Gastric cancer (GC) is the third most lethal and fifth most common cancer in the world. In a variety of cancers, the hexokinase domain component 1 (HKDC1) is carcinogenic. This study was to investigate into how HKDC1 contributes to the development and progression of GC. Three different datasets (GSE103236, GSE13861, and GSE55696) were extracted from the Gene Expression Omnibus (GEO) database and then analyzed using the sva package. The R software was used to identify 411 differentially expressed genes (DEGs) in the pooled dataset. We discovered 326 glycolysis-related genes (glyGenes) in the cancer genome atlas-stomach adenocarcinoma (TCGA-STAD) cohort using gene set enrichment analysis set (GSEA). HKDC1 is one of the most prevalent glyGenes in GC tumor tissues and cells, as seen in the Venn diagram. According to the results of the Cell Count Kit-8 assay, the proliferation of AGS and MKN-45 cells decreased when HKDC1 was knocked down. Lack of HKDC1 in cells enhanced oxygen consumption and decreased glycolytic protein expression while suppressing glucose absorption, lactate production, ATP level, and extracellular acidification ratio. As an oncogene in gastric cancer development, HKDC1 influences cell proliferation and glycolysis.

A yield gap analysis for rapeseed (Brassica napus L.) is critical to meeting the oil demand by identifying yield potential and yield constraints. In this study, potential yield (Yp), attainable yield (Yatt), and actual yield (Yact) for winter rapeseed were determined in five different zones of China. A boundary line approach was adopted to calculate Yp, based on a large-scale field experimental database. A meta-analysis was conducted on the data obtained from 118 published studies to evaluate the effects of agronomic factors on rapeseed yield. The main results indicated that farmers only achieved 37–56% of the yield potential across the zones. The low altitude areas (L-URY) and lower reaches (LRY) of the Yangtze River Basin (YRB), China had high yield levels. The total yield gap was 1893 kg ha–1, due to the agronomic management factors, environmental factors, and socioeconomic factors. The meta-analysis showed that weed control and drainage were the best management practices to improve yields (45.6 and 35.3%, respectively), and other practices improved yields by 17.1–21.6%. Consequently, to narrow the yield gap over the short term, the study could focus on techniques that are easily implemented to farmers.

Background Preoperative prediction of microvascular invasion (MVI) is critical for treatment strategy making in patients with hepatocellular carcinoma (HCC). We aimed to develop a deep learning (DL) model based on preoperative dynamic contrast-enhanced magnetic resonance imaging (DCE-MRI) to predict the MVI status and clinical outcomes in patients with HCC. Methods We retrospectively included a total of 321 HCC patients with pathologically confirmed MVI status. Preoperative DCE-MRI of these patients were collected, annotated, and further analyzed by DL in this study. A predictive model for MVI integrating DL-predicted MVI status (DL-MVI) and clinical parameters was constructed with multivariate logistic regression. Results Of 321 HCC patients, 136 patients were pathologically MVI absent and 185 patients were MVI present. Recurrence-free survival (RFS) and overall survival (OS) were significantly different between the DL-predicted MVI-absent and MVI-present. Among all clinical variables, only DL-predicted MVI status and a-fetoprotein (AFP) were independently associated with MVI: DL-MVI (odds ratio [OR] = 35.738; 95% confidence interval [CI] 14.027–91.056; p < 0.001), AFP (OR = 4.634, 95% CI 2.576–8.336; p < 0.001). To predict the presence of MVI, DL-MVI combined with AFP achieved an area under the curve (AUC) of 0.824. Conclusions Our predictive model combining DL-MVI and AFP achieved good performance for predicting MVI and clinical outcomes in patients with HCC.

Since the vehicle is a complex mechanical system with many subsystems, the influence of the dynamic coupling between the subsystems of the vehicle on shimmy should be taken seriously. Therefore, a 12 degrees-of-freedom dynamic model of vehicle shimmy system with consideration of the dynamic coupling between the vertical motion and the lateral motion of the vehicle is established. In particular, the influence of the vertical load of the tire on the nonlinear cornering force is also considered. Then, the dynamic stability of the shimmy system is discussed with the help of the system eigenvalues, and the influence of the damping of the front suspension on the dynamic stability is also examined from the viewpoint of the energy transfer. On this basis, the damping of the front suspension is selected as a bifurcation parameter, and the two-dimensional center manifold of the high dimensional shimmy system is obtained by means of the center manifold theory. Finally, the Hopf bifurcation characteristic of the shimmy system is analyzed, and the analytical solutions of the shimmy system are derived for different damping of the front suspension. The results show that the increase of the damping of the front suspension is beneficial to attenuate vehicle shimmy and improve the dynamic stability of the vehicle.

