Stomata are small structures present primarily in the leaf epidermis of plants, composed of two guard cells that surround and regulate the opening and closing of the stomatal pore. Stomatal movements regulate atmospheric CO2 input for photosynthesis and transpired water output, being thus important for plant water use efficiency, defined by the ratio of CO2 assimilation to transpired water loss. Recent results indicate that the degradation of sucrose is a mechanism responsible for inducing stomatal opening. Additionally, it has been shown that genetic manipulation of guard cell sucrose metabolism is a potential tool for obtaining plants that consume less water and/or having greater water use efficiency. Within this perspective, this study aimed to evaluate the biotechnological potential of genetic manipulation of guard cells saccharolytic activity in the development of drought tolerant and/or more water use efficient plants. Different biochemical and molecular analyzes coupled with an extensive physiological characterization were developed in order to characterize the phenotype of transgenic Nicotiana tabacum plants antisense for the potato sucrose synthase 3 gene (StSUS3) under the control of the stomatal-specific KST1 promoter. The results demonstrated that the inserted transgene (StSUS3) reduced the expression of tobacco NtSUS2 isoform, which consequently altered substantially guard cell metabolism. Also, the plants decreased stomatal conductance and whole plant transpiration rates and increased the water use efficiency, besides the plants presented a drought avoidance phenotype. Taken together, our results indicate that the NtSUS2 gene is a key regulator of plant transpiration and is therefore an important biotechnological target. Os estômatos são pequenas estruturas presentes principalmente na epiderme foliar das plantas, composto por duas células-guarda que circundam e regulam a abertura e o fechamento do poro estomático. Os movimentos estomáticos regulam a entrada de CO2 atmosférico para a fotossíntese e a saída de água via transpiração, sendo importantes na regulação da eficiência do uso da água, sendo a razão entre a assimilação de CO2 e a perda de água via transpiração. Resultados recentes indicam que a degradação da sacarose é um mecanismo responsável por induzir a abertura dos estômatos. Adicionalmente, foi demonstrado que a manipulação genética do metabolismo da sacarose de células-guarda é uma ferramenta potencial para a obtenção de plantas que consomem menos água e/ou maior eficiência do uso da água. Dentro desta perspectiva, este trabalho teve como objetivo avaliar o potencial biotecnológico da manipulação genética da atividade sacarolítica de células-guarda no desenvolvimento de plantas tolerantes a seca e/ou com maior eficiência do uso da água. Diferentes análises bioquímicas e moleculares somadas à uma extensa caracterização fisiológica foram desenvolvidas a fim de caracterizar o fenótipo de plantas transgênicas de Nicotiana tabacum antisenso para o gene sacarose sintase 3 (StSUS3) de batata sob o controle do promotor KST1 específico de estômatos. Os resultados demonstraram que o transgene inserido (StSUS3) reduziu a expressão da isoforma NtSUS2 de tabaco, que, por consequência, alterou substancialmente o metabolismo de células guarda. Além disso, as plantas diminuíram a condutância estomática e taxas de transpiração da planta inteira e elevou a eficiência do uso da água, além das plantas apresentarem um fenótipo de evitar a seca. Tomados em conjunto, nossos resultados indicam que o gene NtSUS2 é um regulador chave da transpiração vegetal, sendo, assim, um importante alvo biotecnológico.

