Studying scute and fin morphology are advantageous approaches for phylogenetic identification and provide information on biological linkages and evolutionary history that are essential for deciphering the fossil record. Despite this, no prior research has precisely characterized the histological structures of scutes in the common pleco. Therefore, this research investigated the microstructure and organization of bone tissue within the dermal skeleton, including the scutes and fins, in the common pleco, using light microscopy, stereomicroscopy, and scanning electron microscopy. The dermal scutes were organized in a pentagonal shape with denticular coverage and were obliquely aligned with the caudal portion pointing dorsally. The dermal scutes consisted of three distinct portions: the central, preterminal, and terminal portions. Each portion comprised three layers: a superficial bony plate, a basal bony plate, and a mid-plate. Both the superficial and basal bony plates were composed of lamellar bone and lamellar zonal bone, whilst the mid-plate consisted of secondary osteons and woven bone. In the terminal portion, the superficial and basal bony plates became thinner. The pectoral fin consists of spines and rays composed of lepidotrichium (two symmetrical hemi-rays). The spine contained centrifugal and centripetal lamellar and trabecular bones. A centripetal fibrous bone was implanted between the lamellar bones. Besides being oriented in a V shape, the hemi-rays were also composed of thin centrifugal and centripetal lamellar bones and trabecular bones. A fibrous bone was identified between the centrifugal and centripetal bones. The trabecular bone and lamellar bone were made up of bone spicules.

Background Healthcare medical dispatch systems play a fundamental role in the daily operations of prehospital services. This includes facilitating the interpretation of various dispatch-related systems, receiving incident calls, categorizing cases, guiding proper resources deployment, and providing proper instructions before the arrival of healthcare providers. Considering the Kingdom of Saudi Arabia's (KSA's) health sector transformation plan as part of Vision 2030, developing an Emergency Medical Dispatch (EMD) Program as part of essential prehospital services will allow rapid and competent healthcare delivery. In this study, our purpose is to describe the curriculum development of the EMD Program to improve the training of dispatchers and to share the experience in the interest of better prehospital dispatch systems. Methods A selected group of education experts and academics in emergency medical services dispatch were assigned to develop an EMD curriculum over six months. This study aims to describe the approach followed in developing an innovative EMD Program to share the experience and ultimately standardize dispatch-related training programs. The data of this study was collected by reviewing approved documents of the EMD Program including program curriculum, syllabus, logbook, and exam blueprint after approval letters were received from the Health Academy, Saudi Commission for Health Specialties. Results The development of the EMD program utilized a consecutive mixed approach starting with a competency-based with backward design method to ensure the achievement of targeted outcomes followed by the Kern Six-step curriculum development model, namely: (1) problem identification and general need assessment; (2) targeted need assessment; (3) goals and objectives; (4) educational strategies; (5) implementation; and (6) evaluation and feedback. This resulted in four comprehensive modules and seventeen competencies in interpreting various dispatch systems, prioritizing incidents, deploying proper resources, and providing pre-arrival instructions throughout the fourteen-week EMD Program. Conclusion As part of the health sector transformation plan in KSA, EMD services play a fundamental role in the daily operations of prehospital healthcare services. Abstract Clinical biomedical applications of genomic technologies are extensive and provide possibilities to enhance healthcare covering the span of medical talents. Genome disorder prediction is an important issue in biomedical research. Genome disorders cause multivariate diseases such as cancer, dementia, diabetes, Leigh syndrome, etc. Existing machine and deep learning-based methods were introduced to forecast genome disorders. However, the genome prediction outcomes were not sufficient. To address this issue, propose a new method called Quadratic Feynman Polynomial Interpolated and Vector Nearest Centroid-based (QFPI-VNC) for acutely predicting the genome disorder with improved sensitivity and specificity. First, we utilized medical data about children from a public genomes dataset and applied it to Linear Quadratic and Feynman Kac Genome filtering to obtain computationally efficient filtered results. Next, the results are fed to the Concordance Correlated Polynomial Interpolation with the purpose of extracting genome wide data in an accurate manner. Finally, the features extracted are fused and fed to the Support Vector and Nearest Centroid model for genome disorder prediction. Experimental investigations of the proposed method employing the genome dataset confirm that the performance of the proposed method is prospective and in the scope of acceptance with relative to state-of-the-art methods in terms of convergence speed, recognition rate, sensitivity, and specificity. Results suggest that the QFPI-VNC method produces the best performance with a higher genome disease detection rate by 14%, accuracy by 11%, sensitivity by 14% specificity by 12%, and lesser convergence speed by 29% than compared to state-of-the-art methods.

Abstract Biological communication system for neurological disorder patients is similar to the Brain Computer Interface in a way that it facilitates the connection to the outside world in real time. The interdisciplinary field of Electroencephalogram based message depiction is gaining importance as it assists the paralysed person to communicate. In the proposed method a novel approach of feature extraction is done by Empirical Mode Decomposition on non- stationary & non-linear kind of EEG signal. EMD helps in the effective time frequency analysis by disintegrating the EEG signal in the form of six Intrinsic Mode Functions with help of the frequency components. In all nine features are extracted from the decomposed IMFs so as to predict the states or messages of the patient. The above computed features are then served to the Deep Neural Network to perform the classification. The performance of suggested method is studied through applying it to the acquired database generated by the designed hardware as well as also in real time message depiction. The maximum classification accuracy 97% for the acquired database & 85% in real time are obtained respectively by comparative analysis. The command messages generated from the proposed system helps the person suffering from neurological disorder to establish the communication link with the outside world in an efficient way. Thus, the proposed novel method shows better performance in real time message depiction purpose as related to other existing methods.

الملخص: أهداف البحث: السكتة الدماغية هي أحد الأسباب الرئيسية للوفاة وضعف الوظيفة الجسدية. أصبحت عملية استئصال الخثرة داخل الأوعية الدموية الآن هي المعيار المعترف به لعلاج السكتة الدماغية الإقفارية الحادة في الدورة الدموية الأمامية مع انسداد الأوعية الدموية الكبيرة، وخاصة عندما تكون صور الدماغ المواتية واضحة في غضون 24 ساعة من ظهور الأعراض. تهدف هذه الدراسة إلى توضيح الخصائص والملامح السريرية ونتائج المرضى الذين خضعوا لاستئصال الخثرة داخل الأوعية الدموية للسكتة الدماغية الإقفارية الحادة. طريقة البحث: في تصميم مجموعة مستندة إلى السجلات بأثر رجعي، قمنا بمراجعة السجلات الطبية للأفراد الذين خضعوا لاستئصال الخثرة داخل الأوعية الدموية بسبب السكتة الدماغية الإقفارية الحادة في مستشفى الملك فهد العام في جدة، المملكة العربية السعودية. شملت الدراسة، التي أجريت من يوليو 2022 إلى نهاية أكتوبر 2023، تحليلا شاملا للعوامل السريرية والإشعاعية والعوامل المرتبطة بالوقت والنتائج المتنوعة. النتائج: في مجموعة مكونة من 117 مريضا بمتوسط عمر 58.5 عاما، كان 81 مريضا (69.2%) من الذكور. كان ارتفاع ضغط الدم والسكري من أكثر عوامل الخطر شيوعا، حيث أثرا على 48.7% و41% من المجموعة على التوالي. كان لدى ما يقرب من نصف المرضى درجة مواتية وفق برنامج ألبرتا للسكتة الدماغية للتصوير المقطعي المبكر (7-10)، وتلقى 45.3% منهم منشط البلازمينوجين عن طريق الوريد. حقق ما يقرب من ثلثي المرضى درجة 2ب في العلاج المعدل للاحتشاء الدماغي أو أعلى، وأظهر 39.3% انسداد الشريان الدماغي الأوسط-إم1. كان معدل حدوث التحول النزفي المصحوب بأعراض حوالي 13%، بينما حقق ثلث المرضى نتيجة مواتية حسب مقياس رانكين المعدل (0-2). ارتبطت عوامل مثل التقدم في السن، وارتفاع درجات مقياس السكتة الدماغية للمعاهد الوطنية للصحة، والإقامة الطويلة في المستشفى بنتائج أسوأ. وعلى العكس من ذلك، فإن غياب ارتفاع ضغط الدم والرجفان الأذيني، إلى جانب ارتفاع مقياس انحلال الخثرة في احتشاء دماغي، زاد بشكل كبير من احتمالية تحقيق نتائج إيجابية. الاستنتاجات: في دراستنا، أظهر مرضى استئصال الخثرة من الأوعية الدموية الذين تم فحصهم فئة عمرية أصغر سنا، حيث أظهرت النتائج السريرية والشعاعية درجة معينة من التشابه مع المنشورات الأخرى. ومن الجدير بالذكر أن غياب ارتفاع ضغط الدم والرجفان الأذيني، جنبا إلى جنب مع درجة أعلى في اختبار انحلال الخثرة في احتشاء دماغي، كان مرتبطا بزيادة احتمالية تحقيق نتيجة إيجابية Abstract: Background: Stroke is a significant contributor to both mortality and compromised physical function. Endovascular thrombectomy (EVT) is now the recognized standard of care for anterior circulation acute ischemic stroke (AIS) with large vessel occlusion (LVO), especially when favorable brain images are evident within 24 h of symptom onset. This study elucidated the characteristics, clinical profiles, and outcomes of patients who underwent EVT for AIS. Methodology: In this retrospective record-based cohort study, we reviewed the medical records of individuals who underwent EVT for AIS at King Fahad General Hospital in Jeddah, Saudi Arabia. The study, conducted from July 2022 through October 2023, encompassed a comprehensive analysis of clinical, radiological, and time-related factors and diverse outcomes. Results: In a cohort of 117 patients with a mean age of 58.5 years, 81 (69.2%) were male. Hypertension and diabetes mellitus were the most common risk factors, affecting 48.7% and 41% of the cohort, respectively. Approximately half of the patients had a favorable ASPECT Score (7–10), and 45.3% received intravenous tissue plasminogen activator (tPA). Nearly two-thirds achieved a modified thrombolysis in cerebral infarction (mTICI) score of 2b or higher, and 39.3% exhibited M1-middle cerebral artery (MCA) occlusion. The incidence of symptomatic hemorrhagic transformation was about 13%, whereas one-third of the patients attained a favorable mRs outcome (0–2). Factors such as advanced age, higher NIH Stroke Scale (NIHSS) scores, and prolonged hospital stays were associated with poorer outcomes. Conversely, the absence of hypertension and atrial fibrillation, along with higher TICI scores, significantly increased the likelihood of favorable outcomes, with P values of 0.015, 0.0490, and 0.025, respectively. Conclusions: In our study, the examined patients who underwent EVT were younger in age, with clinical and radiological outcomes showing some degree of comparability to other publications. Notably, the absence of hypertension and atrial fibrillation, combined with a higher TICI score, was associated with an increased likelihood of achieving a favorable outcome.

