4. Negotiating a Sea of White: Whiteness and Diversity in Dutch Opera Practices

Abstract

The police murders of George Floyd, Breonna Taylor and Ahmaud Arbery in 2020 sparked large-scale protests and drew worldwide attention to the Black Lives Matter movement. Subsequently, conversations on racism and police violence arose globally. Opera houses, such as the Dutch National Opera (DNO), also discussed racism and promised to diversify. On social media, DNO acknowledges its responsibility to change as a predominantly white organisation. This statement raises the question of how DNO has dealt with their whiteness and measures regarding diversity and inclusion so far. Therefore, in this thesis, I examine how the diversity and inclusion efforts of DNO between 2017 and 2021 reflect upon the organisation's whiteness. First, I position DNO within the contexts of whiteness concerning opera, the Dutch arts sector and Dutch society. The subsequent chapters address diversity and inclusion within the programme, public, personnel, partners and positioning (5 Ps) as the areas in which DNO aims to diversify. The analysis of DNO's 5 Ps reveals how whiteness inhibits the organisation's diversity and inclusion aims. Within the programme, the overwhelming whiteness of the composers, writers, conductors and directors supports whiteness as a mechanism of exclusion through stereotypical casting and whitewashing. The chapter on public indicates that the convergence of both age and diversity within DNO's aims and projects enforces whiteness by centring a young white audience. Personnel analysis shows the workings of whiteness as an orientation, gender advancement at the expense of people of colour, and tokenism. DNO's partners mirror the whiteness of surrounding networks and the Netherlands, as most collaborators struggle with whiteness. Lastly, DNO's positioning strategy reinscribes systemic whiteness. Ultimately, this thesis contributes to the discourse on whiteness and institutional racism within the realm of opera and Dutch society at large and stresses the critical reflectio

Abstract

The present report illustrates the first germination data of Limonium merxmuelleri subsp. merxmuelleri, a pioneer species of mine waste materials of Iglesiente region (South-West Sardinia). After harvesting, seeds were subjected to germination tests at the Sardinian Germplasm Bank (BG-SAR). The results show a high germination capability and germination rate at each tested temperature, with a slight germination reduction at the highest temperature (25°C) that simulates the aridity of the summer period. Abstract

The conservation of endemic and endangered plant species is of great interest to the scientific and research community. In this frame, seed banks play a crucial role when biodiversity preservation and climate change are considered. The study of seed viability and germination during storage conditions provides basic and useful information to ensure successful ex situ conservation. The aim of this study was to evaluate whether storage time and conditions (i.e., base collection at -25°C and active collection at +5°C) affect seed germination in the long term. For these purposes, eight Tyrrhenian endemic vascular plant species (mostly endangered) with orthodox seeds were studied: Brassica insularis, Centranthus amazonum, Dianthus morisianus, Digitalis purpurea var. gyspergerae, Ferula arrigonii, Helicodiceros muscivorus, Iberis integerrima and Verbascum plantagineum. These species were stored in the Sardinian Germplasm Bank (BG-SAR) at -25°C and at +5°C for a time ranging from 2 to 12 years. Germination tests were carried out following the optimal conditions reported in the literature for each species. The results showed, in general terms, the high seed germination capacity of all species stored at both conditions; regarding the time of seed storage, germination in some tested species (such as B. insularis and C. amazonum) slightly decreased over time. We argued that seed dehydration, low seed moisture content during storage and the use of hermetic glass containers can be considered key factors for long-term conservation of these orthodox seeds. In conclusion, this study showed that the conservation of these endemic species is ensured by seed bank storage, according to the general assumption that seed longevity depends on seed lot quality, on well-sealed storage containers and conditions before and during storage. Abstract