Entre esses estímulos, o estresse causado pela seca vem se tornando o principal fator limitante à produção agrícola em todo o mundo. O déficit hídrico é percebido inicialmente pelas raízes, que deve sinalizar eficientemente a parte aérea e, assim, desencadear os mecanismos de adaptação à seca. Por isso, a fim de avaliar o papel das giberelinas (GAs) na comunicação entre a raiz e a parte aérea durante o déficit hídrico, o tomateiro (Solanum lycopersicum L.) mutante procera (pro), com resposta constitutiva à GA, e seu tipo selvagem Micro-Tom (MT), foram utilizados em enxertos recíprocos e cultivados sob condições irrigada e de déficit hídrico. Posteriormente, foi avaliado o crescimento, o estresse oxidativo, a expressão gênica, as relações hídricas e o conteúdo hormonal para fornecer informações sobre os ajustes mediados por GA durante o déficit hídrico. Todas as combinações de enxertia com pro (i.e. pro/pro, MT/pro e pro/MT) não exibiram redução do crescimento sob condições de estresse, embora não tenha evitado o aumento do estresse oxidativo. Além disso, os enxertos com pro induziram maior acúmulo de ácido abscísico (ABA) na parte aérea, independente do genótipo do porta-enxerto. A sensibilidade aumentada à GA no porta-enxerto modulou a condutância estomática (gs) e a eficiência intrínseca de uso da água (EUAi) sob déficit hídrico, ou seja, durante o déficit hídrico a resposta constitutiva às GAs presente no porta-enxerto induziu forte redução da gs e, consequentemente, aumento da EUAi, independentemente do enxerto empregado. Além disso, sob condição de déficit hídrico, verificou-se regulação positiva e negativa dos genes SlDREB2, envolvido na modulação de mecanismos de resposta à seca, e SlDELLA, repressor de GA, respectivamente. O tratamento de folhas destacadas de MT e pro com ABA induziu redução da perda de água de maneira mais pronunciada em folhas de pro. Estes resultados indicam a existência de interação entre a sensibilidade à GA e ABA regulando os mecanismos de reajuste do crescimento e economia de água durante o déficit hídrico. Em outras palavras, a resposta constitutiva às GAs, especialmente nas raízes, modula a sinalização entre a raiz e a parte aérea e, assim, as respostas ao déficit hídrico. Plants are sessile organisms that must perceive and respond to various environmental cues throughout their life cycle. Among these factors, drought stress has become the main limiting factor to crop production around the world. Water deprivation is perceived primarily by the roots, which should efficiently signal the shoot to trigger drought responses in order to maximize the ability of plants to survive. In this study, the procera (pro) mutant tomato (Solanum lycopersicum L.), with a constitutive response to gibberellin (GA), and its near-isogenic line cv. Micro-Tom (MT), were used in reciprocal grafting under well-watered and water stress conditions to evaluate the role of GA signaling in root-to-shoot communication during drought stress. Growth, oxidative stress, gene expression, water relations and hormonal content were measured in order to provide insights into GA-mediated adjustments to water stress. All graft combinations with pro (i.e. pro/pro, MT/pro and pro/MT) prevented the reduction of growth under stress conditions, but without a reduction in oxidative stress. Scions harboring the pro mutation tended to increase the abscisic acid (ABA) content, independent of the rootstock. Moreover, the GA sensitivity of the rootstock modulated stomatal conductance (gs) and intrinsic water use efficiency (WUEi) under drought stress, therefore, during the drought stress the constitutive response to the GAs present in the rootstock induced a strong reduction of the gs and, consequently, increase of the WUEi, regardless of the graft employed. Furthermore, after exposure to drought stress, there was positive and negative regulation of the SlDREB2, involved in the modulation of drought response mechanisms, and SlDELLA genes, GA repressor, respectively. The reduction of water loss was more pronounced on detached leaves of pro after treatment with ABA when compared to MT leaves. These results indicate the occurrence of crosstalk between sensitivity to GA and ABA regulating the mechanisms of growth and water economy readjustment during water deficit. O setor florestal é de grande importância para a economia brasileira, tendo esse setor um PIB de mais de R$ 86 bilhões, correspondendo a 1,3% do PIB brasileiro e 6,9% do PIB Industrial no país. Entre as diversas espécies arbóreas plantadas o eucalipto é a que ocupa maior área no Brasil, com mais de 5 milhões de hectares, e está disperso por quase todo o território nacional, mas principalmente em Minas Gerais, São Paulo e Mato Grosso do Sul. Por conta dessa diversidade de paisagem necessita-se utilizar variedades adaptadas a cada local, para que sejam as mais produtivas possíveis. Em relação ao eucalipto a principal limitação para o seu crescimento é a água. Assim, identificar e compreender os mecanismos responsáveis pela resistência à seca permitirá o desenvolvimento de plantas tolerantes à seca e com maior produtividade. A resposta