Abstract The magmatic complex along the Marsa Alam-Idfu transect, Central-Eastern Desert of Egypt, represents the northern segment of the Arabian–Nubian Shield (ANS), which developed within the framework of the East African Orogen. The basement rocks of the Arabian-Nubian Shield have been developed through three distinct phases of magmatic activity: the island-arc, the syn-orogenic, and the post-orogenic phases. Transitioning of the magmatic phases from the syn-orogenic to the post-orogenic, identifies changing the tectonic regime from a compressional to an extensional setting. The scarcity of comprehensive regional geochronological data that rely on precise isochron methods, such as the zircon U-Pb technique, could limit the comprehensive understanding of this region’s geological and tectonic history. That would raise a number of uncertainties ranging from the timing of the different magmatic activities and timing of changes in the tectonic regime to the existence of the pre-Pan-African crust in the CED. Our study provides new insights into the aforementioned uncertainties through zircon U-Pb dating of different rock units along the Marsa Alam-Idfu transect, CED, Egypt. The resulting ages ranged from 729 ± 3 Ma to 570 ± 2 Ma, constraining the temporal evolution of the ANS in the studied region into (1) the island-arc phase, represented by a metamorphic sample with an age of 729 ± 3 Ma. (2) the syn-orogenic phase, represented by calc-alkaline and alkaline granitic samples with ages ranging from 699 ± 4 Ma to 646 ± 2 Ma. These two phases indicate initiation of the compressional subduction regime in the CED since 729 ± 3 Ma and being dominated till 646 ± 2 Ma. (3) the post-orogenic phase, represented by metavolcanics, volcanic rocks, and alkaline plutonic samples with ages ranging from 623 ± 3 Ma to 570 ± 2 Ma. This phase suggests dominance of the compressional-to-extensional tectonic transition setting from 623 ± 3 Ma to 600 ± 1 Ma along with the Dokhan volcanism and activation of post-collision tensional regime activated at 582 ± 3 Ma. Our findings discourage the proposed dominance of the island-arc and syn-orogenic phases in the CED and the classical restriction of older magmatic activity to calc-alkaline granitic rocks and younger magmatic activity to alkaline granitic rocks. Additionally, we identified evidence of local magmatic sources by dating five grains with Mesoproterozoic (pre-Arabian–Nubian Shield) xenocrysts with ages ranging from 1549 ± 4 to 1095 ± 25 Ma.

Abstract A novel method for discovery of adulteration in edible oil is proposed based on concept of refractive index and electronic sensors. The research work focusses on two distinct methodologies like employing datasets and implementing a fumigation technique that integrates real-time hardware for testing Edible oil Impurities. In the first method, the dataset taken into consideration contains spectral data collected using Advanced ATR-MIR Spectroscopy for pure oil and various levels of adulteration with Vegetable oil. Each and every edible oil has a certain value of refractive index. When such oils are contemned in a change adding adulterants, the value of its refractive indices also changes. This value of refractive index serves as a feature for testing the oil and helps us in detecting the adulteration. If Oil is adulterated with vegetable oils, the refractive index will be lower and with animal fats, the refractive index will be higher than that of pure Oil. While in Fumigation Method a hardware module is develop in which adulterated & pure oil samples are heated at 40–50 °C for 4.66 min and the volatiles that are generated by varying gas concentrations are forcefully passed through to the MEMS Gas Sensor-MISC-2714 and Multichannel Gas sensor. The conductance of the sensors changes according to the gases sensed by the sensors contributes to features extraction. The conductance value serves as a feature for the classifier to determine whether the sample is highly, moderately, or lowly contaminated. Thus, in proposed methods we use different algorithms based on machine learning like KNN, Random Forest, CATBOOST and XGBOOST to accurately reveal the adulteration. Amongst all the applied algorithm Random Forest (RF) Classifier & XGBOOST algorithm outperform well and gives 100% accuracy. The proposed work is used for identifying food adulteration in edible food products which helps us to feed Society with high-quality food.

Abstract The Indian spot-billed duck, Anas poecilorhyncha is a large dabbling and non-migratory breeding bird. The identification and phylogenetic relationship of A. poecilorhyncha remain uncertain due to the presence of overlapping meristic characters and hybridization with closely related species. Molecular data aids when there are challenges in morphological identification. However, genetic characterization of A. poecilorhyncha has been paid less attention. Apart from their functional and physiological role, mitochondrial genome can also be used for various purposes, including species identification, phylogenetic analysis, understanding the domestication history of species etc. Therefore, the present study aimed to sequence the mitochondrial genome of A. poecilorhyncha and its closely related domestic species A. platyrhynchos (mallard duck) to understand their mitochondrial genome structure and phylogenetic relationships. The length of mitochondrial genome of A. poecilorhyncha and A. platyrhynchos was 16,608 and 16,604 bp respectively. Mitochondrial genome contained 37 genes and a non-coding control region. Overall, the characteristics of mitochondrial genome of both species were found to be conserved. The phylogenetic tree exhibited seven major clades (A to G) with a high bootstrap support. Notably, the Indian A. poecilorhyncha population formed a distinct clade (C) whereas the A. poecilorhyncha that were probably sampled from China grouped along with A. zonorhyncha (clade B). Besides, one of the A. poecilorhyncha probably sampled from China was placed in the clade A, which predominantly consisted of A. platyrhynchos. It suggests that Indian A. poecilorhyncha population is genetically different from Chinese A. poecilorhyncha population. Further, it sheds light on the importance of conducting a comprehensive phylogenetic study on these species. The newly sequenced mitochondrial genome of A. poecilorhyncha and A. platyrhynchos would be useful not only to have a better understanding of the phylogeny and evolution of Anas species but also to help in the conservation of A. poecilorhyncha which is under constant threat from rapid urbanization, interspecific hybridization and other human activities.

Abstract Stroke prediction remains a critical area of research in healthcare, aiming to enhance early intervention and patient care strategies. This study investigates the efficacy of machine learning techniques, particularly principal component analysis (PCA) and a stacking ensemble method, for predicting stroke occurrences based on demographic, clinical, and lifestyle factors. We systematically varied PCA components and implemented a stacking model comprising random forest, decision tree, and K-nearest neighbors (KNN).Our findings demonstrate that setting PCA components to 16 optimally enhanced predictive accuracy, achieving a remarkable 98.6% accuracy in stroke prediction. Evaluation metrics underscored the robustness of our approach in handling class imbalance and improving model performance, also comparative analyses against traditional machine learning algorithms such as SVM, logistic regression, and Naive Bayes highlighted the superiority of our proposed method.

Abstract Ovarian cancer is a formidable health challenge that demands accurate and timely survival predictions to guide clinical interventions. Existing methods, while commendable, suffer from limitations in harnessing the temporal evolution of patient data and capturing intricate interdependencies among different data elements. In this paper, we present a novel methodology which combines Temporal Analysis and Graph Neural Networks (GNNs) to significantly enhance ovarian cancer survival rate predictions. The shortcomings of current processes originate from their disability to correctly seize the complex interactions amongst diverse scientific information units in addition to the dynamic modifications that arise in a affected person`s nation over time. By combining temporal information evaluation and GNNs, our cautioned approach overcomes those drawbacks and, whilst as compared to preceding methods, yields a noteworthy 8.3% benefit in precision, 4.9% more accuracy, 5.5% more advantageous recall, and a considerable 2.9% reduction in prediction latency. Abstract Neurodegenerative diseases (NDs) such as Alzheimer disease (AD) and Parkinson disease (PD) are group of diseases affecting the central nervous system (CNS) characterized by progressive neurodegenerations and cognitive impairment. Findings from different studies highlighted the beneficial and detrimental effects of serum uric acid on the development and progression of NDs. Therefore, this mini‐review aims to discuss the beneficial and detrimental effects of uric on NDs. The neuroprotective effect of uric acid is mainly related to the antioxidant effect of uric acid which alleviates oxidative stress‐induced neurodegeneration in AD and PD. However, long‐term effect of hyperuricemia prompts for the development and progression of cognitive impairment. Hyperuricemia is associated with cognitive impairment and dementia, and gout increases dementia risk. In addition, hyperuricemia can cause cerebral vascular injury which is a risk factor for vascular dementia and cognitive impairment. Taken together, the relationship between uric acid and NDs risk remains conflicting. Hence, preclinical and clinical studies are indicated in this regard.