To control the meshing performance of hypoid gear drive, current local conjugate theory and local synthesis method establish a relationship between the relative motion and local geometrical properties of the mating surfaces at one reference point. Theoretically, both local conjugate theory and local synthesis method cannot ensure the contact performance on the entire tooth surface resulting in uncontrolled contact pattern and undesirable transmission error. A global synthesis approach consisting of a machine-tool settings calculation model, a tooth contact analysis model, a meshing quality assessment function, and a swarm intelligence algorithm are proposed. The direction, area, and shape of the contact pattern and the amplitude of the intersection of two adjacent transmission error curves are taken as the evaluation indexes. Three curvatures of the design pitch cone of the pinion are taken as the control variables. A global selection space is then established within the reasonable range of the curvatures. An improved multiple population genetic algorithm is employed to find the optimal set of the curvatures to achieve the target values of the evaluation indexes. To avoid the edge contact and corner contact condition, a feasible region is defined on tooth surface and the contact patterns on the gear and pinion surfaces are all confined within the area. The optimized contact patterns obtained by the loaded tooth contact analysis method are similar to those obtained by the proposed approach, thus demonstrating the effectiveness of this methodology. The main novelty of this approach is to translate the derivation of the mathematical relation between the curvatures of the mating surfaces and the contact properties to solving a multi-objective optimization problem of the meshing quality indexes by the intelligence algorithm.

Poly ADP‐ribose polymerase inhibitors (PARPi) have shown promising therapeutic efficacy in triple‐negative breast cancer (TNBC) patients. However, resistance ultimately develops, preventing a curative effect from being attained. Extensive investigations have indicated the diversity in the mechanisms underlying the PARPi sensitivity of breast cancer. In this study, we found that DNA damage binding protein 2 (DDB2), a DNA damage‐recognition factor, could protect TNBC cells from PARPi by regulating DNA double‐strand break repair through the homologous recombination pathway, whereas the depletion of DDB2 sensitizes TNBC cells to PARPi. Furthermore, we found that DDB2 was able to stabilize Rad51 by physical association and disrupting its ubiquitination pathway‐induced proteasomal degradation. These findings highlight an essential role of DDB2 in modulating homologous recombination pathway activity and suggest a promising therapeutic target for TNBC.

Accurate evaluation of the misalignment sensitivity of hypoid gears is a significant foundation for analysis of its dynamics and for the calculation of machining parameters. A tooth contact analysis (TCA) methodology considering four kinds of misalignments is presented to calculate the contact pattern and transmission error. A sensitivity model of contact pattern to misalignments is established to investigate the effects of different alignment errors on meshing performance. By parameterizing the contact pattern, the influences of offset error, angular error, and the axial error of pinion and gear on the direction, shape, and position features of contact pattern are studied. Coefficients of four evaluation indexes to different misalignments are defined respectively, and the minimum sum of the weighted coefficients is utilized to establish a multi-objective comprehensive sensitivity model. Three curvatures of the pitch cone of the pinion are taken as the control variables, and a global selection space is then built within the reasonable range of those curvatures. An improved multi-population genetic algorithm (MPGA) is used to find the optimal set of curvatures to achieve the minimum synthetic sensitivity. TCA results indicate that the offset error and angular error have the greatest influence on the contact pattern. By adopting this methodology appropriately, the sensitivity of the contact pattern to misalignments can be reduced. The contributions of this paper can be summarized as: (1) an accurate parameterized measurement model of the contact pattern; (2) a comprehensive sensitivity model of the contact pattern to misalignments; (3) an optimization framework consisting of a calculation model of the machining parameters, a TCA model considering misalignments, and a misalignment sensitivity evaluation model.

To investigate specific technique and clinical effects of closed folding top consolidation maneuver combined with splint fixation maneuver for consolidation and cedar bark external fixation splint for the treatment of double fractures of distal ulna and radius in children.From January 2017 to December 2019, 17 children with double fractures of distal ulna and radius were treated with closed folded apex consolidation maneuver, including 13 males and 4 females, aged from 4 to 11 years old with an average of (7.29±2.34) years old. The fractures were fixed with cedar bark splint and followed up for 6 months, and alignment of fracture was evaluated according to the latest X-rays by follow up, and function of the affected limbs was evaluated by Anderson forearm function evaluation criteria.Fifteen of 17 children were successfully reset immediately, and 2 children were successfully reset again. The average fixed time was (25.00±3.35) days. At 6 months of follow up, 12 patients got excellent results, 3 good, 2 fair, and 0 poor according to Anderson forearm function evaluation criteria. The position of all children were larger than 3/4, and 10 children were received anatomical reduction, alignment of 4 children was less than 10°, 3 children was less than 15°. No complications such as fracture displacement, nonunion, compartment syndrome, and forearm rotation dysfunction occurred.Restoration of distal radius double fracture in children with the combination of the closed folding and top fixation maneuver and splint fixation maneuver has advantages of higher success rate, lower complications, which could reduce operating difficultyand pain of patients.