a deficiência hídrica gera mudanças importantes nos processos fisiológicos e morfológicos das plantas, tais como redução da dimensão foliar e biomassa, além de mudanças moleculares como aumento de proteínas envolvidas nos metabolismos de nitrogênio e carboidratos e nos mecanismos de defesa como proteínas heat shock, ascorbato peroxidases e glutationa S-transferases. E devido a essas mudanças, a biologia molecular aparece como uma importante ferramenta na identificação de genes, proteínas e vias metabólicas, que regulam a resposta da planta ao estresse. Nesse trabalho foi usada a proteômica quantitativa label-free para a caracterização do proteoma foliar, caulinar e radicular de dois genótipos de eucalipto contrastantes, em resposta a restrição hídrica, a fim de identificar as diferenças de abundância proteica desses genótipos contrastantes. Também foi realizada a análise dos parâmetros fisiológicos, como fotossíntese, transpiração estomática e condutância estomática das folhas. Dentro de cada genótipo foi feito um grupo controle onde as plantas permaneceram irrigadas. A análise entre os tratamentos controle (C) e estressado (E) de cada tecido, resultou na identificação de proteínas diferencialmente abundantes, sendo, nas folhas, 122 e 74 para Suscetível (S) e Tolerante (T) respectivamente; no caule foram 161 proteínas no S e 180 no T; e na raiz foram 144 e 177 proteínas diferencialmente bundantes no S e T, respectivamente. Dentre as proteínas identificadas, há aquelas relacionadas a processos fisiológicos como fotossíntese, regulação dos estômatos e crescimento e desenvolvimento das plantas, estando em abundância nas plantas submetidas ao estresse por déficit hídrico no genótipo Tolerante. Na análise para cada tecido entre os dois genótipos (S e T), foram identificadas 184, 280 e 256 proteínas estatisticamente significativas para folha, caule e raiz, respectivamente. As plantas tolerantes apresentaram maior abundância de proteínas envolvidas na produção de energia, controle do movimento estomático e crescimento e desenvolvimento das plantas. Assim, o proteoma refletiu a expressão dos genes que influenciam, mais diretamente, a bioquímica e o funcionamento celular, revelando que ambos os tratamentos e genótipos respondem diferentemente à influência do déficit hídrico. Silviculture has been of great importance to Brazilian economy, accounting for a sector PIB with more than R$ 86 billion, wich corresponds to 1,3% of Brazilian PIB and 6.9% of the Industrial PIB in the country. Among several arboreal plant species planted in Brazil, eucalyptus occupies the largest area with more than 5 million hectares, and is spread throughout most of the territory, but mainly Minas Gerais, São Paulo and Mato Grosso do Sul. Due to the different landscapes it is necessary to use varieties adapted to each local, to assure the greatest yields. The main limitation for eucalyptus growth is water. Therefore discovering and understanding the mechanisms responsible for drought resistance allows the development of drought tolerant plants with higher productivity. As a response, water deficit generates important changes in morphological and physiological processes in plants, as it causes the limitation of individual leaf size as well as biomass, also there is changes in molecular processes too, like an accumulation in nitrogen and carbohydrates metabolisms and in the defense mechanisms, with heat shocks, ascorbate peroxidases and glutathione S-transferases. And these changes in the plants makes molecular biology an important tool to identify genes, proteins and metabolic pathways, which regulate the plant response to stress. To characterize the proteome of the leaves, stems and roots of eucalyptus under drought stress of two contrasting genotypes, it was used label-free quantitative proteomics, for identify the differences in protein abundance between the contrasting genotypes, and was made analyses of leaf physiological parameters. For each genotype a control group was established with plants kept irrigated. The analysis between treatments (C and E) of each tissue resulting in the identification of differentially abundant proteins, with in leaves, 122 and 74 in Susceptible (S) and Tolerant (T), respectively; in stems were 161 proteins for S and 180 proteins for T; and in roots were 144 and 177 differentially abundant proteins for S and T, respectively. Among the proteins identified, there is proteins involved in photosynthesis, stomatal regulation and plant growth and development, and were present in abundance in plants submitted to water deficit of Tolerant genotype. In the analysis for each tissue between the genotypes (S and T), 184, 280 and 256 statistical significant proteins for leaf, stem and root were quantified, respectively. And tolerant plants have a greater abundance of proteins involved in energy production, control of stomatal movement and plant growth and development. Thus, the proteome reflected the expression of genes that most directly influence biochemistry and cell function, revealing that both treatments and genotypes respond differently to the influence of water deficit.