Abstract Nuclear receptors (NRs) function as crucial transcription factors in orchestrating essential functions within the realms of development, host defense, and homeostasis of body. NRs have garnered increased attention due to their potential as therapeutic targets, with drugs directed at NRs demonstrating significant efficacy in impeding chronic disease progression. Consequently, these pharmacological agents hold promise for the treatment and management of various diseases. Accumulating evidence emphasizes the regulatory role of exosome-derived microRNAs (miRNAs) in chronic inflammation, disease progression, and therapy resistance, primarily by modulating transcription factors, particularly NRs. By exploiting inflammatory pathways such as protein kinase B (Akt)/mammalian target of rapamycin (mTOR), nuclear factor kappa-B (NF-κB), signal transducer and activator of transcription 3 (STAT3), and Wnt/β-catenin signaling, exosomes and NRs play a pivotal role in the panorama of development, physiology, and pathology. The internalization of exosomes modulates NRs and initiates diverse autocrine or paracrine signaling cascades, influencing various processes in recipient cells such as survival, proliferation, differentiation, metabolism, and cellular defense mechanisms. This comprehensive review meticulously examines the involvement of exosome-mediated NR regulation in the pathogenesis of chronic ailments, including atherosclerosis, cancer, diabetes, liver diseases, and respiratory conditions. Additionally, it elucidates the molecular intricacies of exosome-mediated communication between host and recipient cells via NRs, leading to immunomodulation. Furthermore, it outlines the implications of exosome-modulated NR pathways in the prophylaxis of chronic inflammation, delineates current limitations, and provides insights into future perspectives. This review also presents existing evidence on the role of exosomes and their components in the emergence of therapeutic resistance.

Abstract Hydroxamic acids represent a group of weak organic acids, both naturally occurring and synthetically derived, characterized by the general formula RC(= O)N(R’OH). In this study, we investigated the binding behavior of N-m-tolyl-4-chlorophenoxyaceto hydroxamic acid with calf thymus DNA (ct-DNA) and torula yeast RNA (t-RNA) through a combination of techniques including UV–visible spectroscopy, fluorescence emission analysis, viscometry, and computational simulations using AutoDock4 software. Our findings reveal that the mode of binding between the compound and the nucleic acids is consistent with intercalation. Competitive binding experiments demonstrated that the complex competes effectively with ethidium bromide (EB) for binding to ct-DNA/t-RNA, displacing EB from its binding sites. Additionally, the introduction of the compound into the DNA-EB system resulted in a quenching of fluorescence emission peaks. Analysis of absorption spectra indicated a red shift and hypochromic shift when the compound interacted with DNA, further supporting the intercalative binding mode. The calculated binding constant (Kb) value for the compound is 6.62 × 104 M−1 and 5.40 × 103 M−1 indicating a strong interaction with ct-DNA and t-RNA respectively. We determined the Stern–Volmer constants for ct-DNA and t-RNA as 9.96 × 104 M−1 and 8.13 × 105 M−1, respectively. The binding free energy values for ct-DNA/t-RNA were calculated to be − 3.741 × 107 and − 5.425 × 108 kcal/mol, respectively. Viscometric studies corroborated the UV results, showing a continuous increase in relative viscosity of ct-DNA/t-RNA solutions with the addition of the optimal hydroxamic acid concentration. Furthermore, we assessed the antioxidant activity of the compound using DPPH-radical scavenging and β-carotene linoleic acid assays. Gel electrophoresis results demonstrated the compound's remarkable efficacy in preventing DNA damage. Collectively, all experimental evidence supports the conclusion that N-m-tolyl-4-chlorophenoxyaceto hydroxamic acid binds to ct-DNA/t-RNA through an intercalative mechanism, which is consistent with our molecular docking simulations.

Abstract H. pylori infects approximately 50% of the world’s population that causes chronic gastritis, and may lead to peptic ulcer disease (PUD). H. pylori-induced chronic infections are associated with gastric adenocarcinoma and low-grade gastric lymphoma. In Egypt, H. pylori strains are widespread and became resistant to antimicrobial agents, thus advanced typing methods are needed to differentiate infectious strains that are resistant to antibiotics, and therefore earlier prognosis and infection control. The main objectives were (i) to determine susceptibility of infectious H. pylori strains to some antimicrobial agents that are currently used in eradication therapy in Egypt; (ii) to identify diverse strains commonly detected in the gastrointestinal (GIT) endoscopy units in Egypt through phenotypic and genotypic analyses. In this observational study we isolated 167 isolates from 232 gastric biopsies (antrum and corpus) of patients who were admitted to the upper GIT endoscopy units in five governmental Egyptian hospitals. Antimicrobial susceptibility patterns were investigated using Kirby Bauer disc diffusion and agar dilution Minimum Inhibitory Concentrations (MICs) methods. Phenotypic characterization was based on biotyping and antibiogram typing techniques. Genotypic characterization was carried out using PCR-based Restriction Fragment Length Polymorphism (RFLP) and Enterobacterial Repetitive Intergenic Consensus (ERIC)-PCR analyses. H. pylori isolates were highly resistant to diverse antimicrobial agents including Metronidazole, Fluoroquinolones, Macrolides, Amoxycillin, Tetracycline and Gentamicin. Two factors contributed to the increased resistance of H. pylori to the conventional therapy seen in Egypt: (i) Metronidazole and Amoxycillin are inexpensive and available drugs being abused by patients; (ii) the regional prescribing practice of Macrolids commonly used to treat upper respiratory and urinary tract infections. Five different biotypes were identified depending on the ability of the isolates to synthesize different enzymes. Nine antibiogram types were identified. PCR-RFLP analysis revealed fifteen different fingerprints while ERIC-PCR revealed 22 fingerprints. Biotyping alone or in combination with antibiogram typing are highly useful molecular tools in the prognosis of strain relatedness. PCR-RFLP and ERIC-PCR acquired good discriminatory power for identifying H. pylori infectious sub-types.

The AKT1 oncogene is related to various cancers due to its critical role in the PIC3CA/AKT1 pathway; however, most of the studies screened the hotspot mutation AKT1 (E17K) with various incidences. Low frequency or lack of AKT1 (E17K) mutation was reported in prostate cancer (PC) patients. This study aims to explore genetic alterations in the AKT1 PH domain by extending the sequencing to include AKT1 gene exons 3 and 4. Genomic DNA was extracted from 84 Formalin-Fixed Paraffin-Embedded samples of PC patients in Jordan, and then subjected to PCR and sequencing for the targeted exons. This study revealed the presence of two novel mutations (N53Y and Q59K) and a high frequency of mutations in exon 4, with a lack of mutations in the E17K hotspot. Nine missense and two synonymous mutations were detected in exon 4 (Phe27Tyr, Phe27Leu, Ala58Thr, Ser56Phe, Arg41Trp, Phe35Leu, Asp32Glu, Phe35Tyr, and Gln43Lys) and (Ser56 and Glu40), respectively. Two synonymous mutations were detected in exon 3 (Leu12 and Ser2). It is concluded that there is a high frequency of AKT1 mutation in PC patients in Jordan with two novel missense mutations in the Pleckstrin homology (PH) domain. E17K hotspot mutation was not detected in any tested samples, which underlined the significant role of mutations in other AKT1 exons in PC development.

Cinnamon is a plant with significant medicinal value and used extensively as a spice, flavoring, and fragrance ingredient. Phytochemical characterization via HPLC, antimicrobial activity against various microorganisms, anti-diabetic activity via α-amylase and α-glucosidase assessment, antioxidant activity via 2,2-diphenyl-1-picryl-hydrazyl-hydrate, total antioxidant capacity and Ferric reducing Antioxidant Power methods, besides anticancer activity against MCF-7 (human breast cancer cells) and WI-38 cells (human fetal lung fibroblast cells) of cinnamon bark extract were detected. A promising antimicrobial action towards Enterococcus faecalis, Staphylococcus aureus, Klebsiella pneumoniae, Salmonella typhi, Candida albicans, and Mucor circinelloides with inhibition zones of 25, 23, 18, 19, 29, and 16 mm, respectively was recorded. Cinnamon bark has IC50 of 5.01 and 2.58 µg/mL compared to standard acrobose with their IC50 values 4.32 and 1.99 µg/mL, for α-amylase and α-glucosidase inhibition, respectively. Cinnamon bark extract have IC50 of 77.39 ± 0.84 and 162.67 ± 0.28 μg/mL against MCF-7, and WI-38 cell lines, respectively. The protective effect of cinnamon in accelerating the apoptosis of MCF-7 cells has been verified by flow cytometric evaluation employing Annexin-V and cell cycle kits, besides increasing the amounts of malondialdehyde, hydrogen peroxide and nitric oxide with decreasing glutathione and catalase levels.

