In the central-western and southern regions of Brazil, the cultivation of yerba mate (Ilex paraguariensis) is considered an important source of forest extraction, besides providing an alternative income, especially for families. The yerba mate, like other forest species, is infected by several pathogens, such as fungi of the genus Fusarium, causing root rot. Pathogens have wide genetic variability, even within a single genus and species, resulting from adaptations to their environments. This study aimed at characterizing molecularly isolated Fusarium spp. through the sequencing of the ITS and β-tubulin genome regions, identifying the species level as well as the formation of vegetative compatibility groups (VCG) among these isolates of the root rot pathogen in yerba mate. The sequencing of the ITS and β-tubulin regions, defined the separation between the seven pathogenic isolates of F. solani and F. oxysporum. For the same isolates, the technique to obtain nit mutants was suitable for the phenotypic classification, obtaining five VCGs for species of Fusarium, root rot in yerba mate. Resumo Nas regiões Centro-Oeste e Sul do Brasil, a cultura da erva-mate (Ilex paraguariensis) constitui-se uma importante fonte de extrativismo florestal, além de diversificar a renda, principalmente na agricultura familiar. A erva-mate, como outras espécies florestais, é infectada por diversos patógenos, como o gênero Fusarium, causador da podridão-de-raízes. Os patógenos possuem ampla variabilidade genética, mesmo dentro de um único gênero e dentro de uma única espécie, resultado de adaptações ao meio em que vivem. Este estudo teve como objetivo avaliar o sequenciamento das regiões do genoma ITS e β-tubulina na identificação em nível de espécie dos isolados de Fusarium spp. patogênicos à erva-mate, e identificar a formação de grupos de compatibilidade vegetativa (VCG) entre os isolados de Fusarium spp. patogênicos à erva-mate. O sequenciamento das regiões ITS e β-tubulina consolidou a separação entre os sete isolados patogênicos em F. solani e F. oxysporum. Para esses isolados, a técnica para obtenção de mutantes nit foi adequada para a classificação fenotípica, fundamentais nos testes de VCG para espécies de Fusarium, sendo obtidos cinco VCG.

Avaliou-se com este trabalho, o efeito dos compostos voláteis dos óleos essenciais de alho (Allium sativum), arruda (Ruta graveolens), carqueja (Baccharis trimera) e nim (Azadirachta indica) sobre o crescimento micelial e esporulação de Monilinia fructicola. O fungo foi isolado em meio de cultura BDA (Batata-Dextrose-Ágar) a partir de pêssegos infectados pelo patógeno. Foi aplicado a dose de 20 μL de cada óleo essencial em papel filtro esterilizado com 1,5 cm², fixado no centro da tampa da placa de Petri®. Colocou-se um disco de 0,5 cm de diâmetro contendo micélio e conídios do patógeno no centro da superfície do meio de cultura BDA. Todas as placas foram vedadas e em seguida, incubadas em BOD a 25 ºC e fotoperíodo de 12 h. Após 24 h iniciou-se a mensuração do crescimento micelial de M. fructicola, por 7 dias, neste dia foi avaliado a produção de conídios. Pode-se observar que o óleo essencial de alho liberou compostos voláteis que inibiram totalmente o crescimento micelial de M. fructicola, o que pode ser observado também na produção de conídios. Os demais óleos não diferiram da testemunha., The effect of the volatile compounds of the essential oils of garlic (Allium sativum), rue (Ruta graveolens), carqueja (Baccharis trimera) and neem (Azadirachta indica) on mycelial growth and sporulation of Monilinia fructicola were evaluated. The fungus was isolated in BDA (Potato-Dextrose-Agar) culture medium from peaches infected by the pathogen. The 20 mL dose of each essential oil was applied on 1.5 cm² sterile filter paper, fixed to the center of the lid of the Petri dish. A 0.5 cm diameter disk containing mycelium and conidia of the pathogen was placed in the center of the surface of the BDA culture medium. All plates were sealed and then incubated in BOD at 25 ° C and photoperiod for 12 h. After 24 h the measurement of the mycelial growth of M. fructicola was begun, for 7 days, on this day the production of conidia was evaluated. It can be observed that the essential oil of garlic released volatile compounds that totally inhibited the mycelial growth of M. fructicola, which can also be observed in the production of conidia. The other oils did not differ from the control.