O entendimento das interações do contínuo solo-planta-atmosfera frente às mudanças ambientais do ecossistema é pré-requisito para modelos que preveem o balanço de carbono em florestas tropicais. Neste contexto, este trabalho teve por objetivo caracterizar a influencia de variáveis meteorológicas no Baixio e Platô sobre o potencial hídrico foliar (Ψf), a condutância estomática (gs) e temperatura foliar (Tf) considerando diferentes técnicas, de duas espécies arbóreas respectivamente no dossel de uma floresta primária na Amazônia Central e entender mecanismos bioquímicos como produção de acido abscísico (ABA) e sua relação com a regulação estomática. A pesquisa foi realizada na Reserva Biológica do Cuieiras (2º35’22’’S, 60º06’55’’W) em 2017. As espécies estudadas foram a Eschweilera cyathiformis e Pouteria anomala (no platô) Peltogyne excelsa e Micropholis guyanensis (no baixio). As espécies estudadas no experimento bioquímico foram Cariniana legalis, Handroanthus serratifolius e Astronium fraxinifolium, realizado em Trancoso-BA em 2018. As variáveis meteorológicas foram obtidas a partir da instrumentação instalada em duas torres de monitoramento K34 (platô) e B34 (baixio). O comportamento da progressão diária de Ψf das diferentes espécies na avaliação foi significativo, de modo que a espécie P. anomala e E. ciathiformis na K34 é diferente em relação às espécies P. excelsa e M. guyanensis no baixio, isso pode estar relacionado a um comportamento isohídrico e anisohídrico das espécies no platô e baixio, respectivamente. Para a dinâmica de gs das espécies em resposta aos diferentes períodos chuvoso e seco houve diferença significativa, observou-se que gs diminuiu consideravelmente no período seco quando comparado ao período chuvoso e seguiu o mesmo padrão no platô e baixio. Os resultados das medidas de Tf nas espécies do baixio e platô, e para os períodos chuvoso e seco, mostram diferenças significativas de modo que a medida com o porômetro foi mais significativa quando comparadas com as técnicas do Termovisor (Flir), Termopares e Sensor InfraRed, bem como em relação as medidas de temperatura do ar com Termohigrômetros. Observaram-se variação dos valores ótimos de Tf no platô em relação gs sazonalmente. Para todo o período de coleta se obsreva a configuração de comportamento de histerese entre a condutância estomática e DPV e Tf. No platô, para P. anomala, observou-se que o DPV e Tf não foi suficiente para predizer um maior efeito sobre gs, no período sazonal chuvoso, enquanto no período seco DPV demonstrou maior efeito sobre gs. Já no baixio, para P. excelsa e M. guyanensis, observou-se que o DPV demonstrou ter um maior efeito sobre gs, nos períodos chuvoso e seco. Quanto ao experimento bioquímico observou-se que apenas a espécie Handroanthus serratifolius apresentou concentração de ABA consideráveis na folha e demonstrou variar diurnamente com maiores valores ocorrendo as 2:00PM comparado aos horários de 12:00AM e 18:00PM. A concentração de ABA respondeu mais intensamente ao tratamento não irrigado. Assim, variações específicas da espécie nas sensibilidades de biossíntese de ABA às variáveis ambientais como DPV, Tf, PAR e Ψf, podem ajudar a explicar o comportamento estomático de espécies florestais da Amazônia. Portanto, conclui-se que fatores ambientais e aspectos fisiológicos por si só não explicam a variação diurna no movimento estomático. Assim, é provável que fatores bioquímicos como a produção de ABA, associadas às variações ambientais e suas interações com a fisiologia das árvores de dossel na floresta Amazônica é tema de extrema relevância a ser explorado, para melhorar o entendimento de como as florestas tropicais respondem as