Abstract

Huntington's disease (HD) is a neurodegenerative disorder that severely affects the basal ganglia and regions of the cerebral cortex. While astrocytosis and microgliosis both contribute to basal ganglia pathology, the contribution of gliosis and potential factors driving glial activity in the human HD cerebral cortex is less understood. Our study aims to identify nuanced indicators of gliosis in HD which is challenging to identify in the severely degenerated basal ganglia, by investigating the middle temporal gyrus (MTG), a cortical region previously documented to demonstrate milder neuronal loss. Immunohistochemistry was conducted on MTG paraffin-embedded tissue microarrays (TMAs) comprising 29 HD and 35 neurologically normal cases to compare the immunoreactivity patterns of key astrocytic proteins (glial fibrillary acidic protein, GFAP; inwardly rectifying potassium channel 4.1, Kir4.1; glutamate transporter-1, GLT-1; aquaporin-4, AQP4), key microglial proteins (ionised calcium-binding adapter molecule-1, IBA-1; human leukocyte antigen (HLA)-DR; transmembrane protein 119, TMEM119; purinergic receptor P2RY12, P2RY12), and indicators of proliferation (Ki-67; proliferative cell nuclear antigen, PCNA). Our findings demonstrate an upregulation of GFAP+ protein expression attributed to the presence of more GFAP+ expressing cells in HD, which correlated with greater cortical mutant huntingtin (mHTT) deposition. In contrast, Kir4.1, GLT-1, and AQP4 immunoreactivity levels were unchanged in HD. We also demonstrate an increased number of IBA-1+ and TMEM119+ microglia with somal enlargement. IBA-1+, TMEM119+, and P2RY12+ reactive microglia immunophenotypes were also identified in HD, evidenced by the presence of rod-shaped, hypertrophic, and dystrophic microglia. In HD cases, IBA-1+ cells contained either Ki-67 or PCNA, whereas GFAP+ astrocytes were devoid of proliferative nuclei. These findings suggest cortical microgliosis may be driven by proliferation in HD, supporting the hypothesis of microglial proliferation as a feature of HD pathophysiology. In contrast, astrocytes in HD demonstrate an altered GFAP expression profile that is associated with the degree of mHTT deposition.

Abstract

EUSO-Balloon is a pathfinder mission for the Extreme Universe Space Observatory onboard the Japanese Experiment Module (JEM-EUSO). It was launched on the moonless night of the 25th of August 2014 from Timmins, Canada. The flight ended successfully after maintaining the target altitude of 38 km for five hours. One part of the mission was a 2.5 hour underflight using a helicopter equipped with three UV light sources (LED, xenon flasher and laser) to perform an inflight calibration and examine the detectors capability to measure tracks moving at the speed of light. We describe the helicopter laser system and details of the underflight as well as how the laser tracks were recorded and found in the data. These are the first recorded laser tracks measured from a fluorescence detector looking down on the atmosphere. Abstract

The Telescope Array experiment studies ultra high energy cosmic rays using a hybrid detector. Fluorescence telescopes measure the longitudinal development of the extensive air shower generated when a primary cosmic ray particle interacts with the atmosphere. Meanwhile, scintillator detectors measure the lateral distribution of secondary shower particles that hit the ground. The Middle Drum (MD) fluorescence telescope station consists of 14 telescopes from the High Resolution Fly’s Eye (HiRes) experiment, providing a direct link back to the HiRes measurements. Using the scintillator detector data in conjunction with the telescope data improves the geometrical reconstruction of the showers significantly, and hence, provides a more accurate reconstruction of the energy of the primary particle. The Middle Drum hybrid spectrum is presented and compared to that measured by the Middle Drum station in monocular mode. Further, the hybrid data establishes a link between the Middle Drum data and the surface array. A comparison between the Middle Drum hybrid energy spectrum and scintillator Surface Detector (SD) spectrum is also shown.

Abstract

In this study, we suggested the wave power generator using horizontal motions of the wave for use in the coastal sea. The length of the horizontal movement of the wave in the vicinity of the sea surface is larger than the length of the vertical reciprocating movement of the wave, hence the proposed device has a wave power transmission plate. In addition, because the motion of the wave is maximum to the sea surface, by arranging the buoyancy tanks at the top of the wave power transmission plate, it is always capable of vertical movement in accordance with the sea surface. To confirm the usefulness of the proposed wave power generator, we constructed a mathematical model of the wave power generator and carried out simulation using bondgraph. Furthermore, the efficiency was verified by measuring the degree of electrical energy production through a preliminary experiment.

