Abstract

The Athena mission entered a redefinition phase in July 2022, driven by the imperative to reduce the mission cost at completion for the European Space Agency below an acceptable target, while maintaining the flagship nature of its science return. This notably called for a complete redesign of the X-ray Integral Field Unit (X-IFU) cryogenic architecture towards a simpler active cooling chain. Passive cooling via successive radiative panels at spacecraft level is now used to provide a 50 K thermal environment to an X-IFU owned cryostat. 4.5 K cooling is achieved via a single remote active cryocooler unit, while a multi-stage Adiabatic Demagnetization Refrigerator ensures heat lift down to the 50 mK required by the detectors. Amidst these changes, the core concept of the readout chain remains robust, employing Transition Edge Sensor microcalorimeters and a SQUID-based Time-Division Multiplexing scheme. Noteworthy is the introduction of a slower pixel. This enables an increase in the multiplexing factor (from 34 to 48) without compromising the instrument energy resolution, hence keeping significant system margins to the new 4 eV resolution requirement. This allows reducing the number of channels by more than a factor two, and thus the resource demands on the system, while keeping a 4' field of view (compared to 5' before). In this article, we will give an overview of this new architecture, before detailing its anticipated performances. Abstract

The Athena X-ray Integral Unit (X-IFU) is the high resolution X-ray spectrometer, studied since 2015 for flying in the mid-30s on the Athena space X-ray Observatory, a versatile observatory designed to address the Hot and Energetic Universe science theme, selected in November 2013 by the Survey Science Committee. Based on a large format array of Transition Edge Sensors (TES), it aims to provide spatially resolved X-ray spectroscopy, with a spectral resolution of 2.5 eV (up to 7 keV) over an hexagonal field of view of 5 arc minutes (equivalent diameter). The X-IFU entered its System Requirement Review (SRR) in June 2022, at about the same time when ESA called for an overall X-IFU redesign (including the X-IFU cryostat and the cooling chain), due to an unanticipated cost overrun of Athena. In this paper, after illustrating the breakthrough capabilities of the X-IFU, we describe the instrument as presented at its SRR, browsing through all the subsystems and associated requirements. We then show the instrument budgets, with a particular emphasis on the anticipated budgets of some of its key performance parameters. Finally we briefly discuss on the ongoing key technology demonstration activities, the calibration and the activities foreseen in the X-IFU Instrument Science Center, and touch on communication and outreach activities, the consortium organisation, and finally on the life cycle assessment of X-IFU aiming at minimising the environmental footprint, associated with the development of the instrument. Thanks to the studies conducted so far on X-IFU, it is expected that along the design-to-cost exercise requested by ESA, the X-IFU will maintain flagship capabilities in spatially resolved high resolution X-ray spectroscopy, enabling most of the original X-IFU related scientific objectives of the Athena mission to be retained. 5. Discovery of holoenzyme-disrupting chemicals as substrate-selective CK2 inhibitors.

Abstract

CK2 is a constitutively active protein kinase overexpressed in numerous malignancies. Interaction between CK2α and CK2β subunits is essential for substrate selectivity. The CK2α/CK2β interface has been previously targeted by peptides to achieve functional effects; however, no small molecules modulators were identified due to pocket flexibility and open shape. Here we generated numerous plausible conformations of the interface using the fumigation modeling protocol, and virtually screened a compound library to discover compound 1 that suppressed CK2α/CK2β interaction in vitro and inhibited CK2 in a substrate-selective manner. Orthogonal SPR, crystallography, and NMR experiments demonstrated that 4 and 6, improved analogs of 1, bind to CK2α as predicted. Both inhibitors alter CK2 activity in cells through inhibition of CK2 holoenzyme formation. Treatment with 6 suppressed MDA-MB231 triple negative breast cancer cell growth and induced apoptosis. Altogether, our findings exemplify an innovative computational-experimental approach and identify novel non-peptidic inhibitors of CK2 subunit interface disclosing substrate-selective functional effects.

