Cortical bone loss is intricately associated with ageing and coincides with iron accumulation. The precise role of ferroptosis, characterized by iron overload and lipid peroxidation, in senescent osteocytes remains elusive. We found that ferroptosis was a crucial mode of osteocyte death in cortical bone during ageing. Using a single‐cell transcriptome analysis, we identified activating transcription factor 3 (ATF3) as a critical driver of osteocyte ferroptosis. Elevated ATF3 expression in senescent osteocytes promotes iron uptake by upregulating transferrin receptor 1 while simultaneously inhibiting solute carrier family 7‐member 11‐mediated cystine import. This process leads to an iron overload and lipid peroxidation, culminating in ferroptosis. Importantly, ATF3 inhibition in aged mice effectively alleviated ferroptosis in the cortical bone and mitigated cortical bone mass loss. Taken together, our findings establish a pivotal role of ferroptosis in cortical bone loss in older adults, providing promising prevention and treatment strategies for osteoporosis and fractures. We sought to confirm whether use of Acinetobacter strains Y7 and Y16, both strains of heterotrophic nitrifying bacteria, was practical for removing ammonium (NH-N) from drinking water at low temperatures. To test this, ammonium-containing drinking water was treated with strains Y7 and Y16 at 8 and 2 °C. Continuous ammonium treatment was conducted in order to evaluate the performance of three biologically enhanced activated carbon (BEAC) filters in removing ammonium. The three BEAC filters were inoculated with strain Y7, strain Y16, and a mixture of strains Y7 and Y16, respectively. A granular activated carbon (GAC) filter, without inoculation by any strains, was tested in parallel with the BEAC filters as control. The results indicated that NH-N removal was significant when a BEAC filter was inoculated with the mixture of strains Y7 and Y16 (BEAC-III filter). Amounts of 0.44 ± 0.05 and 0.25 ± 0.05 mg L NH-N were removed using the BEAC-III filter at 8 and 2 °C, respectively. These values were 2.8-4.0-fold higher than the values of ammonium removal acquired using the GAC filter. The synergistic effect of using strains Y7 and Y16 in concert was the cause of the high-ammonium removal efficiency achieved by using the BEAC-III filter at low temperatures. In addition, a high C/N ratio may promote NH-N removal efficiency by improving biomass and microbial activity. Acinetobacter sp. HITLi 7 T showed capacity of heterotrophic nitrification at 2 °C with an optimum growth temperature of 20 °C. The ammonium removal rates were 0.09 and 0.24 mg l −1 h −1 at 2 °C and 20 °C respectively. Cell growth and proliferation affected ammonium removal performance of HITLi 7 T dramatically at low temperature. To investigate the influence of low temperature on protein expression, the response of HITLi 7 T cultivated at 2 °C and 20 °C were characterized and compared by 2-dimensional proteome analyses. A total of 151 proteins were identified to be differentially expressed by the analysis of PDQuest 2D gel analysis software. In comparison to protein expression at 20 °C, 17, 28, and 43 protein spots were found to be up-regulated, down-regulated and only present at 2 °C, respectively. 9 spots, which were overexpressed or induced at low temperature, were successfully identified by peptide mass fingerprinting by matrix-assisted laser desorption ionization-time-of-flight mass spectrometry. The significantly up-regulated proteins at 2 °C were related to metabolism. The proteins only present at 2 °C had functions of protein biosynthesis and refolding. These findings indicated that cold adaption of HITLi 7 T relied on regulation of metabolic pathways, synthesis of proteins and protein refolding. [ABSTRACT FROM AUTHOR]

