3. The impact of signaling pathways on the desmosome ultrastructure in pemphigus.

Abstract

Introduction: The autoantibody-driven disease pemphigus vulgaris (PV) impairs desmosome adhesion in the epidermis. In desmosomes, the pemphigus autoantigens desmoglein 1 (Dsg1) and Dsg3 link adjacent cells. Dsgs are clustered by plaque proteins and linked to the keratin cytoskeleton by desmoplakin (Dp). The aim of this study was to identify the impact of several PV-related signaling pathways on desmosome ultrastructure. Methods: STED microscopy, Dispase-based dissociation assay. Results: As observed using STED microscopy, pemphigus autoantibodies (PV-IgG) reduced desmosome number, decreased desmosome size, increased plaque distance and thickness and caused loss of adhesion. Decreased desmosome number, increased plaque distance and thickness and loss of adhesion correlate with features found for newly assembled immature desmosomes, observed after Ca2+ depletion and repletion. This was paralleled by plaque asymmetry, keratin filament retraction and fragmentation of Dsg1 and Dsg3 immunostaining. Inhibition of each individual signaling pathway investigated here prevented the loss of adhesion and ameliorated keratin retraction. In addition, inhibition of p38MAPK or PLC completely rescued all parameters of desmosomes ultrastructure and increased desmosome number under basal conditions. In contrast, inhibition of MEK1/2 was only partially protective for desmosome size and plaque thickness, whereas inhibition of Src or increase of cAMP decreased desmosome size but increased the desmosome number even in the presence of PV-IgG. Discussion: Alterations of the desmosomal plaque ultrastructure are closely related to loss of adhesion and regulated differently by signaling pathways involved in pemphigus pathogenesis. 4. An Exploration of the Role of Ethnic Identity in Students' Construction of 'British Stories'

Abstract

Much of the research into history teaching and ethnicity concludes that the historical narratives that children from minority ethnic groups construct differ significantly from 'mainstream' or official national narratives and are often accompanied by a sense of disengagement or even alienation from the dominant history narratives taught in schools. Our research suggests that in England (or more specifically in London) the picture is more complicated than this. First, we did not find compelling evidence that students from black and minority ethnic backgrounds feel alienated or disengaged from the British history they are taught. Second, we found a surprising similarity across the narratives that different ethnic groups chose to tell us about British history, suggesting amongst other things some curriculum inertia in schools. Where differences did exist, we suggest that these can be explained as much by gender and broad cultural influences as by ethnicity. Third, whilst "what" students chose to include in their narratives was broadly similar across different ethnic groups, the reasons for including them "did" differ. Finally, and perhaps most positively, we conclude that students of all ethnic groups are keen to engage more critically with British narratives and would relish more opportunities to do so than current school curricula appear to encourage.

AbstractObjective:Method:Results:Conclusion:Research on nonverbal synchrony (NVS) as a core element in the therapeutic relationship has substantially increased and suggests that NVS influences therapeutic alliance and outcomes.Studies on NVS regarding body movements, vocal pitch, peripheral physiological measures, and hormonal states were included. A random-effects multilevel meta-analysis was performed on 23 publications from 13 trials.There was no significant overall association between NVS and alliance/outcome (r = .03, p = .644). Across modalities, there was a marginally significant association between higher NVS and smaller interpersonal problems (r = .10, p = .084) and a nonsignificant effect for the association between NVS and alliance (r = .06, p = .316), symptomatic outcome (r = -.06, p = .320), and other outcomes (r = .07, p = .255). There was significant heterogeneity between studies (I2 = 56.47). Moderator analyses revealed that vocal pitch synchrony was negatively associated with alliance/outcome (r = -.20, p = .011), while NVS of peripheral physiological parameters was positively correlated with alliance/outcome (r = .32, p = .006).Findings suggest that specific modalities of NVS show specific associations with outcomes. Simple Summary: The Wnt signaling pathway plays a pivotal role in governing developmental processes and maintaining stem cell characteristics, while also exhibiting significant implications in cancer pathogenesis. While initially characterized in colorectal cancer, aberrations in Wnt signaling are pervasive across various cancer types. Both intrinsic and extrinsic factors modulate Wnt signaling, mainly influencing the strictly coordinated kinase cascade within Wnt signaling. Understanding the complex interplay of canonical and non-canonical Wnt pathways and their potential dysregulation in disorders holds promise for the development of innovative therapeutic strategies in cancer treatment. Since the initial identification of oncogenic Wnt in mice and Drosophila, the Wnt signaling pathway has been subjected to thorough and extensive investigation. Persistent activation of Wnt signaling exerts diverse cancer characteristics, encompassing tumor initiation, tumor growth, cell senescence, cell death, differentiation, and metastasis. Here we review the principal signaling mechanisms and the regulatory influence of pathway-intrinsic and extrinsic kinases on cancer progression. Additionally, we underscore the divergences and intricate interplays of the canonical and non-canonical Wnt signaling pathways and their critical influence in cancer pathophysiology, exhibiting both growth-promoting and growth-suppressing roles across diverse cancer types. Abstract