We conducted a pooled analysis of two phase III trials, RV-MM-EMN-441 and EMN01, to compare maintenance with lenalidomide-prednisone vs. lenalidomide in newly diagnosed transplant-eligible and -ineligible myeloma patients. Primary endpoints were progression-free survival, progression-free survival 2 and overall survival with both regimens. A secondary aim was to evaluate the impact of duration of maintenance on overall survival and on outcome after relapse. A total of 625 patients (lenalidomide-prednisone arm, n = 315; lenalidomide arm, n = 310) were analyzed. The median follow-up was 58 months. Median progression-free survival (25 vs. 19 months; p = 0.08), progression-free survival 2 (56 vs. 49 months; p = 0.9) and overall survival (73 months vs. NR; p = 0.08) were not significantly different between the two arms. Toxicity profiles of lenalidomide-prednisone and lenalidomide were similar, with the exception of neutropenia that was higher in the lenalidomide arm (grade ≥ 3: 9% vs. 19%, p < 0.001), without an increase in the rate of infections. Overall survival (median NR vs. 49 months, p < 0.001), progression-free survival from relapse (median 35 vs. 24 months, p = 0.004) and overall survival from relapse (median not reached vs. 41 months, p = 0.002) were significantly longer in patients continuing maintenance for ≥2 years. We showed that the addition of prednisone at 25 or 50 mg every other day (eod) to lenalidomide maintenance did not induce any significant advantage.

Abstract

An updated inventory of the vascular flora alien to Italy, providing details on the occurrence at regional level, is presented. The checklist includes 1597 species, subspecies, and hybrids, distributed in 725 genera and 152 families; 2 taxa are lycophytes, 11 ferns and fern allies, 33 gymnosperms, and 1551 angiosperms. 157 taxa are archaeophytes and 1440 neophytes. The alien taxa currently established in Italy are 791 (570 naturalized and 221 invasive), while 705 taxa are casual aliens, 4 are not assessed, 7 are of unknown regional distribution, 47 have not been confirmed in recent times, 3 are considered extinct or possibly extinct in the country, and 40 are doubtfully occurring in Italy. This checklist allows to establish an up-to-date number (9792) of taxa constituting the whole (native and alien) Italian flora.

Abstract

La Commissione Europea (CE) ha inserito ad oggi 36 taxa esotici vegetali nella lista delle specie esotiche invasive di rilevanza unionale ai sensi del Regolamento (UE) n. 1143/2014 del Parlamento Europeo e del Consiglio, recante disposizioni volte a prevenire e gestire l’introduzione e la diffusione delle specie esotiche invasive. La lista delle specie di rilevanza unionale viene periodicamente aggiornata e include quelle specie che rappresentano una grave minaccia per la biodiversità, ma anche per la salute dei cittadini e le attività economiche nei territori dell’Unione Europea e che necessitano di una gestione concertata a livello comunitario. La CE vigila sullo stato di ogni taxon grazie anche a periodiche rendicontazioni da parte dei paesi dell'Unione. In vista di tali report, tra il 2020 e il 2021 è stata definita e integrata la distribuzione di queste specie in Italia.

26. Pomalidomide in patients with multiple myeloma: potential impact on the reconstitution of a functional T-cell immunity.

Abstract

Background: Pomalidomide, a third-generation oral immunomodulatory drug, exhibits efficacy in patients with relapsed multiple myeloma or those refractory to bortezomib and lenalidomide (RRMM)., Methods: In this clinical context, we employed flow cytometry and CDR3 spectratyping to monitor the dynamics of the T-cell repertoire during Pomalidomide treatment, aiming to investigate its potential to reverse the immunological abnormalities characteristic of RRMM., Results: By flow cytometry at baseline we found a significant decrease in CD4 + frequency in MM patients, while CD8 + frequency were significantly higher in patients when compared to controls. Most T cell populations remained stable across all time points, except for CD4 + frequency, which notably decreased from t1 to subsequent assessments. Our investigation revealed as most relevant finding the notable increase in CD4 + expansions and the growing prevalence of patients manifesting these expansions. This pattern is even more evident in patients receiving their treatment until t3 and therefore still responding to treatment with Pomalidomide. We also conducted a comparison of spectratyping data before and after treatment, substantially demonstrating a relatively stable pattern throughout the course of Pomalidomide treatment., Conclusions: These observations imply that Pomalidomide treatment influences the T-cell repertoire, particularly in the CD4 + subpopulation during the later stages of treatment, raising speculation about the potential involvement of these lymphocyte expansions in mechanisms related to antitumor immunity., (© 2024. 27. New Alien Plant Taxa for Italy and Europe: An Update.