Background: Radioactive iodine-125 ( 125 I) microparticle therapy is a new type of internal radiation therapy that has shown unique advantages in the treatment of malignant tumors, especially hepatocellular carcinoma. Patients with hepatocellular carcinoma frequently experience portal vein embolism, which exacerbates the difficulty and complexity of treatment. 125 I particles, used in local radiotherapy, can directly act on tumor tissue and reduce damage to surrounding healthy tissue. Through retrospective analysis, this study discussed the efficacy and safety of radioactive 125 I particles in portal vein embolization patients with hepatocellular carcinoma in order to provide more powerful evidence supporting clinical treatment., Aim: To investigate the effect of transcatheter arterial chemoembolization combined with portal vein 125 I particle implantation in the treatment of primary liver cancer patients with portal vein tumor thrombus and its influence on liver function., Methods: The clinical data of 96 patients with primary liver cancer combined with portal vein tumor thrombus admitted to our hospital between January 2020 and December 2023 were retrospectively analyzed. Fifty-two patients received treatment with transcatheter arterial chemoembolization and implantation of 125 I particles in the portal vein (combination group), while 44 patients received treatment with transcatheter arterial chemoembolization alone (control group). The therapeutic effects on tumor lesions, primary liver cancer, and portal vein tumor embolisms were compared between the two groups. Changes in relevant laboratory indexes before and after treatment were evaluated. The t test was used to compare the measurement data between the two groups, and the χ 2 test was used to compare the counting data between groups., Results: The tumor lesion response rate in the combination group (59.62% vs 38.64%) and the response rate of patients with primary liver cancer complicated with portal vein tumor thrombus (80.77% vs 59.09%) were significantly greater than those in the control group ( χ 2 = 4.196, 5.421; P = 0.041, 0.020). At 8 wk after surgery, the serum alpha-fetoprotein, portal vein main diameter, and platelet of the combined group were significantly lower than those of the control group, and the serum alanine aminotransferase, aspartate aminotransferase, and total bilirubin were significantly greater than those of the control group ( t = 3.891, 3.291, 2.330, 3.729, 3.582, 4.126; P < 0.05). The serum aspartate aminotransferase, alanine aminotransferase, and total bilirubin levels of the two groups were significantly greater than those of the same group 8 wk after surgery ( P < 0.05), and the peripheral blood platelet, alpha-fetoprotein, and main portal vein diameter were significantly less than those of the same group before surgery ( P < 0.05)., Conclusion: In patients with primary liver cancer and a thrombus in the portal vein, transcatheter arterial chemoembolization plus portal vein 125 I implantation is more effective than transcatheter arterial chemoembolization alone. Renal cell carcinoma (RCC) is often diagnosed in advanced stages and a third of patients have distant metastasis at diagnosis. Metastasis may be the first evidence of clear cell RCC in many cases. RCC most often metastasises to the lung, liver, bone, brain and thyroid; however, metastatic disease to the oral cavity, especially the mandible, is rare. The purpose of this study is to report a case of clear cell RCC metastatic to the mandible and review the literature. The mandible lesion underwent radical excision in this case. Notably, no metastatic lesions were detected in the lungs and liver in this patient until 15 months after the mandibulectomy. The patient lived for around 2.5 years after the diagnosis of RCC.

Herb-partitioned moxibustion can effectively mitigate visceral pain, a major symptom in inflammatory bowel disease, but the analgesic mechanism is still unclear. Moreover, extracellular signal-regulated kinase, substance P, and neurokinin-1 are involved in formation of central hyperalgesia. Thus, we postulated that the analgesic effect of herb-partitioned moxibustion may be associated with these factors. Accordingly, in this study, we established an inflammatory bowel disease visceral pain model in rat by enema with a mixed solution of 5% trinitrobenzenesulfonic acid and 50% ethanol. Bilateral Tianshu (ST25) and Qihai (CV6) points were selected for herb-partitioned moxibustion. Our results showed that herb-partitioned moxibustion improved visceral pain and down-regulated extracellular signal-regulated kinase, substance P, and neurokinin-1 protein and mRNA expression in dorsal root ganglia. These results indicate that down-regulation of extracellular signal-regulated kinase, substance P, and neurokinin-1 protein and mRNA may be a central mechanism for the analgesic effect of herb-partitioned moxibustion., Competing Interests: Conflicts of interest: None declared.