Objective: Chemoresistance has been a major problem in cancer chemotherapy. The present study aimed to investigate the effect of Rosmarinic acid (RA) on chemoresistance to 5-Fu and its molecular mechanism in gastric carcinoma. Methods: CCK8 cell proliferation and apoptosis assay were used to evaluate the effect of RA on chemoresistance to 5-Fu in GC cells. RNA microarray was used to identify miRNA involved. Expression level of miRNA in GC cells was determined by RT-PCR. Down- or up-regulating of miRNA in the GC cells was performed by transfection of RNA interference or expression vectors in the GC cells. Double luciferase reporter assay was used to verify miRNA target genes. Expression of P-glycoprotein and Bax was analyzed with Western blot. Results: RA treated SGC7901/5-Fu cells showed significant increased chemosensitivity to 5-Fu. Background PRKAA1 encodes α-subunit of 5-AMP-activated protein kinase (AMPK), which has been implicated in the pathogenesis of carcinoma of the stomach. Previous works have suggested that polymorphisms in the PRKAA1 may be associated with the risk of non-cardiac gastric cancer (NCGC), but whether PRKAA1 polymorphisms are related to clinical pathologic characteristics of gastric cancer and its clinical outcome is largely unknown. Methods We carried out a case-control study including a total of 481 gastric cancer patients and 490 healthy controls. The genotypes of enrolled polymorphisms were identified with Sequenom MassARRAY platform. Results This study showed that rs10074991 GG genotype (adjusted OR = 1.44, 95%CI:0.99–2.09, p = 0.056) has a borderline significantly increased risk for gastric cancer, which was consistent with the result of additive model (adjusted OR = 1.21, 95%CI:1.01–1.46, p = 0.042). In similar, an increased risk of gastric cancer was also observed for rs13361707 TC genotype (adjusted OR = 1.47, 95%CI: 1.01–2.14, p = 0.043; additive model: adjusted OR = 1.22, 95%CI: 1.02–1.47, p = 0.033). Furthermore, the rs154268 and rs461404 were also found associated with increased gastric cancer risk, which may be influenced by age, tumor type and differentiation, and tumor stage. Haplotype analysis indicated A-G-C-T-C-G haplotype (rs6882903, rs10074991, rs13361707, rs3805490, rs154268 and rs461404) is associated with increased risk of gastric cancer (OR = 1.29, 95%CI: 1.02–1.62, p = 0.035). The univariate analysis for overall survival (OS) revealed that both of rs10074991 and rs13361707 variants are associated with poor OS in patients with NCGC. Conclusion This case-control study provided the evidence thatrs13361707CC, rs10074991GG, rs461404GG, and rs154268CC are associated with increased gastric cancer risk, especially for NCGC, and that patients with rs10074991 G or rs13361707 C allele have a poor OS.

MicroRNA (miRNA) is an important factor in the regulation of gene transcription.

In eukaryotic cells, autophagy is a process associated with programmed cell death. During this process, cytoplasmic proteins and organelles are engulfed by double‐membrane autophagosomes, which then fuse with lysosomes to form autolysosomes. These autolysosomes then degrade their contents to recycle the cellular components. Autophagy has been implicated in a wide variety of physiological and pathological processes that are closely related to tumorigenesis. In recent years, an increasing number of studies have indicated that nonsteroidal anti‐inflammatory drugs, such as celecoxib, meloxicam, sulindac, aspirin, sildenafil, rofecoxib, and sodium salicylate, have diverse effects in cancer that are mediated by the autophagy pathway. These nonsteroidal anti‐inflammatory drugs can modulate tumor autophagy through the PI3K/Akt/mTOR, MAPK/ERK1/2, P53/DRAM, AMPK/mTOR, Bip/GRP78, CHOP/ GADD153, and HGF/MET signaling pathways and inhibit lysosome function, leading to p53‐dependent G1 cell‐cycle arrest. In this review, we summarize the research progress in autophagy induced by nonsteroidal anti‐inflammatory drugs and the molecular mechanisms of autophagy in cancer cells to provide a reference for the potential benefits of nonsteroidal anti‐inflammatory drugs in cancer chemotherapy.