Abstract

Despite the wide amount of scientific contributions published on alien plant species, their diffusion dynamics, and their interactions with native taxa, it is increasingly difficult to slow down their spreading and their negative impact on habitats. Last recent years, in fact, a sharp rise in the number of new alien plant taxa introduced in Italy and Europe has been recorded. The aim of this work is to investigate most of the Italian territory in order to verify whether this alarming trend is still underway. Specimen collections and/or observations of alien plants have been performed in as many as 12 Italian regions. All the collected specimens are stored in public or private herbaria. Taxa have been identified according to the literature from the countries of origin of the investigated taxa, while the nomenclature followed the current international references. Updates on 106 taxa are reported. In particular, among 117 new records, 89 are first records, 27 are changes to status and there is 1 extinction. Seven new taxa for Italian alien flora are reported, two of which are new to Europe. The administrative regions with the highest number of records are Calabria (48), Sardegna (17) and Sicilia (15). Five of the surveyed taxa, for the first time, have been considered invasive aliens to Italian territory. The unfrequent amount of original results provided by this work, over the simple importance of data itself, proves how floristic investigation, still today, represents one of the most effective tools in broadening the current knowledge about alien taxa and their dynamics.

28. The Effect of Heat Shock on Seed Dormancy Release and Germination in Two Rare and Endangered Astragalus L. Species (Fabaceae).

Abstract

Many Astragalus species exhibit seeds with physical dormancy (PY), but little is known about the ecological context of this dormancy. We focused on A. maritimus and A. verrucosus , two threatened Sardinian endemic species inside the subgenus Trimeniaeus Bunge. Fresh seeds collected from the only two respective known populations were used to investigate the effect of mechanical scarification, heat shock, and water imbibition processes on PY release and germination. PY can be overcome through mechanical scarification of the water-impermeable seed coats, while no dormancy break was detected, nor a subsequent increase in seed germination due to fire-induced heat. This suggests that fire does not trigger dormancy release and seed germination in these species. The seeds tolerate relatively high heat shock temperatures (up to 120 and 100 °C for A. verrucosus and A . maritimus , respectively), but after 120 °C for 10 min, the number of dead seeds increases in both species. These facts suggest the capacity to develop a soil seed bank that can persist after fires and delay germination until the occurrence of optimal conditions. As regards water imbibition, both Astragalus species did not show the typical triphasic pattern, as germination started without further water uptake. This study emphasizes the significance of understanding germination processes and dormancy in threatened species. In fire-prone ecosystems, PY dormancy plays a crucial role in soil seed bank persistence, and it may be selectively influenced by post-fire conditions. Understanding such adaptations provides useful insights into conservation strategies.

29. Using Mediterranean Native Plants for the Phytoremediation of Mining Sites: An Overview of the Past and Present, and Perspectives for the Future.

Abstract

Mining exploitation in the Mediterranean Basin has left evident scars on the environment, and poses serious risks for human health and biodiversity, especially when mine wastes are left abandoned. This review analysed the main issues of metal(loid)s pollution related to mine exploitation in the Mediterranean Basin. Here, a list of Mediterranean native plant species studied for phytoremediation is given and, considering their biological forms, vegetational types, and ecology, we categorised them into halotolerant and hydro/hygrophilous vegetation, annual and perennial meadows, garrigues and maquis, and high maquis and woods. The main conclusions of the review are as follows: (1) plant communities established on mine environments are often rich in endemic taxa which ensure a high biodiversity and landscape value, and can help in the psychophysical health of local inhabitants; (2) political and land management should take greater account of the use of native plants for the remediation of contaminated soils; (3) a multidisciplinary approach that includes, among others, studies on biochemical response to metal(loid)s as well as the application of innovative soil amendments gives better results; (4) phytoextraction applications require a detailed recovery plan that takes into consideration several issues, including the negative influence on biodiversity due to extensive use of monotypic plantations, disposal of harvested hazardous plants, and the risk of phytoextracts entering the food chain; and (5) more studies are necessary to increase knowledge and to detect suitable species-especially halophytic ones-for phytoremediation purposes.