RESUMO Cordia americana (L.) Gottschling & J. S. Mill. (guajuvira) é uma espécie florestal amplamente distribuída no território brasileiro e que apresenta importância econômica e ambiental devido à qualidade de sua madeira e utilização em reflorestamentos. Com o objetivo de avaliar a patogenicidade de Fusarium sp. associado às sementes, foram caracterizados, morfológica e molecularmente, quatro isolados do fungo que foram inoculados nas sementes de guajuvira. A caracterização morfológica foi realizada com o auxílio de uma chave de identificação para o gênero Fusarium e para a identificação molecular foram sequenciadas as regiões genômicas Internal Transcribed Spacer (ITS) e Fator de elongação 1 - alpha (TEF1-α). Os resultados do sequenciamento de DNA concordaram com os da caracterização morfológica e permitiram identificar as espécies Fusarium acuminatum e Fusarium verticillioides. No teste de patogenicidade, verificou-se que ambas as espécies do fungo foram patogênicas a Cordia americana, causando tombamento em pré e pós-emergência, além de apodrecimento de raízes e necrose no hipocótilo. ABSTRACT Cordia americana (L.) Gottschling & J. S. Mill. (guajuvira) is a forest species widely distributed in Brazil and represents environmental and economic importance due to the quality of its timber and use in reforestation. In order to evaluate the pathogenicity of Fusarium sp. associated with seeds, characterizing the isolates morphologically and molecularly in order to identify them on the species level, was obtained four fungal isolates that were inoculated in seeds to demonstrate their pathogenicity. The morphological characterization was performed using an identification key for the genus Fusarium and, for the molecular identification, genomic regions Internal Transcribed Spacer (ITS) were sequenced with its elongation factor 1 alpha (TEF1-α). The results of the molecular identification agreed with the morphological characterization and allowed to identify the species Fusarium acuminatum and Fusarium verticillioides, both being pathogenic to Cordia americana, causing damping-off in pre and post emergence, and rotting of roots and necrosis of hypocotyl.

Produtos alternativos para o controle de doenças têm como objetivo substituir os químicos, proporcionando aos frutos mesma qualidade, sem agredir saúde e meio ambiente. O objetivo deste trabalho foi avaliar o potencial dos óleos essenciais,extratos hidroalcoólicos (álcool 70%) e extratos vegetais aquosos a frio e quentede alecrim (Rosmarinus officinalisL.), alho (Allium sativum), cravo-da-índia (Sizygium aromaticumL.),losna (Artemisia absinthium), orégano (Origanum vulgare L.) e pitangueira (Eugenia uniflora) no controle in vitro por contato direto e indireto do fungo Monilinia fructicolae na pós-colheita aplicados em pêssegoscultivar“Dourado”. Foram realizados dois estudos. No Estudo I avaliou-se os óleos essenciais e extratos vegetais por contato direto e indireto,analisando o crescimento micelial, índice de crescimento micelial, inibição do crescimento micelial, concentração mínima inibitória, concentração fungicida mínima, indução de fitoalexinas, toxicologia dos produtos em Artemia salina e microscopia eletrônica de varredura. No Estudo II avaliou-se extratos vegetais e óleos essenciais por contato indireto e direto em pós-colheita de pêssegos, analisando a incidência e severidade da podridão parda nos frutos, além da indução de resistência, com análise das enzimas fenilalanina amônia-liase, quitinase e -1,3-glucanase. O delineamento experimental foi inteiramente casualizado, sendo a concentração utilizada para osóleos essenciais1%e extratos vegetais 10%. No Estudo I contato direto e indireto dos óleos essenciais de alho, cravo-da-índia e orégano; contato direto do extrato frio de alho; extrato quente de alecrim, alho e cravo-da-índia; extrato hidroalcoólico de alho e cravo-da-índia obtiveram os melhores resultados quanto as avaliações realizadas. No Estudo II, os óleos essenciais de alho por contato indireto e orégano por contato direto apresentaram potencial para controlar a incidência e severidade da podridão parda na pós-colheita de pêssegos. Esses óleos também induziram resistência pela ativação das enzimas FAL e quitinase. Desta forma, os óleos essênciais cravo-da-índia e orégano obtiveram resultados satisfatórios nos Estudos I e II e,se tornam promissores para mais estudos no controle da M. fructicola. The objective of this work was to evaluate the potential of essential oils and hydroalcoholic extracts (70% alcohol) and hot and cold aqueous plant extracts of rosemary (Rosmarinus officinalisL.), garlic (Allium sativum), cloves (Sizygium aromaticumL.), wormwood (Artemisia absinthium), oregano (Origanum vulgareL.) and Surinam cherry (Eugenia uniflora) in the in vitro control by direct and indirect contact of the fungus Monilinia fructicolaand in the Dourado peach variety post-harvest. Two studies were carried out. In Study I, essential oils and plant extractswere evaluated by direct and indirect contact, analyzing mycelial growth, mycelial growth index, inhibition of mycelial growth, minimum inhibitory concentration, minimum fungicidal concentration, phytoalexin induction, toxicology of products in Artemia salineand scanning electron microscopy. In Study II, plant extracts and essential oils were evaluated by indirect and direct contact in post-harvest peaches, analyzing the incidence and severity of brown rot in fruits, in addition to resistance induction, with analysis of the enzymes phenylalanine ammonia lyase, chitinase and -1,3-glucanase. The experimental design was completely randomized, with the concentration used for essential oils 1% and plant extracts 10%. In Study I direct and indirect contact withessential oils of garlic, cloves and oregano; direct contact of the cold garlic extract; hot extract of rosemary, garlic and cloves; hydroalcoholic extract of garlic and cloves obtained the best results regarding the evaluations performed. In Study II, the essential oils of garlic by indirect contact and oregano by direct contact had the potential to control the incidence and severity of brown rot in the post-harvest of peaches. These oils also induced resistance by activating the enzymes FAL and chitinase. Thus, clove and oregano essential oils obtained satisfactory results in Studies I and II and are promising for further studies in the control of M. fructicola.