mudanças climáticas. Understanding the interactions of the continuous soil-plant-atmosphere to ecosystem environmental changes is a prerequisite for models that predict carbon balance in tropical forests. In this context, the objective of this study was to characterize the influence of meteorological variables in the Baixio and Platô on leaf water potential (Ψf), stomatal conductance (gs) and leaf temperature (Tf) considering different techniques of two tree species, respectively in the canopy of a primary forest in Central Amazonia and to understand biochemical mechanisms such as abscisic acid production (ABA) and the relationship with stomatal regulation. The species studied were the Eschweilera cyathiformis and Pouteria anomala (in the plateau), and Peltogyne excelsa and Micropholis guyanensis (in the Baixo). The species studied in the biochemical experiment were Cariniana legalis, Handroanthus serratifolius and Astronium fraxinifolium, performed in Trancoso-BA in 2018. The meteorological variables were obtained from the instrumentation installed in two monitoring towers K34 (plateau) and B34 (Baixo). The behavior of the daily progression of Ψf of the different species in the evaluation was significant, so that the species P. anomala and E. ciathiformis in K34 is different in relation to the species P. excelsa and M. guyanensis in the Baixo, this may be related to isohydric and anisohydric behavior of the species in the plateau and shallow, respectively. For the gs dynamics of the species in response to the different rainy and dry periods there was significant difference, it was observed that gs decreased considerably in the dry period when compared to the rainy season and followed the same pattern in the plateau and shallow. The results of the measurements of Tf in the species of the basin and plateau, and for the rainy and dry periods, show significant differences so that the measurement with the porometer was more significant when compared with the techniques of Thermovisor (Flir), Thermocouples and Sensor InfraRed, as well as in relation to the air temperature measurements with Thermohygrometers. There was variation of the optimal values of Tf in the plateau relative to gs seasonally. For the entire collection period, the configuration of hysteresis behavior between stomatal conductance and VPD and Tf is obscured. In the plateau, for P. anomala, it was observed that the VPD and Tf was not sufficient to predict a greater effect on gs in the rainy season, while in the dry period VPD showed a greater effect on gs. In the Baixo, for P. excelsa and M. guyanensis, it was observed that the VPD showed to have a greater effect on gs in the rainy and dry periods. As for the biochemical experiment, it was observed that only the species Handroanthus serratifolius presented a considerable ABA concentration in the leaf and showed a diurnal variation with higher values occurring at 2:00 PM compared to the times of 12:00 AM and 6:00 PM. The concentration of ABA responded more intensely to non-irrigated treatment. Thus, speciesspecific variations in ABA biosynthesis sensitivities to environmental variables such as VPD, Tf, PAR and Ψf may help to explain the stomatal behavior of Amazonian forest species. Therefore, we conclude that environmental factors and physiological aspects alone do not explain the diurnal variation in stomatal movement. Thus, it is likely that biochemical factors such as ABA production, associated to environmental variations and their interactions with the physiology of canopy trees in the Amazon forest, are of extreme relevance to be explored to improve the understanding of how tropical forests respond to climate changes.