Abstract

Cancer is a complex disease including approximately 200 different entities that can potentially affect all body tissues. Among the conventional treatments, radiotherapy and chemotherapy are most often applied to different types of cancers. Despite substantial advances in the development of innovative antineoplastic drugs, cancer remains one of the most significant causes of death, worldwide. The principal pitfall of successful cancer treatment is the intrinsic or acquired resistance to therapeutic agents. The development of more effective or synergistic therapeutic approaches to improve patient outcomes and minimize toxicity has become an urgent issue. Inula viscosa is widely distributed throughout Europe, Africa, and Asia. Used as a medicinal plant in different countries, I. viscosa has been characterized for its complex chemical composition in order to identify the bioactive compounds responsible for its biological activities, including anticancer effects. Sesquiterpene lactones (SLs) are natural, biologically active products that have attracted considerable attention due to their biological activities. SLs are alkylating agents that form covalent adducts with free cysteine residues within enzymes and key proteins favoring cancer cell cytotoxicity. They are effective inducers of apoptosis in several cancer cell types through different molecular mechanisms. This review focuses on recent advances in the cytotoxic effects of I. viscosa and SLs in the treatment of neoplastic diseases, with a special emphasis on their proapoptotic molecular mechanisms.

Abstract

The authors present the case of a patient who underwent the removal of a small bluish lesion of the cheek. After discharge, the patient presented with profuse bleeding and hematoma of the cheek. Blood tests revealed severe secondary immune thrombocytopenia (SITP). SITP was probably triggered by the anti-SARS-CoV-2 Pfizer vaccine, which was inoculated to the patient 3 days before the lesion appeared and 12 days before surgery. The authors' aim is to inform colleagues about this possible, rare, adverse effect of the vaccine. Abstract

Multiple myeloma (MM) is an aggressive B cell malignancy. Substantial progress has been made in the therapeutic context for patients with MM, however it still represents an incurable disease due to drug resistance and recurrence. Development of more effective or synergistic therapeutic approaches undoubtedly represents an unmet clinical need. Tomentosin is a bioactive natural sesquiterpene lactone extracted by various plants with therapeutic properties, including anti‑neoplastic effects. In the present study, the potential antitumor activity of tomentosin was evaluated on the human RPMI‑8226 cell line, treated with increasing tomentosin concentration for cytotoxicity screening. The data suggested that both cell cycle arrest and cell apoptosis could explain the antiproliferative effects of tomentosin and may result in the inhibition of RPMI‑8226 cell viability. To assess differentially expressed genes contributing to tomentosin activity and identify its mechanism of action, a microarray gene expression profile was performed, identifying 126 genes deregulated by tomentosin. To address the systems biology and identify how tomentosin deregulates gene expression in MM from a systems perspective, all deregulated genes were submitted to enrichment and molecular network analysis. The Protein‑Protein Interaction (PPI) network analysis showed that tomentosin in human MM induced the downregulation of genes involved in several pathways known to lead immune‑system processes, such as cytokine‑cytokine receptor interaction, chemokine or NF‑κB signaling pathway, as well as genes involved in pathways playing a central role in cellular neoplastic processes, such as growth, proliferation, migration, invasion and apoptosis. Tomentosin also induced endoplasmic reticulum stress via upregulation of cyclic AMP‑dependent transcription factor ATF‑4 and DNA damage‑inducible transcript 3 protein genes, suggesting that in the presence of tomentosin the protective unfolded protein response signaling may induce cell apoptosis. The functional connections analysis executed using the Connectivity Map tool, suggested that the effects of tomentosin on RPMI‑8226 cells might be similar to those exerted by heat shock proteins inhibitors. Taken together, these data suggested that tomentosin may be a potential drug candidate for the treatment of MM.