MicroRNAs (miRNAs) are a class of short highly-conserved non-coding endogenous RNA molecules of 18-25 nucleotides in length. By incompletely or completely complementary to 3'-untranslated region (3'-UTR) of mRNAs, miRNAs can suppress the transcription of target genes, and degrade mRNA. This article mainly reviewed the relevant research progress of miRNAs in gastric cancer. Abnormal miRNAs regulated by epigenetics participate in the proliferation, apoptosis, metastases and invasion of gastric cancer cells by regulating different target genes. In the meantime, NF-kB and AKT signal pathways highly associated with gastric cancer progression might be activated or inhibited by miRNAs. MiRNAs involving in change of drug resistance of tumor cells can provide new targets for the treatment of gastric cancer. Abnormal expression of miRNAs in the blood provides new biological markers for noninvasive diagnosis of gastric cancer. Moreover, miRNAs also participate in carcinogenic process of gastric cancer caused by helicobacter pylori (HP) infection. In conclusion, miRNAs play vital important roles in the occurrence, progression, diagnosis, treatment and prognosis of gastric cancer, which provides new insight into gene therapy for gastric cancer.

Fertilization is essential for oilseed rape because it is sensitivity to nutrient deficiency, especially for winter oilseed rape (Brassica napus L.). To investigate regional nutrient efficiency and nutrient uptake-yield relationship of winter oilseed rape in an intensive cropping system, this study used data from 619 site-year on-farm experiments carried out in the winter oilseed rape planting area of the Yangtze River Basin, China from 2005 to 2010, with large yield in the range of 179–4 470 kg ha−1. Currently recommended application rates of N, P and K fertilizers increased rapeseed yield at different levels of soil indigenous nutrient supply (INS) in this region. Boundary values of plant nutrient uptake were established to analyze the nutrient uptake-yield relationship of winter oilseed rape (internal nutrient efficiency), i.e., 128 kg N ha−1, 24 kg P ha−1, and 122 kg K ha−1. The internal nutrient efficiency declined by 48.2%–64.1% when nutrient uptake exceeded the boundary value, resulting in excessive nutrient uptake (i.e., low yield response with high nutrient uptake), especially for K. In the intensive cropping system, agronomic efficiencies of N, P, and K were 5.9, 3.4, and 3.6 kg kg−1, and recovery efficiencies of N, P, and K were 35.6%, 24.1%, and 36.8%, respectively. These findings showed that the fertilization rate should be optimized by considering INS, nutrient status, and nutrient efficiency of winter oilseed rape. In this study, considering the lower yield improvement to high K uptake levels and low K fertilizer efficiency, application rate of K fertilizer should be reduced since soil K deficiency has already been mitigated.

In the current study, we tested expression and potential function of Ca 2+ /calmodulin-dependent protein kinase phosphatase (Ppm1E), an AMPKα phosphatase, in human gastric cancers. Ppm1E expression was elevated in human gastric cancer tissues ( vs. normal tissues), which was correlated with AMPK (p-AMPKα, Thr-172) dephosphorylation and mTOR complex 1 (mTORC1) activation. Ppm1E upregulation, AMPK inhibition and mTORC1 activation were also observed in human gastric cancer cell lines (AGS, HGC-27, and SNU601). Intriguingly, Ppm1E knockdown by shRNA induced AMPK activation, mTORC1 inactivation, and proliferation inhibition in AGS cells. On the other hand, forced over-expression of Ppm1E induced further AMPK inhibition and mTORC1 activation to enhance AGS cell proliferation. Remarkably, microRNA-135b-5p (“miR-135b-5p”), an anti-Ppm1E microRNA, was downregulated in both human gastric cancer tissues and cells. Reversely, miR-135b-5p exogenous expression caused Ppm1E depletion, AMPK activation, and AGC cell proliferation inhibition. Together, Ppm1E upregulation in human gastric cancer is important for cell proliferation, possible via regulating AMPK-mTOR signaling.

