21. Influence of culture conditions on extracellular polymeric substances production by the white rot fungi Phanerochaete chrysosporium

Abstract

The extracellular polymeric substances of white rot fungi play an important role in the adsorption of heavy metals, but the influence of culture conditions on extracellular polymeric substances production is still unknown. In this paper, we researched on the influence of temperature, incubation time, the rotational speed and the inoculation volume on the yield of extracellular polymeric substances produced by Phanerochaete chrysosporium, a model strain of white rot fungi. The results show that the optimum culture conditions for Phanerochaete chrysosporium to produce extracellular polymeric substances was culturing at 40 °C, incubating for 5 d, rotating at 100 rpm, and inoculating 0.5 ml of spore suspension with concentration of 2.5×106 spores/ml. The highest yield of EPS was 234.65 mg/g when the fungi was cultured at 100 rpm, 40 °C and incubated for 5 days. This study can provide useful information for the follow-up experiments related to extracellular polymeric substances of white rot fungi

Abstract

Translation and bend of the spine in the sagittal plane during high-speed quadruped running were investigated. The effect of the two spine motions on slip between the foot and the ground was also explored. First, three simplified sagittal plane models of quadruped mammals were studied in symmetric bounding. The first model’s trunk allowed no relative motion, the second model allowed only trunk bend, and the third model allowed both bend and translation. Next, torque was introduced to equivalently replace spine motion and the possibility of foot slip of the three models was analyzed theoretically. The results indicate that the third model has the least possibility of slip. This conclusion was further confirmed by simulation experiments. Finally, the conclusion was verified by the reductive model crawling robot.

Abstract

Energy efficiency is important in the performance of quadruped robots and mammals. Flexible spine motion generally exists in quadruped mammals. This paper mainly explores the effect of flexible spinal motion on energy efficiency. Firstly, a planar simplified model of the quadruped robot with flexible spine motion is introduced and two simulation experiments are carried out. The results of simulation experiments demonstrate that both spine motion and spinal flexibility can indeed increase energy efficiency, and the curve of energy efficiency change along with spinal stiffness is acquired. So, in order to obtain higher energy efficiency, quadruped robots should have flexible spine motion. In a certain speed, there is an optimal spinal stiffness which can make energy efficiency to be the best. Secondly, a planar quadruped robot with flexible spine motion is designed and the conclusions drawn in the two simulation experiments are verified. Lastly, the third simulation experiment is carried out to explore the relationship between the optimal spinal stiffness, speed and total mass. The optimal spinal stiffness increases with both speed and total mass, which has important guiding significance for adjusting the spinal stiffness of quadruped robots to make them reach the best energy efficiency.