Abstract Biopolymers embedded with nanoparticles of metal oxides (MOs) demonstrate a wide range of bio-functions. Chitosan-incorporated MOs are an interesting class of support matrices for enhancing the biological function, compared to other support matrices. Therefore, the importance of this study lies in exploiting chitosan as a carrier not of one metal as in previous studies, but of two metals in the form of a nanocomposite to carry out several biological functions. The coprecipitation approach was employed to synthesize chitosan/Fe2O3/ZnO-nanocomposite in the present research. The characterization of chitosan/Fe2O3/ZnO-nanocomposite was performed to find out the morphology and dispersion properties of chitosan/Fe2O3/ZnO-nanocomposite. The X-ray diffraction (XRD) investigation revealed that these were crystalline. Fourier transforms infrared (FTIR) spectrum bands were viewed at 400/cm and 900/cm, due to the stretching vibration of Fe and Zn oxygen bond. TEM showed that chitosan/Fe2O3/ZnO-nanocomposite was of 20–95 nm in size. chitosan/Fe2O3/ZnO-nanocomposite exhibited inhibitory potential against Staphylococcus aureus, Bacillus subtilis, Escherichia coli, and Candida albicans with inhibition zones of 25 ± 0.1, 28 ± 0.2, 27 ± 0.1, and 27 ± 0.2 mm, respectively while didn’t inhibited Aspergillus niger. MIC value of nanocomposite was 15.62 ± 0.33 µg/mL for C. albicans, B. subtilis and E. coli, while it was 62.50 ± 0.66 µg/mL for Pseudomonas aeruginosa. Ranged values of nanocomposite MBC (15.62 ± 0.33 to 125 ± 1 µg/mL) were attributed to all tested bacteria. Different concentrations of chitosan/Fe2O3/ZnO-nanocomposite MBC (25, 50, and 75%) reflected anti-biofilm activity against E. coli (85.0, 93.2, and 96.0%), B. subtilis (84.88, 92.21, and 96.99%), S. aureus 81.64, 90.52, and 94.64%) and P. aurogenosa (90.11, 94.43, and 98.24%), respectively. The differences in the levels of antimicrobial activities may depend on the type of examined microbes. Antioxidant activity of chitosan/Fe2O3/ZnO-nanocomposite was recorded with excellent IC50 values of 16.06 and 32.6 µg/mL using DPPH and ABTS scavenging, respectively. Wound heal by chitosan/Fe2O3/ZnO-nanocomposite was achieved with 100% compared to the untreated cells (76.75% of wound closer). The cytotoxicity outcomes showed that the IC50 of the chitosan/Fe2O3/ZnO-nanocomposite was 564.32 ± 1.46 µg/mL normal WI-38 cells. Based on the achieved findings, the chitosan/Fe2O3/ZnO-nanocomposite is a very promising agent for perform pharmacological activities.

Abstract Background PPHN is a common cause of neonatal respiratory failure and is still a serious condition and associated with high mortality. Objectives To compare the demographic variables, clinical characteristics, and treatment outcomes in neonates with PHHN who underwent ECMO and survived compared to neonates with PHHN who underwent ECMO and died. Methods We adhered to the Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) guideline and searched ProQuest, Medline, Embase, PubMed, CINAHL, Wiley online library, Scopus and Nature for studies on the development of PPHN in neonates who underwent ECMO, published from January 1, 2010 to May 31, 2023, with English language restriction. Results Of the 5689 papers that were identified, 134 articles were included in the systematic review. Studies involving 1814 neonates with PPHN who were placed on ECMO were analyzed (1218 survived and 594 died). Neonates in the PPHN group who died had lower proportion of normal spontaneous vaginal delivery (6.4% vs 1.8%; p value > 0.05) and lower Apgar scores at 1 min and 5 min [i.e., low Apgar score: 1.5% vs 0.5%, moderately abnormal Apgar score: 10.3% vs 1.2% and reassuring Apgar score: 4% vs 2.3%; p value = 0.039] compared to those who survived. Neonates who had PPHN and died had higher proportion of medical comorbidities such as omphalocele (0.7% vs 4.7%), systemic hypotension (1% vs 2.5%), infection with Herpes simplex virus (0.4% vs 2.2%) or Bordetella pertussis (0.7% vs 2%); p = 0.042. Neonates with PPHN in the death group were more likely to present due to congenital diaphragmatic hernia (25.5% vs 47.3%), neonatal respiratory distress syndrome (4.2% vs 13.5%), meconium aspiration syndrome (8% vs 12.1%), pneumonia (1.6% vs 8.4%), sepsis (1.5% vs 8.2%) and alveolar capillary dysplasia with misalignment of pulmonary veins (0.1% vs 4.4%); p = 0.019. Neonates with PPHN who died needed a longer median time of mechanical ventilation (15 days, IQR 10 to 27 vs. 10 days, IQR 7 to 28; p = 0.024) and ECMO use (9.2 days, IQR 3.9 to 13.5 vs. 6 days, IQR 3 to 12.5; p = 0.033), and a shorter median duration of hospital stay (23 days, IQR 12.5 to 46 vs. 58.5 days, IQR 28.2 to 60.7; p = 0.000) compared to the neonates with PPHN who survived. ECMO-related complications such as chylothorax (1% vs 2.7%), intracranial bleeding (1.2% vs 1.7%) and catheter-related infections (0% vs 0.3%) were more frequent in the group of neonates with PPHN who died (p = 0.031). Conclusion ECMO in the neonates with PPHN who failed supportive cardiorespiratory care and conventional therapies has been successfully utilized with a neonatal survival rate of 67.1%. Mortality in neonates with PPHN who underwent ECMO was highest in cases born via the caesarean delivery mode or neonates who had lower Apgar scores at birth. Fatality rate in neonates with PPHN who underwent ECMO was the highest in patients with higher rate of specific medical comorbidities (omphalocele, systemic hypotension and infection with Herpes simplex virus or Bordetella pertussis) or cases who had PPHN due to higher rate of specific etiologies (congenital diaphragmatic hernia, neonatal respiratory distress syndrome and meconium aspiration syndrome). Neonates with PPHN who died may need a longer time of mechanical ventilation and ECMO use and a shorter duration of hospital stay; and may experience higher frequency of ECMO-related complications (chylothorax, intracranial bleeding and catheter-related infections) in comparison with the neonates with PPHN who survived.

Abstract Abu Marawat area in the Central Eastern Desert of Egypt is a very promising mineralization district located in the Golden Triangle area. The current study provides an integrated approach from multisource datasets including; remote sensing, airborne geophysical spectrometry and magnetic data supported by field studies and spectroscopic analyses for delineating potential mineralization localities. Several remote sensing techniques were adopted including; Band Ratios, Relative Band Depth, Mineralogical Indices, Spectral Angle Mapper, and Constrained Energy Minimization. These techniques showed that the alteration mineral assemblage is mainly, kaolinite, sericite, and iron oxides, with less abundant chlorite, epidote, and carbonates. In addition, the radiometry data were processed to map the localities with the highest possibility of potassic alteration abundance by integrating the potassium distribution, K/eTh ratio, and the F-parameter maps. The surface and subsurface linear structural features were also mapped using Digital Elevation Model (DEM) and aeromagnetic data, respectively. The surface linear structures were found exhibiting E-W and NE-SW trends, while, the subsurface structures showed dominant NW–SE trend. All the depicted fault trends match well with the local and regional geological and tectonic setting of the study area suggesting structural control on the mineralization in this area. Integration between the results obtained from both the remote sensing and the geophysical data was conducted by a GIS weighted overlay model. The obtained mineralization potentiality map highlights eight potential localities for mineralization. The accuracy of the adopted methodology was demonstrated through fieldwork and spectral analyses; several alteration indicators were observed, including quartz veins, iron oxides, kaolinite, malachite, montmorillonite, chlorite, talc, and sericite alteration indicator minerals. The adopted remote sensing-geophysical approach showed being very effective for mapping the hydrothermal gold-related alteration zones, and is recommended for other similar investigations.

BACKGROUND: Research has showed a link between patients with methamphetamine dependence and the risk of impulsiveness, aggression, and the risk of suicide. But, this link has not been studied in patients abusing methamphetamine, and it is unknown how impulsiveness, aggression, and the risk of suicide affect them. MATERIALS AND METHODS: This cross-sectional study included 130 adult patients diagnosed with the disorder of the use of amphetamine, methamphetamine, cannabinoids, alcohol, other substances, and polysubstance admitted in the Addiction Department for Mental Health. Participants were interviewed for detailed psychiatric history using a structured questionnaire comprising of structured clinical interview for diagnosis I, Arabic version of the Barratt Impulsiveness Scale-11 (BIS), Beck Scale for Suicidal Ideation for the evaluation of suicidal ideation and behavior, and the Aggression and Hostility scale for adolescents and youth. SPSS was used for data analysis; Initial analysis included descriptive statistics: frequencies and percentages for categorical variables and mean and standard deviation for continuous variables. Chi-square test/Fisher’s exact test assessed for association between categorical variables, whereas one-way analysis of variance (ANOVA)/ Kruskal–Wallis test was used to compare continuous variables. RESULTS: Patients who used methamphetamine either alone (23%) or with polysubstance (42.6%) were associated with higher suicidal risk than patients who did use other substances than methamphetamine (36.1%). A comparison of the three groups on impulsivity, showed significant difference regarding total scores, motor preservation, and non-planning self-control. No significant differences found between three groups on the aggression scores. CONCLUSIONS: There was a higher rate of impulsivity and suicidal risk in patients with methamphetamine dependence in comparison to dependence on other substances, while there was no difference with regard to aggression between patients with methamphetamine dependence and those dependent on other substances. This finding raises the issue of methamphetamine use disorder as a new substance of dependence.

Background and Aim: Brucellosis is a highly contagious, neglected zoonotic disease of major importance worldwide. The disease is endemic in many countries, burdening healthcare systems and the livestock industry and representing a persistent public health concern in these countries. Brucellosis is considered an important occupational hazard for livestock workers. Limited studies have investigated human brucellosis in Saudi Arabia. Therefore, this study aimed to estimate the prevalence of brucellosis among employees of high-risk brucellosis professions, including veterinarians, animal herders, and abattoir workers in Madinah, Saudi Arabia, and to determine the associated risk factors. Materials and Methods: A cross-sectional study was conducted in Madinah, Saudi Arabia, during the period of January–March 2023. Ninety blood samples were collected from individuals occupationally at risk of exposure to Brucella infections. Serum samples were examined for immunoglobulins (Ig)M and IgG antibodies against Brucella using an indirect enzyme-linked immunosorbent assay. Before sample collection, a predesigned online questionnaire was used to collect the participants’ sociodemographic characteristics and the probable risk factors for human brucellosis. A Chi-square test was used to compare the differences among groups; p < 0.05 were considered statistically significant. Results: Among the 90 participants among the high-risk individuals, Brucella IgM and IgG seropositivity were found in 8 (8.8%) and 11 (12.12%) cases, respectively. IgM mono antibody positivity was observed in 4 (4.44%) and 7 (7.77%) of the study population who tested positive for IgG only. Dual positivity for IgM and IgG antibodies was observed in 4 (4.44%) participants. No significant association was determined between seropositivity and age, urbanicity, education, occupation, and duration of exposure (p > 0.05). Conclusion: Brucellosis is a high-risk occupational disease among workers with close contact with livestock. This study demonstrates that the seroprevalence of brucellosis among occupationally high-risk individuals in Madinah, Saudi Arabia, is relatively low compared to other countries in the region. Nevertheless, educational programs should be implemented to improve knowledge regarding brucellosis, particularly among high-risk individuals.