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior - CAPES The cultivation of Yerba-mate (Ilex paraguariensis) has intensified in the state of Rio Grande do Sul, due to the valorization of the price paid to the producer. However, the demand for studies related to the pathosystem Fusarium x yerba-mate is still insufficient, so that many producers have faced significant losses in the production of the plantations. Given the above, the aim of this study was to characterize the causal agent of rot-of-roots in Yerba-mate (I. paraguariensis). The following specific objectives were established: a) to select efficient morphological characters in the separation of the pathogenic isolates by groups of similarity b) to evaluate the sequencing of different regions of the genome in the identification of the pathogenic isolates at a species level c) to identify the formation of vegetative compatibility groups (VCG) among the pathogenic isolates d) to investigate the enzymatic arsenal of the pathogenic isolates. Therefore, samples were taken in five municipalities of the state, for the pathogen isolation. In addition, soil samples were collected for the area characterization and also for the geo-referencing of collection points. Fusarium spp. isolates were tested for their pathogenicity through substrate inoculation. It was not possible to associate the chemical properties of the soil of the collected areas with the pathogenicity of the isolates in commercial substrates. The 39 isolates were morphologically characterized through the variables of mycelial growth, sporulation, length and width of macroconidia, shape of microconidia, pigmentation of the colonies and formation of chlamydospores. The variables used in the morphological characterization were efficient in the discrimination for seven groups of isolates, especially length of macroconidia associated with sporulation. There were identified seven pathogenic isolates of Fusarium spp. The pathogenic isolates were also identified molecularly by sequencing the ITS, TEF-1 and β-tubulin regions. The sequencing of the ITS, TEF-1α and β-tubulin regions, consolidated the separation between the pathogenic isolates in F. solani and F. oxysporum. For the same isolates, the technique to obtain nit mutants is suitable for henotypic classification, which is fundamental in the VCG tests for Fusarium species. Five VCGs were obtained. The pathogenic isolates of Fusarium spp. produced extracellular enzymes catalase, laccase, cellulase, caseinase, amylase, protease, lipase, polygalacturonase and pectate lyase. O cultivo da erva-mate (Ilex paraguariensis) tem se intensificado no estado do Rio Grande do Sul, fruto da valorização do preço pago ao produtor. Conduto, a demanda de estudos relacionados ao patossistema Fusarium x erva-mate ainda são insuficientes, de modo que muitos produtores têm enfrentado perdas significativas na produção dos ervais. Diante do exposto, o objetivo geral deste trabalho foi caracterizar o agente causal da podridão-de-raízes da erva-mate (I. paraguariensis). Como objetivos específicos estabeleceram-se os seguintes: a) selecionar caracteres morfofisiológicos eficientes na separação dos isolados em grupos de similaridade; b) avaliar o sequenciamento de diferentes regiões do genoma na identificação dos isolados patogênicos em nível de espécie; c) identificar a formação de grupos de compatibilidade vegetativa (VCG) entre os isolados patogênicos; d) investigar o arsenal enzimático dos isolados patogênicos. Para tanto, foram realizadas coletas em cinco municípios do estado, para isolamento do patógeno. Além disso, foram coletadas amostras de solo para a caracterização da área e também o georreferenciamento dos pontos de coleta. Os isolados de Fusarium spp., foram testados quanto a sua patogenicidade através da inoculação em substrato. Não foi possível associar as propriedades químicas do solo das áreas de coleta com a patogenicidade dos isolados em substrato comercial. Os 39 isolados foram caracterizados morfologicamente através das variáveis crescimento micelial, esporulação, comprimento e largura dos macroconídios, formato dos microconídios, pigmentação das colônias e formação de clamidósporos. As variáveis utilizadas na caracterização morfológica foram eficientes na discriminação em sete grupos de isolados, especialmente comprimento dos macroconídios associado a esporulação. Foram identificados sete isolados patogênicos de Fusarium spp. Os isolados patogênicos também foram identificados molecularmente através de sequenciamento das regiões do gene ITS, TEF-1 e β-tubulina. O sequenciamento das regiões ITS, TEF-1α e β-tubulina, consolidou a separação entre os isolados patogênicos em F. solani e F. oxysporum. Para os mesmos isolados a técnica para obtenção de mutantes nit é adequada para a classificação fenótipica, fundamentais nos testes de VCG para espécies de Fusarium. Foram obtidos cinco VCG‟s. Os isolado patogênicos de Fusarium spp. produziram as enzimas extracelulares catalase, lacase, celulase, caseinase, amilase, protease, lipase, poligalacturonase e pectato liase.