Purpose The purpose of this paper is to propose a novel strategy of optimal parameters configuration and placement for sensitive equipment. Design/methodology/approach In this study, clamped thin plate is considered as the foundation form, and a novel composite system is proposed based on the two-stage isolation system. By means of the theory of mechanical four-pole connection, the displacement amplitude transmissibility from the thin plate to precision equipment is derived. For the purpose of performing optimal design of the composite system, a novel multi-objective idea is presented. Multi-objective particle swarm optimization (MOPSO) algorithm is adopted as an optimization technique, which can achieve a global optimal solution (gbest), and selecting the desired solution from an equivalent Pareto set can be avoided. Maximum and variance of the four transmitted peak displacements are considered as the fitness functions simultaneously; the purpose is aimed at reducing the amplitude of the multi-peak isolation system, meanwhile pursuing a uniform vibration as far as possible. The optimization is mainly organized as a combination of parameter configuration and placement design, and the traversal search of discrete plate is performed in each iteration for the purpose of achieving the global optimum. Findings An important transmissibility based on the mechanical four-pole connection is derived, and a composite vibration isolation system is proposed, and a novel optimization problem is also defined here. This study reports a novel optimization strategy combined with artificial intelligence for parameters and placement design of precision equipment, which can promote the traditional view of two-stage vibration isolation. Originality/value Two-stage vibration isolation systems are widely applied to the vibration attenuation of precision equipment, but in these traditional designs, vibration participation of foundation is often ignored. In this paper, participation of foundation of equipment is considered, and a coherent new strategy for equipment isolation and foundation vibration is presented. This study shows a new vision of interdisciplinary including civil engineering, mechanical dynamics and computational science.

In order to explore temporal-spatial variability of farmland soil pH at Enshi Antonomous Prefecture, Hubei, China, soil pH during the past three decades was analyzed, using the datasets of the Second National Soil Survey (1980-1983) and the Cultivated Land Quality Evaluation (2010-2013). The natural and human factors inducing the change of soil pH were evaluated to provide theoretical guidance for further soil acidification management. Results showed that acidic soil (i.e., pH＜6.5) and neutral and alkaline soil (i.e., pH 6.5-8.5) were accounted for 98.4% and 1.6% in the farmland during the period of 2010-2013, respectively. The ratio increased 61.4% for the acidic soil but decreased 61.2% for the neutral and alkaline soil as compared with the period of 1980-1983. In addition, there was no alkaline soil (pH8.5) in the region in 2010-2013. According to the dataset of the Second National Soil Survey (1980-1983), acidic soil was mainly distributed at Laifeng, Lichuan, Xuanen and Xianfeng counties, with the area ratio of 74.4%, 63.5%, 61.3% and 60.7%, respectively. For the period of 2010-2013, the ratio of acidic soil enhanced widely which was above 96% for each county. At Enshi Autonomous Prefecture, farmland soil showed an obvious acidification trend during the past three decades, with spatial variation of higher in the eastern part and lower in the western part of the region. Furthermore, soil pH decline occurred among different land use types in different areas. Overall, farmland soil pH declined 0.90 on average, with 1.14 decrease for upland and 0.87 for paddy soil, respectively. Clearly, upland soil acidification was severe than paddy soil. Factors related to soil acidification in the Enshi Autonomous Prefecture were mainly human factors such as unreasonable fertilizer combination, fertilizer ratio change, and more base cations taking away by high crop yield.以第二次土壤普查(1980—1983年)和耕地质量评价(2010—2013年)两个不同时期恩施州土壤pH数据为基础,对恩施州耕地土壤pH近30年时空变化进行了研究,明确了恩施州耕地土壤pH值变化特征,并初步探讨了导致土壤pH变化的自然因素和人为因素,以期为制定土壤酸化控制对策提供理论指导.结果表明: 2010—2013年,恩施州pH＜6.5的酸性土壤占耕地面积的98.4%,6.5～8.5中性偏碱性土壤仅占耕地面积的1.6%;与1980—1983年相比,酸性土壤(pH＜6.5)面积比例提高了61.4%,中性偏碱性土壤(pH在6.5～8.5)面积比例减少了61.2%,pH＞8.5的强碱性耕地土壤在全州已基本消失.1980—1983年,pH＜6.5的耕地土壤主要分布在来凤县、利川市、宣恩县和咸丰县,面积比例分别为74.4%、63.5%、61.3%和60.7%,至2010—2013年,恩施州酸性土壤分布更加广泛,各县市酸性土壤(pH＜6.5)面积比例均达到96%以上.近30年来,恩施州土壤呈酸化趋势,土壤pH空间分布呈现东高西低格局.不同区域、不同耕地利用类型土壤pH值均发生不同程度的下降.耕地土壤pH整体下降了0.90,其中,旱地土壤pH下降了1.14,水田土壤pH下降了0.87,旱地土壤酸化程度大于水田.恩施州土壤酸化主要受肥料结构、配比变化及高产作物收获带走大量盐基养分等人为因素影响.