Merrill), uma das principais culturas oleaginosas, é muito sensível a fatores ambientais, sendo a deficiência hídrica um fator particularmente limitante para seu desenvolvimento e produção. Algumas substâncias podem ser utilizadas para minimizar os efeitos danosos do estresse hídrico. Bioreguladores, tais como ácido γ-aminobutírico (GABA) e Acetilcolina (Ach) atuam na defesa de plantas em respostas ao estresse, desempenhando um papel osmoprotetor e na regulação do movimento estomático, respectivamente. O objetivo deste estudo foi de analisar diferentes modos de aplicação exógena desses dois bioreguladores (isoladamente ou em combinação) e estudar o seu efeito na regulação dos processos de resposta de soja ao deficit hídrico. Para tanto, foi realizada a aplicação combinada dos dois bioreguladores na dose de 2,0 mM em plantas de soja cv. Intacta sob diferentes regimes hídricos. Os fatores estudados foram: 1) da aplicação dos bioreguladores (i) nas sementes- S (ii) via foliar F; (iii) nas semente e via foliar - SF; (iv) controle sem aplicação - C; e 2) regimes hídricos: (i) 100% da capacidade de campo (CC) e (ii) suspensão da irrigação (SI). Foram realizadas análises de trocas gasosas foliares e biomassa para caracterização fisiológica das plantas, das atividades das enzimas anti-oxidantes (CAT e APX) e das alterações nas respostas transcricionais dos genes induzidos por deficiência hídrica, P5CS, LEA e ABA2. Para os tratamentos de déficit hídrico, foi observada uma queda de 93% da capacidade fotossíntécia (PN) no sexto dia após a suspensão da irrigação, assim como diminuição da biomassa, além de apresentar maior expressão dos genes induzidos por deficiência hídrica. O tratamento da combinação das substâncias GABA e Ach aplicados na semente e na folha sob déficit hídrico, destacou-se por apresentar menor queda da fotossíntese, assim como, aumento para condutância estomática e biomassa. Para as análises transcricionais a aplicação dacombinação de GABA e Ach em semente e folha apresentaram menor expressão dos genes P5CS, LEA e ABA2 e maior atividade das enzimas SOD, CAT e APX, em relação ao tratamento controle Em nosso estudo foi possível observar que a aplicação combinada das substâncias GABA e Ach na semente e folha a 2,0 mM promoveu uma melhora do desempenho das plantas por meio da açãode Ach promovendo um aumento sobre a condutância estomática e um efeito osmoprotetor de GABA sobre as plantas em situação de estresse. Soybean (Glycine max (L.) Merrill), a major oil crop, is very sensitive to environmental factors, and water deficiency is a particularly limiting factor for its development and production. Some substances can be used to minimize the harmful effects of water stress. Bioregulators, such as γ-aminobutyric acid (GABA) and Acetylcholine (Ach) act in the defense of plants in response to stress, playing an osmoprotective role and regulating stomatal movement, respectively. The objective of this study was to analyze different modes of exogenous application of these two bioregularators (alone or in combination) and to study their effect on the regulation of soybean response processes to the water deficit. Therefore, it was used in combination of two bioregulator dose of 2.0 mM in soybean plants cv. Intact under different water regimes. The factors studied were: 1) the application of bioregulators (i) in seeds- S (ii) in leaf L ; (iii) in the seed and leaf - SL; (iv) control without application - C; and 2) water regimes: (i) 100% of field capacity (CC) and (ii) suspension of irrigation (SI). Leaf gas exchange and biomass analyzes were performed for the physiological characterization of plants, the activities of the antioxidant enzymes (CAT and APX) and the changes in the transcriptional responses of genes induced by water deficiency, P5CS, LEA and ABA2. For water deficit treatments, a 93% decrease in the photosynthetic capacity (PN) was observed on the sixth day after the suspension of irrigation, as well as a decrease in biomass, in addition to a greater expression of genes induced by water deficiency. The treatment of the combination of the GABA and Ach substances applied to the seed and the leaf under water deficit was highlighted by lower photosynthesis decrease, as well as an increase in stomatal conductance and biomass. For the transcriptional analysis the application of GABA and Ach in seed and leaf showed lower expression of the genes P5CS, LEA and ABA2 and greater activity of the SOD, CAT and APX enzymes, in relation to the control treatment. In our study it was possible to observe that the combined application of the GABA and Ach substances in the seed and leaf at 2.0 mM promoted an improvement of the performance of the plants through the action of Ach promoting an increase on the stomatal conductance and an osmoprotective effect of GABA on plants in a situation of stress.