Plants represent a great source of medicines, and for their components to be discovered, extraction processes must be developed, especially methods based on green technology. Supercritical fluid extraction (SFE) was employed as a green method for Moringa oleifera extraction in the present investigation. The maximum yield of extraction was obtained at 25 MPa. Moreover, the extraction at 25 MPa induced the release of various phenols and flavonoids, as analyzed via high-performance liquid chromatography. The investigation revealed the concentrations of chlorogenic, gallic, rosmarinic, and coumaric acids to be 150.59, 89.90, 44.75, and 29.41 µg/mL, respectively at 25 MPa. However, their concentrations were 0.73, 1.53, 0.24, and 0.04 µg/mL, respectively at 15 MPa; vs. 4.73, 2.62, 1.06, and 0.50 at 35 MPa, respectively. Totals of saponin, flavonoid, phenolic, tannins, and alkaloid were recorded in maximum yield at 25 MPa. Moringa oleifera extracted at 35 MPa reflected highest inhibition zones of 27 ± 0.1, 30 ± 0.2, and 30 ± 0.1 mm against C. glabrata, C. tropicalis, and C. albicans, correspondingly. α-Amylase and α-glucosidase activities were greatly suppressed by the M. oleifera extract at 25 MPa with less IC50 (12.97 µg/mL and 6.0 µg/mL), than the IC50 (53.46 and 22.02 µg/mL) at 15 MPa, compared with acarbose IC50 (5.52 and 2.64 µg/mL), correspondingly.

Efficacies of plant metabolites are known to be dependent on their extraction methods. Yields and compositions of phytoconstituents in the extract were evaluated following supercritical fluid extraction (SFE) of Vitex agnus-castus leaves, static extraction times (SET) for 30 min, subsequently dynamic extraction time (DET) for 30 min (condition A) and SET for 0 min followed by DET for 60 min (condition B). The extract exposed to condition B gave an extraction yield of 0.169 g compared to 0.115 g for condition A. High-performance liquid chromatography analysis revealed compounds including cinnamic acid, kaempferol, ferulic acid, rutin, and caffeic acid, in high concentrations in the extract exposed to condition B. Staphylococcus aureus, Pseudomonas aeruginosa, Klebsiella pneumoniae, Enterococcus faecalis, and Candida albicans were more affected by the condition B with 32 ± 0.1, 20 ± 0.2, 32 ± 0.2, 35 ± 0.2, and 40 ± 0.1 mm inhibition zones, respectively. Less MIC and MBC were noticed of the exposed extract to condition B than to condition A against C. albicans and bacteria. The IC50 of the extract exposed to condition B was high against ovarian tumor cells. Presently the efficacy of the exposed extract to condition B for wound healing process was documented.

Green routes for the bio-designing of bicomponent nanocomposites and their utilizations have attracted many investigators. Bio-designing of CuO@ZnO nanocomposites was performed using spent mushroom substrate (SMS). Ultraviolet-spectrophotometry, transmission electron microscopy, Fourier transform infrared (FT-IR), and energy dispersive X-ray (EDX), besides X-ray diffraction (XRD) were exploited to characterize the synthesized CuO@ZnO. The dimensions of CuO@ZnO nanocomposites ranged from 31.4 and 95.9 nm. Both FT-IR and EDX analyses displayed the presence of some organic constituents from the SMS that joined to the surface of the fabricated CuO@ZnO nanocomposite. CuO@ZnO nanocomposite succeeded in inhibiting Candida albicans with an inhibition zone of 33.5 ± 2 mm. C. albicans biofilm was affected by CuO@ZnO nanocomposite with biofilm inhibition of 25.08, 68.70, and 88.56% at 25, 50, and 75% of minimum inhibitory concentration, respectively. Molecular docking studies showed substantial binding affinities, as well as common hydrogen bonds. Optimum binding sites for CuO and ZnO nanoparticles were found to have binding affinities of interactions with 4YDE, 3DRA, and 1EAG proteins of C. albicans, resulting in, respectively, -2.7942, -3.30097, and -2.52129 kcal/mol, and -3.78244, -4.6029, and -4.1352 kcal/mol values. The findings suggest that CuO@ZnO nanocomposite can effectively suppress C. albicans growth.

Abstract Hip fractures are common orthopedic injuries that have significant impacts on patients and healthcare systems. Previous studies have shown varying outcomes for hip fracture management in different settings, with diverse postoperative outcomes and complications. While teaching hospital settings have been investigated, no studies have specifically examined hip fracture outcomes in teaching hospitals in Jordan or the broader Middle East region. Therefore, the aim of this study was to investigate this important outcome. A cohort comprising 1268 patients who underwent hip fracture fixation from 2017 to 2020 was analyzed for nine distinct outcomes. These outcomes encompassed time to surgery, ICU admissions, perioperative hemoglobin levels, length of hospital stay, readmission rates, revision procedures, and mortality rates at three time points: in-hospital, at 6-months, and at 1-year post-surgery. Abstract Porous asphalt mixture is conventional hot mix asphalt (HMA) with substantially decreased fines, which produces an open-graded mixture that enables the water to flow through an interconnected void space. Porous asphalt is a permeable system that has a lot of benefits. However, because of its open structure, the durability of this mixture decreases, and both its stability and resilient modulus are much lower compared to the dense conventional asphalt mixtures. Also, the high void percentage may lead to an increase in the draindown proportion. Fibers (cellulose or mineral) and polymer-modified binders are recommended for porous asphalt mixtures, especially in hot and moderate climates. The objective of this study is to improve the porous asphalt mixture's performance by using ethylene–vinyl acetate (EVA) polymer-modified bitumen. Two types of fibers (cellulose fibers and glass wool fibers) were used, separately to determine the control mixture. Four different proportions of EVA polymer were added to the bitumen (1%, 2%, 3%, and 4%) and Scanning Electron Microscopy (SEM) was used for better investigating of the bitumen microstructure, then The Marshall mix design was used to determine the optimum EVA content (OEC) for the porous asphalt mixture. Several performance tests were conducted to investigate the characteristics of the porous asphalt mixture, such as the infiltration rate, binder draindown, the wheel track and the cantabro abrasion tests. The findings of the study conclude that the addition of EVA polymer to the porous asphalt mixtures enhances the performance as it increases stability by 20.8% and the infiltration rate by 20.6%. It decreases binder draindown proportion by 33.3%, cantabro abrasion loss by 25.1% and the rut depth at 5,000 cycles and 10,000 cycles by 29.8% and 19.7%, respectively.

This study discusses the feasibility of diversifying the scope of application of lead-free Sr3ZBr3 (Z = As, Sb) as promising materials for their enhanced electronic, mechanical, optical, thermodynamic, and thermoelectric properties using first-principles calculation based on Density Functional Theory (DFT). Both compounds have cubic crystal structures, and both materials' dynamic stability is validated by the lack of negative frequencies in their phonon dispersion spectra. Besides ground state physical characteristics, we also examined the impact of hydrostatic pressure (0–21 GPa) on electronic, mechanical, and optical properties. The lattice parameters exhibit a linear decrease from 6.27 to 5.61 Å for Sr3AsBr3 and 6.45 to 5.71 Å for Sr3SbBr3 under increasing pressure, attributed to bond length reduction, which alters their physical properties. Initially observed direct band gap (Γ-Γ) of Sr3AsBr3 and Sr3SbBr3 were 2.35 and 2.30 eV using modified Becke-Johnson (mBJ) functional which reduces to 1.15 and 0.82 eV as the compounds go from 0 to 21 GPa pressure. This study reveals enhanced optical properties under pressure, with broader spectral response, increased sensitivity, and improved dielectric constant. Optical absorption and conductivity also shift toward lower-energy photons. The investigation further shows that the compounds show brittle to ductile transition under high pressure. Their ductility and anisotropy nature are further enhanced with pressure along with other mechanical features. The thermodynamic properties indicate their ability to withstand and perform well at high temperatures and pressures. Lastly, the thermoelectric properties of Sr3ZBr3 (Z = As, Sb) were assessed through electrical and thermal conductivity, Seebeck coefficient, power factor (PF), and figure of merit (ZT) as a function of temperature. Both compounds exhibit high PF and near-unity ZT. These findings underscore their potential as strong candidates for optoelectronic and thermoelectric devices, owing to their direct bandgap and exceptional properties.

This study presents a comprehensive account of geochemical data on the granitoids of Ambaji-Sendra Terrain and associated mafic dykes. These granitoids, interpreted as S–type, I-type and A-type, lack a consensual interpretation probably due to the study of only those rocks which are exposed in Sendra region. We identified that these granitoids are A-type (A2- subtype) granites. There are two petrogenetic groups of the granitoids having exclusive crustal and crust-mantle interactive origins which are supported by their mineralogy and structure. The associated dykes also display two distinct geochemical groups one with within-plate character and other with typical arc signatures. Whole body of evidence suggests that magma for the granitoids and dykes was generated during the closure of Delhi Ocean. The simultaneous emplacement of granitoids started during subduction process and culminated after the collision or/ accretion of Delhi arc with eastern crustal block, consequent to the Rodinian assembly mechanism.