This large, international dataset contains survey responses from N = 12,570 students from 100 universities in 35 countries, collected in 21 languages. We measured anxieties (statistics, mathematics, test, trait, social interaction, performance, creativity, intolerance of uncertainty, and fear of negative evaluation), self-efficacy, persistence, and the cognitive reflection test, and collected demographics, previous mathematics grades, self-reported and official statistics grades, and statistics module details. Data reuse potential is broad, including testing links between anxieties and statistics/mathematics education factors, and examining instruments’ psychometric properties across different languages and contexts.

Abstract

The HIV-protease inhibitor nelfinavir has shown broad anticancer activity in various preclinical and clinical contexts. In patients with advanced, proteasome inhibitor (PI)–refractory multiple myeloma, nelfinavir-based therapy resulted in 65% partial response or better, suggesting that this may be a highly active chemotherapeutic option in this setting. The broad anticancer mechanism of action of nelfinavir implies that it interferes with fundamental aspects of cancer cell biology. We combined proteome-wide affinity-purification of nelfinavir-interacting proteins with genome-wide CRISPR/Cas9–based screening to identify protein partners that interact with nelfinavir in an activity-dependent manner alongside candidate genetic contributors affecting nelfinavir cytotoxicity. Nelfinavir had multiple activity-specific binding partners embedded in lipid bilayers of mitochondria and the endoplasmic reticulum. Nelfinavir affected the fluidity and composition of lipid-rich membranes, disrupted mitochondrial respiration, blocked vesicular transport, and affected the function of membrane-embedded drug efflux transporter ABCB1, triggering the integrated stress response. Sensitivity to nelfinavir was dependent on ADIPOR2, which maintains membrane fluidity by promoting fatty acid desaturation and incorporation into phospholipids. Supplementation with fatty acids prevented the nelfinavir-induced effect on mitochondrial metabolism, drug-efflux transporters, and stress-response activation. Conversely, depletion of fatty acids/cholesterol pools by the FDA-approved drug ezetimibe showed a synergistic anticancer activity with nelfinavir in vitro. These results identify the modification of lipid-rich membranes by nelfinavir as a novel mechanism of action to achieve broad anticancer activity, which may be suitable for the treatment of PI–refractory multiple myeloma. Significance: Nelfinavir induces lipid bilayer stress in cellular organelles that disrupts mitochondrial respiration and transmembrane protein transport, resulting in broad anticancer activity via metabolic rewiring and activation of the unfolded protein response.

Abstract

A prominent theory assumes that the approximate number system (ANS), which is thought to be measured by non-symbolic magnitude comparison tasks, constitutes the basis for development of numerical skills. Recent studies showing influences of inhibition and visual strategies raise the question of what is measured with this task. Elderly people show age-related inhibitory decline, which can be intensified in Parkinson’s disease (PD) where inhibitory and visuo-spatial deficits occur. Therefore, PD-immanent inhibitory and visuo-spatial deficits in some patients allow for a quasi-experiment studying the influence of these cognitive domains on the performance in a typical ANS task. This enables inferring conclusions for the theory of number processing from this clinical population.

Abstract

The Spatial-Numerical Association of Response Codes (SNARC) effect – i.e., faster responses to small numbers with the left compared to the right side and to large numbers with the right compared to the left side – suggests that numbers are associated with space. However, it remains unclear whether the SNARC effect evolves from a number’s magnitude or the ordinal position of a number in working memory. One problem is that, in different paradigms, the task demands influence the role of ordinality and magnitude. While single-task setups in which participants judge the parity of a displayed number indicate the importance of magnitude for the SNARC effect, evidence for ordinal influences usually comes from experiments where ordinal sequences have to be memorized or setups in which participants possess pre-existing knowledge of the ordinality of stimuli. Therefore, in this preregistered study, we employed a SNARC task without secondary ordinal sequence memorization. We dissociate ordinal and magnitude accounts by carefully manipulating experimental stimulus sets. The results indicate that even though the magnitude model better accounts for the observed data, the ordinal position seems to matter as well. Hence, numbers are associated with space in both a magnitude- and an order-respective manner, yielding a mixture of both compatibility effects. Moreover, a multiple coding framework may most accurately explain the roots of the SNARC effect.