A ocorrência de potenciais de ação e neurotransmissores, componentes principais do sistema nervoso animal, em plantas, bactérias e fungos mostra a universalidade dos princípios de sinalização e transmissão de informações na forma de sinais químicos e elétricos em todos os organismos. Esses tópicos de estudo, juntamente com inteligência em plantas e transporte vesicular de auxina, constituem as linhas de pesquisa principais da recém-criada Neurobiologia Vegetal. Entre os neurotransmissores encontrados em plantas, a acetilcolina atua, entre outras situações, no crescimento e desenvolvimento controlado pelo fitocromo e na permeabilidade iônica de membranas. Nesse contexto, foi sugerido que a acetilcolina pode desempenhar um papel importante na regulação do movimento estomático, tendo efeito estimulatório na abertura dos estômatos além de poder atuar na sinalização entre raiz e parte aérea. Dessa forma, foi proposto identificar a presença deste neurotransmissor em plantas de milho hidratadas e submetidas a estresse hídrico, com o objetivo de correlacionar a presença de acetilcolina com as respostas estomáticas de tais plantas. Além disso, foi objetivo do trabalho avaliar parâmetros fisiológicos como potencial hídrico, condutância estomática, transpiração e fotossíntese líquida e suas possíveis relações com a acetilcolina em três folhas das plantas hidratadas e estressadas. Para tanto, foi montado um experimento em blocos casualizados em esquema fatorial (2x3). Os fatores foram: água, nos níveis de hidratação e estresse, e idade das folhas nos níveis folha 4 (mais velha), folha 5 (idade intermediária) e folha 7 (mais jovem). As plantas foram divididas em 20 blocos, contendo uma planta hidratada e uma sob estresse cada e as análises fisiológicas feitas nas três folhas. As plantas foram colocadas em câmara de crescimento tipo BOD com controle de iluminação e temperatura. Após análises fisiológicas, as folhas foram utilizadas para extração e determinação de acetilcolina. Os extratos purificados e secos foram submetidos à pirólise e cromatografia gasosa e as substâncias identificadas por espectrometria de massas. Não foi detectada acetilcolina nas plantas, apesar de estudos anteriores demonstrarem sua ocorrência em folhas e sementes de milho. Hipóteses foram levantadas para explicar tal fato. Quanto as variáveis fisiológicas, o déficit hídrico reduziu em aproximadamente 59% a transpiração, em 65% a condutância estomática e em 59% a fotossíntese das plantas. Condutância estomática e transpiração, condutância e fotossíntese, e transpiração e fotossíntese apresentaram intensa correlação. Já o potencial hídrico teve baixa correlação com essas variáveis. Quanto ao fator idade, folhas 7 apresentaram maiores valores de fotossíntese, condutância e transpiração que as folhas 4 e 5. The occurrence of action potential and neurotransmitters, the major components of animal nervous system, in plants, bacteria and fungi, shows the universality of signaling principles and information transmission in the way of chemical and electrical signals in all organisms. These study topics, along with plant intelligence and vesicular-based auxin transport, constitute the major research lines of the newly created Plant Neurobiology. Among the neurotransmitters found in plants, the acetylcholine plays a role in phytochromecontrolled growth and development and in membrane ion permeability. In this context, it was suggested that acetylcholine can play an important role in the regulation of stomatal movements, having stimulatory effect in the stomatal opening. In addition it can play a role in root-to-shoot signaling process. Therefore, it was proposed to identify the presence of this neurotransmitter in maize plants hydrated and under water stress, with the aim of correlating the presence of acetylcholine with the stomatal responses of such plants. Moreover, another aim of the study was to evaluate physiological parameters like water potential, stomatal conductance, transpiration rate and net photosynthetic rate and their possible relationship with the acetylcholine in three leaves of hydrated and stressed plants. Therefore, an experiment was set up in randomized block design in 2x3 factorial. The factors were: water, in the levels of hydration and stress, and leaves age in the levels leaf 4 (older), leaf 5 (intermediary age) and leaf 7 (younger). The plants were divided in 20 blocks, and each one has had one hydrated plant and one stressed plant and the physiological analysis was made in three leaves. The plants were placed in B.O.D. growth chamber under controlled conditions of light and temperature. After the physiological analysis, the leaves were used to extraction and determination of acetylcholine. The dried and purified extracts were subjected to pyrolysis and gas chromatography and the substances identified by mass spectrometry. The acetylcholine was not detected in plants, although earlier studies have had demonstrated its occurrence in maize leaves and seeds. Hypotheses were elaborated to explain such fact. Regarding the physiological variables, water stress reduced the plants transpiration rate in 59%, stomatal conductance in 65% and net photosynthesis in 59%. Stomatal conductance, transpiration and photosynthesis were strongly related. On the other hand, the water potential showed weak correlation with that variable. As for the age factor, leaves 7 had higher photosynthetic rates, conductance and transpiration than the leaves 4 and 5.
