 2.0 × 105 W m−2·K on the whole. The interface makes the theoretical thermal conductivity of cast iron drop down 34.6%. Abstract

(Ti, Zn)-co-doped Bi0.97Nd0.03FeO3(BNFTZ) thin films with different thicknesses were prepared directly on indium tin oxide/glass substrates deriving from the precursor solutions with different solution concentrations. The X-ray diffraction, scanning electron microscopy and insulating/ferroelectric/dielectric measurements were utilised to characterise the BNFTZ thin films. Both BNFTZ thin films with the thicknesses of 160 and 320 nm are crystallised into phase-pure perovskite structure without second phase. Compared to the film of 160 nm, a lower leakage current can be obtained in BNFTZ film of 320 nm due to due to its denser microstructure with smaller grain size. The film with 320 nm thickness possesses a high energy-storage efficiency of 87%, which can be ascribed to the slimP–Eloop. At 100 kHz, the film of 320 nm owns lower dielectric constant (~91) and dissipation factor (~0.10). The possible contributions are discussed to explain the difference in the related performances for BNFTZ thin films with different thicknesses. [ABSTRACT FROM AUTHOR]

Abstract

OBJECTIVE: To analyze how changes in the levels of brain-derived neurotrophic factor (BDNF) and neuroglobin (NGB) affect learning and memory in rats with intracerebral hemorrhage. MATERIALS AND METHODS: Thirty male Sprague-Dawley rats were randomly divided into the control group, sham operation group and model group with 10 rats each. The rats in the control group were untreated, while those in the sham operation group were treated with sterile saline instead of type VII collagenase injection in the globus pallidus. The model of cerebral hemorrhage was established according to the methods described by Rosenberg. The expression of perihematomal BDNF mRNA was measured by Real-time quantitative PCR (RT-PCR) for 7 days consecutively. Perihematomal NGB-positive cells were detected by immunohistochemistry. The Morris water maze was used to test the spatial learning and memory of rats. RESULTS: Compared with the control group and sham operation group, the expression of BDNF mRNA and number of NGB-positive cells in the model group were significantly higher. Furthermore, the escape latency was significantly prolonged (p < 0.05). The NGB and BDNF mRNA levels and escape latency were positively correlated. The correlation coefficients were as follows: rs (NGB) = 1.1838 (p = 0.008); rs (BDNF) = 0.5948 (p = 0.012). CONCLUSIONS: Cerebral hemorrhage significantly inhibited the spatial learning and memory ability of rats. The mechanism may be related to decreased cerebral expression of BDNF and NGB. Abstract

Concrete cover separation is a common failure mode of RC beams strengthened with a fiber-reinforced polymer (FRP) plate externally bonded to the tension face (FRP-plated RC beams). This failure mode initiates at the critical plate end and then propagates at the level of steel tension reinforcement in the direction of increasing moment. Plate-end anchorage by FRP U-jackets has been specified in some design guidelines as a mitigation measure to delay or suppress concrete cover separation, although its effectiveness is far from clear. This mitigation method is more attractive than other options because the same strengthening material is used and the installation procedure is simple. This paper presents the first systematic experimental study on the use of FRP U-jackets for mitigating plate-end concrete cover separation failure, with particular attention to the effect of inclination angle between the U-jacket and the beam axis. A total of 10 fullscale FRP-plated RC beams were tested. The test results show that both the load-carrying capacity and the ductility of an FRP-plated RC beam can be significantly enhanced by FRP U-jacketing at the critical plate end. Inclined U-jacketing at 45° to the beam axis was found to be much more effective than vertical U-jacketing (i.e., at 90° to the beam axis) in mitigating plate-end cover separation failure. Abstract

As an ideal material for the cylinder head of high power density diesel engine, vermicular graphite cast iron (VGI) with higher thermal conductivity was received more and more attention recently. In this article, finite element (FE) software ANSYS was applied to investigate the changes of thermal conductivity of VGI with the vermicularity and the FE-models are carried out based on the real metallographic images of VGI. The results indicate that there is a big difference between the numerical simulation and the reality without the consideration of the thermal contact resistance of interface between matrix and graphite. The numerical simulation result is close to the reality when the contact thermal conductance is set at 0.1 W/(m²K). In addition, the equivalent thermal conductivity along X direction kequ,x rises from 34.437 W/mK to 38.902 W/mK and increases by 13% or so when the vermicularity is improved from 40% to 90% according to the numerical simulation. Abstract

