1. Evaluation of Stay-Green QTL Introgression Lines for Drought Tolerance in Sorghum [Sorghum bicolor (L). Moench]

Abstract

Stay-green QTL introgression lines (plus the parents and checks) were evaluated during the 2004/05 and 2005/06 post-rainy sorghum cropping season at the International Crops Research Institute for the Semi Arid Tropics (ICRISAT), Patancheru, India. The staygreen introgression lines (BC2F2/BC1F3) were field evaluated for their agronomic and stay-green related characters under well watered and water-stress conditions. The introgression lines had high green leaf area (%) and chlorophyll content compared with the senescent lines showing the relationships among these traits. Moreover, many of the introgression lines had grain yields statistically similar to grain yields of the recurrent parent (R16). The introgression lines such as RSG04001 were good both in terms of green leaf area (%) and grain yield indicating the contribution of stay green to grain yield. This has also been evidenced by the highly significant (R2 = 0.47 in 2004/05 and R2 = 0.77 in 2005/06) correlations of relative grain yield to relative green leaf area (%) in the moisture stress environments. Overall, the results showed promise for utilization of the stay-green trait for better grain yield in drought-prone environments.&nbsp

Abstract

Pearl millet [Cenchrus americanus (L.) Morrone] is a staple food for more than 90 million farmers in arid and semi-arid regions of sub-Saharan Africa, India and South Asia. We report the ∼1.79 Gb draft whole genome sequence of reference genotype Tift 23D2B1-P1-P5, which contains an estimated 38,579 genes. We highlight the substantial enrichment for wax biosynthesis genes, which may contribute to heat and drought tolerance in this crop. We resequenced and analyzed 994 pearl millet lines, enabling insights into population structure, genetic diversity and domestication. We use these resequencing data to establish marker trait associations for genomic selection, to define heterotic pools, and to predict hybrid performance. We believe that these resources should empower researchers and breeders to improve this important staple crop.

Abstract

International audience; Pearl millet is the main component of traditional farming systems and a staple grain in the diet of sub-Saharan Africa and India. To facilitate breeding work in this crop, a genetic map consisting of single nucleotide polymorphism (SNP) markers was constructed using an F2 population of 93 progenies, from a wild × cultivated pearl millet cross. We used a modified genotyping-by-sequencing (GBS) protocol involving two restriction enzymes (PstI–MspI) and PCR amplification with primers including three selective bases to generate 3,321 SNPs. Of these, 2,809 high-quality SNPs exhibited a minor allele frequency ≥0.3. In total, 314 non-redundant haplotypes and 85 F2 individuals were used to construct a genetic map spanning a total distance of 640 cM. These SNPs were evenly distributed over seven linkage groups ranging considerably in size (62–123 cM). The average density for this map was 0.51 SNP/cM, and the average interval between SNP markers was 2.1 (±0.6) cM. Finally, to establish bridges between the linkage groups of this and previous maps, 19 SSR markers were examined for polymorphism between the parents of this population. We could only tentatively suggest a correspondence between four of our linkage groups and those of previous maps. Overall, GBS enabled us to quickly produce a genetic map with a density and uniformity of markers greater than previously published maps. The availability of such a map will be useful for the identification of genomic regions associated with Striga resistance and other important agronomic traits.

Abstract

There is substantial genetic variation for drought adaption in pearl millet in terms of traits controlling plant water use. It is important to understand genomic regions responsible for these traits. Here, F7 recombinant inbred lines were used to identify quantitative trait loci ( QTL) and allelic interactions for traits affecting plant water use, and their relevance is discussed for crop productivity in water-limited environments. Four QTL contributed to increased transpiration rate under high vapour pressure deficit ( VPD) conditions, all with alleles from drought-sensitive parent ICMB 841. Of these four QTL, a major QTL (35.7%) was mapped on linkage group ( LG) 6. The alleles for 863B at this QTL decreased transpiration rate and this QTL co-mapped to a previously detected LG 6 QTL, with alleles from 863B for grain weight and panicle harvest index across severe terminal drought stress environments. This provided additional support for a link between water saving from a lower transpiration rate under high VPD and drought tolerance. 863B alleles in this same genomic region also increased shoot weight, leaf area and total transpiration under well-watered conditions. One unexpected outcome was reduced transpiration under high VPD (15%) from the interaction of two alleles for high VPD transpiration ( LG 6 (B), 40.7) and specific leaf mass and biomass ( LG 7 (A), 35.3), (A, allele from ICMB 841, B, allele from 863B, marker position). The LG 6 QTL appears to combine alleles for growth potential, beneficial for non-stress conditions, and for saving water under high evaporative demand, beneficial under stressful conditions. Mapping QTL for water-use traits, and assessing their interactions offers considerable potential for improving pearl millet adaptation to specific stress conditions through physiology-informed marker-assisted selection.
