1. Mitigation of 3-MCPDE and GE in palm oil in Indonesia

Abstract

Among consumed-vegetable oils, RBDPO (refined, bleached and deodorized palm oil) is indicated containing higher level of 3-MCPDE and GE, which have been classified as potentially carcinogenic and/or genotoxic to human. Those contaminants in RBDPO are formed during deodorization process with high temperature and also correlate with chloride content and quality of CPO as raw material. The maximum limit for 3-MCPDE and GE content in vegetable oil has been set at 2500 and 1000 µg kg-1, and as low as 750 and 500 µg kg-1 when used for production of baby food and cereal-based foods for infants and young children. The mitigation of 3-MCPDE and GE in RBDPO is quite challenging for palm oil refineries in producing a good quality of palm oil in term of sensory, stability, safety, and nutritional value, especially in Indonesia. It has been proven that modification of conventional refining process can reduce the 3-MCPDE and GE content. They are including pre-treatment of CPO by water washing to reduce the chloride content, alkali neutralization to remove any acids present in the oil prior to deodorization, using neutral bleaching earth, adding antioxidants after bleaching to reduce free radical-mediated formation of 3-MCPDE and GE, lowering the deodorization temperature with a longer time, and even double bleaching and/or double deodorization of the palm oil. Post-refining treatment using specific adsorbents is also applied to remove the 3-MCPDE and GE. However, palm oil refineries have to apply combination of several mitigation strategies to adequately reduce the levels of 3-MCPDE and GE, including using good quality of CPO with low FFA, chloride, and oxidized components content.

Abstract

Crude Palm Oil (CPO) is palm oil obtained from the extraction or compression process of oil palm fruit flesh and has not been purified. Palm oil is usually used for food, cosmetic industry, chemical industry, and animal feed industry. Increasingly open insight and increasing consumer awareness of the importance of food safety and quality assurance supported by technological developments and laboratory analysis methods, new types of contaminants in food products are also found. Currently, the Indonesian government is facing challenges regarding the issue of 3-monochloro-1,2-propanediol ester (3-MCPD Ester) and glycidyl ester (GE) in palm oil which can hamper Indonesia's palm oil trade in the future. The practice in the field affects the quality of CPO and the retrieval of low-quality CPO is mixed with high-quality CPO. And the length of time the process of pumping CPO from reclaimed tanks to VCT affects the quality of CPO. So this study aims to modify the pumping time and storage time of CPO from Reclaimed tanks to VCT by mixing samples from VCT (fresh CPO) and samples from Reclaimed tanks (lower quality CPO).

8. Development of resistance of mutans streptococci and Porphyromonas gingivalis to chlorhexidine digluconate and amine fluoride/stannous fluoride-containing mouthrinses, in vitro

Abstract

Objective: The aim if this study was to determine the minimal inhibitory concentrations of chlorhexidine digluconate and an amine fluoride/stannous fluoride-containing mouthrinse against Porphyromonas gingivalis and mutans streptococci during an experimental long-term subinhibitory exposition. Material and methods: Five P. gingivalis strains and four mutans streptococci were subcultivated for 20-30 passages in subinhibitory concentrations of chlorhexidine digluconate or an amine fluoride/stannous fluoride-containing mouthrinse. Results: Pre-passaging minimal inhibitory concentrations for chlorhexidine ranged from 0.5 to 2mg/l for mutans streptococci and from 2 to 4mg/l for the P. gingivalis isolates. For the amine fluoride/stannous fluoride-containing mouthrinse minimal inhibitory values from 0.125 to 0.25% for the mutans streptococci and from 0.063 to 0.125% for the P. gingivalis isolates were determined. Two- to fourfold increased minimal inhibitory concentrations against chlorhexidine were detected for two of the five P. gingivalis isolates, whereas no increase in minimal inhibitory concentrations was found for the mutans streptococci after repeated passaging through subinhibitory concentrations. Repeated exposure to subinhibitory concentrations of the amine fluoride/stannous fluoride-containing mouthrinse did not alter the minimally inhibitory concentrations of the bacterial isolates tested. Conclusion: Chlorhexidine and the amine fluoride/stannous fluoride-containing mouthrinse are effective inhibitory agents against the oral bacterial isolates tested. No general development of resistance against chlorhexidine or the amine fluoride/stannous fluoride-containing mouthrinse was detected. However, some strains showed potential to develop resistance against chlorhexidine after prolonged exposure. Clinical relevance: The use of chlorhexidine should be limited to short periods of time. Abstract

OBJECTIVE To assess the relationship between electrical stimulation administered to patients undergoing bilateral electroconvulsive therapy (ECT) and subsequent measures of cognitive function and depression severity. METHODS Stimulus dose titrated patients receiving bilateral ECT were assessed with the Cambridge Neuropsychological Test Automated Battery (CANTAB) Spatial Recognition Memory test and Montgomery Asberg Depression Rating Scale (MADRS) at baseline, after 4 ECT treatments and on course completion. Changes in CANTAB and MADRS scores were assessed in relation to electrical dosage, initial stimulus dose, and demographic variables using linear mixed models. RESULTS Data pertained to 143 patients (mean age, 56.85 [SD, 14.94], 43% male). Median change in CANTAB score was -10% (-20% to 5%) after 4 ECT treatments and -10% (-20% to 5%) at course completion. Median change in MADRS score was -22 (-33 to -13) after 4 ECT treatments and -14 (-25 to -7) at course completion. Electrical dosage had no effect on CANTAB or MADRS change scores either after 4 treatments or course completion. Improvement in CANTAB score at end of course was associated with female sex (P < 0.05), higher intelligence quotient (P = 0.01), and age. After 4 treatments, improvement in CANTAB score was associated with younger age (P < 0.001) and higher intelligence quotient (P < 0.01). Improved MADRS score at course completion was associated with older age (P < 0.001 at end of course and after 4 treatments). CONCLUSIONS Electroconvulsive therapy has significant antidepressant and cognitive effects which are not associated with the total electrical dose administered. Other, unalterable variables, such as age and sex, have an influence on these effects.