Abstract

(1) Tomentosin is the most representative sesquiterpene lactone extracted by I. viscosa. Recently, it has gained particular attention in therapeutic oncologic fields due to its anti-tumor properties. (2) In this study, the potential anticancer features of tomentosin were evaluated on human Burkitt's lymphoma (BL) cell line, treated with increasing tomentosin concentration for cytotoxicity screening. (3) Our data showed that both cell cycle arrest and cell apoptosis induction are responsible of the antiproliferative effects of tomentosin and may end in the inhibition of BL cell viability. Moreover, a microarray gene expression profile was performed to assess differentially expressed genes contributing to tomentosin activity. Seventy-five genes deregulated by tomentosin have been identified. Downregulated genes are enriched in immune-system pathways, and PI3K/AKT and JAK/STAT pathways which favor proliferation and growth processes. Importantly, different deregulated genes identified in tomentosin-treated BL cells are prevalent in molecular pathways known to lead to cellular death, specifically by apoptosis. Tomentosin-treatment in BL cells induces the downregulation of antiapoptotic genes such as BCL2A1 and CDKN1A and upregulation of the proapoptotic PMAIP1 gene. (4) Overall, our results suggest that tomentosin could be taken into consideration as a potential natural product with limited toxicity and relevant anti-tumoral activity in the therapeutic options available to BL patients.

Abstract

Burkitt lymphoma is a very aggressive B-cell non-Hodgkin lymphoma. Although remarkable progress has been made in the therapeutic scenario for patients with Burkitt lymphoma, search and development of new effective anticancer agents to improve patient outcome and minimize toxicity has become an urgent issue. In this study, the antitumoral activity of Inula viscosa , a traditional herb obtained from plants collected on the Asinara Island, Italy, was evaluated in order to explore potential antineoplastic effects of its metabolites on Burkitt lymphoma. Raji human cell line was treated with increasing Inula viscosa extract concentration for cytotoxicity screening and subsequent establishment of cell cycle arrest and apoptosis. Moreover, gene expression profiles were performed to identify molecular mechanisms involved in the anticancer activities of this medical plant. The Inula viscosa extract exhibited powerful antiproliferative and cytotoxic activities on Raji cell line, showing a dose- and time-dependent decrease in cell viability, obtained by cell cycle arrest in the G2/M phase and an increase in cell apoptosis. The treatment with Inula viscosa caused downregulation of genes involved in cell cycle and proliferation (c-MYC, CCND1) and inhibition of cell apoptosis (BCL2, BCL2L1, BCL11A). The Inula viscosa extract causes strong anticancer effects on Burkitt lymphoma cell line. The molecular mechanisms underlying such antineoplastic activity are based on targeting and downregulation of genes involved in cell cycle and apoptosis. 36. Lenalidomide Maintenance with or without Prednisone in Newly Diagnosed Myeloma Patients: A Pooled Analysis.