Present study examined the influence of different types of slow/controlled release urea on rice yield and annual greenhouse gas emissions in a paddy field, and assessed the greenhouse gas intensity (GHGI, equivalent to global warming potential GWP/rice yield). Plane-wave pseudo-potential methods based on density functional theory are used to study the electronic structures, magnetic properties, and half-metallicity of new predicted Ti2-based full Heusler alloys Ti2RhZ (Z = Al, Ga, In). The CuHg2Ti-type structure is observed to be more favorable than the AlCu2Mn-type structure for these alloys. They are half-metallic ferrimagnets with total magnetic moment of 2.00 μ B per formula unit and follow the Slater-Pauling rule. Also, results show that these Ti2RhZ (Z = Al, Ga, In) alloys have an indirect band gap with values of 0.577, 0.696, and 0.616 eV, respectively, in the minority spin channel. Two types of structural changes are used to test the sensitivity of half-metallicity. Moreover, the whole alloys investigated in the current work have a negative formation energy, which means they are thermodynamically stable.

The d 0 half-metallic properties of a new and promising compound, CsMgS 2, are investigated by means of first-principle calculations within the framework of density functional theory. We find that CsMgS 2 is a d 0 half metal with a Cu 2MnAl-type Heusler structure in the ferromagnetic ground state. The half-metallicity is robust against isotropic stress and is suitable to be fabricated in the form of thin films.

Use of the conventional cancer chemotherapy (i.e. vincristine) is limited in tumor cells exhibiting pre-existing or acquired resistance. Here, we found that C6 ceramide (C6) dramatically sensitized vincristine's activity. In vitro, C6 and vincristine coadministration induced substantial necrosis and apoptosis in multiple human cancer cell lines, which were accompanied by a profound AMP-activated protein kinase (AMPK) activation, subsequent p53 activation, mTORC1 inactivation and Bcl-2/HIF-1α downregulation. Such synergistic effects were attenuated by AMPK inactivation through genetic mutation or short hairpin RNA silencing. Coadministration-activated p53 translocated to mitochondria, and formed a complex with cyclophilin-D, leading to mitochondrial permeability transition pore opening and cell necrosis. Disrupting p53-Cyp-D complexation through pharmacological or genetic means reduced costimulation-induced cytotoxicity. In vivo, a liposomal C6 was synthesized, which dramatically enhanced the antiproliferative activity of vincristine on HCT-116 or A2780 xenografts. Together, C6 sensitizes vincristine-induced anticancer activity in vivo and in vitro, involving activating AMPK-p53 signaling.

In the present paper, a database composed of 1035 on-farm observations collected from 2005 to 2010 across the major winter oilseed rape production regions in China was used to evaluate internal nutrient efficiencies (kg seed per kg nutrient in plant dry matter); then the Quantitative Evaluation of the Fertility of Tropical Soils (QUEFTS) model for winter oilseed rape was developed to describe the nutrient uptake-yield relationship of oilseed rape over a wide range of environmental conditions and predict the nutrient requirements for a target yield. Direct-sowing establishment method has great significance in improving winter oilseed rape (Brassica napus L.) production and guaranteeing edible oil security in China. However, nutrient responses on direct sown winter oilseed rape (DOR) performance and population development dynamic are still not well understood. Therefore, five on-farm experiments were conducted in the reaches of the Yangtze River (RYR) to determine the effects of nitrogen (N), phosphorus (P), and potassium (K) deficiencies on population density, dry matter production, nutrient uptake, seed yield, and yield components of DOR plants. Four fertilization treatments included the balanced NPK application treatment (NPK, 180 kg N, 39.3 kg P, 100 kg K, and 1.8 kg borax ha−1) and three nutrient deficiency treatments based on the NPK treatment, i.e., –N, –P, and –K. The results indicated that DOR population density declined gradually throughout the growing season, especially at over-wintering and pod-development stages. Nutrient deficiency decreased nutrient concentration in DOR plants, limited dry matter production and nutrient uptake, and thereby exacerbated density reduction during plants growth. The poor individual growth and reduced population density together decreased seed yield in the nutrient deficiency treatment. Averaged across all the experiments, seed yield reduced 61% by N deficiency, 38.3% by P deficiency, and 14.4% by K deficiency. The negative effects of nutrient deficiency on DOR performances followed the order of –N>–P>–K, and the effects were various among different nutrient deficiencies. Although N deficiency improved DOR emergence, but it seriously limited dry matter production and nutrient uptake, which in turn led to substantial plants death and therefore resulted in a very low harvested density. The P deficiency significantly reduced initial density, limited plants growth, and exacerbated density reduction. The K deficiency mainly decreased individual growth and yield, but did not affect density dynamic. Our results highlighted the importance of balanced NPK application in DOR production, suggesting that management strategy of these nutrients should be comprehensively considered with an aim to build an appropriate population structure with balanced plant density and individual growth.