In the present study, the antioxidant potential and angiotensin-I converting enzyme (ACE) inhibitory activity of cow's milk paneer supplemented with pastes (10 % of milk) from indigenous date varieties were monitored during storage. The prepared paneer was also evaluated for its physicochemical characteristics, microbiological and sensorial quality during storage period (0–12 days, 5 °C). It was observed that the ESFs of Ajwa paste showed significantly (p < 0.05) higher antioxidant potential and ACE-inhibition than other investigated date pastes. Moreover, all of the investigated paneer showed significantly (p < 0.05) increasing trend concerning sensory evaluation until 9 days of storage. The paneer supplemented with date pastes showed significantly (p < 0.05) higher ferric reducing antioxidant power (FRAP) (3150.70–5776.00 μmol equivalent of Trolox (TE)/g of ethanol soluble fraction (ESF)) and DPPH (1634.70–1645.70 μmol TE/g ESF)/ABTS (83.26–102.43 mg TE /100 g ESF) radical scavenging activity than control paneer. Moreover, paneer (3 days of storage) supplemented with Ajwa paste showed significantly (p < 0.05) high ACE-inhibition (66 %) and lower inhibitory concentration (IC)50 (9 mg/mL) as compared to all other investigated samples. Hence, it may be concluded that the paneer supplemented with date pastes offers considerable antioxidant potential and ACE-inhibitory activity.

Valproic acid and its derivatives are common drugs used in the treatment of epilepsy, a result of CNS disorders. Because the molecular structure of valproic acid, as a fatty acid, does not contain any chromophore groups, its peak response demanded special conditions to be detected in HPLC, ensuring the best precision results. Still, the current approach illustrates simple, validated procedures for conduction. The conducted chromatographic system consists of the BDS Hypersil C8, 150 × 4.6 mm, 5 µm column using a mobile phase of acetonitrile: phosphate buffer (4:6) at a detection wavelength of 215 nm at room temperature. A full method validation study was conducted and approved to ensure precise, repeatable, and accurate results by implementing system suitability parameters. In microbiological terms, a suitability test is used to validate microbiological testing methods, ensuring their efficacy despite potential interference from antimicrobial properties in the tested materials. The HPLC-UV developed and validated analytical method was evaluated, and it was found to be sensitive for use in the detection of low concentrations of methylparaben, propylparaben, and valproate with an optimum run time of six minutes. limit of detection (LODs) were statistically estimated and found to be 2.27 µg/mL, 39.77 ng/mL, and 1.84 µg/mL for methylparaben, propylparaben, and valproate, respectively. Additionally, the method demonstrated a high recovery of methylparaben, propylparaben, and valproate with an excellent closed-accuracy range (98.84% - 101.34%). An excellent regression coefficient (r) of 0.99924, 0.99998, and 0.99997 for methylparaben, valproate, and propylparaben, respectively, was achieved. Assay determination of various pharmaceutical dosage forms in the local market was implemented, yielding admirable results.

Brain tumors, characterized by abnormal cell growth, pose a significant challenge in clinical imaging due to their complex and diverse structures. Early and accurate identification, classification, localization, and segmentation of these tumors are critical to reducing mortality. However, the extensive data generated by MRI scans makes manual segmentation time-consuming and impractical for clinical use. To address these challenges, we propose, a hybrid deep learning model that precisely segmented tumor regions with U-Net to enable YOLOv8s to efficiently detect, classify and localize tumors. The model was trained and validated using The Cancer Imaging Archive (TCIA) dataset, which includes MRI images of brain tumors, and the Cancer Genome Atlas (CGA) low-grade glioma dataset, which includes data from 110 patients with FLAIR aberrant segmentation masks. The proposed hybrid model was evaluated using several performance metrics, including F1 score, specificity, recall, precision, accuracy, and ROC-AUC score. Hybrid proposed performed highly, achieving a precision of 97.8 %, accuracy of 98.6 %, recall of 95.2 %, F-1 score of 96.3 %, specificity of 89.1 %, and ROC-AUC score of 98.5 %. The integration of YOLOv8s and U-Net in Enhanced TumorNet offers a powerful solution for the automated analysis of brain tumors in MRI scans, significantly improving detection and segmentation accuracy. This hybrid approach holds great potential for clinical applications, enhancing the efficiency and effectiveness of brain tumor diagnosis.

IntroductionOne of the most prevalent and recurrent infectious diseases that can range from moderate to fatal is urinary tract infection (UTI). Broad-spectrum antibiotics are the only management strategy for UTIs in ambulators and hospital stays. Due to the ongoing emergence of antibiotic resistance among uropathogens, there is a need for proper selection of antibiotics for empirical therapy against UTIs. This study aimed to compare the etiological profiles and antibiotic susceptibility patterns between diabetic and non-diabetic UTI female patients from the Maternity and Children Hospital in Jeddah, Saudi Arabia.MethodsUrine samples from different age categories of female UTI patients were collected from January 2021 to June 2023. The positive urine cultures with a single pathogen were selected and all bacterial isolates were identified by MALDI-TOF/MS system. Antibiotic susceptibility testing was done using VITEK-2. Our study included 2,245 female patients, of which 1825 (81%) were non-diabetic and 420 (19%) were diabetic.ResultsThe results showed a significant relationship (p = 0.00063) between the average age and the number of diabetic UTI patients. Undoubtedly, urbanization has improved human living conditions. However, it has also altered the natural landscape, leading to negative consequences such as increased Land Surface Temperature (LST) and Urban Heat Island (UHI) due to the expansion of Impervious Surface (IS). Much research has been conducted in other countries on the effects of changing urban landscape structures on LST and UHI formation. However, in Sri Lanka, only a few studies have been available on this topic, primarily concentrating on major cities like Colombo. Impervious Surface is absorbing high amounts of solar energy as well, which accelerates the magnitude of UHI in urbanized areas. Remote Sensing indices such as the Normalized Difference Vegetation Index (NDVI), Normalized Difference Buildup Index (NDBI), UHI, and Environmental Criticality Index (ECI) can effectively be used to quantify the intensity of the UHI phenomenon. This study aimed to investigate the effect of spatiotemporal variations in IS and Green Surface (GS) on UHI, LST, and the environmental criticality in Galle Municipal Council (MC), Sri Lanka employing multi-temporal Landsat-5 and 8 data from four different periods: 1996, 2005, 2014, and 2022. Different geospatial techniques including supervised image classification, Urban-Rural Gradient Zone (URGZ) analysis, grid-based analysis, UHI profiles, and regression analysis were used in the study. The findings revealed that Impervious Surface increased by 42.3% (7.34 km2) while Green Surface had a decline of 22.5% (3.91 km2) during the concerned period. This landscape transition led to a 2.74 C increase in mean surface temperature in the study area, along with a 9.5 C increase in the UHI index during 26 years. The results further revealed that Impervious Surface rapidly developed within 4 km (URGZ1-19) from city center, while Green Surface decreased. Newly built-up areas within the 1.5 km gradient (URGZ1-URGZ8) were more affected by increased LST. A positive correlation was identified between NDBI and LST, especially in the year 2022, with an R2 of 0.457, while NDVI and LST reported a negative R2 of 0.257. The grid-based analysis demonstrated an increasingly positive relationship between mean LST and the fraction of Impervious Surface, highlighting the role of built-up areas in raising LST and UHI in the MC. As a result, very high environmental critical areas have been concentrated in and around high-density Impervious Surface. Thus, it can be predicted that the UHI effect and Environmental Criticality (EC) may increase further in the future. In this context, planning agencies should prioritize green urban planning strategies, such as implementing green belts and urban agriculture in the study area, particularly in and around areas with high LST and high environmental criticality. This approach may help protect the natural environment and sustainably ensure the health of the urban community.

Identifying lithology is crucial for geological exploration, and the adoption of artificial intelligence is progressively becoming a refined approach to automate this process. A key feature of this strategy is leveraging population search algorithms to fine-tune hyperparameters, thus boosting prediction accuracy. Notably, Bayesian optimization has been applied for the first time to select the most effective learning parameters for artificial neural network classifiers used for lithology identification. This technique utilizes the capability of Bayesian optimization to utilize past classification outcomes to enhance the lithology models performance based on physical parameters calculated from well log data. In a comparison of artificial neural network architectures, the Bayesian-optimized artificial neural network (BOANN) demonstrably achieved the superior classification accuracy in validation and significantly outperformed a non-optimized wide, bilayer, and tri-layer network configurations, indicating that incorporating Bayesian optimization can significantly advance lithofacies recognition, thus offering a more accurate and intelligent solution for identifying lithology.

Pemphigus is a group of autoimmune intra-epidermal blistering diseases that affect both the skin and mucous membranes. Characteristic histological features of pemphigus include intra-epidermal blisters and acantholysis, which is the loss of connections between epidermal cells. A cross-sectional hospital-based study was conducted in the period between January 2016 and January 2019. A total of 549 cases were selected; 92 cases were clinically diagnosed as pemphigus, and direct immunofluorescence (DIF) was applied for immunoglobulin (Ig) A, IgG, IgM, and C3 to confirm clinical diagnosis. Bullous pemphigoid (BP) was the most common autoimmune skin condition, comprising 29.3% of cases, predominantly affecting females (19.6%). Pemphigus vulgaris (PV) followed with 32.6% of cases, evenly distributed between males and females. Lichen planus pemphigoid accounted for 17.4%, with a higher prevalence in females (13.0%). Other conditions, such as pemphigus gestationis and bullous lupus erythematosus, were rare, each affecting only one gender. Epidermolysis bullosa acquisita was seen only in males, while linear IgA bullous dermatosis was slightly more common in females. Pemphigus foliaceus (PF) and pemphigus vegetans were infrequent, with the first one more common in males and the second one more common in females. Paraneoplastic pemphigus (PNP) and IgA pemphigus had the lowest incidence, with PNP affecting one male and IgA pemphigus equally distributed between genders. The study found PV (32.6%) to be the most common autoimmune blistering disease, followed by BP (29.3%), with females more frequently affected. DIF was effective in confirming clinical diagnoses across various subtypes.

