8. Calycosin activates Nrf2/Keap1 signaling to ameliorate hydrogen peroxide‐induced spinal cord neuron death and mitochondrial dysfunction.

Abstract

Oxidative stress is a hallmark of secondary injury of spinal cord injuries. Controlling oxidative stress is crucial for mitigating secondary injury and promoting functional recovery after spinal cord injuries. Calycosin is an O‐methylated isoflavone with antioxidant activity. To evaluate the effect of calycosin on spinal cord neurons under oxidative stress and clarify the molecular mechanism underlying the effect, we tested the neuroprotective activity of calycosin in a primary spinal cord neuron culture model. We found that calycosin protected neurons from H2O2‐induced neuronal death in a dose‐dependent manner. Further experiments revealed that calycosin decreased H2O2‐induced mitochondrial fragmentation and mitochondrial membrane potential loss, and subsequently reduced H2O2‐triggered release of mitochondrial cytochrome c into the cytoplasm. In addition, calycosin inhibited H2O2‐induced reactive oxygen species generation and activation of NF‐κB signaling in spinal cord neurons. Furthermore, the expression of several antioxidant enzymes such as HO‐1, NQO1, GCLC, GCLM, TrxR1, and Trx1 was significantly promoted by calycosin. More importantly, we revealed that the Nrf2/Keap1 signal is crucial for the effect of calycosin, because calycosin increased the amount of nuclear Nrf2 while decreasing the amount of cytoplasmic Nrf2. Nrf2 knockdown with siRNA transfection abolished the neuroprotective effect of calycosin. Taken together, this study disclosed a novel mechanism by which calycosin combats oxidative stress. In this study, we characterized the genomic feature of an IMP-4-producing Klebsiella pneumoniae ST1873 strain, a rare sequence type (ST) isolated from an infant with a bloodstream infection in China.Patients and Methods: K. pneumoniae strain, BKP19, was collected from a bloodstream infection in an infant who was hospitalized at the department of paediatrics. The whole genome sequence of the strain was sequenced using the Illumina NovaSeq 6000 platform and long-read MinION sequencer. Multilocus sequence typing, antimicrobial resistance gene identification, plasmid and phylogenetic relationship analysis of the strain were analysed by various bioinformatics approaches.Results: K. pneumoniae BKP19 was resistant to multiple antimicrobials, including carbapenems. Eleven antimicrobial resistance genes corresponding to beta-lactam resistance, quinolone resistance, phenicol resistance and fosfomycin resistance could be identified in the genome. Abstract

Background: Few biomarkers can predict the efficiency of PD-1 blockade in patients with hepatocellular carcinoma (HCC). This study aimed to investigate the prognostic role of AFP and PIVKA-II in HCC patients receiving anti-PD-1 immunotherapy.Methods: A total of 235 HCC patients treated with PD-1 blockade were enrolled. Serum AFP and PIVKA-II levels were collected before and after treatments. The patients were divided into groups based on the reduction in AFP and PIVKA-II: AFP reduction ≤50% vs AFP reduction > 50% and PIVKA-II reduction ≤50% vs PIVKA-II reduction > 50%. The primary endpoints included objective response rate (ORR), progression-free survival (PFS) and overall survival (OS). Binary logistic regression analyses were used to explore the related factors of ORR. A Cox proportional hazards model was employed to identify the potential prognostic factors of PFS and OS.Results: Among all the patients, 34.9% (82/235) achieved a complete or partial response. There was a positive correlation between AFP reduction > 50% or PIVKA-II reduction> 50% and the ORR of PD-1 blockade (P < 0.001 and = 0.003). PFS was significantly improved in patients with AFP reduction > 50% and PIVKA-II reduction > 50% (p < 0.001 and = 0.021). In addition, AFP reduction > 50% and PIVKA-II reduction> 50% were positively correlated with longer OS (p = 0.003 and 0.006).Conclusion: Early reductions in AFP and PIVKA-II can be predictors of the efficacy of PD-1 blockade in HCC patients. Abstract

Magnesium hydride has attracted great attention because of its high theoretical capacity and outstanding reversibility, nevertheless, its practical applications have been restricted by the disadvantages of the sluggish kinetics and high thermodynamic stability. In this work, an unexpected high reversible hydrogen capacity over 8.0 wt% has been achieved from MgH2metal hydride composited with small amounts of LiBH4and Li3AlH6complex hydrides, which begins to release hydrogen at 276 °C and then completely dehydrogenates at 360 °C. The dehydrogenated MgH2+LiBH4/Li3AlH6composite can fully reabsorb hydrogen below 300 °C with an excellent cycling stability. The composite exhibits a significant reduction of dehydrogenation activation energy from 279.7 kJ/mol (primitive MgH2) to 139.3 kJ/mol (MgH2+LiBH4/Li3AlH6), as well as a remarkable reduction of dehydrogenation enthalpy change from 75.1 kJ/mol H2(primitive MgH2) to 62.8 kJ/mol H2(MgH2+LiBH4/Li3AlH6). The additives of LiBH4and Li3AlH6not only enhance the cycling hydrogen capacity, but also simultaneously improve the reversible de/rehydrogenation kinetics, as well as the dehydrogenation thermodynamics. This notable improvement on the hydrogen absorption/desorption behaviors of the MgH2+LiBH4/Li3AlH6composite could be attributed to the dehydrogenated products including Li3Mg7, Mg17Al12and MgAlB4, which play a key role on reducing the dehydrogenation activation energy and increasing diffusion rate of hydrogen. Meanwhile, the LiBH4and Li3AlH6effectively destabilize MgH2with a remarkable reduction on dehydrogenation enthalpy change in terms of thermodynamics. In particular, the Li3Mg7, Mg17Al12and MgAlB4phases can reversibly transform into MgH2, Li3AlH6and LiBH4after rehydrogenation, which contribute to maintain a high cycling capacity. This constructing strategy can further promote the development of high reversible capacity Mg-based materials with suitable de/rehydrogenation properties.