Objective Esophageal cancer is one of the deadliest cancers worldwide occurring at upper gastrointestinal tract. This study aimed to explore the possible role of long non-coding RNA X Inactive Specific Transcript (XIST) in the development of esophageal cancer. Material and methods The lncRNA XIST expressions both in esophageal cancer tissues and in cells were investigated. The TE-1 and SKGT-4 cells were transfected with LV-sh-XIST and LV-scramble for the further detection of the effects of XIST expression on cell biological processes, including proliferation, apoptosis and the expression of apoptosis-related proteins, migration, invasion and the expression of epithelial-mesenchymal transition markers. Additionally, the regulatory relationships between lncRNA XIST and miR-494, between miR-494 and CDK6, between miR-494/CDK6 and JAK2/STAT3 pathway were investigated. Results LncRNA XIST was overespressed in esophageal cancer tissues and cells. Suppression of XIST significantly inhibited the proliferation, migration and invasion, but induced apoptosis of two kinds of cells, TE-1 and SKGT-4. Moreover, miR-494 was down-regulated in esophageal cancer tissues and cells. XIST could sponge miR-494 and inhibition of miR-494 reversed the effects of XIST suppression on the malignant behaviors of TE-1 cells. Also, CDK6 was a target of miR-494 and CDK6 knockdown reversed the effects of miR-494 inhibition on the malignant behaviors of TE-1 cells. Besides, the expression of p-JAK2 and p-STAT3 was increased after miR-494 inhibition, which was reversed after inhibition of miR-494 and CDK6 at the same time. Conclusions The data presented in this study revealed that XIST abnormal expression may play an oncogenic role in the development of esophageal cancer via miR-494/CDK6 axis through JAK2/STAT3 signal pathway. This study may provide theoretical basis for the molecular mechanism investigation of esophageal cancer.

Tissue engineering technology is applicable for study of nerve regeneration after spinal cord injury. Many natural and artificial scaffold are not applicable because of poor mechanical properties and cell compatibility. Polypeptides with fine three-dimensional structure and cell compatibility and are widely used in tissue engineering research. The purpose of this study was to verify the neuronal differentiation of neural stem cells by using self-polymerize dendritic polypeptide for spinal cord tissue engineering. Neural stem cells were isolated from cerebral cortex of neonatal SD rats.Conventional media was triggered the 1wt% nano peptide solution self polymerizated to formed a nano gel. The gel was tested by scanning electron microscope and transmission electron microscope. Neural stem cells were inoculated onto gel or on Polylysine-coated slides with fetal bovine serum or not. SD rats were randomized divided into four groups. neural stem cells and self-polymerized peptide were transplanted into spinal cord injury models. Then we test the Density of NF-positive axons in the spinal cord injury area at 8 weeks after surgery and MS score of the locomotive function of hind limbs among mice of four groups. Neural stem cells were showed anti Nestin (+), anti NSE (+), anti GFAP (+). The gel tested by scanning electron microscope was showed thick wall structure, another one tested by transmission electron microscope was showed self-polymerized dendritic nanofibers, which contains several spacings. The cells in serum group were differentiate into neurons, but non serum group were not. These results suggest that the self-assembling peptide nanofiber scaffold(SAPNS) were cytocompatible to neural stem cells which were differentiated into neurons. A large number of axonal regeneration and recovery of joint function of hind limb were appeared. The self-polymerized Peptide maybe used as practical tissue engineering materials as future.

San Leng powder extract has been used as medicinal compound for the prevention and treatment of cancers. The antitumor activity of SLPE was determined by treating BALB/C mice harboring a human gastric cancer xenograft with SPLE for 17 days. Mice were also treated with fluorouracil (5-Fu, 25 mg/kg) or a combination of SLPE and 5-Fu. Our results indicate that the inhibition of tumor growth by SLPE might be due to a block in the cell cycle and the induction of apoptosis. These results suggest that SLPE might be useful in the treatment of gastric cancer.

Although the function of the anterolateral stabilizing structures of the knee in the anterior cruciate ligament (ACL) injuries has been recognized since many years, most of orthopedic surgeons do not take the anterolateral structure into consideration when performing an ACL reconstruction. Anatomic ACL reconstruction will improve knee stability, but a small subset of patients may experience some residual anteroposterior and rotational instability. For this reason, some researchers have paid attention to the anterolateral aspects of the knee, especially the anterolateral ligament. We don't know the best time to perform ACL and ALL reconstruction. And we lack the evidence to prove which technique is the best one. So we look forward to more random controlled trial.