Histone deacetylase 8 (HDAC8) is a member of class I histone deacetylases (HDACs) that catalyzes the deacetylation of both histone and non-histone proteins. Dysregulation and overexpression of HDAC8 are implicated in the development of various complex diseases, including cancer and neurodegenerative disorders. HDAC8 plays a significant role in cancer progression, contributing to cancer cell proliferation, metastasis, immune evasion, and drug resistance. The available HDAC8-targeting inhibitors suffer from poor target engagement and low tolerability, and demonstrate off-target toxicity due to limited selectivity, leading to adverse effects in patients, and thus urging for the identification and development of new molecules. Drug repurposing is a useful strategy for identifying useful drugs for predefined targets which can be exploited here for identifying promising drug molecules against HDAC8. This study involved an integrated virtual screening against HDAC8 using the DrugBank database to identify repurposed drugs capable of inhibiting HDAC8 activity. The process started by selecting the top 10 drug molecules based on their binding affinity. The drug profiling and biological function of selected molecules were then evaluated, showing anti-cancer and anti-neurological properties with a high probability of being active. Interaction analysis revealed crucial binding of radotinib and sertindole molecules with the HDAC8 protein. Both molecules showed higher binding affinity than reference inhibitor droxinostat. The elucidated molecules were further evaluated for 500 ns long-run molecular dynamics (MD) simulation with HDAC8. Structural deviation, compactness, folding behavior, hydrogen bonds analysis, and secondary structure content profiling revealed complex stability formed by HDAC8 and the selected compounds. Principal component analysis and Gibbs free energy calculations strongly recommend that both complexes were highly stable during the simulation. Overall, the results indicate that radotinib and sertindole can be promising candidates as HDAC8-targeting repurposed drugs against cancer and neuropathological conditions.

Background: Dental stem cells with neurosphere-forming abilities are a promising cell source for the treatment of neural diseases and injuries. This scoping review aimed to systematically map the existing literature on dental sphere formation assays and their characteristics associated with neural regeneration potential. Methods: The Web of Science, EMBASE, SCOPUS, and PubMed databases were systematically searched for in vitro, animal, and clinical studies and reviews focusing on stem cells isolated from the oral cavity, subsequently cultured as spheres with neural regeneration potential. Data were extracted and evidence was synthesized according to the predetermined variables in the registered protocol. Results: A total of 35 articles (31 in vitro, 1 combined in vitro and in vivo, and 3 reviews) were included. The predominant method utilized for sphere formation was low-attachment culture. Spheres were characterized using assessment of neural marker expression via confocal microscopy, immunohistochemistry, RT-qPCR, or western blotting. Overall, the synthesized results indicate a lack of in vivo studies investigating the utility of dental neurospheres for neural regeneration, with dental pulp stem cells being the most investigated for their neural regenerative potential. Conclusion: Dental stem cell spheres demonstrate significant potential for neural regeneration. Several assays and characterizations have been performed to characterized the mechanisms underlying dental sphere formation. Furthermore, in vivo studies are imperative to deduce the neural regenerative potential of stem cells in complex biological environments.

IntroductionOver-the-counter (OTC) medications are those obtained without a medical prescription from a healthcare professional. With the increasing availability of information from various sources, including social media, pharmacy students may be exposed to unreliable or inaccurate data. Incorrect medication use is particularly concerning due to its potential risk of causing adverse health effects.” Hence, this study aims to determine students’ knowledge and attitudes at Taif University’s pharmacy college.MethodsThis research utilized a cross-sectional online questionnaire-based study, employing data from a sample of 450 pharmacy students from Taif University in Saudi Arabia. Descriptive analysis included descriptive and differential analysis. The data were analyzed using statistical package for social sciences (SPSS) Version 27.ResultsThe majority of participants, 297 (88.2%), were aware that inappropriate use of over-the-counter medications might have negative implications. A total of 233 participants (51.8%) reported having previously used an OTC medication. Also, 293 (65.1%), were aware that using OTC medications beyond their expiration date was harmful. A total of 280 participants (62.2%) had a high knowledge of OTC medication, whereas 170 respondents (37.8%) had a low level of knowledge. A significant correlation was found between age, year of study, and the use of OTC medication p-values 0.05).ConclusionThe study found positive attitudes toward OTC medications. Due to increased pharmaceutical exposure and self-medication, upper-year students and OTC course graduates comprehend OTC medications better. The examination found safety protocol violations in expiration dates, prescription label interpretation, and storage. Therefore, the study provides useful information for future attempts. Also, this study may contribute to the literature and guide future research to fill knowledge gaps.

Skin cancer is a widespread and perilous disease that necessitates prompt and precise detection for successful treatment. This research introduces a thorough method for identifying skin lesions by utilizing sophisticated deep learning (DL) techniques. The study utilizes three convolutional neural networks (CNNs)—CNN1, CNN2, and CNN3—each assigned to a distinct categorization job. Task 1 involves binary classification to determine whether skin lesions are present or absent. Task 2 involves distinguishing between benign and malignant lesions. Task 3 involves multiclass classification of skin lesion images to identify the precise type of skin lesion from a set of seven categories. The most optimal hyperparameters for the proposed CNN models were determined using the Grid Search Optimization technique. This approach determines optimal values for architectural and fine-tuning hyperparameters, which is essential for learning. Rigorous evaluations of loss, accuracy, and confusion matrix thoroughly assessed the performance of the CNN models. Three datasets from the International Skin Imaging Collaboration (ISIC) Archive were utilized for the classification tasks. The primary objective of this study is to create a robust CNN system that can accurately diagnose skin lesions. Three separate CNN models were developed using the labeled ISIC Archive datasets. These models were designed to accurately detect skin lesions, assess the malignancy of the lesions, and classify the different types of lesions. The results indicate that the proposed CNN models possess robust capabilities in identifying and categorizing skin lesions, aiding healthcare professionals in making prompt and precise diagnostic judgments. This strategy presents an optimistic avenue for enhancing the diagnosis of skin cancer, which could potentially decrease avoidable fatalities and extend the lifespan of people diagnosed with skin cancer. This research enhances the discipline of biomedical image processing for skin lesion identification by utilizing the capabilities of DL algorithms.

BackgroundTransition to electronic prescribing (e-prescribing) systems, such as Wasfaty, represents a significant advancement in healthcare. Introduced by the Saudi Arabian Ministry of Health in 2019, Wasfaty aims to enhance medication availability and streamline healthcare delivery. This study explores the challenges and enablers community pharmacists face when using the Wasfaty system in Saudi Arabia. This study uses the Theoretical Domains Framework (TDF) to analyse behavioural and contextual factors influencing pharmacists’ adoption of electronic prescriptions. TDF identifies key challenges and enablers across multiple behavioural domains, making it well-suited for understanding complex healthcare implementation processes.MethodsA qualitative study design was employed using the Theoretical Domains Framework (TDF) to understand factors affecting Wasfaty use. Participants were purposively sampled, focusing on community pharmacists experienced with Wasfaty prescriptions. Recruitment and interviews occurred from June to August 2024. Data saturation was achieved at 18 interviews. Transcripts were coded and mapped into TDF domains to identify barriers and enablers.ResultsFive key TDF domains influenced Wasfaty use: environmental context and resources; social influences; beliefs about capabilities; social/professional role and identity; and knowledge. Challenges included high prescription volumes, medication shortages, technical difficulties, resistance from elderly patients, and inconsistent training. Some pharmacists reported inadequate training as a barrier, while others highlighted effective programs as enablers.ConclusionSignificant barriers, such as insufficient training and high prescription volumes, impede effective Wasfaty use. However, continuous training, prior experience with electronic systems, and organisational support were crucial enablers. Addressing these challenges through targeted interventions can enhance e-prescription efficiency, improving patient care and outcomes. Findings underscore the importance of ongoing professional development and supportive work environments in fostering pharmacists’ competence in electronic systems.

A cross-sectional study was conducted to assess antibiotic usage, residues, resistance and drivers of their emergence in fish aquaculture in Northern Nigeria between 2019 and 2020. A structured questionnaire was administered to 151 randomly selected farmers. Fish, column and wastewater samples were analysed with enzyme link immunosorbent assay (ELISA) for residue detection and quantification. We performed descriptive and analytic statistical analyses. All selected farmers participated, 78.1% used antibiotics in ponds. Majority (77.1%) of the farmers did not know what antibiotic misuse entailed and 73.3% mentioned that antibiotic misuse and overuse cannot predispose to residues and resistance. The most frequently used antibiotics were tetracyclines (TCs) (99.2%). Significant risk routes for residue and resistance spread were: contaminated fish and its products ingestion, direct contact with fish and fomites contaminated as well as water and aerosols contaminated in the pond environment. Significant social and cultural that influenced residue and resistance development include antibiotics misuse/overuse (Odds Ratio [OR] = 3.8; 95% confidence interval [CI]: 1.62–8.74) as well as poor education and expertise of the farmers (OR = 2.9; 95% CI: 1.24–6.94). The mean TCs residues level in column and wastewater was 123.6 ± 18.2 μg/kg−1. Poor knowledge and attitudes regarding antibiotic usage were identified. Adequate antibiotic stewardship should be promoted through farmers’ education. Identified social and cultural factors can be mitigated through the ‘One Health’ approach.