Abstract

: We conducted a pooled analysis of two phase III trials, RV-MM-EMN-441 and EMN01, to compare maintenance with lenalidomide-prednisone vs. lenalidomide in newly diagnosed transplant-eligible and -ineligible myeloma patients. Primary endpoints were progression-free survival, progression-free survival 2 and overall survival with both regimens. A secondary aim was to evaluate the impact of duration of maintenance on overall survival and on outcome after relapse. A total of 625 patients (lenalidomide-prednisone arm, n = 315; lenalidomide arm, n = 310) were analyzed. The median follow-up was 58 months. Median progression-free survival (25 vs. 19 months; p = 0.08), progression-free survival 2 (56 vs. 49 months; p = 0.9) and overall survival (73 months vs. NR; p = 0.08) were not significantly different between the two arms. Toxicity profiles of lenalidomide-prednisone and lenalidomide were similar, with the exception of neutropenia that was higher in the lenalidomide arm (grade ≥ 3: 9% vs. 19%, p < 0.001), without an increase in the rate of infections. Overall survival (median NR vs. 49 months, p < 0.001), progression-free survival from relapse (median 35 vs. 24 months, p = 0.004) and overall survival from relapse (median not reached vs. 41 months, p = 0.002) were significantly longer in patients continuing maintenance for ≥2 years. Abstract

Background: Cognitive training (CT) is a non-pharmacological intervention based on a set of tasks that reflect specific cognitive functions. CT is aimed at improving cognition in patients with cognitive impairment, though no definitive conclusions have yet been drawn on its efficacy in Alzheimer's disease (AD)., Objective: To assess the effectiveness of a CT program designed to improve cognition in AD patients., Method: This is a randomized, controlled, single-blind, longitudinal trial with a no-treatment control condition in mild-to-moderate AD. Treated patients received in-group CT twice a week for six months, whereas controls did not. CT consisted of tasks ranging from paper-and-pencil to verbal-learning exercises. Participants' cognitive levels were assessed at baseline, post-intervention and 6 months later by means of a complete neuropsychological test battery. Repeated measures ANOVA was used to analyze the effect of time on the outcome measures, as well as to compare treated and untreated patients over time, with demographic data considered as covariates., Results: Of the 140 patients enrolled, 45 in the treated group and 85 controls concluded the study. The CT significantly improved treated subjects' cognitive functions immediately after the CT. Six months later, some test scores remained stable when compared with those obtained at baseline. The control group performed significantly worse than the treated group at each time-point, displaying a progressive cognitive decline over time., Conclusion: Our results suggest that CT may improve cognitive functions in patients with AD and may help to temporarily slow their cognitive decline., (Copyright© Bentham Science Publishers; For any queries, please email at epub@benthamscience.org.)

40. The Impact of Frailty on the Risk of Conversion from Mild Cognitive Impairment to Alzheimer's Disease: Evidences from a 5-Year Observational Study.

Abstract

The frailty construct has increasingly been adopted in the field of cognitive disorders. The aim of the present study was to measure frailty in a cohort of individuals with mild cognitive impairment (MCI) and to explore whether frailty measures may consent to predict the risk of conversion to dementia. We retrospectively reviewed the clinical charts of outpatients with amnesic MCI (aMCI) consecutively recruited at our Department, and followed-up for 5 years. Individual frailty status was measured by means of a frailty index (FI) consisting of 39 deficits (including signs, symptoms, diagnoses, and disabilities). Univariate analyses were used to compare the socio-demographic and clinical characteristics between subjects converting or not converting to probable Alzheimer's disease (AD) dementia over the follow-up. Risk for conversion to AD dementia was assessed using Cox regression models. Ninety-one subjects with aMCI (mean age 72.7, SD 7.1 years; women 49.5%) were consecutively recruited over a period of 12 months. Low levels of frailty were documented in the sample (mean FI score 10.0, SD 5.3). A statistically significant correlation between age and FI was observed. Overall, 58 participants converted to AD dementia over time. The Cox regression analysis showed that age (HR: 1.04, 95% CI: 1.00-1.08), male sex (HR: 0.52, 95% CI: 0.30-0.91), Mini-Mental State Examination score (HR: 0.85, 95% CI: 0.77-0.94), and FI (HR: 1.11, 95% CI: 1.05-1.18) were all significantly associated with the probability of MCI conversion. Individual's frailty status may increase the risk of conversion from a condition of MCI to overt AD dementia. The adoption of constructs comprehensively reflecting the biological decline of the aging subject may add useful estimates and information in the clinical approach to cognitive disorders.