Abstract

By the end of 2018, 42 years after the landing of the two Viking seismometers on Mars, InSight will deploy onto Mars' surface the SEIS (Seismic Experiment for Internal Structure) instrument; a six-axes seismometer equipped with both a long-period three-axes Very Broad Band (VBB) instrument and a three-axes short-period (SP) instrument. These six sensors will cover a broad range of the seismic bandwidth, from 0.01 Hz to 50 Hz, with possible extension to longer periods. Data will be transmitted in the form of three continuous VBB components at 2 sample per second (sps), an estimation of the short period energy content from the SP at 1 sps and a continuous compound VBB/SP vertical axis at 10 sps. The continuous streams will be augmented by requested event data with sample rates from 20 to 100 sps. SEIS will improve upon the existing resolution of Viking's Mars seismic monitoring by a factor of ∼2500 at 1 Hz and ∼200000 at 0.1 Hz. An additional major improvement is that, contrary to Viking, the seismometers will be deployed via a robotic arm directly onto Mars' surface and will be protected against temperature and wind by highly efficient thermal and wind shielding. Based on existing knowledge of Mars, it is reasonable to infer a moment magnitude detection threshold of Mw∼3 at 40∘ epicentral distance and a potential to detect several tens of quakes and about five impacts per year. In this paper, we first describe the science goals of the experiment and the rationale used to define its requirements. We then provide a detailed description of the hardware, from the sensors to the deployment system and associated performance, including transfer functions of the seismic sensors and temperature sensors. 14. In College and in Recovery: Reasons for Joining a Collegiate Recovery Program

Abstract

Objective: Collegiate Recovery Programs (CRPs), a campus-based peer support model for students recovering from substance abuse problems, grew exponentially in the past decade, yet remain unexplored. Methods: This mixed-methods study examines students' reasons for CRP enrollment to guide academic institutions and referral sources. Students (N = 486) from the 29 CRPs nationwide operating in 2012 completed an online survey in 2013. Results: Students were somewhat older than traditional age (mean age = 26). Now sober for 3 years (mean), they had experienced severe dependence on multiple substances. One third reported they would not be in college were it not for a CRP, and 20% would not be at their current institution. Top reasons for joining a CRP were the need for same-age peer recovery support and wanting to "do college sober," recognizing that college life challenges sobriety. Conclusions: CRPs appear to meet their mission of allowing recovering students to pursue educational goals in "an abstinence hostile environment." Findings emphasize the need for more institutions to address the support needs of students in recovery.

Abstract

Mars’s seismic activity and noise have been monitored since January 2019 by the seismometer of the InSight (Interior Exploration using Seismic Investigations, Geodesy and Heat Transport) lander. At night, Mars is extremely quiet; seismic noise is about 500 times lower than Earth’s microseismic noise at periods between 4 s and 30 s. The recorded seismic noise increases during the day due to ground deformations induced by convective atmospheric vortices and ground-transferred wind-generated lander noise. Here we constrain properties of the crust beneath InSight, using signals from atmospheric vortices and from the hammering of InSight’s Heat Flow and Physical Properties (HP3) instrument, as well as the three largest Marsquakes detected as of September 2019. From receiver function analysis, we infer that the uppermost 8–11 km of the crust is highly altered and/or fractured. We measure the crustal diffusivity and intrinsic attenuation using multiscattering analysis and find that seismic attenuation is about three times larger than on the Moon, which suggests that the crust contains small amounts of volatiles.

21. Counting Nemo: anemonefish Amphiprion ocellaris identify species by number of white bars

Abstract

The brilliant colors of coral reef fish have received much research attention. This is well exemplified by anemonefish, which have distinct white bar patterns and inhabit host anemones and defend them as a territory. The 28 described species have between 0 and 3 white bars present, which has been suggested to be important for species recognition. In the present study, we found that Amphiprion ocellaris (a species that displays three white bars) hatched and reared in aquaria, when faced with an intruder fish, attacked their own species more frequently than other species of intruding anemonefish. Additionally, we explicitly tested whether this species could distinguish models with different numbers of bars. For this, 120 individuals of A. ocellaris were presented with four different models (no bars, and 1, 2 and 3 bars) and we compared whether the frequency of aggressive behavior towards the model differed according to the number of bars. The frequency of aggressive behavior toward the 3-bar model was the same as against living A. ocellaris, and was higher than towards any of the other models. We conclude that A. ocellaris use the number of white bars as a cue to identify and attack only competitors that might use the same host. We considered this as an important behavior for efficient host defense.