Recent research has shown that droughts have intensified in South Asia over the past two decades. As a natural disaster, this has severely impacted people’s livelihoods, especially in the dry zones of Sri Lanka. Thus, to ensure human well-being, security of water resources, and ecosystem health, it is essential to minimize the impacts of droughts using reliable information. Remote sensing (RS) data and techniques help bridge the gap by enabling the analysis of drought phenomena through a diverse array of indices developed in the fields. However, until now, there has been a lack of systematic monitoring and reliable data for accurately characterizing droughts in the study area. As the first comprehensive analysis, we tried to evaluate the spatial–temporal dynamics of drought conditions in two Divisional Secretariat Divisions of the Anuradhapura district, Sri Lanka using eight standardized remote sensing-derived indices over a decade (2013–2023) including the Standardized Precipitation Index (SPI). SPI values indicated that the region has experienced notably below-average precipitation. According to SPI results, a significant portion of the Medawchchiya area experienced arid conditions in 2023. All other indices proved that 2018 was the driest year and 2013 was the wettest year among the three time points, as reflected by their low and high index values. However, according to NVSMI and LST, the wettest year is 2023, with only 1.78 % of areas experiencing severe drought and a maximum LST of 31.4 °C. LULC change detection revealed that 14.3 % of agricultural lands and 5.1 % of forest areas were converted into barren lands over a decade. Overall, this conversion may be another leading factor contributing to increased LST and dryness in the area during the concerned period while increasing the mean LST of barren land by around 4.7 °C per decade. Land surface-related and vegetation-related indices, such as NDWI, NDMI, LST, and NDVI, exhibited a more pronounced impact on short-term drought occurrences. The findings revealed that average precipitation coincides with short-term drought episodes in the area, with 2018 standing out as having the least rainfall and the driest year. The study’s findings may provide additional insights for planning authorities, supporting environmental protection and enhancing agricultural production by mitigating droughts’ impacts through short- and long-term strategies. Although, the study focused on a small area, a similar approach could be extended to other areas by incorporating advanced machine learning techniques and additional drought indices in the future.

Smoking declines are uneven around the world, and we have few studies on the correlates of youth smoking in contexts like Saudi Arabia, where declines have been slowest. Using a broadly socio-ecological framework and network data, we report on one of the few studies to simultaneously examine peer, family, and school features associated with smoking in the Saudi context. We find strong and consistent peer and family associations with both occasional and regular smoking via direct modeling (level of peers/family that also smoke) and substantive interactions (lying to parents in the family domain, engaging in unsupervised youth-centric activities, or seeking popularity in the peer domain). Although our design precludes causal claims, our results are consistent with smoking initiation being driven by occasional use surrounding attempts to gain youth social status, whereas regular use depends on implicit family and peer acceptance.

Background: Increasing student numbers and logistical challenges in pharmacy education limit patient counselling and clinical placement opportunities. Computer-based simulation (CBS) offers scalable, interactive learning but faces integration barriers.Objective: To explore global perceptions of CBS implementation in pharmacy education among educators and students. Methods: An online cross-sectional survey was developed based on literature review and expert feedback. The survey was distributed globally through academic pharmacy organisations, social media, and the authors’ networks. It included 20 questions targeting pharmacy educators and students.Results: Responses from 152 educators across 38 countries and 392 students from 46 countries, spanning six WHO regions (AFRO, AMRO, EMRO, EURO, SEARO, and WPRO) were analysed using descriptive and inferential statistics. The majority of educators (90.1%, n = 137) and students (84.2%, n = 330) expressed comfort with using CBS and implementing it in their curriculum. Despite this, CBS was perceived as underutilised by 53.5% (n = 81) of educators and 63.7% (n = 250) of students. Students valued CBS for enhancing communication and problem-solving skills, while educators highlighted its relevance to community pharmacy practice. Both groups supported CBS use in assessments. All educators (100%) identified workload reduction as a key priority, hoped CBS could assist in this area. Educators also reported barriers such as financial constraints (56.6%, n = 86) and insufficient technical support (53.3%, n = 81). On the other hand, students were less optimistic about institutional support, with only a few (7.4%, n = 29) believed institutional leaders would actively support CBS adoption. Regional differences emerged, with SEARO (Southeast Asia) and AFRO (Africa) showing the lowest CBS usage rates. Educators in SEARO, AFRO, and EMRO (Eastern Mediterranean) raised concerns about technical support, while those in SEARO, AFRO, and WPRO (Western Pacific, including Australia, New Zealand, and Singapore) expressed financial concerns. Educators in AFRO and WPRO, however, reported being 100% comfortable with using CBS.Conclusion: Both students and educators recognised the potential of CBS in pharmacy education, with strong support for its integration. Addressing barriers such as educator workload, financial constraints, and technical support is crucial for broader adoption. Improved resource allocation and targeted training for educators are essential to effectively incorporate CBS into the pharmacy curriculum.

Here, first-principles calculations have been employed to make a comparative study on structural, mechanical, electronic, and optical properties of new Ca3MF3 (M = As and Sb) photovoltaic compounds under pressure. The findings disclose that these two systems possess a direct band gap, showcasing a large tunable range under pressure, effectively encompassing the visible light spectrum. Adjusting various levels of hydrostatic pressure has effectively tuned both the band alignment and the effective masses of electrons and holes. Both compounds were initially identified as brittle materials at 0 GPa pressure; however, as the pressure increases, they transform, becoming highly anisotropic and ductile. Due to the material's mechanical robustness and enhanced ductility, as evidenced by its stress-induced mechanical properties, the Ca3MF3 (M = As and Sb) material shows potential for use in solar energy applications. Furthermore, as the influence of external pressure increases, the absorption edge seems to move slightly towards lower energy region. Optical properties show that the materials studied might be used from several optoelectronic devices in the visible and ultraviolet range area. Our findings show that pressure considerably influences the physicochemical properties of Ca3MF3 (M = As and Sb) compounds, which is a promising feature that can be useful for optoelectronic and photonic applications, for instance, light-emitting diodes, photodetectors, and solar cells.

Commonly used antimicrobial agents are no longer effective due to their overuse or misuse. In addition, many medicinal plant extracts can combat infectious diseases due to their main active constituents or secondary metabolites. The current study aimed to assess the bioactivities of Launaea nudicaulis (LN) leaf extract (LE) against different multi-drug resistant (MDR) Pseudomonas aeruginosa (P. aeruginosa) isolates. The ethyl acetate extract of a Launaea nudicaulis (LN) leaf was analyzed using GC–MS, which identified 27 key bioactive compounds. The major constituents found were as follows: 7-acetyl-6-ethyl-1,1,4,4-tetramethyltetralin, isopropyl myristate, thiocarbamic acid, N,N-dimethyl, S-1,3-diphenyl-2-butenyl ester, hahnfett, cyclopentane acetic acid, 3-oxo-2-pentyl-, methyl ester, hexadecanoic acid, and dotriacontane. Our study demonstrated that the LN leaf was a rich source of other important phytochemicals, including phenolic acids, tannins, saponins, and steroids. The relative biosafety of the L. nudicaulis LE was determined from the elevated inhibitory concentration 50 (IC50) of 262 μg/mL, as calculated from the cytotoxicity assay against the Wi-38 normal cell line. Conversely, 12.7 and 24.5 μg/mL were the recorded low IC50 values for the tested extract against the MCF-7 and Hep-G2 cancerous cell lines, respectively, reflecting its potent activity against the tested cancerous cell lines. Microbiologically, the susceptible P. aeruginosa isolates to the tested extract showed a growth inhibition zone diameter, in the well diffusion assay, ranging from 11.34 ± 0.47 to 26.67 ± 0.47 mm, and a percent inhibition (PI) value of 50–106.2%, reflecting its acceptable activity. In addition, the broth microdilution assay recorded minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) in the ranges of 15.625–1,000 μg/mL and 125–1,000 μg/mL, respectively. In conclusion, the L. nudicaulis LE revealed showed promising activity and high selectivity against P. aeruginosa. Moreover, the extract exhibited natural anticancer activities with safe low concentrations, indicating its potential as a superior candidate for future studies of its active constituents.

Probiotics have recently gained significant interest for their possible therapeutic effects in treating numerous health conditions. Probiotics containing Bacillus subtilis have been shown to have several health benefits, most notably in preventing diarrhea and gastrointestinal problems. A novel probiotic strain, Bacillus subtilis (NMCC-path-14), isolated from the rumen of a Nilli Ravi Buffalo, was evaluated for 28-day repeated dose toxicity in Balb/c mice. The NMCC-path-14 in low dose (1 × 108 CFU/ml) and high dose (1 × 1010 CFU/ml) was administered to the mice through gavage regularly. After 28 days of treatment, it was discovered that the no-observed-adverse-effect level (NOAEL) for NMCC-path-14 wasgreater than 1 × 1010 CFU/animal/day. This study also revealed no treatment-related changes in clinical parameters, body weight, gross pathology, or histology. Food consumption, hemoglobin, hematocrit, red blood cell counts, and colon length increased, while total/differential leukocyte count and platelets remained unchanged. The administration of NMCC-path-14 also resulted in decreased bilirubin and creatinine levels. Furthermore, NMCC-path-14 also displayed a promising antioxidant potential by increasing the antioxidant enzymes (GST, GSH, and CAT) and decreasing oxidant enzyme (MDA and NO) levels in vital organs like the liver, kidneys, spleen, and colon. TheNMCC-path-14also decreased the pathogenic bacterial population while increasing the beneficial population. Given the lack of adverse effects observed after NMCC-path-14 treatment, this strain is safe and must be considered as a potential probiotic in humans.

This study explores the foundational iterative processes of fractional calculus, focusing on Θ-conformable fractional derivatives (Θ-CFD). We introduce novel fractional operators and define their associated function spaces. Additionally, we establish a series of theorems that enhance our understanding of these operators within the context of fractional calculus.