Objective: To explore the relationship between amniotic fluid and peripheral blood inflammatory factors and the pregnancy outcomes after emergency cervical cerclage, and to identify effective indicators for predicting adverse pregnancy outcomes after the procedure. Methods: A case-control study was conducted, including pregnant women who were hospitalized at Sun Yat-sen Memorial Hospital, from January 1, 2013, to July 31, 2019, and underwent emergency cervical cerclage due to cervical dilatation at gestational age between 16 and 28 weeks. A total of 85 pregnant women who underwent amniocentesis for the detection of amniotic fluid inflammatory factors during the perioperative period were included. Based on whether their baby was perinatal death, the participants were divided into the case group (28 cases with perinatal death) and the control group (57 cases with live births). Univariate logistic regression analysis was performed to identify risk factors associated with adverse pregnancy outcomes, followed by multivariate logistic regression analysis to establish a regression model and nomogram. Results: (1) The levels of tumor necrosis factor α (TNF-α), interleukin (IL)-1β, IL-6, IL-8, IL-10 in the amniotic fluid during the perioperative period and postoperative serum C-reactive protein (CRP) were significantly higher in the case group compared to the control group (all P <0.05). The case group underwent emergency cervical cerclage at an earlier gestational age compared to the control group, and their cervical dilation was greater than that of the control group (all P <0.05). However, there were no significant differences in the white blood cell counts, neutrophil percentage, and the level of preoperative CRP in the peripheral blood of pregnant women during the perioperative period (all P >0.05). (2) Univariate logistic regression analysis showed that the levels of amniotic fluid WBC, TNF-α, IL-1β, IL-2 receptor (IL-2R), IL-6, IL-8, IL-10, postoperative CRP in the peripheral blood, gestational age at cerclage and cervical dilation were associated with adverse pregnancy outcomes (all P <0.05). Multivariate regression analysis indicated that only the levels of amniotic fluid WBC and TNF-α were independent risk factors for perinatal death. (3) Based on clinical practice, a multivariate logistic regression model was constructed including the levels of amniotic fluid TNF-α, WBC, gestational age at cervical cerclage, and cervical dilation. A nomogram and calibration curve were plotted, which suggested its good predictive value for adverse pregnancy outcomes. Conclusions: During the perioperative period of emergency cervical cerclage, the levels of amniotic fluid WBC, TNF-α, IL-1β, IL-2R, IL-6, IL-8, IL-10 are associated with adverse pregnancy outcomes, with amniotic fluid WBC and TNF-α showing the closest relationship. However, there is no significant correlation between maternal peripheral hemogram during the perioperative period and adverse pregnancy outcomes. A model constructed by amniotic fluid TNF-α, WBC, cervical cerclage gestational age, and cervical dilation has a good predictive effect on adverse pregnancy outcomes.

Abstract

Outbreaks caused by highly industrialized food companies are characterized by cross-border, trans-regional, rapid and unpredictable, related to serious disease and economic burden. A cluster of cases with monophasic s almonella enterica serovar Typhimurium ST34 infection suspected to be associated with consumption of contaminated chocolate products have been reported in several Europe countries since December 2021. After retrospective investigations, the buttermilk circuit in the Belgian factory was suspected to be the point of origin of the contamination. This outbreak could provide a reference for the risk management of foodborne pathogens contamination in China. The objective of this paper was to summarize the process and characteristics of the outbreak of monophasic S. Typhimurium caused by contaminated chocolate products, analyze the characteristics of ST34 monophasic S. Typhimurium and the microbial management measures in the process of chocolate products, and systematically discuss the suggestions for the risk management of foodborne pathogens contamination and countermeasures for the rapid development of industrialization of food enterprises in China, in order to provide scientific and technological support for the prevention and control, prediction and early warning of sudden cases in China.

Abstract

Objective: To explore the incidence and risk factors of postoperative surgical site infection (SSI) after colon cancer surgery. Methods: A retrospective case-control study was performed. Patients diagnosed with colon cancer who underwent radical surgery between January 2016 and May 2021 were included, and demographic characteristics, comorbidities, laboratory tests, surgical data and postoperative complications were extracted from the specialized prospective database at Department of General Surgery, Peking Union Medical College Hospital. Case exclusion criteria: (1) simultaneously multiple primary colon cancer; (2) segmental resection, subtotal colectomy, or total colectomy; (3) patients undergoing colostomy/ileostomy during the operation or in the state of colostomy/ileostomy before the operation; (4) patients receiving natural orifice specimen extraction surgery or transvaginal colon surgery; (5) patients with the history of colectomy; (6) emergency operation due to intestinal obstruction, perforation and acute bleeding; (7) intestinal diversion operation; (8) benign lesions confirmed by postoperative pathology; (9) patients not following the colorectal clinical pathway of our department for intestinal preparation and antibiotic application. Univariate analysis and multivariate analysis were used to determine the risk factors of SSI after colon cancer surgery. Results: A total of 1291 patients were enrolled in the study. 94.3% (1217/1291) of cases received laparoscopic surgery. The incidence of overall SSI was 5.3% (69/1291). According to tumor location, the incidence of SSI in the right colon, transverse colon, left colon and sigmoid colon was 8.6% (40/465), 5.2% (11/213), 7.1% (7/98) and 2.1% (11/515) respectively. According to resection range, the incidence of SSI after right hemicolectomy, transverse colectomy, left hemicolectomy and sigmoid colectomy was 8.2% (48/588), 4.5% (2/44), 4.8% (8 /167) and 2.2% (11/492) respectively. Univariate analysis showed that preoperative BUN≥7.14 mmol/L, tumor site, resection range, intestinal anastomotic approach, postoperative diarrhea, anastomotic leakage, postoperative pneumonia, and anastomotic technique were related to SSI (all P <0.05). Multivariate analysis revealed that anastomotic leakage (OR=22.074, 95%CI: 6.172-78.953, P <0.001), pneumonia (OR=4.100, 95%CI: 1.546-10.869, P =0.005), intracorporeal anastomosis (OR=5.288, 95%CI: 2.919-9.577, P <0.001) were independent risk factors of SSI. Subgroup analysis showed that in right hemicolectomy, the incidence of SSI in intracorporeal anastomosis was 19.8% (32/162), which was significantly higher than that in extracorporeal anastomosis (3.8%, 16/426, χ(2)=40.064, P <0.001). In transverse colectomy [5.0% (2/40) vs. 0, χ(2)=0.210, P =1.000], left hemicolectomy [5.4% (8/148) vs. 0, χ(2)=1.079, P =0.599] and sigmoid colectomy [2.1% (10/482) vs. 10.0% (1/10), χ(2)=2.815, P =0.204], no significant differences of SSI incidence were found between intracorporeal anastomosis and extracorporeal anastomosis (all P >0.05). Conclusions: The incidence of SSI increases with the resection range from sigmoid colectomy to right hemicolectomy. Intracorporeal anastomosis and postoperative anastomotic leakage are independent risk factors of SSI. Attentions should be paid to the possibility of postoperative pneumonia and actively effective treatment measures should be carried out.