Aussi, nous avons mené une étude rétrospective avec pour objectifs : (1) d’évaluer si la vancomycine topique diminuait le taux d’infections péri-prothétiques ; (2) étudier son influence sur d’éventuelles complications cicatricielles.

24. ZebRA: An overview of retinoic acid signaling during zebrafish development.

Abstract

Retinoic acid (RA), the main active vitamin A derivative, is crucial for embryo development, regulating cellular processes, embryo patterning and organogenesis. Many studies performed in mammalian or avian models have successfully undertaken the investigation of the role played by RA during embryogenesis. Since the early 1980s, the zebrafish ( Danio rerio ) has emerged as a powerful developmental model to study the in vivo role of RA during embryogenesis. Unlike mammalian models, zebrafish embryogenesis is external, not only allowing the observation of the translucent embryo from the earliest steps but also providing an easily accessible system for pharmacological treatment or genetic approaches. Therefore, zebrafish research largely participates in deciphering the role of RA during development. This review aims at illustrating different concepts of RA signaling based on the research performed on zebrafish. Indeed, RA action relies on a multitude of cross-talk with other signaling pathways and requires a coordinated, dynamic and fine-regulation of its level and activity in both temporal and spatial dimensions. This review also highlights major advances that have been discovered using zebrafish such as the observation of the RA gradient in vivo for the first time, the effects of RA signaling in brain patterning, its role in establishing left–right asymmetry and its effects on the development of a variety of organs and tissues including the heart, blood, bone and fat. This review demonstrates that the zebrafish is a convenient and powerful model to study retinoic acid signaling during vertebrate embryogenesis. Abstract

Most marine organisms have a biphasic life cycle during which pelagic larvae transformed into radically different juveniles. In vertebrates, the role of thyroid hormones (TH) in triggering this transition is well known, but how the morphological and physiological changes are integrated in a coherent way with the ecological transition remains poorly explored. To gain insight into this question, we performed an integrated analysis of metamorphosis of a marine teleost, the false clownfish Amphiprion ocellaris. We show how TH coordinate a change in color vision as well as a major metabolic shift in energy production, hence highlighting how it orchestrates this transformation. By manipulating the activity of LXR, a major regulator of metabolism, we also identify a tight link between metabolic changes and metamorphosis progression. Strikingly, we observed that these regulations are at play in the wild, explaining how hormones coordinate energy needs with available resources during life cycle.

L’objectif de ce travail était donc d’obtenir des informations sur le ressenti des équipes officinales en Aveyron concernant les outils mis à disposition par les différents centres hospitaliers ayant une activité de cancérologie (lors de la réception d’une primo-prescription de MAVO), et d’évaluer l’impact de ces dispositifs sur leur pratique.

28. Combinaison de deux technologies innovantes (bras robotisé et capteur de force per opératoire) dans l’équilibrage ligamentaire lors de la pose de prothèse totale de genou

Abstract

Un bon équilibrage ligamentaire lors de la pose d’une prothèse totale de genou est supposé améliorer les résultats cliniques, mais l’obtention d’un équilibre ligamentaire est très dépendant du chirurgien sans aide technologique. Aussi nous proposons une technique présentée associant l’utilisation du bras robotisé MAKO (Stryker, Kalamazoo, Michigan, USA) comme seul moyen d’équilibrer la tension ligamentaire par des recoupes osseuses sous le contrôle du capteur de force VERASENSE (Orthosensor, Inc, Dania Beach, Florida, USA), lors de la mise en place d’une prothèse totale de genou. Sur cette série préliminaire de 29 patients, à la fin de la procédure, 27 cas (93%) étaient jugés comme équilibrés en extension et 23 (79%) en flexion et en extension, cela sans avoir pratiqué aucun geste d’allongement des parties molles péri-articulaires. Le rôle du capteur de force est d’optimiser, après les premières coupes osseuses, l’analyse de l’équilibre ligamentaire et guider une éventuelle recoupe osseuse (fémorale ou tibiale). Cette technique permet un équilibre quantifiable et maitrisé de la tension ligamentaire lors de la pose d’une prothèse totale de genou. La collecte des données objectives per-opératoires permettra de faire avancer les connaissances sur la mise en place d’une prothèse totale de genou.