Abstract

Experiment 1 was a feeding trial in which 75 (Landrace ⅹ Yorkshire) ⅹ Duroc pigs with average initial bodyweight (BW) of 26 ± 1 kg were used. It was assigned to three pigs/pen and five pens/treatment. Experiment 2 was a metabolic trial in which 25 (LandraceⅹYorkshire)ⅹDuroc pigs with average initial bodyweight (BW) of 36 ± 1 kg were used. The basal diet consisted of maize (57.71 %), soybean meal (32.45 %), and wheat bran (5 %). Treatments consisted of i) CON (control diet); ii) OE (organic acid + essential oils) (Aviplus-S ® ), that is, CON + 0.05 % OE; iii) OC (organic charcoal) (Olga Black ® ), that is, CON + 0.1 % OC; iv) AE (anise extracts) (RESQ ® ), that is, CON + 0.015 % AE; and v) PB (probiotics) (Bonvital ® ), that is, CON + 0.1 % PB. Average daily gain (ADG) and gain/feed ratio (G : F) were significantly higher in the OE and AE groups than in the others. Average daily feed intake (ADFI) for OE, OC, and AE treatments was significantly higher than that for CON and PB treatments. Dry matter (DM) digestibility was significantly higher in AE than CON. Crude protein (CP) was significantly higher in OE, AE, and PB than CON. CON and other treatments did not cause significant differences in blood l-density lipoprotein (LDL), and glucose. NH 3 emissions were significantly lower in all treated groups than in CON. Among the treatments, NH 3 emissions were lowest in OE and AE. H 2 S emissions were significantly lower in OE, AE, and PB than in CON. Among the treatments, OE and AE were most effective at reducing H 2 S emissions. The emissions of volatile fatty acids (VFAs) showed no significant difference between treatments and CON. In conclusion, this experiment was shown to be more effective on growth performance, malodour emission, blood profiles, and nutrient digestibility in OE and AE than other additives. Abstract

Background Premenopausal women with MBC have a distinct tumor biology, and more aggressive behavior. In the BOLERO-2 trial, adding EVE to exemestane (EXE) significantly prolonged progression-free survival (PFS) compared with EXE alone in postmenopausal women with recurrent/metastatic HR+, HER2– breast cancer. Methods Patients (pts) with progression or prior exposed to tamoxifen with or without GnRH agonist, either sequentially or concurrently, in adjuvant or metastatic setting were randomly assigned (2:1) to receive LEUP+LET+EVE (arm A) or LEUP+LET (arm B) until disease progression or unacceptable toxicity. Randomization was stratified by the presence of visceral metastases and sensitivity to endocrine therapy. The primary end point was PFS. Secondary end points included overall survival (OS), objective response rate (ORR), and safety. Results Between January 2014 and October 2018, 137 pts were enrolled in this study. Baseline characteristics were well balanced between the two groups. The median age was 44 years (range 24-54), 74.5% had endocrine-sensitive disease, and 60.6% had visceral metastasis. At a median follow-up of 32.4 months, the median PFS was 18.1 months in arm A and 13.8 months in arm B, respectively (HR 0.73; 95% CI, 0.48 to 1.11; P = 0.137), and predominant in pts with visceral metastases (16.4 vs. 9.5 Mo) (HR 0.58; 95% CI, 0.34 to 0.99; p = 0.048). The median OS was not reached in both arms. ORR was 46.9% and 28.2% in arms A and B, respectively (P = 0.051). The most common grade 3/4 adverse events were neutropenia (9.8% in arm A vs. 0% in arm B), AST elevation (5.4% vs. 0%), enterocolitis (4.3% vs. 0%), and anemia (3.3% vs. 0%), without grade 3/4 stomatitis in both arms. Five pts discontinued treatment (1 AKI, 1 pneumonitis, 1 liver dysfunction, 2 unknown) but no treatment related death. Conclusions Adding EVE to LET demonstrated numerically improved PFS (D 4.3 Mo, median PFS), higher ORR. Significant improved PFS was noted in patients with visceral metastases. Abstract

Inclusive photoproduction of D*+- in ep collisions at HERA has been measured with the ZEUS detector for photon-proton centre of mass energies in the range 115 < W < 280 GeV and photon virtuality Q**2 < 4 GeV**2. The cross section sigma(ep --> D*X) integrated over the kinematic region pt(D*) > 3 GeV and -1.5 < eta(D*) < 1.0 is (10.6+-1.7(stat.)+1.6(syst.)-1.3(syst.)) nb. Differential cross sections as functions of pt(D*), eta(D*) and W are given. The data are compared with two next-to-leading order perturbative QCD predictions. For a calculation using a massive charm scheme the predicted cross sections are smaller than the measured ones. A recent calculation using a massless charm scheme is in agreement with the data., 18 pages including 4 figures

Abstract

The gluon momentum density xg ( x , Q 2 ) of the proton was extracted at Q 2 = 20 GeV 2 for small values of x between 4 × 10 −4 and 10 −2 from the scaling violations of the proton structure function F 2 measured recently by ZEUS in deep inelastic neutral current ep scattering at HERA. The extraction was performed in two ways. Firstly, using a global NLO fit to the ZEUS data on F 2 at low x constrained by measurementsfrom NMC at larger x ; and secondly using published approximate methods for the solution of the GLAP QCD evolution equations. Consistent results are obtained. A substantial increase of the gluon density is found at small x in comparison with the NMC result obtained at larger values of x .

Abstract

Summary: Microglia are the primary innate immune effectors of the central nervous system. Although numerous protocols have been developed to isolate fetal mouse microglia, the isolation of adult mouse microglia has proven more difficult. Here, we present a simple, widely accessible protocol to isolate pure microglia cultures from 4- to 14-month-old mouse brains using their adherent properties in vitro.