To compare the clinical effect of Endobutton plates combined with an anchor and clavicle hook plate in the treatment of acromioclavicular dislocation.From January 2012 to August 2014, 83 patients with Rockwood type III acromioclavicular dislocation underwent surgical treatments. Among them, 34 patients were treated with Endobutton plate and anchor repair(Endobutton group), including 23 males and 11 females, and the mean age was(39.0±6.3) years old (26 to 51 years old); the average time from injury to operation was(4.1±1.3) days(3 to 7 days);the injured side:14 left, 20 right; the dislocation in 28 patients dues to fall, 6 patients dues traffic accident. There were 49 patients treated with clavicular hook plate(hook plate group), including 33 males and 16 females;the mean age was(37.9±6.3) years old (27 to 53 years old); the average time from injury to operation was(4.1±1.1) days (2 to 7 days);the injured side: 18 left, 31 right;the dislication in 36 patients dues to fall, 13 patients dues traffic accidents. The indexes such as intraoperative bleeding volume, operation time, incision size, postoperative complication and postoperative coracoclavicular space, shoulder joint function, and life quality were compared between two groups.In the hook plate group with 49 patients, the plates in 43 patients were removed at the secondary operation, and 32 patients had shoulder pain or limited active range. Thirty four patients in the Endobutton group had no pain symptoms and limited active range. All the patients did not suffer acromioclavicular dislocation again. There was no significant difference between the two groups in operation time, and intraoperative bleeding volume(Endobutton plate combined with an anchor can effectively fix Rockwood type III or more acute acromioclavicular dislocation. The method has less complications, avoiding secondary removal of internal fixation.

This paper reports a novel optimization strategy combined with artificial intelligence for parameters and location design of precision equipment and provides a broader view for traditional passive vibration isolation. It also considers "precision equipment-vibration isolators-thin plate foundation" as a composite passive vibration isolation system and the clamped thin plate. The vibration isolation system is considered as four-point support, and the displacement amplitude transmissibility from the thin plate to precision equipment is derived and based on the analysis of influencing factors of transmissibility; subsequently, multi-objective optimization of the composite system is performed. A novel swarm intelligence multi-objective optimization method—a multi-objective particle swarm optimization (MOPSO) algorithm is adopted in this paper which can achieve a global optimal solution and by selecting the desired solution from an equivalence relation, the whole Pareto set can be avoided. The maximum and variance of the four transmitted peak displacements are simultaneously considered as fitness functions, and the purpose is to reduce the amplitude of the multi-peak isolation system, and in the meantime, to allow the plate to be uniformly vibrated as far as possible. Moreover, the presented idea is validated numerically, and the location research of the precision equipment mounted on the plate is also conducted.

To explore the treatment of chronic calcaneal osteomyelitis with bone defect after debridement and evaluate its clinical outcomes.From June 2009 to June 2011, 52 patients with chronic calcaneal osteomyelitis were treated with stage-one compound grafting of antibiotic-impregnated calcium sulfate and autogenous cancellous bone,including 12 females and 40 males with an average age of 43 years old ranging from 18 to 67. According to Cierny-Mader classification, there were 34 cases with stage III and 18 with stage IV. There were 32 cases on right side and 20 on left,with a course of 6 months to 3 years. The area of soft tissue wound ranged from 3.0 cm x l.5 cm to 23.0 cm x l2.0 cm. The clinical effects were evaluated according to infection controlling, calcium sulfate absorption,bone defect repair and heel functional recovery.All patients were followed up for 2 to 3.8 years (averaged 2.8 years). Primary healing was achieved in 52 patients. Two cases of recurrence were found post-operatively, 1 case in 3 months and another in 5 months,which were cured after a second operation. Bone repair healing was gained in 1.5 to 3.5 months (averaged 2.5 months). Complete radiological absorption of calcium sulfate was found in 1.2 to 3 months(averaged 2.2 months). Local exudation after removal of drainage tube had been persisting in 10 patients for 2 to 3 months, which was consistent with the time when cacium sulfate were totally absorbed. Flap had partial necrosis in 4 cases,and the wounds were closed after appropriate treatment finally. The mean Maryland score was 88.15±7.70. There were excellent results in 32 cases, good in 14, fair in 6.A satisfactory short-term clinical results can be gained by one-stage compound grafting of antibiotic-impregnated calcium sulfate and autogenous cancellous bone in chronic calcaneal osteomyelitis, but the long-term results need further follow-up. And much more study is also demanded to reduce the exudation of calcium sulfate.
